Methods : The MTS assay was used to confirm the inhibition of cancer cell proliferation in A375 cells. And the MUSE analyzer was used to determine the ability of OJEF to induce cell cycle arrest. Western blotting was used to determine the changes in protein expression in A375 cells after treatment with OJEF. Results : OJEF showed cytotoxicity to A375 cells. And cell cycle arrest occurred in G1 phase and G2/M phase owing to inhibition of CDK1, cyclin B1, CDK4, and cyclin D, which are related to cell cycle regulation and cell division control. Conclusion : OJEF is effective in regulating cell cycle of human melanoma cells and thus can be a good theraputic agent to treat patients with melanoma. 2 한방안이비인후피부과학회지제 32 권제 4 호 (2019 년 11 월 ) Ⅰ. 서론 최근생활습관및환경이변화되면서각종유해물질이나자외선의노출기회가빈번해짐에따라전세계적으로피부암발생빈도가증가되었고, 피부암은공중보건의심각한문제를야기하고있다 1). 그중흑색종은멜라닌세포가존재하는어느부위에나발생할수있으며, 다른피부암과달리성장속도가매우빨라발생초기에다른장기로전이율이높아악성도가높은종양으로알려져있다 2). 또한국소재발이높은빈도로일어나고, 화학요법이나방사선치료에도저항성이강하여치료가가장어려운악성종양중의하나이다 3). 지금까지의항암물질에대한연구는암세포를사멸시키는단편적인결과물에치중되어왔으나, 이에대한분자생물학적수준에서의기작연구가불충분한것이현실이다. 따라서항암효과가기대되는천연물에대한명확한기작연구가진행되어야한다 4). 서양의학에서피부암의치료는외과적절제술이가장일반적인치료방법이며동결요법, 소파술, 전기건조법, 광역동치료, 방사선요법, 화학요법, 면역요법등의다양한치료방법이있다 5). 그러나흑색종이다른피부암보다더치명적인이유는항암화학요법에대한저항성을가지고있어 20% 정도의환자에서만치료효능을보이기때문이다 6). 약물저항성에대한기전연구는아직밝혀지지않았지만세포의죽음기전과연관되어있다고생각되고있으며, 또한이것이질병의진행과전이에연관되어있다고알려져있어세포의죽음에관한기전연구가절실히요구되고있다 7,8). 세포는세포가 mitosis 하는시기 (M phase) 와염색체 DNA 를 synthesis 하는 S phase, 그리고이들의준비를행하는시기 (G1 and G2 phase) 를반복하여증식하는데, 이반복을세포주기라한다 9). 세포의증식과 Corresponding author : Deok-Seon Ryu, Department of Biomedical Laboratory Science, College of Medical Sciences, Soonchunhyang University. (Tel : , hilde0922@sch.ac.kr) Received 2019/9/2 Revised 2019/10/17 Accepted 2019/10/24 고사는세포주기에의해결정되는데, 세포주기는 regulatory cyclins subunits, cyclin dependent kinases 그리고 their inhibitors 같은세포주기조절인자에의해조절된다 10). 암이라는것은정상적으로세포활동이유지되지못하고성장과분열이제어가되지않아서무절제한방식으로세포가증식하는현상을말하는데, 이러한세포주기제어능력을확인하는것이항암활성을검증하는지표로사용된다 11). 와송은돌나무과 (Crassulaceae) 에속한다년생육질초본인바위솔 (Orostachys japonica Berger) 의전초로여름, 가을에채취하여쇄건하여사용한다. 性味는凉酸苦하여肝, 肺經에歸入하며, 주로凉血止血, 淸熱解毒, 收濕斂瘡하는효능이있어吐血, 衄血, 便血, 血痢, 熱淋, 月經不調및疔瘡癰腫등의병증에적용한다 12). 