Background : Malignant cell nuclei, in general, have increased amounts of heterochromatin and decreased electron densities of euchromatin, making the chromatin pattern coarser than that of benign cell nuclei. The chromatin pattern in benign and malignant cells, however, is barely explained in terms of molecular structure. In this study, the chromatin pattern of metaplastic and carcinomatous squamous cells of the uterine cervix was correlated with transcriptional activity by ultrastructural autoradiography. Methods : Punch-biopsied tissues were cultured with 3 H-uridine for 5 minutes and processed for electron microscopy. Thin sections of the tissues on nickel grids were covered with photosensitive emulsion and kept cold in a dark room for 10 to 16 weeks. After development and staining, the tissues were observed by electron microscopy. Results : The nuclei of the metaplastic squamous cells consisted mostly of euchromatin. A few silver grains were observed, mainly at the periphery of the nuclei. The nuclei of the carcinomatous cells had increased amounts of heterochromatin along the nuclear membrane, and also in the euchromatin area. Silver grains were observed mainly at the boundary between the heterochromatin and euchromatin. Conclusions : These findings suggest that an increased amount of heterochromatin in carcinomatous cells results in an increase of the boundary area between the heterochromatin and euchromatin, an area which may be a transcriptionally active site. Key Words : Autoradiography, Carcinoma, Chromatin, Transcription, Genetic 일반적으로악성종양세포의핵은정상세포의핵에비하여크고, 다양한형태를보이며, 뚜렷한핵소체를가진다. 또한악성종양세포핵은이염색질 (heterochromatin) 의증가때문에거친염색질을보인다. 실제로악성종양세포핵의이와같은특징때문에자궁경부암종등의세포학적진단이가능하다. 1 악성종양세포핵의형태학적특징중에서도염색질패턴은매우중요한소견으로새롭게인식되고있다. 2 염색질은인간의 eye-brain system으로는객관성있게인지하기가어려우므로화상분석학적방법에의해분석되고있다. 3 Juhng 등 4 은자궁경부의상피내종양세포핵의전자현미경사진을대상으로한화상분석결과, 암종세포로진행함에따라이염색질의양이증가하고진염색질 (euchromatin) 의전자밀도가상대적으로낮아진다고보고하였다. 그러나이와같은변화가무엇을의미하는지는현재로서는불분명하다. 핵내의유전자가발현되기위하여서 는전사기구가유전자조절부위에작용하여야하므로암종세포에서진염색질의전자밀도가상대적으로낮아지는것을전사기구의접근성 (accessibility) 과연관지어볼수는있다. 그러나이것은확인이필요하다. 이것이확인된다면암종세포뿐만아니라일반적인진핵세포 (eukaryotic cell) 에서의전사조절 (transcription regulation) 에대한지견을얻을수있을것으로기대된다. 지금까지전사조절에대한많은연구보고가있었으나주로특정유전자의추출된 DNA나 transferred DNA를대상으로하는연구보고이며, 5 형태학적구조를유지한상태에서의전사조절에대한보고는찾아보기어렵다. 저자들은형태학적구조를유지한상태의암종세포를대상으로이염색질과진염색질이만드는염색질양상과전사의산물인새로합성된 mrna의소재부위와의상호관계를검토하고자본연구를시도하였다. 93

전자현미경용절편제작배양한조직을 4 의 Karnovsky 용액으로 4시간고정한후 1% osmium tetroxide로 2시간더고정하였다. 고정된조직을 ethanol로탈수시키고 propylene oxide를거쳐 epoxy resin으로포매하였다.

