Methods : We used 2 kinds of cancer cell lines such as colon cancer cells(ht-29), human oral epitheloid carcinoma cells(kb). MTT assay was performed to examine the efficacy of SM extracts on the cytostaticity of cancer cells in proportion to time and doses. Apoptosis was evaluated by DNA laddering and DAPI nuclei staining. Results : The MTT absorbances against HT-29 and KB of SM extracts were significantly decresed. DNA ladders could be identified in KB of SM extracts. The morphological change were observed and number of cells were decreased by SM extracts. Conclusions : SM extracts is considered to be effective to induce apoptosis and inhibit cancer cell proliferation. 2 대한한의학방제학회지제 22 권제 2 호 (2014 년 12 월 ) Herbal Formula Science(HFS) 2014;22(2):35~43 5) Ⅰ. 서론암은환경오염과식생활습관, 흡연, 음주등으 로인하여발병하며, 개체를구성하는정상세포가어떠한자극에의해유전자의형질전환이발생하고그결과세포의형태학, 생물학, 물리학, 면역학적행동이변한변형세포가유전적으로대를이어무절제한증식을하여발생한다 1). 암에대한치료법은수술요법, 방사선요법, 면역요법, 약물요법등으로알려져있는데, 현재사용하고있는항암제는세포독성을이용하여암세포의사멸과증식억제를이용하는방법으로암세포의성장을효과적으로억제하기도하지만정상세포와면역계세포에도독성을발현하는문제점을나타내고있다 2). 따라서부작용을감소시키면서암세포에대한특이적독성을나타내는항암제를합성물질이아닌천연물질에서개발하려는노력이시도되고있고, 한의학계에서도기존의항암치료의한계를극복하기위한노력으로한약제의항암효과와면역증진및항암제의부작용억제와항암제와의병용요법등에관한연구들을진행해오고있다 3). 암에대한한의학의인식은 黃帝內經 에서 石瘕, 腸覃 등으로언급한이래, 癌, 腫瘍, 乳巖, 石疽, 石瘕 등으로인식하였고 4,5), 治法에대해서는歷代로부터 堅者削之, 結者散之, 留者攻之, 損者益之 의 4대방법이설정되어왔고, 近來에는理氣化血, 通經活絡, 化痰利濕, 軟堅散結, 解毒止痛, 益氣養血, 健脾和胃, 滋補肝腎등의治法이활용되고있다 6). 단삼 (Salviae miltiorrhizae Radix) 은꿀풀과에속한다년생초본으로活血祛瘀, 調經止痛, 養血安神, 凉血消癰의효능이있어癥瘕積聚, 癰瘡腫毒, 月經不調, 心腹疼痛, 煩燥不安등의병증을치료한다 7). 단삼에대한연구로항암작용 8,10) 및항균, 항염증작용,9,11), 심혈관질환에대한응용 12,13) 등다양한연구가이루어졌고, 사람암세포주에대한항암작용및세포독성에대한연구 8,9,20) 도이루어졌으나암세포주의 apoptosis에의한세포증식억제효과를보여주는실험은미비하였다. 이에저자는암세포증식억제효과를규명하고자대장암세포주 (HT-29) 및구강상피세포암세포주 (KB) 세포를대상으로 MTT assay, DNA laddering 등을수행하여유의한결과를얻었기에이에보고하는바이다. Ⅱ. 재료및방법 1. 재료 1) 약물추출단삼 (Salviae miltiorrhizae Radix) 100 g에증류수 600 ml를가한후대웅약탕기 (DWP-1800 T, 한국 ) 로 3시간동안전탕한다음여과하고상층액 100 ml을얻었다. 저온순환수조 (COOL ACE CA-1500) 에서 1차동결한다음동결건조기 (Samwon, SFDSMO06, 한국 ) 로동결건조하여 8 g의분말을얻었다. 2. 