와송에는 Friedelin, epi-friedlanol, grutinone, glutinol, triterpenid, kaempferol, quercetin, flavonoidβ-sitosterol, cam-pesterol, fatty acid ester, quercetin, flavonoid 등다양한약리적성분이존재하는것으로밝혀졌으며 13,14), 전통적으로소염제, 해열제, 지혈제, 해독제및항암의목적으로사용되었다고알려져있다 15). 또한항균 16), 항당뇨 17) 효능과위암 15), 간암 18), 대장암 19) 등여러질환에서유효한작용을나타내는것으로밝혀졌다. 이전연구들에서는와송의생리활성물질이여러종류의암에서항암효과가있다고알려져왔지만, 아직까지와송이흑색종에미치는세포사멸과항암활성에관한연구는없다. 이에본연구에서는와송유래생리활성물질에의한인체흑색종 A375SM 세포에대한항암효과를세포주기조절의관점에서확인하고이의분자적기전을밝히고자한다. Ⅱ. 재료및방법 1. Cell line and reagents A375 human melanoma cell lines 은 Korean 2

3 류덕현외 1 인 : 인체흑색종세포에대한와송추출물의세포주기억제를통한항암효과와기전연구 cell line bank(kclb, Seoul, Korea) 로부터구입하여사용하였다. 세포배양과정에서는 Dulbecco s modified Eagle s medium(dmem; Corning, Manassas. VA, USA), fetal bovine serum(fbs; Corning, Manassas. VA, USA), penicillin-streptomycin antibiotic solution(hyclone, Logan, UT, USA) 을사용하였다. MTS(3-[4,5-dimethylythiazol-2-yl]-5-[3-carbpxymethoxyphenyl]-2-[4-sulfophenyl]-2H-tetrazoliu m, innersalt) 용액인 CellTier 96 non-radioactive cell proliferation assay kit(promega, Madison, WI, USA) 를이용하였고, Apoptosis 유도능과 cell cycle arrest 정도를확인하기위하여 MUSE R Annexin V and Dead Cell Assay Kit(Merck KGaA, Darmstadt, Germany) 와 MUSE R Cell Cycle Kit(Merck KGaA, Darmstadt, Germany) 를사용하였다. 또한 4,6-diamidino-2-phenylindole(DAPI) solution(vectashield mounting medium with DAPI, Burlingame, CA, USA) 으로핵을염색하여형광현미경으로확인하였다. Western blotting 을위하여사용한 primary antibodies 인 NF-κB, Bcl-2, pro-caspase-3, cleaved caspase-3, CDK1, CDK4, Cyclin B1, Cyclin D1, p-erk, p-jnk, p-p38, ERK, JNK, p38은 cell signaling technology (Danver, MA, USA) 로부터구입하여사용하였고, Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) 는 AbFrontier(Seoul, Korea) 에서구입하여사용하였다. 그리고 Mouse anti-rabbit IgG-HRP conjugated secondary antibodies는 Santa Cruz Biotechnology(Dallas, TX, USA) 에서구입하였다. 2. Plant materials and extraction O. japonicus은 Geobugiwasong 농장 (Miryang, Korea) 으로부터제공받았다. O. japonicus 는자연건조시켜, 믹서기를이용하여잘게분쇄하여 powder 로만들었다. Round bottom flask 안에 200g 의 powder O. japonicus 과 95% ethyl alcohol(etoh) 1l를현 탁한다음 reflux condenser(scilab R, Seoul, Korea) 를이용하여 3h, 3회끓였다. Crude O. japonicus extract를 rotary evaporator(ika-werke GmbH & Co. KG, Staufen, Germany) 를사용하여 40 에서 EtOH 를완전히증발시켰다. 