Symptoms included wilting of foliage, drying and withering of older leaves, and stunting of the plants. The infected plants eventually died during growth. Colonies on potato dextrose agar were pinkish white, and felted with cottony and aerial mycelium. Macroconidia were falcate to almost straight, thin walled and usually 3-septate. Microconidia were usually formed abundantly in false-heads on short monophialides on the hyphae and were hyaline, smooth, oval to ellipsoidal, aseptate or medianly 1-septate, very occasionally 2-septate, slightly constricted at the septa, mm. On the basis of the morphological characteristics and phylogenetic analyses of molecular markers (internal transcribed spacer rdna and translation elongation factor 1a), the fungus was identified as Fusarium oxysporum. Pathogenicity of a representative isolate was proved by artificial inoculation, fulfilling Koch s postulates. To our knowledge, this is the first report on the occurrence of Fusarium oxysporum on kohlrabi in Korea. 2 28 Research in Plant Disease Vol. 21 No 년 4월에 전라북도농업기술원의 노지 시험연구포장에 정식한 콜라비(품종 아삭콜)에서 처음으로 시들음병이 발생 하였다. 이병주에서 병원균을 순수분리하여 균주를 확보하 였다. 균학적 특성 및 DNA 염기서열을 분석한 결과 Fusarium oxysporum으로 동정되었으며, 콜라비 유묘에 인공접종하여 병 원성을 확인하였다. 따라서 본 연구는 콜라비 시들음병의 병징, 균학적 특징, 병원성 검정 및 염기서열 분석결과를 보고하고자 한다. 발병 및 병징. 콜라비 종자를 105공 트레이에서 30일간 육 묘하여 노지포장에 정식하였는데, 그후 약 20일이 경과하면서 시들음병이 발생하기 시작하였다. 병징은 생육초기에는 건전 주에 비해 생육이 더디고 신초부터 시들음 증상이 나타나기 시 작하였으며, 오래된 잎은 마르고 뒤틀리다가 더 이상 생육을 하 지 않고 황화되고 결국에는 고사하였다. 이병주의 지제부는 땅 에 누운 상태로 주저 앉았으며, 병이 진전됨에 따라 부패하기 시작하여 곰팡이가 피고 건전주에 비해 쉽게 뽑혔다(Fig. 1A, B). 지제부를 자르면 병원균에 감염된 도관부가 갈색으로 변색되 어 있으며, 낮에는 시들고 밤에는 다소 회복되는 증상이 반복되 었다. 그러나 박과작물에서처럼 지제부에서 끈적끈적한 진갈 색 수액이 병든 조직으로부터 유출되지는 않았다. 이와 같은 증 상은 2014년 9월 익산의 가을재배하는 다른 농가포장에서도 동일하게 나타났다. 발병된 농가에 따르면 같은 증상이 매년 반 복되었으며, 수량감소에 가장 크게 영향을 미치는 병해로 인식 하고 있었다. 익산의 서로 다른 콜라비 재배포장에서 조사된 시 들음병의 발병률은 %이었다. 병원균의 분리 및 병원성 검정. 병원균을 분리하기 위하여 발병포장에서 이병주를 채집하여 실험실에서 건전부와 이병 부의 경계조직을 5 5 mm 크기로 잘라 1% NaOCl 용액에서 1분 동안 표면살균하고 멸균수로 2-3회 세척한 후 멸균된 여과 지에서 물기를 제거한 다음 물한천배지에 치상하였다. 25oC 항 온기에서 3일간 암배양한 후에 균사체 선단부를 떼어내 감자한 천배지(Difco, MD, USA)에 옮겼으며, 25 oc 항온기 내에서 7일간 배양하며 병원균의 균학적 특성을 조사하였다. 감염된 표본은 고려대학교 표본실(Korea University Herbarium, KUS) 에 보존하였다. KUS-F27847로부터 대표 균주가 분리되었으며, 대표 균주는 농업유전자원정보센터(Korean Agricultural Culture Collection)에 기탁(기탁번호 KACC47747)하였으며, 병원 성 검정 및 염기서열 분석에 사용하였다. 콜라비에 대한 병원성 검정은 아삭콜 품종으로 실시하였다. 병원성 검정용 식물체는 종자를 파종하여 105공 트레이에 1달간 생육한 후 건전한 개체 를 20주 선별하여 250 ml 삼각플라스크에 1주씩 넣고 스폰지로 고정하여 준비하였다. 접종원은 KACC47747 균주를 감자한처 Fig. 1.
