In addition, VEGF can mediate the inflammatory response in tumors. VEGF increases the level of neutrophil migration by upregulating interleukin-8 (IL-8) in endothelial cells in vitro. However, it is unclear if VEGF can mediate IL-8 production in vivo. Methods: To address this issue, this study examined the effect of VEGF on IL-8 production in vivo using an adenovirus transduction and mouse ear assay. Results: Adenovirus-encoded VEGF (VEGF-Ad) increased the level of IL-8 production in endothelial cells in vitro compared to the control-adenovirus (CTL-Ad). The mouse ear assay showed that VEGF-Ad increased the level of IL-8 production in the endothelium. Immunohistochemistry showed that the IL-8 proteins were expressed in the vasculature within a human glioblastoma, which is known to strongly express VEGF. Conclusion: These results suggest that VEGF can mediate the inflammatory response in endothelial cells in vivo via the up-regulation of IL-8. 즉, VEGF는내피세포표면에있는 VEGFR-2와결합하여내피세포의성장 (proliferation), 생존 (survival), 흡착 (adhesion), 이동 (migration), 맥관- 형성 (tube formation) 등혈관생성의알려진거의모든단계에관여한다.

2 200 Korean J Hematol Vol. 44, No. 4, December, 2009 을발휘하기위해서 VEGF는내피세포의표면에존재하는수용체인 VEGFR-1 (Flt-1), VEGFR-2 (KDR/Flk- 1) 혹은 neuropilin-1 (NRP-1) 등과결합하는것으로알려져있다. 8) 이들수용체중에서 VEGFR-2가 VEGF 에의한내피세포의신호전달에핵심역할을하는것으로보고되었다. 즉, VEGF는내피세포표면에있는 VEGFR-2와결합하여내피세포의성장 (proliferation), 생존 (survival), 흡착 (adhesion), 이동 (migration), 맥관- 형성 (tube formation) 등혈관생성의알려진거의모든단계에관여한다. 실제로 VEGFR-2의인산화활성 (tyrosine kinase activity) 을억제하는화합물들은혈관신생을강력히억제하는것으로밝혀졌고, 이들억제제들은암환자들에대한임상실험에서그효능이증명되고있다. 9) 한편 VEGF는종양에의한생체내염증유발에도관여하는것으로알려져있다. 10) 암환자들에서혈전의위험성이증가하는것은염증과깊은상관관계가있을것으로보고되었다. 11) 염증을유발하는인자들중에는케모카인 (chemokine) 으로알려져있는저분자단백질들이있다. 지금까지알려진케모카인들은 44종류이고그수용체는 21개정도로알려져있는단백질군집을이루고있다. 12) VEGF는이들케모카인의일종인 SDF-1과 MCP-1의발현을증가시켜백혈구등의면역세포를염증유발자리로이동을유도하는것으로알려져있다. 13) 더욱이 VEGF는혈관내피세포에서 IL-8의발현을증가시켜호중구의이동을유도하는것으로밝혀졌다. 14) 하지만 VEGF가혈관내피세포에서 IL-8의발현을증가시키는것은오직 in vitro에서만보고되었다. 본연구에서는 VEGF가생체에서실제로 IL-8의발현에관여하는지여부를연구하는것을주요목표로설정하였다. 서의 IL-8의발현을조직염색법을이용하여연구를수행했다. 3. 연구방법 1) VEGF 발현아데노바이러스 (Ad) VEGF-Ad 와 CTL-Ad 는 Harvard Medical School 의 Harold Dvorak 박사로부터분양받았다. 2) 세포배양 HUVEC (human umbilical vein endothelial cells) 은 Cambrex사에서구입하였고 EBM-2 배지에서배양되었다. 3) Western blot 분석 전기영동법은기존에보고된방법에의해수행되었다. 15) 4) ELISA 분석법에의한 IL-8 정량 HUVEC을 6-well 판에서 subconfluent하게배양하였다. 그후성장인자가제거된 EBM-2 배지에 5시간배양한후 VEGF-Ad 혹은 control-ad 10 MOI (multiplicity of infection) 를처리하였다. 3일후배양액을수거 대상및방법 1. 연구계획 VEGF 발현아데노바이러스를이용하여 VEGF가생체에서 IL-8의발현을유도함을증명하고실제질환에서 VEGF와 IL-8 발현의상관성을연구함을목적으로한다. 2. 연구대상선택생쥐의양쪽귀에아데노바이러스를주입하는방법을통하여생체에서의 VEGF에의한 IL-8 발현검증과 VEGF가과량발현하는것으로알려진악성뇌종양에 Fig. 1. 3 신가희 외: 혈관내피성장인자에 의한 혈관에서 Interleukin-8 발현 한 후 R&D Systems사의 IL-8 정량용 ELISA 키트를 사 용하여 IL-8 양을 측정하였다. 5) 생체내 IL-8 유도 생쥐의 귀에 VEGF-Ad 혹은 CTL-Ad 100 MOI를 미 세주사기를 이용하여 주입하였다. 7일 후 생쥐를 안락 사 한 후 귀를 절개하여 동결한 상태에서 5 μm로 절 편한 후 조직염색법으로 IL-8 발현을 확인하였다. 6) 조직 염색법 생쥐의 귀조직을 100% 원액의 차가운 메탄올로 10분 간 고정시킨 후 차단용액(Dako Inc., Carpinteria, CA) 으로 비특이적 결합부위들을 차단하였다. 그 후 IL-8, 201 VEGF, von Willebrand factor (vwf) 항체를 1 100으 로 처리한 후 밤 새 반응시켰다. 생리식염수로 세 번 세척한 후 2차 항체를 1시간동안 반응시켰다. 생리식 염수로 세 번 세척한 후 표본 용액으로 고정한 후 현미 경으로 관찰하였다. 결과 및 고찰 1. 혈관내피세포(HUVEC)에서 VEGF에 의한 IL-8 발현 제조된 VEGF-Ad가 VEGF를 발현시키는지 조사하 Fig. 2. 1A에서 보 이는 것처럼 10 MOI의 VEGF-Ad를 감염시킨 HUVEC 에서의 VEGF의 발현이 CTL-Ad에 비해 현격히 증가 함을 알 수 있었다.