동일플라스크에 1l의 double distilled water(h 2 O) 를넣고충분히흔들어준후, 동량의 n-hexane(hexane), dichloromethane (DCM), ethylacetate(etoac), n-butanol(buoh), H 2 O순으로 systematic solvent fractionation을수행하였다 (Fig. 1). 각각회수된 solvent fraction 은 rotary evaporator 을사용하여용매를완전히제거하고, 유리용기에옮겨담은후, silica gel 이들어있는밀폐된용기에서충분히건조시키고가루로분쇄하여 2 0 냉동보관하였다. 본연구에사용한용매분획물은이전의연구에서항암효과가가장뛰어난것으로밝혀진 15) EtOAc 분획물로서실험전세포실험전용 0.1% dimethyl sulfoxide(dmso; applichem GmbH, Darmstadt, Germany) 용액에녹여서사용하였다. Cell culture 본연구에사용한세포주는 A375 human melanoma cell로서 10% FBS, 1% penicillin-streptomycin이들어있는 DMEM 배지를이용하여 37, 5% CO 2 incubator 에서배양하였다. 세포가 90-95% confluency 를보일때, 4-5일마다계대배양하였고, 1-2일마다 1 PBS 를사용하여 2번세척하고배지를교체해주었다. 이때 10% trypsin(corning, Manassas. VA, USA) 을이용하여 Dish 에부착된세포주를떼어내었다. 3

4 한방안이비인후피부과학회지제 32 권제 4 호 (2019 년 11 월 ) 세포주는모든실험에서 cells/ ml농도로사용하였고, 농도별 EtOAc fraction(0, 20, 40, 60, 80, 100 μg / ml ) 을처리하기전, 1% penicillin-streptomycin 만들어있는 serum free(sf) media 를이용하여 6h 이상 starvation 하였다. 4. Cell viability assay A375 human melanoma cells 에대한 EtOAc fraction 의농도별, 시간별세포생존율을확인하기위하여 CellTier 96 non-radioactive cell proliferation assay kit를사용하였고, 실험방법은제조사에서지시하는대로수행하였다. 96 well plates 에 cells/ ml농도로세포를분주한후, 18h 37, 5% CO 2 incubator 에서배양하였다. 배양후 SF media 로 washing하고, 6h starvation 시킨후, EtOAc fraction을농도별 (0, 20, 40, 60, 80, 100μg / ml ) 로처리하였다. 시간별 6, 12, 24h 배양후, 상층액을모두제거하고 MTS 시약 100μl를분주하였다. 37, 5% CO 2 incubator에서 1h 배양후, 470nm에서 Multiskan sky microplate spectrophotometer (ThermoFisher Scientific, Waltham, MA, USA) 를이용하여흡광도를측정하였다. MTS를이용한 cell viability assay 는 3번반복실험하여평균결과및 standard deviation 수치를산출하였다. 5. Cell cycle analysis OJEF의 A375 human melanoma cells 에대한 cell cycle arrest 유도능을확인하기위하여 MUSE analyzer를이용하여 G0/G1, S, G2/M phase의 % 를각각측정하였다. A375 human melanoma cells 에대한 EtOAc fraction 의농도별세포주기제어능을확인하기위하여 MUSE R Cell Cycle Kit 를사용하였고, 실험방법은제조사에서지시하는데로수행하였다. 6 well plates 에 cells/ ml농도로세포를분주한후, 18h 37, 5% CO 2 incubator 에서배양하였다. 배양후 SF media 로한번 washing 하고, 6h starvation 시킨후, EtOAc fraction 을농도별 (0, 20, 40, 60, 80, 100μg / ml ) 로처리하였다. 6h 배양후, 상층액을모두제거하고, 1 PBS 를사용하여 2번세척하였다. 그리고 trypsin 100 μl를분주하여 plate 에부착된세포를분리하였고, 1.5ml e-tube 에세포를회수하였다. 