As a result, growth of cancer cells as assessed by MTT assay decreased in a concentration-dependent manner. Chromatin condensation by DAPI staining significantly increased in a concentration-dependent manner, indicating apoptosis. The level of p53-protein increased while those of anti-apoptotic molecules (Bcl-2, p-gsk3β) decreased in the western blot. Tumor size decreased in cells treated with 10 mg/kg of delphinidin compared with the control group in vivo. Cell apoptosis assessed by TUNEL assay significantly increased, and tumor inhibitory effect was confirmed. Therefore, delphinidin can be developed for cancer preventive medicine due to its growth inhibitory effects and induction of apoptosis in human breast cancer cells. Key words: delphinidin, anthocyanin, anticancer, breast cancer, p53 서 우리나라는최근 10년간암에의한사망률이점차증가하는추세이며, 그중에서도유방암은여성에게발병률이높은암으로보고되고있다 (1). 국내유방암발생빈도는 2011년발표된중앙암등록본부자료에의하면 2009년전체암발생의 7% 로 6위, 여성의암중에서 2위를차지하였다 (2). 유방암은원발부위로부터림프절이나혈관을타고다른곳으로쉽게이동하여 2차성암을형성하는전이능력을가지며특히선택적으로뼈나폐로전이가잘된다고알려졌는데이러한전이성유방암환자들은수술과여러치료요법으로도개선되기힘들다 (3,4). 최근까지개발된외과적기술, 방사선치료와약물치료, 호르몬요법등과같은유방암치료방법은암환자의생존율을다소향상시키기는하지만일반적으로알려진항암치료는강한독성으로인해부작용이크고재발위험도큰것 론 Received 27 August 2013; Accepted 14 January 2014 Corresponding author. wangza@kongju.ac.kr, Phone: 으로보고되고있다. 그에따라항암제를통해치료하는기존의화학요법과달리부작용이적으면서인체에미치는고통을최소화하는암예방및치료요법을개발하는데사람들의관심이증가되었다. 더불어자연으로부터쉽게얻을수있고부작용은적으면서암을효율적으로치료할수있는, 식물유래의천연물질들에많은관심을두게되었다. 대표적으로블루베리의항암효과가대중매체를통해주목받고있다. 블루베리에포함된안토시아닌 (anthocyanin) 은식물계에널리분포되어있는페놀화합물중의하나로과실, 줄기, 잎, 뿌리등식물체각부위에폭넓게분포되어있는적색, 자색, 청색등의매력적인색을띠는수용성색소이다 (5). 기능성인측면에서안토시아닌은항산화활성 (6,7), 신경계질환 (8), 심장독성 (9), 항염증 (10) 등에효과가있으며암발생억제 (7,11), 항바이러스작용 (12,13), 지질대사조절 (14,15), 시력 (16) 및난소기능증강효과 (17) 등생리활성연구가보고되어있다. 특히채소및과실류에서안토시아닌중 cyanidin이 50% 정도로가장많으며 delphinidin, pelargonidin, peonidin이각각 12% 정도, petunidin과 malvidin은 7% 로분포하는데,

Chemical structure of delphinidin. 그중본연구에쓰인 delphinidin(fig. 1) 은담자색, 적자색및청색을나타내고항산화작용을비롯한콜레스테롤저하와항암및항염증 (18), DNA 손상의보호 (19,20) 등에효과가있는것으로알려졌다. 또한시력과기억력이좋아지고비만을억제하여심장, 뇌혈관질환과당뇨병및관절염에효과가있다고알려졌다 (21). 채소과실류에가장많이분포하고있는 cyanidin에관한연구는활발하게진행되고있는반면에 delphinidin의생리적기능성효과가 petunidin과 malvidin에비하여더우수한것으로알려졌음에도불구하고현재이에대한연구는미비한실정이다. 그리하여본연구에서는블루베리에서항암효과를나타낼것으로기대되는 delphinidin이인간유래의 MDA-MB- 231 유방암세포의증식을억제하는지확인하고, in vivo 실험에서도항암효과가나타나는지알아보고자하였다. 