2 94 김미숙 정상우 재료와방법재료염색질양상이악성종양세포식별의중요한기준으로정립되어있는자궁경부의편평상피내암종으로진단된 20예를연구대상으로하였다. 대조군으로자궁경부의편평화생성조직 10예를포함하였다. 조직배양생검으로절취된신선조직을 Dulbecco's modified Eagle's medium (DMEM) 이든배양접시에놓고무균의작은조직편으로만든다음, DMEM과 10% fetal bovine serum이든배양액으로 37 에서 7% CO2로배양하였다. 배양시 3 H-uridine (100 Ci/L) 을첨가하여 5분간배양하였다. 전자현미경용절편제작배양한조직을 4 의 Karnovsky 용액으로 4시간고정한후 1% osmium tetroxide로 2시간더고정하였다. 고정된조직을 ethanol로탈수시키고 propylene oxide를거쳐 epoxy resin으로포매하였다. 병변을확인하기위하여 epoxy resin 포매괴에서부터약 750 nm 두께의절편을만들어 toluidine blue로염색하였고광학현미경으로관찰하였다. 자기방사법확인된병변부위에서 80 nm 두께의절편을만들어 nickel grid 위에올리고암실에서감광유제 (Ilford L4) 의얇은막을입힌후, 10주-16 주동안 4 의암실상태에두어 3 H-uridine의방사선으로감광시켰다. 감광시킨절편을 Kodak D19로 20 에서 4분간현상하였고, 6 현상정지액 (1% acetic acid) 에서 10초간둔후고정액 (sodium thiosulfate) 으로 2분간고정시켰다. 전자현미경적관찰현상한전자현미경표본을 lead citrate와 uranyl acetate로이중염색한후, JEM 100CXII 전자현미경 (JEOL) 으로가속전압 80 kv 하에서관찰하여염색질양상과새로합성된 RNA 를의미하는은입자의소재를검색하였다. 결과편평상피화생성세포의핵은대체로반원형이었고윤곽이평활하였다. 염색질은주로진염색질로구성되었으며핵막을따라소량의이염색질이출현하였다. 편평상피화생성세포의소수에서핵소체를관찰할수있었다. 세포질은풍부하였으며장원섬유 (tonofilament) 와교소체 (desmosome) 가뚜렷하였다. 은입자는소수였으며주로핵내에서관찰되었으나세포질내에서관찰되기도하였다. 세포질내에출현하는은입자와특정한소기관과의관계를규정하기는어려웠다. 핵내의은입자는주로핵막과인접한핵의가장자리에위치하였으나핵의안쪽에위치하는경우도상당수있었다 (Fig. 1). 은입자를볼수없는세포도상당수있었다. 상피내암종세포의경우정상세포보다핵과세포질의비율이증가되었고핵의모양이불규칙하였다. 이염색질의양은많았으 Fig. 1. 진염색질과이염색질의전자밀도차이가커져서이염색질이보다뚜렷하게관찰되었다. 뚜렷한핵소체를갖는세포의수도더많았다. 장원섬유와교소체도쉽게확인할수있었다. 편평상피화생성세포에비해은입자의수는다소더증가되었다. 은입자는진염색질부위에출현하기도하였으나주로진염색질과이염색질의경계부위에출현하였다. 은입자가핵막에걸쳐서출현하기도하였다.

A fibroblast and a myofibroblast show silver grains at the border between the heterochromatin and euchromatin ( 8,400). 며, 이염색질은주로핵막을따라나타났으나핵의중앙부에서도볼수있었다. 진염색질과이염색질의전자밀도차이가커져서이염색질이보다뚜렷하게관찰되었다. 뚜렷한핵소체를갖는세포의수도더많았다. 장원섬유와교소체도쉽게확인할수있었다. 편평상피화생성세포에비해은입자의수는다소더증가되었다. 은입자는진염색질부위에출현하기도하였으나주로진염색질과이염색질의경계부위에출현하였다. 은입자가핵막에걸쳐서출현하기도하였다. 세포질내에출현하는은입자의수는적었으며세포질내의소기관과연관짓기는어려웠다 (Fig. 2, 3). 은입자를전혀볼수없는세포도소수있었다. 상피내암종조직의표면에위치하는세포는세포질내소기관의종창이심하고염색질이미만성으로응집되는퇴행성변화를보이 는경우가많았다. 그러한세포의핵내에서도은입자를간혹볼수있었으며, 은입자는응집된염색질의가장자리에출현하였다 (Fig. 4). 편평상피화생성세포나상피내암종세포의핵내에은입자가관찰된경우에도핵소체에서는은입자를볼수없는경우가많았으며 (Fig. 3), 핵소체내에은입자가출현하는경우에도핵과핵소체내은입자의출현부위를연관지을수없었다 (Fig. 5). 화생성편평상피조직이나상피내암종조직의아래에있는간질조직내에침윤되어있는염증세포와혈관내피세포또는섬유모세포에서도간혹은입자를관찰할수있었다. 은입자는대부분핵내부에서관찰되었으며주로진염색질과이염색질의경계부위에위치하였다 (Fig. 6).