회수한세포는 5,000rpm 에서 5분간원심분리를수행하여 cell pellets 를수집하였고, 수집한 cell pellets 은 70% cold EtOH 에현탁하여 4h이상 -20 냉동고에서 fixation 시켰다. MUSE analyzer를이용하여분석하기직전에 EtOAc 분획물의농도별시료를전처리하였다. 먼저 5000rpm 에서 5분간원심분리를수행하여 EtOH 이남지않도록상층액을제거하고, 1 cold PBS 로세척하고 1 cold PBS 100μl에현탁한후, cell cycle-muse solution 100μl추가하고어두운실온에서 30분간 incubator 시켰다. 대조군및실험군에대한농도별 cell cycle arrest 정도를 G1, S, G2/M phase 로구분지어서 MUSE analyzer(merck KGaA, Darmstadt, Germany) 을이용하여측정하였다. MUSE analyzer 를이용한 cell cycle analysis 는 3번반복실험하여평균결과및 standard deviation 수치를산출하였다. 6. Western blot analysis A375 human melanoma cells 에대한 EtOAc fraction 의농도별, 시간별단백질발현정도를확인하기위하여 Western blotting 을수행하였다. 6 well plates에 cells/ ml농도로세포를분주한후, 18h 37, 5% CO 2 incubator 에서배양하였다. 배양후 SF media로한번 washing 하고, 6h starvation 시킨후, EtOAc fraction 을농도별 (0, 20, 40, 60, 80, 100μg / ml ) 로처리하였다. 3, 12, 24h 배양후, 상층액을모두제거하고, cold 1 PBS 를사용하여 2번세척하였다. Lysis buffer(cell signaling technology, Danver, MA, USA) 를 100 μl를분주하고 scrapper 을이용하여세포를회수하였다. 회수한세포는 ice에서 4

5 류덕현외 1 인 : 인체흑색종세포에대한와송추출물의세포주기억제를통한항암효과와기전연구 30분간 incubation 시킨후단백질정량은 BCA시약 (Pierce, Rockford, IL, USA) 를이용하여농도를정량한후, 8-15% SDS-polyacrylamide gel에전기영동 (Bio-Rad, Philadelphia, PA, USA) 으로분리하였다. 전기영동후단백질을 PVDF membrane(bio-rad, CA, USA) 으로 transfer하고, membrane은 5% nonfat milk 로 2h blocking 하였으며, 1:1,000 으로희석한 1차 antibody 와 4 에서 18h 이상반응시켰다. 반응 membrane 을 1 PBST buffer 에 5분간 3번세척하고, 세척후 membrane 은 1:500 으로희석한 2 차 antibody 와 4 에서 2h 반응시키고다시 1 PBST 에세번세척하였다. 표적단백질은 ECL detection kit(santa Cruz, CA, USA) 를이용하여 X-ray film에감광시켜확인하였고, band 는 ImageJ software 를이용하여분석하였다. 7. Statistical analysis 모든실험결과는 3번이상반복실험하여평균결과값및 standard deviation 수치를산출하였다. 통계처리는 p<0.05 수준에서 Student's t-test 에의해유의차를검증하였다. Ⅲ. 결과 1. Inhibition of cell growth A375 세포에대한암세포증식억제효과를 MTS assay 를통하여확인하였다. 암세포의증식이 OJEF 의처리농도와배양시간에의존적으로감소하는것을알수있었다 (Fig. 2). OJEF를농도별 (0, 0.1%DMSO, 20, 40, 60, 80, 100, 120, 140, 150μg / ml ) 로처리하고, 12h 후회수하여보라색의 formazan 형성정도를 470 nm흡광도에서측정하고, 대조군을 100% 생존율로보고각농도별실험군의결과값을 % 으로환산한값을그래프로나타내었다 (Fig. 2A). 