재료및방법시험물질 Delphinidin chloride는 Sigma-Aldrich(St. Louis, MO, USA) 에서구입하여 in vitro 실험에서는 DMSO에희석하여사용하였고, in vivo 실험에서는 D W증류수에희석하여사용하였다. MDA-MB-231 유방암세포배양실험에사용된 MDA-MB-231 유방암세포는 American Type Culture Collection(Rockville, MD, USA) 에서구입하였다. MDA-MB-231 유방암세포배양을위해 RPMI-1640배지, 5% fetal bovine serum(fbs; Life Technologies Inc., Gaithersburg, MD, USA) 과항생제 (1% penicillin-streptomycin ml) 를첨가하여 37 C, 5% CO 2 가유지되는배양기 (Thermo Electron Corp., Marietta, OH, USA) 내에서세포를배양하였다. 세포가 80~90% 정도 flask를덮으면 phosphate buffered saline solution(pbs) 으로 2번씻어내고 trypsin-edta를처리하여세포를계대배양하고배지는 2~3일마다교환하였다. 12.5, 25, 50 µm 농도조건으로처리하였다. 37 C, 5% CO 2 가유지되는배양기내에서약 24시간동안배양하였다. 그후각 well당 40 µl의 PBS에 2 mg/ml의 MTT를녹인용액을넣고호일로감싼후 37 C 배양기에 2시간동안방치하였다. 다음으로 MTT 용액을제거하고 well당 100 µl의 DMSO를첨가하여형성된 formazan을녹인다. 완전히용해된 formazan의흡광도를 ELISA-reader(Bio-Rad Laboratories Inc., Hercules, CA, USA) 로 595 nm에서측정하였다. DAPI stain 체외배양중인 MDA-MB-231 유방암세포의블루베리처리시세포의핵변화를시각화하기위하여 DNA염기서열에특이적으로반응하는 DAPI 핵염색을수행하여핵의변화를분석하였다. 96 well plates에 MDA-MB-231 유방암세포 (2 10⁵ cells/ml) 를넣은후 37 C 배양기에서배양하였다. 처리시간이끝나면배양기에서배양접시를상온으로꺼낸후 PBS로조심스럽게세척한후에 4% paraformaldehyde solution으로 15분간고정하였으며, PBS로다시세척후 PBS로 10배희석한 DAPI stain 용액을 500 µl/well 처치하였다. 처치후암실에서형광현미경 (Zeiss fluorescence microscope, Thornwood, NY, USA) 으로관찰하였다. Western blot 175T flask에 MDA-MB-231 유방암세포를 24시간배양한후 delphinidin을 0, 25 µm 농도로 24시간동안처리하였다. 그후 trypsin-edta를처리하여세포를부유상태로만들어원심분리 (1,000 rpm, 5 min, 4 C) 하였다. 차가운 PBS로세포를세척하고원심분리하여 cell pellet에 cell lysis buffer(invitrogen, Carlsbad, CA, USA) 를첨가하여 4 C에서 30분간반응시켰다. Lysate는 13,000 rpm에서 5 분동안원심분리하여상등액을취해 cell lysate로사용하였다. 추출한단백질의농도는 Bradford protein assay를이용해측정하였다. 단백질을 12% sodium dodecyl sulfate polyacrylamide gel electrophoresis(sds-page) 로크기별로분리한후 nitrocellulose membranes(bio-rad Laboratories Inc.) 에이동시켰다. Membrane은 5% skim milk로 1시간동안 blocking한후 anti-bcl-2, anti-p53, anti-p-gsk3β, anti-β-actin의 1차항체를각각첨가하여 4 C에서 overnight 하였다. 그후 anti-rabbit IgG를첨가하여 1시간반응시켰다. 각 protein band는 ECL detection reagents(pierce, Rockford, IL, USA) 를이용하여실험결

