HEMA (2-hydroxyethyl methacrylate), NVP (N-vinyl pyrrolidone), MMA (methyl methacrylate) 가담긴몰드에백금나노입자를첨가한후농도조건을다양화하여 70 o C에서약 40분, 80 o C에서약 40분, 100 o C에서약 40분동안의열처리공정을각기거쳐공중합하였다. 제조된콘택트렌즈의물리적특성을측정한결과, 산소투과율은 9 ~ cm/s ml O 2/mL mmhg, 함수율은 ~ 35.52%, 굴절률은 ~ 1.435, 가시광선투과율은 88.3 ~ 91.2% 그리고인장강도값은 ~ Kgf을나타내었다. 제조된콘택트렌즈는여러가지색을띠어인공착색제를사용하지않고서도콘택트렌즈에착색효과를나타내었으며, 콘택트렌즈로사용가능한물리적특성에부합되어안의료용기능성소재로의응용이충분히가능할것으로판단된다. 주제어 : 콘택트렌즈, 백금나노입자, 착색제 ABSTRACT. Platinum nanoparticles were added to a mixture of HEMA (2-hydroxyethyl methacrylate), NVP (N-vinyl pyrrolidone) and MMA (methyl methacrylate) in a mould at various concentrations. The resulting mixture was copolymerized by heating at 70 o C for 40 min, 80 o C for 40 min, and 100 o C for 40 min, respectively. The physical properties of contact lens were then measured. The oxygen transmissibility of 9 ~ cm/s ml O 2/mL mmhg, water content of ~ 35.52%, refractive index of ~ 1.435, visible transmittance of 88.3 ~ 91.2% and tensile strength of ~ kgf were obtained. The addition of platinum nanoparticles to the polymer allowed the contact lens to have various colors without artificial coloring agents. The polymer materials satisfied the physical properties required to produce contact lenses, making the material suitable to be applied as a functional material for ophthalmological purposes. Keywords: Contact lens, Platinum nanoparticle, Coloring material 서론 나노의작고미세한크기의입자는동일무게의물질을넓은부피로사용가능하게하여그효과를증가시킬수있는장점을가지고있으며, 나노물질을활용한나노기술은물질을분석하고이를첨가하여새로운성질과특성을얻는기술이다. 이러한나노기술의파급효과는사회전반에걸쳐영향을미치고있고, 앞으로도더욱커질것으로예상되며, 현대산업에서매우다양한분야에서응용및적용되고있다. 1,2 또한최근건강에대한관심증가로나노기술은전방위산업뿐아니라의류와생활용품등과같은실생활에도적극활용되고있다. 3,4 그중백금은 Ru(ruthenium), Rh(rhodium) 그리고 Pd(palladium) 등과함께백금족에속하는원소로특히촉매로높은 효율을갖는특성을가지고있어공업적으로많이사용되고있다. 5,6 또한백금은강한전기전도성과열전도성을가지고있으며, 7-9 특히부식에대한저항이강하여귀금속으로도많이사용되고있다. 이와같은많은특성중나노백금은나노은, 나노금등과같이항균성을가지고있는것으로알려져나노백금의사용도확대되어가고있다 또한금속나노입자들은입자크기에따라띠간격 (bandgap) 의차이로특유의색을가지고있어, 이를활용한연구도진행이가능한부분중하나로볼수있다. 콘택트렌즈는안경이갖는단점이보안된안의료기구로그착용률이점차증가해왔고, 앞으로도증가할것으로예상된다. 그러나안경이갖는단점을부분적으로보완하였지만눈에직접착용해야함에있어콘택트렌즈만이갖는문제점을야기시켰다. 실제로눈은생리적으로적절한온도와습 99