그결과, Control 에비해각농도별 (20, 40, 60, 80, 100, 120, 140, 150μg / Fig. 2. The values are expressed as the mean ± SD. *Significantly different from the control at p<0.05. ml ) 결과값은순서대로 ±1.38, ±1.46, 98.05±0.90, 97.1±2.42, 74.59±1.24, 58.32±3.82, 57.39±8.58, 36.49±2.93이었고, 100μg / ml에서부터유의적으로감소하였다. OJEF 를농도별로처리하고, 24h 후회수하여 MTS assay 를수행한결과, 농도별 (20, 40, 60, 80, 100, 120, 140, 150μg / ml ) 로 92.21±0.85, 93.55±5.65, 80.32±0.93, 68.78±0.80, 47.34±0.12, 39.88±1.78, 28.67±0.88, 27.47±0.37 이었고, 이중 20μg / ml와 60μg / ml에서부터유의적으로감소하였다 (Fig. 2B). OJEF 를농도별로처리하고, 48h 후회수하여 MTS assay 를수행한결과, 농도별 5

6 한방안이비인후피부과학회지제 32 권제 4 호 (2019 년 11 월 ) (20, 40, 60, 80, 100, 120, 140, 150μg / ml ) 로 76.97±1.56, 68.15±1.38, 51.85±1.38, 51.85±5.88, 41.74±1.34, 28.18±0.46, 21.31±0.71, 19.87±2.42, 18.38±1.15 이었고, 20μg / ml에서부터유의적으로감소하였다 (Fig. 2C). 2. Induction of cell cycle arrest OJEF 의 A375 세포에대한 cell cycle arrest 유도능을확인하기위하여 MUSE analyzer 를이용하여 G0/G1, S, G2/M phase의 % 를각각측정하였다. 농도별 (0, 0.1% DMSO, 20, 40, 60, 80, 100μg / ml ) 로 OJEF를처리하고 6h 배양후세포주기를확인한 histogram 결과, 농도의존적으로 G0/G1 phase peak 가높아지는것이관찰되었으며, 반대로 G2/M phase peak 는낮아지는현상이관찰되었다 (Fig. 3). Fig. 3의 histogram 을 % 으로변환한결과, G0/G1 phase 의경우 44.17±6.71 인 control 과비교하여농도별 (20, 40, 60, 80, 100μg / ml ) 로 48.30±4.82, 50.10±7.00, 57.47±3.50, 59.47±4.48, 61.80±6.26 이었고, 60μg / ml에서부터유의하게증가하였다. S기는전체농도에서큰차이를보이지않았으며, G2/M phase 인경우 27.90±5.21 인 control 과비교하여농도별 (20, 40, 60, 80, 100μg / ml ) 로 23.63±6.29, 21.13±7.60, 19.10±2.65, 15.83±3.58, 11.43±2.17 이었고, 60μg / ml에서부터유의적으로감소하였다 (Table 1). 3. 7 류덕현외 1 인 : 인체흑색종세포에대한와송추출물의세포주기억제를통한항암효과와기전연구 arrest 에의해서세포증식이억제되는것을확인하였다. 이와관련하여각세포주기를조절하는인자에대하여단백질수준에서 western blotting 을통하여확인하였다. MUSE analyzer 를이용하여실험한 cell cycle 결과에서 G1 phase 는유의적으로 arrest가증가하였고, G2/M phase으로의진행은유의적으로감소하였다. G1 phase 에서는세포의크기가 DNA 복제에적당한지확인하는시기인데, 이때관여하는단백질은 CDK 4와 cyclin D1 complex 이다. 또한분열기로진행하기전에 DNA 복제가종결되었는지확인하는시기는 G2/M phase 로서관여하는단백질은 CDK1 과 cyclin B1 complex 이다. Fig. 4는 OJEF 처리후, 농도 (0, 0.1% DMSO, 20, 40, 60, 80μg / ml ) 및시간 (12 및 24h) 에따른 CDK4 와 cyclin D1의발현정도를확인한결과로서, 농도및시간의존적으로단백질발현량이감소하는것을확인할수있었다. 각농도별 band 강도를 house keeping gene 인 GAPDH 와비교하여수치로변환한것을그래프로나타낸결과를보면, 12h 의 CDK 4 경우, 1.93±1.82 인 control 과비교하여농도별 (20, 40, 60, 80μg / ml ) 로 0.73±0.25, 0.46±0.22, 0.66±0.20, 0.29±0.05으로감소하였고, cyclin D1의경우, 1.37±0.47인 control 과비교하여농도별 (20, 40, 60, 80μg / ml ) 로 0.70±0.13, 0.37±0.04, 0.29±0.06, 0.14±0.07으로감소하는경향을보였으며, 40μg / ml에서부터유의적으로감소하였다 (Fig. 4A). 