Xenograft 37 C, 5% CO 2 가유지되는배양기에서 5% FBS를첨가한 RPMI-1640 배지에 MDA-MB-231 유방암세포를배양하였다. 75T flask에 80~90% 의세포가배양되었을때 trypsin-edta를첨가하여세포를부유시켜원심분리 (1,200 rpm, 5 min, 4 C) 하였다. 그후 PBS로세척후다시한번원심분리 (1,200 rpm, 3 min, 4 C) 하여 cell pellet을얻어 1 10⁷ cells/ml가되도록하여 RPMI-1640 배지에분주하였다. ( 주 ) 오리엔트 (Seoul, Korea) 에서 5주령의 nude mice 암컷 10마리를구입하여 1주일의순화기간을보낸 nude mice 등에 200 µl( cells/ml) 씩 MDA-MB- 231 유방암세포를주입하여종양을유발시켰다. 종양조직이어느정도자라면 nude mice를마취시켜종양을적출한후 1 mm 3 크기로절편하여다시 nude mice에주입하였다. 그다음종양사이즈가군별로일정하게나오도록군분류를한후실험군 3마리는 delphinidin을선행연구논문 (22) 에서생체실험에서효과가있는것으로판단되는최저농도를선정하여 D W증류수 200 µl에희석시켜매주 3번 10 mg/kg 을같은시간에 4주간경구투여하였고, 대조군은 D W증류수 200 µl를매주 3번같은시간에 4주간경구투여하였다. 투여기간동안마우스의일반적인상태를살피고종양사이즈는주 2회측정하였다. TUNEL assay Dead End Colorimetric TUNEL System(Promega, Madison, WI, USA) 를이용한 TUNEL assay를통해 apoptosis 세포를확인하였다. 먼저 xylene을이용해탈파라핀화를거친후 ethanol을 100, 95, 85, 70, 50% 순서로이용하여함수시켰다. 그후 PBS로세척후 Proteinase K를각슬라이드에떨어트려실온에서 15분간반응시키고, equilibration buffer와 Biotinylated Nucl. Mix, rtdt를섞어각슬라이드에처리하여 37 C에서 1시간반응시켰다. 그후 0.3% hydrogen peroxide를 PBS와섞어 5분간반응후 Streptavidin HRP를각슬라이드에처리하였고, 각슬라이드에 3,3'-diaminobenzidine tetrahydrochloride(dab) solution을 10분반응시켰다. 마지막으로 methyl green으로염색후광학현미경으로관찰하였다 ( 200). 통계처리모든실험결과는평균치와표준오차를사용하여나타내고각군간비교는 one-way ANOVA에이은 t-test분석을실시하였다. 대조군과비교하여 P 값이 0.05 미만일때를통계학적으로유의성이있다고판정하였다. The significance was determined by Student's t-test ( * P<0.05 compared with untreated control). 결과및고찰 Delphinidin 이 MDA-MB-231 유방암세포생존에미치는영향 Delphinidin이 MDA-MB-231 유방암세포생존에미치는영향을확인하기위하여 delphinidin을농도 (0, 12.5, 25, 50 µm) 별로처리하여 MTT assay을통해관찰하였다. Delphinidin을각각 0, 12.5, 25, 50 µm로 24시간처리하였을때각각농도별세포생존율은 100, 100, 80, 20% 로대조군에비해 25 µm의농도에서부터농도의존적으로세포생존에대하여유의적인감소를보였다 (Fig. 2). 따라서 delphinidin이 MDA-MB-231 유방암세포에 25 µm부터농도의존적으로세포사멸을유도하는것으로판단되었다. Yun 등 (23) 의연구에의하면 HCT116 cell에 delphinidin 을 30, 60, 120 µm로처리하였을때농도의존적으로감소한다고나타났으며, Kim 등 (6) 에서는 Jurkat T cell에서 100 µg/ml, MCF-7 cell에서 125 µg/ml 감소하는것으로나타났다. Pal 등 (24) 의연구에서도 A549, NHBE, NCI- H441, SK-MES-1 cell에 delphinidin을농도별로처리한결과 NHBE cell을제외하고나머지 cell에서 20 µm의농도에서부터농도의존적으로감소를보였다. 이러한결과는본연구에서 delphinidin이 MBA-MD-231 유방암세포에농도의존적으로세포생존율을감소시킨것과같은결과로나타나 delphinidin이암세포에억제효과를보이는것으로사료된다. Delphinidin 에의한 MDA-MB-231 유방암세포의 apoptosis 효과 MTT assay을통해관찰된세포사멸이 apoptosis에의해나타난결과인지확인하기위해 DAPI stain을실시하였다. MDA-MB-231 유방암세포에 delphinidin을농도 (0, 12.5, 25, 50 µm) 별로 24시간처리후 DAPI 염색을이용하 * *

그결과 delphinidin 을 25 µm 처치한투여군에서부터 apoptosis 발현세포의증가가관찰되었으며, MTT assay 결과에서농도의존적으로세포의성장이감소한것과동일하게 DAPI 염색에서도대조군과비교하였을때 delphinidin을처리한군에서세포수가줄어들었고세포질의위축, 염색사의응축, apoptotic body와같은 apoptosis형태가관찰되었다.