방법 1) 세포배양사람대장암세포주 (HT-29) 및구강상피세포암세포주 (KB) 세포를 RPMI medium 1640(Invitrogen, Carlsbad, CA, USA), 10% heat-inactivated Fetal Bovine Serum(FBS; Invitrogen, Carlsbad, CA, USA) 배지를이용하여 5% CO2, 37 조건에서계대배양을하여실험에이용하였다. * Corresponding author : Chang-won Choi, PhD. DNA laddering MTT assay에서가장뛰어난효과를보인 KB 세포주 1x10 6 개를직경 6cm dish에분주하여 24시간배양한후, 단삼을 72시간처리하고 DNA laddering Kit (ApopLadder Ex TM, TAKARA, Japan) 를이용하여 DNA를추출하였다. 추출된 DNA를 1.5% agarose gel에서전기영동을실시한후 SYBR safe DNA gel stain (Invitrogen, Oregon, USA) 을이용하여염색을시행하였다. Ⅲ. 결과 1. MTT assay 1) 1일 HT-29 세포주와 KB 세포주를 24시간배양한후 MTT assay를통해서얻은세포의밀도는 Fig. 1과같다. HT-29 세포주에서는단삼추출액의농도가 1.0 mg / ml까지는세포의밀도변화가없었으나, 단삼 1.5 mg / ml투여군에서는대조군에비하여유의성 (p<0.05) 있게감소하였다 (Fig. 1). KB 세포주에서는단삼추출액의농도가 1.0 mg / ml부터세포의밀도가감소하였으며, 단삼 1.5 mg / ml투여군에서는대조군에비하여유의성 (p<0.05) 있게감소하였다 (Fig. 1). 4. Apoptosis 1x10 4 개의 HT-29, KB세포주를 Multicell 8 plus TM (CTRLbio, Korea) 에각각분주하여 24시간배양한후, 단삼 1.5 mg / ml를 72시간처리하였다. 이어 4% paraformaldehyde에 10분간고정하고 4',6-diamidino-2-phenylindole(DAPI) 에 10분간염색하고봉입한후 LSM 고촛점현미경 (Carl Zeiss, Standort Gottingen-Vertrieb, Germany) 하에게관찰하였다. 5. 통계분석 MTT assay에서얻은흡광도를 one-way ANOVA 로시행하였으며각군간의통계분석법은 95% 신뢰수준으로 student-newman-keuls 방법을사용하였다. HT-29 세포주에서는단삼추출액의농도가 0.5 mg / ml부터세포의밀도가감소하기시작하였으며, 단삼 1.5 mg / ml투여군에서는대조군에비하여약 55% 감소하였고통계적으로유의성 (p<0.05) 을보였다 (Fig. 2). KB 세포주에서는 0.1 mg / ml의단삼추출액을투여한군에서부터미약한감소가있었으며, 1.0 mg / ml의단삼추출액투여군에서는약 10% 세포밀도가감소하였고, 1.5 mg / ml투여군에서는대조군에비하여약 50% 감소하였으며통계적으로유의성 (p<0.05) 을보였다 (Fig. 2). 3) 5일 HT-29 세포주와 KB 세포주를 5일간배양한후 MTT assay를통해얻은세포의밀도는 Fig. 3 과같다. Apoptosis 성장을 하거나 다른 부위로 확산과 전이를 일으 1) HT-29 세포주 켜 정상적인 조직기관을 파괴할 뿐만 아니라, 숙 대조군의 HT-29 세포주에서는 대부분의 암세 주에게 일련의 대사장애와 영양장애 등을 일으켜 포들이 DAPI-염색 결과 핵 모양이 둥글게 관찰 항병력을 약화시켜 대부분 사망에 이르게 하는 되었으나(Fig. 5a), 3일간 단삼 1.5 / 를 처리한 질병이다14). 최근 암 발생률은 산업의 급격한 발 결과 많은 세포의 핵들에서 세포고사 변화(apoptotic 달, 지구 생태계 및 식생활의 변화 등으로 과거에 change)를 보여주었다(Fig. 5b). 비해 급격히 증가하고 있으나, 아직도 암의 발생 기전이 불명확하여 난치성 질환으로 알려져 있다2). 2) KB 세포주 암에 대한 치료법은 수술요법, 항암화학요법, 정상 대조군의 KB 세포주의 핵들은 둥근 모양 방사선요법, 면역요법 및 유전자요법 등이 알려져 으로 관찰되었으나(Fig. 6a), 단삼 1.5 / 를 3일 있는데 그 중 사용빈도가 높은 화학요법과 방사 간 처리한 실험군에서 다수의 세포들에서 핵분절 선요법은 정상세포까지 독성을 나타내어 소화 장 (nuclei fragmentation) 이 관찰되었다(Fig. 6b). 애, 혈액학적 변화, 면역 기능 저하, 조혈장애, 유 39