그리고 24h 의 CDK 4 경우, 1.31±0.43 인 control 과비교하여농도별 (20, 40, 60, 80μg / ml ) 로 0.82±0.23, 0.22±0.11, 0.03±0.03, 0 으로감소하는경향을보였으며, 40μg / ml에서부터유의적으로감소하였다. cyclin D1의경우, 1.43±0.63 인 control 과비교하여농도별 (20, 40, 60, 80μg / ml ) 로 1.34±0.79, 1.52±1.27, 1.44±1.05, 0.48±0.42으로감소하였다 (Fig. 4B). CDK 1과 cyclin B1 complex 의경우에서도농도및시간의존적으로단백질발현이감소하는것을확인하였다 (Fig. 5). 12h 의 CDK 1 경우, 3.66±1.31 인 control 과비교하여농도별 (20, 40, 60, 80μg / ml ) 로 2.99±1.22, 1.41±0.84, 0.76±0.45, 0.24±0.26 으로감소하는경향을보였으며, 60μg / ml에서부터유의적으로감소하였다. cyclin B1의경우, 0.71±0.53 인 control 과비교하여농도별 (20, 40, 60, 80μg / ml ) 로 0.37±0.48, 0.13±0.13, 0.03±0.06, 0으로감소하였다 (Fig. 5A). 그리고 24h 의 CDK 1 경우, 1.74±0.22 인 control 과비교하여농도별 (20, 40, 60, 80μg / ml ) 로 1.94±0.76, 1.79±1.14, 1.09±0.77, 0.94±0.80 으로감소하였고, cyclin B1의경우, 0.54±0.16인 control 과비교하여 Table 1. Effect of OJEF Treatment on Protein Expression of CDK1 and Cyclin B1 in A375 Cells The cells were treated with the indicated concentration(0, 0.1% DMSO, 20, 40, 60, and 80 / ) for (A) 12 h and (B) 24 h. The expression of CDK1 and cyclin B1 was examined by western blotting. The band density was quantitated and plotted on the bar graph. CDK1, 34 kda; cyclin B1, 55 kda. GAPDH was used as an internal control. The intensities of bands were determined by using Image J software for three separate experiments with similar results. The values are expressed as the mean ± SD. *Significantly different from the control at p<0.05. 농도별(20, 40, 60, 80 / )로 0.49±0.26, 0.36±0.34, 0.12±0.15, 0으로 감소하는 경향을 보였 8 으며, 60 / 에서부터 유의적으로 감소하였다(Fig. 5B).

9 류덕현외 1 인 : 인체흑색종세포에대한와송추출물의세포주기억제를통한항암효과와기전연구 Ⅳ. 고찰 약리성분과관련하여와송은대량의초산을함유하고있다고하였고 20), 와송의성분에대한연구에서 Friedelin, epi-friedlanol, grutinone, glutinol, triterpenid, kaempferol, quercetin, flavonoidβ -sitosterol, cam-pesterol, fatty acid ester, quercetin, flavonoid 등다양한약리적성분이존재하는것으로밝혀졌으며 13,21), 박등 22) 은와송의 Triterpene 및 Steroid 성분으로 taraxerone, stigmast-4-3-one and ergost-4-ene-3-one을분리하였다. 와송의항암효능에대한관심은비교적오래되어한의학계에서는 1966 년에초보적인실험이나마항암작용과독성에대한고찰이이루어졌으며 23), 최근에와서는위암 11), 대장암 19,24), 간암 18), 전립선암 25), 난소암과자궁경부암 26) 및백혈병 27) 등에서항암효능이밝혀져있다. 그러나분자생물학적수준에서의항암기작연구 18,19,28) 는부족한실정이다. 