MDA-MB-231 cells were treated with delphinidin for 24 hr and stained with DAPI. 여형광현미경으로관찰하였다 (Fig. 3). 그결과 delphinidin 을 25 µm 처치한투여군에서부터 apoptosis 발현세포의증가가관찰되었으며, MTT assay 결과에서농도의존적으로세포의성장이감소한것과동일하게 DAPI 염색에서도대조군과비교하였을때 delphinidin을처리한군에서세포수가줄어들었고세포질의위축, 염색사의응축, apoptotic body와같은 apoptosis형태가관찰되었다. Hafeez 등 (25) 의연구에서도 PC3 cell에대한 delphinidin의 apoptosis를현미경으로관찰한결과 60 µmol/l에서 apoptosis의발현을관찰하였고 120, 180 µmol/l로농도가증가할수록발현이증가한것으로보고하였다. 이러한결과로부터 delphinidin이 MDA-MB-231 유방암세포에대한세포사멸이 apoptosis 유도에의해이루어지는것으로여겨진다. Delphinidin 이 apoptosis 의경로단백질발현에미치는영향이전실험에서 MDA-MB-231 유방암세포에 delphinidin을 25 µm 처치한투여군에서부터 apoptosis 양성세포의유의적인증가가관찰되었으므로 Western blot을통해 apoptosis를조절하는 p53, Bcl-2, p-gsk3β 단백질의유전자발현정도를관찰하여 delphinidin이 apoptosis 발현유전자의역할에어떠한영향을미치는지확인하였다. p53 은종양억제유전자의산물로서세포증식을억제하며유전자가손상되었을경우복구하거나괴사를유도하고, apoptosis를유발하는종양억제유전자로알려졌다 (26). Western blot을실시하여 delphinidin 처리에따른 p53 단백질의발현여부를관찰한결과, p53 단백질은대조군에비해 delphinidin의처리농도 25 µm에서증가하였고, anti-apoptotic 인자로서 apoptosis 유발을억제하는 Bcl-2, p-gsk3 β 단백질의비율은 25 µm에서유의적으로감소하였다 (Fig. 4). Yun 등 (23) 의연구에서는 HCT116 cell에 delphinidin 을처리하였을때 30 µm에서부터농도의존적으로 p53 단백질이증가하였고 Bcl-2 단백질은감소한것으로나타났으며, Hafeez 등 (25) 의연구에서는 PC3 cell에 delphinidin 을 60, 120, 180 µmol/l로처리하였을때 p21, p27은농도의존적으로증가하였고 pnf-κb/p50과 pnf-κb/p65가농도의존적으로감소하였다. 결과적으로 delphinidin 이 MDA (μm) p-gsk3β p53 Bcl-2 β-actin Fig. 4. MB-231 유방암세포의 apoptosis 유도에매우중요한역할을하고있음을알수있다. Delphinidin 이 xenograft 를이용해이식한종양에미치는영향 Xenograft를실시한마우스의 MDA-MB-231 유방암세포증식에 delphinidin이어떠한영향을미치는지관찰하였다. 종양사이즈는주 2회측정하였고, delphinidin(10 mg/ kg) 은 D W증류수 200 µl에희석시켜주 3회같은시간에 4주경구투여하였고, 대조군은 D W증류수 200 µl를주 3 회같은시간에 4주경구투여하였다. 그결과경구투여 11일째부터대조군과비교하였을때, 종양의사이즈가감소하는경향을보였다. 25일째대조군에비해 delphinidin을처리한군에서 43% 정도의억제율을보였다 (Fig. 5A). 최종종양의크기는대조군은평균 3,953 mm 3, delphinidin 투여군은 1,566 mm 3 이었으며, 종양의무게는대조군 g, delphinidin 투여군은 g으로 delphinidin 투여군의종양이감소하는경향을보였다 (Fig. 5B). Scale bar size: 10 μm. phinidin을주 3회, 2 mg을복강투여하여 8주동안관찰한결과최종종양의크기는대조군은 1,200 mm 3, delphinidin 투여군은 361 mm 3 로높은억제율을나타내었고, delphinidin 투여군의종양크기가 12주가되었을때대조군의 1,200 mm 3 와비슷해졌다. 