Intrinsic and extrinsic factors induced decrease of collagen expression and increase of MMP1 expression by these factors. Especially, decrease of collagen induces skin aging and formation of wrinkles. Sericin, major ingredient protein of silk synthesized by silk worm, induce increase of collagen and decrease of MMP1 and used collagen substitute in cell culture. In addition, alpha-mangostin is one of major ingredients in mangostin. In our study, we investigated whether alpha-mangostin induced synergetic effects with sericin on collagen type 1a1 expression and anti-wrinkle effects. In vitro study, sericin only induced collagen type 1a1 expression in human dermal fibroblasts (HDFs). However, cotreatment with alpha-mangostin reinforced increase of collagen type 1a1 expression in sericin-treated HDFs. In addition, to evaluate efficacy on aging skin phenomena, clinical efficacy trials were conducted using Antera 3D. Improvement of wrinkle was assessed using ANTERA 3D. Results of clinical efficacy shown that sericin and alpha-mangostin contained mask decreased wrinkle depth of crow s-feet in using time dependent manner. Overall, sericin and alphamangostin synergistically improves ECM structure using increase of collagen expression. These results suggest combination with sericin and alpha-mangostin as a new, effective anti-aging cosmetic ingredient. Keywords: Sericin, alpha-mangostin, Collagen, Normal human dermal fibroblasts, Wrinkle 피부의노화는발생요인은크게내적요인과외적요인으로나 눠진다. 내적요인으로는호르몬의변화, 대사작용에인한활성산소의축적, teromere의감소가있고, 이로인해자연노화가발생된다 (Rittié & Fisher, 2002). 외적요인은다른말로환경적요인으로예를들어자외선, xenobiotics, 물리적자극이있 Both authors contributed equally to this work. *Corresponding author: In-Sook An, 2nd Floor, Venture Center II, Osongsaengmyeong 1-ro, Osong-eup, Heungdeok-gu, Cheongju-si 28160, Chungcheongbuk-do, Republic of Korea Tel: , Fax: anis@skinresearch.or.kr Received August 8, 2015; Revised November 25, 2015; Accepted December 6, 2015; Published December 30, 2015 고, 대표적으로광노화를유발시킨다 (Fisher et al., 2002). 이러한피부노화는표피, 진피, 피하지방, 세포외기질에서전반적으로진행되어나타나는현상이지만, 특히모든피부노화에서공통적으로발생되는주름은진피층의세포외기질에변형에의해서발생된다고잘알려져있다 (Baumann, 2007). 진피층의세포외기질은콜라겐 type I, III, V, VII와 elastin, proteoglycans, fibronectin 등으로구성되어있는데, 그중콜라겐 type I이대부분을차지한다 (Bateman et al., 1996; Uitto, 1986). 건강한피부에서는진피섬유아세포를통해서콜라겐 type I이합성되어진피층의세포외기질의구조를유지하지만, 노화가진행되는피부에서는콜라겐 type I의발현량이감소하기때문에세포외기질의구조를유지하지못하고세포외기질의변형이발생되는것으로알려져있다 (Bateman et al., 1996; Uitto, 1986). 콜라겐 type I의발현감소외에노화가진행되는피부에서는 matrix metalloproteases (MMPs) 729