악성흑색종은림프절을통해전이되고전체피부종양에의한사망자의약 80% 를차지하는악성피부종양이다 29). 멜라닌세포가존재하는어느부위에나발생할수있으며, 다른피부암과달리성장속도가매우빨라발생초기에다른장기로전이율이높아악성도가높은종양으로알려져있다 6). 백인들의경우미국, 호주, 유럽등지에서발생빈도가점차증가하는추세이며, 과도한자외선노출과유전적인요인등이악성흑색종의발병에중요한원인이다 30). 흑색종이다른피부암보다더치명적인이유는항암화학요법에대한저항성을가지고있기때문인데그기전은아직밝혀지지않았지만세포사멸기전과연관되어있다고생각되고있으며, 또한이것이질병의진행과전이에연관되어있다고알려져있어세포사멸에관한기전연구가절실히요구된다. 세포주기조절의관점에서암세포는세포주기의비정상화에기인한질병으로서특정시기의세포주기억제는세포주기 checkpoint 각각의시기에요구되는세포 주기조절인자의발현및활성여부에따라조절된다 31,32). 첫번째는 restriction point로써 G1기에세포크기가정상인지 DNA 복제에필요한물질들을제대로합성했는지검사하는시기이며, 이지점에관여하는세포주기유전자는 CDK2/4/6 와 cyclin D/E 이다 33). 두번째는 G2/M transition 으로써세포분열을진행하기전에 DNA 복제가종결되었는지를확인하는지점이며, 이지점에관여하는세포주기유전자는 CDK1 과 cyclin B이다 34). 세번째는 M기내의 metaphaseanaphase transition 으로써방추사가제대로부착했는지확인하는지점이다. 세포분열이일어나기위해서반드시필요한인자를 mitosis promoting factor (MPF) 라고하며, CDK1 과 cyclin B 결합형효소를말한다 35). 정상세포에서는 DNA 손상시세포주기가 G1/S 기또는 G2/M 기에서정체되어손상된 DNA 를수복할수있는 DNA 수복기전이존재한다. 그러나과다한 DNA 손상이야기되어 DNA 수복이어려워지게되면 G1/S 또는 G2/M check point 에서이를감지하여 apoptosis 가유도되어게놈상의안전성을유지하고있다 36). 본연구에서는 OJEF 처리시농도의존적으로 G1 phase 는유의적으로 arrest 가증가하였고, G2/M phase 으로의진행은유의적으로감소하였다. OJEF 를농도별로처리하고 6h 배양했을때 100μg / ml처리군에서 control 에비해 G0/G1 phase peak 의 % 는 1.4배증가하였고, 같은농도에서 G2/M phase peak 의 % 는 control 에비해 2.44 배감소하였다. 이와같은결과를통하여 OJEF 가 A375 human melanoma cells 의 G1 phase 를 arrest 시킴으로서암세포증식을억제함을확인하였다. 이는 G1기이행단계를조절하는 CDK4 와 cyclin D1의단백질발현량이농도및시간의존적으로감소한것과깊은관련이있을것으로판단된다. 또한 CDK 1과 cyclin B1가농도및시간의존적으로감소한것은 G2/M phase 로의진행을늦추는것과연관된것으로보인다. 이를통하여 OJEF 가 A375 흑색종세포의세포주기정지를일으켜세포분열이제어됨으로써증식이억제될것으로사료된다. 9

10 한방안이비인후피부과학회지제 32 권제 4 호 (2019 년 11 월 ) 이상의결과를종합하면 OJEF는흑색종세포주인 A375 cells 에선택적으로시간및농도의존적인세포독성을보였으며세포주기조절과관련된 CDK1, cyclin B1, CDK4, cyclin D 억제로 G1 phase와 G2/M phase 에서세포주기정지가일어나세포분열이제어되었음을확인하였다. 따라서와송유래생리활성물질의흑색종에대한항암효과가확인되었으므로앞으로와송추출물의항암제로의개발을위해서는세포자멸사에대하여보다정확한신호전달기전연구와 in vivo 연구가필요하리라사료되며이를바탕으로다양한질병으로고통받는환자들을치료하는데도움이될치료제로서개발가능성이매우높다고사료된다. Ⅴ. 결론 A375 인체흑색종세포에대한와송의항암효과를실험적으로규명하기위하여 OJEF를투여한후암세포증식억제효과와 cell cycle arrest 유도능을측정하고 western blotting을통하여 apoptosis 관련유전자및단백질의발현을측정하여다음과같은결과를얻었다. A375 인체흑색종세포의증식이 OJEF 의처리농도와배양시간에의존적으로감소하는것을알수있었고세포주기조절을보면농도의존적으로 G1 phase 는유의적으로 arrest 가증가하였고, G2/M phase 으로의진행은유의적으로감소하였다. CDK4 와 cyclin D1, CDK 1과 cyclin B1의단백질발현량이농도및시간의존적으로감소하였다.