이러한결과는본연구에서나타난결과와일치하여 delphinidin이 MDA-MB-231 유방암세포에대한억제효과를보이는것으로판단되어진다. Delphinidin 이미치는조직수준에서의 apoptosis 관찰 Xenograft 부검을통해채취한종양조직에대하여 apoptosis 유도에의한항암효능을확인하고자 Tunel assay를실시하였다. 그결과대조군보다 10 mg/kg delphinidin을투여한군에서 apoptosis가일어난세포의유의적증가를관찰할수있었다 (Fig. 5C). Anthocyanin 성분들의암에대한연구중가장분포가많은 cyanidin에대한연구로, Chu 등 (27) 의연구에서는 cyanidin이 MDA-MB-231 유방암세포의이동성과침윤성에미치는영향을조사했는데 cyanidin의첨가농도가세포증식에영향을주지않는 0, 5, 10, 20 μm에서이동성및침윤성이유의적으로감소되었고, Zhang 등 (28) 의연구에서는 AGS cell, HCT-116 cell, MCF-7 cell, NCI-H460 cell, SF-268 cell에 cyanidin을 100 μm 이상으로처리했을때유방암세포인 MCF-7 에서세포증식이유의적으로감소되었다고보고하였다. Cyanidin, malvidin에관한 Choi(29) 의연구에서는 cyanidin과 malvidin이산화적스트레스에의한 WI-38 cell의지질과산화생성을유의적으로감소시켰으며이를통해 cyanidin, malvidin의산화적스트레스개선을통한항노화효과가뛰어남을확인할수있었다. 그리고 Kim 등 (6) 의연구결과에따르면 anthocyanin 함량중특히 cyanidin 함량은타계통보다 2배이상높았고, 암세포에대한독성효과로 JurKat T 세포와 MCF-7 세포에 cyanidin, delphinidin, petunidin을농도별로처리한결과 petunidin은 cyanidin에비해조금떨어지지만, delphinidin은 cyanidin에뒤지지않는암세포독성효과가있는것으로나타났다. 이러한결과는본연구에서 delphinidin이 MDA-MB-231 유방암세포에농도의존적으로세포생존율을감소시킨것과같은결과로나타나 delphinidin이암세포에억제효과를보이는것으로사료된다. 그동안 delphinidin, petunidin에비해 cyanidin의효과가부각된것은대부분식물이함유하고있는 anthocyanin 이 cyanidin인까닭에그동안의연구결과는 cyanidin 중심으로이루어진것으로생각된다. 따라서 cyanidin을포함한 anthocyanin의개별성분에대한더깊은연구가필요할것으로판단된다. 본연구는 anthocyanin의성분중항암

6 236 장혜연 이송희 안인정 이해님 김혜리 박영석 박병권 김병수 김상기 조성대 남정석 최창순 정지윤 가능성이있을것으로여겨지는 delphinidin이 MDA-MB- 231 유방암세포에미치는영향을알아보았고, delphinidin 이 MDA-MB-231 유방암세포에대해 apoptosis를유발시켜암세포증식을억제한다는것을확인하여 delphinidin이암예방제나치료제로개발될수있을것으로사료된다. 요 본연구에서는블루베리에포함된 anthocyanin 중 delphinidin이인간유래의 MDA-MB-231 유방암세포의성장을억제시키고 apoptosis를유발하는지살펴보고 in vivo 실험에서도항암효과가나타나는지확인하였다. 그결과 cell viability를보기위한 MTT assay에서는농도의존적으로암세포의성장을억제시켰으며, apoptosis의확인을위한 DAPI stain에서농도의존적으로 chromatin condensation 이유의적으로증가하는것을확인하였다. 또한 western blot에서암억제유전자인 p53 단백질이증가하였고, anti-apoptotic 분자인 Bcl-2 단백질과 p-gsk3β 단백질은감소하였다. In vivo 실험에서는대조군과비교해 10 mg/kg delphinidin을투여한군에서종양의크기가감소하였으며, TUNEL assay를통해 apoptosis 세포또한통계학적으로유의적인증가가관찰되어종양억제효과를확인하였다. 이상의결과들로볼때, MDA-MB-231 유방암세포에서 delphinidin은암세포의증식을억제시키고, apoptosis를유발시키는효과를보이므로암예방제나치료제로개발될수있을것으로사료된다. 약 감사의글 본연구는농촌진흥청의연구비지원으로수행된연구결과의일부이며 (Project NO.