2 100 예기훈 성아영 도를가지고있어많은균들로부터쉽게노출되어있다. 이로인해안과질환은급성적으로빠르게그리고자주발생한다. 이러한균에의한안과질병으로세균결막염 (bacterial conjunctivitis) 이있다. 세균성결막염은급성점액농성결막염 (acute mucopurulent conjunctivitis), 만성세균성결막염 (chronic bacterial conjuncticitis), 가성막과막결막염 (pseudomembranous and membranous conjunctivitis) 등이있으며, 이는우리가일상에서쉽게접하는포도구균과연쇄구균등과같은병원균등으로인해발생하게된다 따라서본실험에서는나노백금의항균성과고유의색을이용하여의료용콘택트렌즈에응용하였다. 또한콘택트렌즈의최근경향은미용을목적으로하는칼라가첨가된미용칼라콘택트렌즈사용이급증하고있다. 현재사용되는미용칼라콘택트렌즈는안료를사용하여콘택트렌즈에착색시키는방식이사용되고있다. 그러나나노백금재료를사용하여금속고유의색을자연스럽게얻어내어콘택트렌즈에응용하여다양한색의착색이유도될수있을것으로판단된다. 본실험에서는나노백금을안의료기구인콘택트렌즈에주로사용되는 HEMA, NVP, MMA, EGDMA 등과함께공중합하여색을가지며, 항균기능성도가질수있는의료용고분자를제조하였으며, 또한제조된고분자의함수율 (water content), 굴절률 (refractive index), 광투과율 (optical transmittance), 인장강도 (tensile strength) 등의물리적특성을실험하여, 콘택트렌즈응용으로의적합성을확인하였다. 실험 한색은제조된고분자에도다소영향을주었으며, 함량에따라다양한회색빛을나타내었다. 교반후준비된재료를열중합방식으로고분자를제조하였다. 제조과정은교반된모너머를콘택트렌즈몰드에주입후 70 o C에서약 40분, 80 o C에서약 40분그리고마지막으로 100 o C에서약 40분의열처리를공정을거쳐렌즈를제조하였다. 제조된콘택트렌즈는건조된무게를측정하고 0.9% 의염화나트륨생리식염수에약 24시간동안함수시킨후, 수화된무게와표면검사및광투과율과인장강도등의측정을통해제조된렌즈가기존콘택트렌즈의물리적특성에부합되는정도를알아보았다. 제조된콘택트렌즈의물리적특성은각각 3번씩측정하여오차를줄였으며, 이에대한평균값을나타내었다. 측정기기및분석함수율함수율은공중합된고분자의건조되었을때무게와 0.9% 의염화나트륨 (NaCl) 생리식염수에약 24시간동안함수시킨후수화된무게를각각측정하여중량측정법을통하여함수율을산출하였다. w H2 O (watercontent, %) = a W.H.L is the weight of hydrated lens b W.D.L is the weight of dried lens a W.H.L b W.D.L W.H.L 시약및재료본실험에사용된 HEMA(2-hydroxyethyl methacrylate) 와개시제 AIBN(azobisisobutyronitrile) 은 JUNSEI사제품을, MMA (methyl methacrylate) 는 Crown Guaranteed Reagents사제품을, NVP(N-vinyl pyrrolidone) 와 EGDMA(ethylene glycol dimethacrylate) 는 Acros사제품을구입하여사용하였다. 또한실험에사용된나노백금입자들은 2 ~ 5 nm 크기의 colloid 형태로사용하였다. 굴절률굴절률은 ABBE Refractormeter(ATAGO NAR 1T, Japan) 를사용하여측정하였다. 측정방법은중합을통해수화된 sample 을프리즘부에위치시킨후, 측정핸들을돌려경계선에위치될때눈금을읽어굴절률을측정하였다. 인장강도인장강도는 AIKOH Engineering사의 Model-RX series를 실험방법고분자중합을위해사용된모너머는콘택트렌즈의주재료인 HEMA (9.62 g), NVP (2.50 g), MMA (1.00 g) 그리고교차결합제로 EGDMA (0.3 g) 를사용하였다. 개시제로는 AIBN (0.001 g) 을사용하였으며, 각각의시료를배합하여약 30분동안교반하여 reference를준비하였다. 또한이와같은조합에 platinum colloid를각각 1% 에서 10% 까지농도를다양화시킨후첨가하여교반하였다. 실험에사용된 platinum colloid는약 100 ppm의농도를사용하였다. 교반후나타난시료의색변화를농도별로 Fig. 1의 (a), (b), (c) 에각각나타내었다. Percent composition of samples unit : % HEMA NVP MMA EGDMA Platinum colloid Ref PN PN PN PN PN (a) PN1-2 Valley, California, USA) 를사용하였다. 측정을위해온도를일정하게유지하기위한발열상자 (heated box) General Incubator (LIB-030M, Labtech, Korea) 를사용하여 35 o C ± 0.5 o C를유지시킨후시험시료를고정장치에의해센서의음극과맞닿아고정하여평행을이룰때의전류값을측정하였다. 측정된전류값을계산식에대입하여 Dk/t( 산소투과율 ) 를결정하였다. 결과및고찰 Fig. 2. Distribution chart of water content. 사용하여측정하였다. 