2 의발현이높게유지되는것으로알려져있다. MMPs는피부진피층의세포외기질을분해하는단백질인데, 그중 MMP1 은콜라겐 type I을분해하여피부노화및진피층의세포외기질의변형을유도하는단백질로잘알려져있다 (Fisher & Voorhees, 1998). 때문에화장품분야에서는콜라겐 type I과 MMP1의발현조절경로를연구하고, 이를통해서피부노화를개선시키려는노력을하였다 (Groß et al., 1999; Varani et al., 2001; Dy et al., 1999; Angel et al., 2001). 항노화용스킨케어제품에는주로 vitamins C&E, coenzyme Q10, ferulic acid 및다양한천연추출물등을이용하여피부의노화를억제한다 (Lupo, 2001). 이중 sericiin은누에고치에서나오는 silk의주요성분중하나로피부의보습기능을상승시키고, 자외선및산화적스트레스에인한세포사멸을억제하고, wound healing 을유도한다고알려져있다 (Padamwar et al., 2005; Dash et al., 2008; Aramwit & Sangcakul, 2007; Takahashi et al., 2003). 또한피부진피세포에서는콜라겐 type I의발현을증가시키기때문에여러화장품및화장의료품에서 sericin을사용하고있다 (Zhang, 2002; Thitinan et al., 2013). 또한망고스틴은열대상록수인 Garcinia mangostana L. (Clusiaceae) 의과육으로, 과피추출물을이용하여기능성원료로사용하고있다. 항산화, 세포독성항염증및항암에도효과있는활성물질을다량함유하는것으로알려져있고, 이러한활성성분중 xanthone계열의 α-mangostin이대표적으로알려져있다 (Jung et al., 2006; Balunas et al., 2008; Han et al., 2009; Chairungsrilerd et al., 1996; Chomnawang et al., 2009). 때문에본논문에서는 sericin과 α-mangostin에의한피부개선효과를보고이에대한상승효과가나타나는지확인하고, 이를통해서주름기능제품에적용하여그효능을검증하고자한다. 1. 세포배양및시료인간진피섬유아세포는 Lonza사에서구매하여사용하였느며, 세포배양액은 10% fetal bovine serum (FBS; Gibco), 10% penicillin (100 unit/ml) 과 streptomycin (100 μg/ml) 가들어가 Dulbeco s Modified Eagle s Medium (DMEM; Gibco, USA) 을이용하였다. 배양조건은 37, 5% CO 2, 습윤한상태에서배양하였다. 2. 세포생존률측정 WST-1 assay를통해서세포의생존율을측정하였다. 인간진피섬유아세포를 96 well plate에 개를 24 h 배 양후, 각각의실험조건을처리하였다. 이후 24 h 후 WST- 1 solution을 10배희석하여가하고, 30 min 후 plate reader (Bio-Rad, USA) 로 595 nm의파장에서흡광도를측정하였다. 3. MMP1과 Col1a1의 mrna 발현측정 MMP1과 Col1a1의 mrna 발현량을보기위해서 Quantitative real time polymerase chain reaction (qrt- PCR) 을이용하여측정하였다. Total RNA를추출하기위해서 Trizol (Invitrogen, USA) 을이용하였다. 추출된 RNA 2 μg을 2 μg의 oligo dt와함께 94 에서 5 min간 denaturation 시킨후, 2X reverse transcriptase mix (Enzynomics, Korea) 를넣고, 37 로 1 h 반응시켜각각의샘플에대한 cdna를합성하였다. 합성된 cdna는 0.2 μm primers, 50 mm KCl, 20 mm Tris/HCl ph8.4, 0.8 mm dntp, 0.5 U Extaq DNA polymerase, 3 mm MgCl 2, 1X SYBR green (Invitrogen) 조건에서 LinegeneK (BioER, China) 을이용하여 qrt-pcr 을수행하였다. MMP1의 primer는 MMP1 forward primer, 5 -CTTTCTGGAAGGGCAAGGAC-3 ; MMP1 reverse primer, 5 -TTGCCTCCCATCATTCTTCA-3 ; Col1a1 forward primer, 5 -GRGCTAAAGGTGCCAATGGT-3 ; Col1a1 reverse primer, 5 -CTCCTCGCTTTCCTTCCTCT-3 을사용하였으며, β-actin는 β-actin forward primer, 5 -CGACAGGATGCAGAAGGAG-3 ; β-actin reverse primer, 5 -ACATCTGCTGGAAGGTGGA-3 를사용하였다. Primer는 Primer 3 (bioinfo.ut.ee/primer /primer3) 를이용하여 design하였다. β-actin은샘플간의 normalization 을위해서사용하였다. 