6 대한한의학방제학회지제 22 권제 2 호 (2014 년 12 월 ) Herbal Formula Science(HFS) 2014;22(2):35~43 전자손상등의부작용을초래하고있다 1,2). 최근에는서양의학적인암치료법의한계를극복하기위하여기존의치료에한약을병행하는치료를함으로써암치료의효율을높이고생존율을증가시키는연구가이루어지고있으며, 화학요법이나방사선요법의부작용을줄이기위한노력들이이루어지고있다 15,16). 암에대한한의학의인식은 黃帝內經 에서 石瘕, 腸覃 등으로언급한이래, 癌, 腫瘍, 乳巖, 石疽, 石瘕 등으로인식하였고 4,5), 癰疽와癌의관계는浮腫, 發熱, 疼痛하면서肌肉間에발생하는것을癰이라하고, 腫根이堅固하면서熱이없고暗紫色을띠는것을疽라하며, 紫黑色을띠고堅硬하며潰瘍이됨으로써巖穴과같은상태가되는것을癌이라하여癌이癰疽形態의하나라고분류하였다. 治法에대해서는歷代로부터 堅者削之, 結者散之, 留者攻之, 損者益之 의 4대방법이설정되어왔고, 近來에는理氣化血, 通經活絡, 化痰利濕, 軟堅散結, 解毒止痛, 益氣養血, 健脾和胃, 滋補肝腎등의治法이활용되고있다 6). 본실험에사용된단삼 (Salviae miltiorrhizae Radix) 은꿀풀과 (Labiatae) 에속한다년생초본으로活血祛瘀, 調經止痛, 養血安神, 凉血消癰의효능이있어癥瘕積聚, 癰瘡腫毒, 月經不調, 心腹疼痛, 煩燥不安등의병증을치료한다 7). 약리작용으로는관상동맥확장작용이있어서혈류량을현저히증가시키고, 지질대사를활성화시켜콜레스테롤을강하시킨다. 또한혈압강하, 간기능활성화, 진정작용, 항염증작용, 항암작용, 항균작용이있을것으로보고되고있다 17). 단삼의주요화학성분으로는 tanshinoneⅠ, ⅡA, ⅡB등을포함하는 diterpene 화합물과, danshensu(salvianic acid A), protocatechuic aldehyde, salvianolic acid B 등을포함하는 phenolic 화합물등, 그외에 baicaline, b-sitosterol, ursolic acid, vitamin E와 tannin 등이알려져있다 18). 단삼에대한실험적인연구로는남 19,20) 등이세포독성을억제한다고하였으며, 성 21) 등은항암효과와활성물질분리에대한연구를하였고, 정 8) 등은단삼의추출물중에암세포증식에대한억제 효과를가진성분이존재한다고보고하였다. 이외에항균, 항염증작용,9,11), 심혈관질환에대한응용 12,13), 항산화 22), 항돌연변이 23) 등다양한연구가이루어졌고, 사람의암세포주에대한항암작용및세포독성에대한연구 8,9,20) 도이루어졌으나암세포주의 apoptosis에의한세포증식억제효과를보여주는실험은미비하였다. 이에저자는암세포증식억제효과를규명하고자대장암세포주 (HT-29) 및구강상피세포암세포주 (KB) 세포를대상으로 MTT assay, DNA laddering 등을수행하였다. 암세포증식억제효과를분석하기위해서암세포를배양기에서배양하면서다양한농도의단삼추출액을투여하여세포의증식정도를알수있는 MTT assay를통하여객관적으로분석하였다. Tetrazolium-based colorimetric(mtt) assay 는 Mosmann 24) 이개발하고 Kotnik 25) 등이변형시킨방법으로많은시료를간단하고빠르게판독할수있어배양된세포의세포독성에관한연구에주로사용되었다. 이방법은대사과정이온전한세포의미토콘드리아탈수소효소가노란색수용성 tetrazolium salt 3-[4,5-Dimethylthiazol-2-yl]-2,5-diphenyltetrazol ium bromide 를자주색을띄는비수용성의 formazan 결정으로환원시키는원리를이용하여 570nm의파장에서흡광도를측정함으로써대사적으로왕성한세포의농도를반영한것이다. 실험의결과는 HT-29, KB 세포주에서단삼추출액을농도별로처리한후 1일, 3일, 5일배양하여 MTT assay를통해세포의밀도를측정한결과단삼추출액의농도에따라세포밀도의감소정도가차이가있었으나단삼추출액농도의증가에따라세포의밀도가감소하는경향을보였고, 단삼 1.5 mg / ml투여군에서는가장현저한감소를보였으며통계적으로유의성 (p<0.05) 있는결과를나타내었다. 이는단삼추출액이 HT-29, KB 세포주의암세포증식을억제하는효과가있다고사료되었다. 그러나정확한직접투여량과투여기간에대한유의성은아직불명확하여이에대한추가적인검증이필요할것으로사료된다. 기존에사용되고있는항암제의대부분은암세 40