그 결과 VEGF-Ad는 CTL-Ad 에 비해 IL-8의 발현을 현격히 증가시킴을 관찰할 수 있었다(Fig. 1B).

이 결과는 VEGF가 생체의 혈관조직에서 IL-8의 발현을 유도한다는 것을 명확히 2.

Fig. 1A에서 보 이는 것처럼 10 MOI의 VEGF-Ad를 감염시킨 HUVEC 에서의 VEGF의 발현이 CTL-Ad에 비해 현격히 증가 함을 알 수 있었다. 다음으로 VEGF-Ad에 감염된 HUVEC의 상층액을 수집하여 ELISA방법을 이용하여 IL-8의 양을 측정하였다. 그 결과 VEGF-Ad는 CTL-Ad 에 비해 IL-8의 발현을 현격히 증가시킴을 관찰할 수 있었다(Fig. 1B). 이 결과는 재조합 VEGF 단백질이 혈 관내피세포에서 IL-8의 발현을 유도한다는 이전보고 14) 와 일치하였다. 확인할수 있었다(Fig. 3). 이 결과는 VEGF가 생체의 혈관조직에서 IL-8의 발현을 유도한다는 것을 명확히 2. 생쥐의 귀조직에서 VEGF에 의한 IL-8 발현 VEGF가 생체에서 IL-8의 발현을 증가시키는지를 조사하기위해 VEGF-Ad와 CTL-Ad 100 MOI를 생쥐 의 귀에 주입하였다. 7일후 생쥐의 귀를 절개한 후 동 결절편을 만들어서 조직 염색을 시도하였다. 5 신가희외 : 혈관내피성장인자에의한혈관에서 Interleukin-8 발현 203 보여주는것으로사료된다. 3. 아교모세포종 (glioblastoma) 에서 IL-8 발현 다음으로악성뇌종양의하나인아교모세포종조직에서 IL-8의발현을조사하였다. 아교모세포종은 VEGF를과량으로발현하는암으로알려져있을뿐만아니라종양조직에서염증을유발하는면역세포들이과량존재한다고보고되었다. 16) 따라서악성뇌종양의염증과 IL-8의발현의상관관계를연구하기위해아교모세포종에서 IL-8의발현을조사하였다. Fig. 4에서보이는것처럼아교모세포종은 VEGF를과량으로발현하였고종양의혈관에서 IL-8이발현됨을관찰하였다. 이상의결과는 VEGF가생체내혈관에서 IL-8의발현에직접적으로관여함을보임과동시에 VEGF가 IL-8의발현을통해염증을일으킬수있음을보여주는것으로사료된다. 이전연구에의하면 VEGF는혈관내피세포에서 IL-8의발현을유도하여호중구의화학주성을증가시키는것으로보고하였다. 14) 하지만실제생체에서 VEGF가 IL-8의발현에관여한다는보고는아직연구되지않은실정이다. 본연구는 VEGF가생체의혈관에서 IL-8의발현을유도하는것을처음으로관찰하였다. 염증반응은암세포의생체내성장에도밀접히관여하므로 VEGF의기능을억제하는것은혈관신생억제와더불어염증반응억제를통해암세포의성장을억제할수있음을본연구는제시하고있다. 요 배경 : 혈관내피성장인자 (vascular endothelial growth factor, VEGF) 는암세포들에의해이루어지는혈관-신생 (angiogenesis) 에주요한역할을한다고알려져있다. 또한 VEGF는종양에의한생체내염증유발에도관여하는것으로알려져있다. VEGF는혈관내피세포에서 IL-8의발현을유도하여호중구의이동에관여한다. 그러나 VEGF가실제생체에서 IL-8의발현에관여하는지는아직연구되지않은실정이다. 방법 : VEGF가생체에서 IL-8의발현에관여하는것을증명하기위해아데노바이러스를이용한 VEGF 발현시스템을이용하여생쥐의귀조직에서 VEGF와 IL-8 발현의상관성을연구하였다. 결과 : VEGF 발현아데노바이러스는시험관에서배양된혈관내피세포에서 VEGF를발현하였고동시에 IL-8발현도유도하였다. VEGF 발현아데노바이러스 약 를생쥐의귀조직에주입하였을경우 VEGF 발현부위에서강한 IL-8 발현이관찰되었다. 더욱이 IL-8은혈관에서발현하는것이관찰되었다. VEGF를과량으로발현하는악성뇌종양의일종인아교모세포종조직에서 IL-8의발현을조사한결과혈관조직에서특이적으로발현하는것이관찰되었다. 결론 : 본연구결과는 VEGF가생체에서 IL-8의발현을유도하여염증반응에관여함을제시해주고있다. 참고문헌 1) Hicklin DJ, Ellis LM.