4 96 김미숙 정상우 고찰암세포는일차적으로형태학적소견으로인지되는데, 그중에서도염색질양상은매우중요한소견으로새롭게인식되고있다. 염색질양상은자궁경부암종의세포학적진단 2 외에도갑상선종양의감별, 7 항암제에대한암세포의내성획득여부의판별, 8 실험적간암종의발생단계추적, 3 자궁경부암종의발달과정추적 4 등에적합하다고보고되어있다. 그럼에도불구하고암세포의다른형태학적소견과마찬가지로염색질패턴의변화가어떻게일어나고어떤의미를갖는지에대해서는아직도대부분모르고있다. 주로 DNase I hypersensitivity에의한시험관내의실험결과에바탕을두고추정되는바로는, 진염색질은 DNA가응축되어있지않으므로전사가일어난다면이부위에서일어나리라는것이다. 그러나최근의학의여러분야에서새로운해석을가능하게해주고있는분자생물학적방법에의해형태학적소견을재조명하여볼필요가있다. 전사활동의핵내소재를조사하기위해서본연구에서는 3 H-uridine을이용하여 RNA의합성장소를자기방사법으로관찰하였다. 3 H-uridine은 mrna 외에 rrna와 trna에도삽입되지만, rrna는핵소체내에출현하며 trna는전체 RNA 의약 3% 만을차지하므로 9 핵소체외의염색질에출현하는은입자는주로새로합성된 mrna를의미한다고볼수있다. 따라서본연구결과는암종세포핵의이염색질과진염색질의경계부위에서 mrna가주로출현한다고해석할수있다. 합성된 mrna는 5 -capping, polyadenylation 및 splicing의과정을거쳐핵공 (nuclear pore) 을통해세포질로이동하므로, 본연구에서관찰된 mrna의위치와합성장소가다를가능성도완전히배제하기는어려울것이다. 그러나조직배양중에 3 H- uridine을짧은시간 (5분) 동안첨가한후바로고정시켰기때문에본연구에서관찰한 mrna의위치와합성장소는같거나매우근접해있을것으로생각한다. 본연구는암종세포핵에서새로합성된 mrna가주로이염색질과진염색질의경계부위에소재한다는점외에도핵내에서매우국소적으로출현함을보여주고있다. 이는핵내의전사활동이한시점에서는극히제한적으로이루어지고있음을시사해주는소견이다. 일반적으로핵내의전사가진염색질에서이루어지리라는것은추정되어왔지만세포활동의한시점에서국소적으로만전사가이루어질것이라는보고는찾아보기어렵다. 국소적인전사활동은핵내의전사활동에대한조절이순차적으로일어나게끔고도로조정되고있을가능성을암시하고있다. 본연구성적상많은암종세포의 mrna 은입자가이염색질과진염색질의경계부위에출현하는현상에대하여더관찰할필요가있다. 이러한현상은자궁경부의편평세포암종세포외에도일반적으로관찰되는현상일가능성이크다. Berman 등 10 은 myotube 를구성하는근세포에서 acetylcholine 수용체 mrna의 intron이핵막을따라분포하는것을형광 in situ hybridization 법으로관찰하였고, Xing 등 11 은섬유모세포의 fibronectin mrna가핵막의가장자리에출현하는것을역시형광 in situ hybridization 법으로관찰하였다. 이러한현상을설명하기위한가정중의하나는, 이염색질과진염색질의경계부위에서염색질의응축과이완에관여하는물질들에의해 DNA의이완이일어나며이부위에서 mrna가합성되리라는것이다. 이는염색질의응축과이완에관여하는물질들에대한소재를관찰함으로써확인할수있으며, 진핵세포에서의전사기작에대한단서도제공할수있을것으로생각된다. 일부세포에서는 mrna의은입자가이염색질부위에도출현하였다. 이는이염색질의전사활동을의미할수도있으나, 세포의입체적구조를생각한다면은입자의위또는아래에이염색질이중첩되어있기때문이라고생각할수도있다. 극히제한된경우에서는이염색질의전사활동도보고되어있으므로양자의가능성이모두있다고생각된다. 정상세포에비하여자궁경부암종세포핵에서이염색질의양이많아지고진염색질의전자밀도가낮아지는현상 4 을본연구성적만으로설명하기는어렵다. mrna의소재부위가주로이염색질과진염색질의경계부위라는본연구성적과연관지어생각하여보면, 암종세포핵에서는이염색질과진염색질의경계부위의 DNA가전사기구의접근이용이하게끔특별히이완된상태로존재하고있을가능성을시사하여준다. 즉암종세포핵에서는, 특정유전자의전사활동은활발하나그외의 DNA는주로응축되어이염색질을형성할것이라고추정할수있다. 현대의분자생물학분야에서도유전자전사조절에관한연구는중심과제의하나이다. 유전자조절은주로시험관내방법이나 transfection에의한생체내의방법에의하여연구되고있다. 즉, footprinting, 12 poly (ADP-ribosylation), 13 DNA에대한 acridine orange binding, 14 supercoiling, 15 DNA 응축, 16,17 DNA methylation, 18 또는 histone acetylation 등을조사함으로써유전자조절을연구하거나 DNA transfer를이용하여전사조절을연구하는경향이다. 그러나본연구에서와같이염색질양상을전사기구와결부시켜세포핵의형태를유지한상태에서유전자조절을연구하는것이궁극적으로는필요하다. 염색질양상과전사활동과의관계가규명된다면병리학적진단상중요한기준중의하나인염색질양상을분자생물학적으로해석할수있게되어, 염색질패턴의변화를보다이론적으로뒷받침하는진단기준을제시할수있을것이다. 또한전사활동의핵내분포를조사함으로써진핵세포의전사조절에대한생체내상태에서의정보를제공할수있을것이며, 시험관내가아닌생체내에서적용되는분자생물학적기법을찾게되어현상의기술에머물러있는기술형태학 (descriptive morphology) 을해석형태학 (inferential morphology) 으로확장시키는하나의예가될수있을것이다.

5 염색질경계면에서의 mrna 합성 97 본연구에서는새로합성된 mrna의소재를전자현미경적자기방사법으로관찰하였으나, laser confocal microscope나면역전자현미경방법을병용하여전사에관여하는물질들을관찰한다면염색질양상과전사활동과의관계를보다명확하게밝힐수있게될것이며염색질양상이갖는생물학적의의를보다더잘이해할수있게될것으로기대한다. 참고문헌 1.