7 양의호외 2 인 : 단삼추출물이암세포주에미치는세포독성효과 Yang et al., Cytotoxic Effect of the Salvia miltiorrhiza Bunge Extracts 포의 apoptosis를유도하여암세포의증식을막는것으로알려져있다. apoptosis 여부를판단하기위해많은방법들이사용되는데, 형태적인방법에는 HematoxylinEosin staining, Hoechst3342 형광염색법, TUNEL 정량법, AnexinV-FITC and PI staining 등이있고생화학적인방법으로 DNA laddering이있으며 cell cycle 분석에의한 Flow cytometry 이용법등이주로이용되고있다 26,27). 본실험에서는단삼이암세포의성장억제효과를나타내는지확인하기위해서세포자연사의가장특이적인현상으로알려진 DNA laddering 방법을사용하였다. DNA laddering이일어나는것은 apoptosis 가일어나면서분비가증가되는 endonuclease에의해 nucleosome이라는반복적인구조를가지는 DNA에 nucleosome 단위로 fragmentation이일어나게됨으로써관찰되어진다. 따라서전기영동후 gel상에서 DNA ladder를관찰할수있으며이는 apoptosis를판정할가장흔히사용되는방법이다 28). 단삼에투여후암세포의성장억제에가장뚜렷한억제효과를보였던구강상피세포암주 (KB cell line) 의 3일군에서 DNA laddering을실시하였다. 그결과대조군에서는하나의 band가관찰되었으나단삼 1.5mg / ml를투여한결과여러개의 band가관찰되었다. 이와같은결과는단삼의추출물이암세포의핵에영향을미치어 apoptosis에의한 cell death를유발할것으로사료되었다. Apoptosis를관찰하기위해 HT-29와 KB 세포주에 3일간단삼 1.5mg / ml를처리하고 DAPI 염색한후관찰한결과 HT-29 세포주의많은세포핵에서세포고사변화를관찰할수있었고 (Fig. 5b), KB 세포주에서는다수의핵분절 (nuclei fragmentation) 이관찰되었다 (Fig. 6b). 이는단삼추출물이암세포 apoptosis에의한 cancer cell death를유발하였다고판단되지만, apoptosis가 cell death receptor 를통하는지 mitochondrial pathway 를통하는지는추후연구가더필요하다고사료된다. 이상의실험결과를요약하여보면대장암세포주 (HT-29) 및구강상피세포암세포주 (KB) 를대상으로 MTT assay, DNA laddering 등을수행한결과암조직에서유래한세포의 apoptosis를유도 하여암세포증식을억제하는효과가있다고유추해볼수있다. 그러나단삼추출물의암세포증식억제를유도하는최적의농도와생체내의유효성및처리시간에따른효과의증감등의추가적인연구가필요할것으로사료된다. Ⅴ. 결론단삼추출물의항암활성과 apoptosis 효과를확인하기위해대장암세포주 (HT-29) 및구강상피세포암세포주 (KB) 세포를대상으로 MTT assay, DNA laddering 등의실험을하여다음과같은결론을얻었다. 1. MTT assay 결과, HT-29 및 KB세포에서단삼추출액농도의증가에따라세포의밀도가감소하는경향을보였고, 1.5 mg / ml투여군에서가장현저한감소를보였으며유의성있는결과를나타내었다. 2. KB 세포 3일군에서 DNA laddering 을실시한결과대조군에서는하나의 band가관찰되었으나 1.5mg / ml투여군에서여러개의 band가관찰되었다. 3. Apoptosis 를관찰한결과, HT-29 세포주의많은세포핵에서 Apoptosis변화를관찰할수있었고 KB 세포주에서는다수의핵분절 (nuclei fragmentation) 이관찰되었다. 이러한결과들을통하여단삼추출물은 apoptosis 를유도하여암세포증식을억제하는효과가있다고판단되며향후지속적인연구가필요할것으로생각된다. 참고문헌 1.