측정방법은생리식염수에약 24시간동안함수시킨후, 수화된고분자를 0에서 10초시간동안 kgf에서 kgf의힘을콘택트렌즈에가해파괴될때의값을인장강도로나타내었다. 표면분석표면분석을위해 JEOL사의 JSM-7500F+EDS(Oxford) 을사용하여공중합한 sample 표면형상을관찰하였다. 산소투과율산소투과율측정은 Rehder single-chamber system - O2 permeometer model 201T(Rehder Development Company, Castro Polymer 표면분석기존콘택트렌즈의재료에 platinum colloid를첨가하여공중합한후콘택트렌즈를제조하여주사전자현미경 FE-SEM을통해분석한결과를 Fig. 2 (a), (b) 에나타내었다. SEM image 분석을통해표면에생성된나노입자의존재를확인할수있었다. 물리적특성재료의배합비수산기를포함하고있어친수성의성질을가지며, 소프트콘택트렌즈의주재료로사용되는 HEMA와락탐고리가포함되어생체친화력이뛰어나고, 높은친수성을갖는 NVP 그리고하드재료로광학적이뛰어나고, 렌즈의형상을견고히하는 MMA와가교역할을하는교차결합제로 EGDMA를사용하여배합하였다. HEMA (96.20%), NVP (2.50%), MMA (1.00%), EGDMA (0.30%) 를기본조합으로하고, platinum colloid의양을일정량증가시켜배합하였다. 이에대한배합비를 Table 1 에요약하여나타내었다. 또한이를각각농도별로분류하여 Ref. PN1, PN3 와 PN5, PN7 그리고 PN10 으로명명하였다. 함수율건조무게와수화무게그리고중량측정법에의해얻어진평균함수율을 Table 2에나타내었으며, 측정된함수율의경향과분포를 Fig. 3에나타내었다. Platinum colloid를첨가하지않은 Reference 는중합후얻어진건조무게와함수후얻어진수화무게의차이가 g 차이를, 이에따른평균함수율은 34.22% 를나타내었다. PN1은 g 차이와 34.33% 의평균함수율을, PN3은 g과 34.82%, PN5는 g과 35.19%, 그리고 PN7은 g 차이와 35.52% 의평균함수율을나타 2010, Vol. 54, No. 1

Reference는 1.433, PN 를, PN , PN 을그리고 PN10은 1.432를나타내었다. 이결과로볼때나노백금의첨가가생성된고분자의굴절률에는큰영향을주지않는것으로나타났다. 광투과율중합하여얻어진콘택트렌즈의평균광투과율을 Table 4에 나타내었으며, 대표적으로 sample PN2 의측정결과를 Fig. 5 에 나타내었다. Ref. 의 UV-B, UV-A 그리고 visible transmittance 는각각 88.5%, 89.2%, 91.2% 의평균광투과율을나타내었다. PN1의경우 82.3%, 87.9%, 91.2% 를 PN3은 82.2%, 86.8%,

Avg PN1 Avg PN3 Avg PN5 Avg PN7 Avg PN10 Avg 산소투과율중합한고분자를사용하여제조된콘택트렌즈의산소투과율을측정하였으며, 대표적으로 sample PN3의결과를 Fig. 7 에나타내었다. 측정된전류값은 3.3 Ampere, 산소투과율은 cm/s ml O 2/mL mmhg 을나타내었다. 결론 [S] Fig. 6. Tensile strength of PN % 를, PN5는 81.1%, 86.6%, 89.2% 를나타내었으며, PN7 은 82.2%, 86.1%, 89.6% 그리고 PN10은 80.2%, 87.3%, 88.3% 를각각나타내었다. 이결과로볼때생성된고분자의가시광선영역에대한전체적인광투과율은비교적높은값을나타내는것으로판단된다. 인장강도중합한고분자를사용하여제조된콘택트렌즈의평균인장강도를 Table 5에나타내었으며, 대표적으로 sample PN1 의결과를 Fig. 6에나타내었다. 모든 sample에대한인장강도는 kgf ~ kgf의범위에서얻어졌다. 본연구는나노백금입자를여러가지모너머와함께중합하여의료용소재인콘택트렌즈에적용시켜기능성고분자소재를제조하였다. 제조후기존콘택트렌즈에해당하는물리적특성에부합되는지를함수율, 굴절률, 광투과율, 인장강도를통해알아보았다. 그결과 platinum colloid는함수율에영향을주는것으로나타났으며, Ref. 와비교하여 PN1~7 은 platinum colloid의양이점차증가할수록함수율이 1.30% 증가하는경향을나타냈다. 그러나 PN10에서는 34.76% 로다소감소하는경향을나타내었다. 평균굴절률은 ~ 의범위를, 평균광투과율 UV-B, UV-A, Visible transmittance 에서각각 80.2% ~ 82.3%, 86.1% ~ 87.9%, 88.3% ~ 91.0% 의범위, 또한평균인장강도의경우 kgf ~ kgf의범위를나타내어, reference sample과많은차이를나타내지않았다. 본실험결과생성된고분자는착색효과를지니면서동시에항균성을포함한콘택트렌즈및기능성의료용고분자에적용이가능할것으로판단된다.