4. 피험자선정본연구는한국피부임상과학연구소의연구윤리심의위원회의승인은받았다. 피시험자의선정기준은자발적으로모집된만 30세이상의서울및수도권에거주하는성인여성중에서피부질환을포함하는급 만성신체질환이없고, 자발적으로동의서를작성하고서명한사람이면서선정제외기준에해당사항이없는여성 23명을최종피시험자로선정하였다. 이들에게는본인체적용시험과관련된모든정보를충분히알렸으며, 피시험자는자의에따라동의서를작성하고시험에참가하였다. 피시험자선정제외기준임신또는수유중인여성과임신가능성이있는여성, 스테로이드가포함된외형제를 1개월이상사용한여성, 민감성 과민성피부를가진여성, 동일한시험부위에유사효능이있는화장품및의약품을 3개월이내에사용한여성, 연구시작 6개월내에시험부위에시술을받은여성, 피부이상소견이있는여성은피시험자선정에제외시켰다

(A) Sericin, (B) alpha-mangostin 5. 피험자눈꼬리주름깊이측정피시험자의피부상태분석시측정값의오차를최소화하기위해, 피시험자는측정할안면부위를동일한세안제로세안후 30 min간항온항습실 ( 온도 : 22±1, 습도 : 45±5%) 에서안정을취하도록하고눈꼬리주름, 볼광대리프팅, 피부톤, 피부결변화를측정하였다. 측정기기는학계에서가장보편적으로많이사용되고있는 ANTERA 3D (Miravex, Ireland). 눈꼬리주름깊이측정피시험자 23명을대상으로눈꼬리주름깊이변화에대한분석을실시하였다. 기기측정은시험제품사용전과 1주사용후에이루어졌으며, ANTERA 3D를적용하여눈가주름깊이개선평가를확인하였다. 측정부위는피험자의오른쪽눈꼬리부위였으며, 측정한이미지는 ANTERA 3D 전용소프트웨어인 ANTERA pro software를이용하여분석하였다. 측정및분석의단위는 mm이며측정된수치가감소할수록눈가주름이개선된것을의미한다. 6. 통계처리모든실험에서는 Student t-test방법을이용하여실험결과간의통계적유의성을판단하였고, 임상실험의경우특이성에따라 Paired Student t-test 를진행하였다. t-test 결과 p value 가 0.05 이하인경우값을유의성있다고판단하였다. 1. 인간진피섬유아세포에서 sericin과 alpha-mangostin의세포독성인간진피섬유아세포에서 sericin과 alpha-mangostin의독성을알아보기위해서 0, 40, 80, 120, 160, 200 μg/ml의 sericin과 alpha-mangostin를인간진피섬유아세포에처리하고 24 h 뒤에서세포독성을측정하였다. Figure 1에서와같이 sericin과 alpha-mangostin이 μg/ml의농도구간에서세포의성장을저해하지않는것으로확인이되어서, 안전성을가지는 200 μg/ml이하에서실험을진행하였다. 2. Collagen type 1a1의 mrna발현에서 sericin과 alphamangostin의상승작용 Sericin은이전의보고에서 collagen type1a1의발현을높이고, MMP1의발현을감소키는것으로알려져있고, alphamangostin에대한보고는아직없다 (Thitinan et al., 2013). 때문에 Sericin과 alpha-mangostin에의한각각의 collagen 731

Figure 2에서와같이 sericin과 alpha-mangostin를각각처리하였을때 sericin에의해서 collagen type1a1이농도의존적으로 50, 100, 150, 200 μg/ml sericin에서 , , , % 증가하는것을확인할수가있었지만, alpha-mangostin에의해서증가하는것은확인할수없었다. 또한동시에 sericin과 alpha-mangostin를처리한결과, sericin에의해서증가되는 collagen type1a1의 mrna발현이 alpha-mangostin에의해서증가폭이높아지는것을확인하였다. 때문에 sericin과 alpha-mangostin이상승효과가있는것을확인할수있었다 (Figure 3). 때문에실제피부에서 collagen의발현을유도하여피부의주름을감소시킬것으로예상할수있다. 3. Sericin과 alpha-mangostin의함유한마스크를이용한임상적용시험마지막으로 sericin과 alpha-mangostin이동시에들어간마스크를제작하였고, 이를이용하여사람피부에서임상실험을통해서 sericin과 alpha-mangostin에의한상승작용이주름에미치는영향을확인하였다. 임상실험은 sericin 과 alphamangostin을함유한마스크팩사용전과후를비교하여주름감소효능을확인하였다. Figure 4에서보는것과같이 23명의임상피험자에서마스크팩사용전에눈꼬리주름의깊이를측정한결과평균적으로 mm로측정되었다. 동일한피험자를대상으로 sericin과 alpha-mangostin이동시에들어간마스크를사용하고 2주, 4주후에동일한위치의눈꼬리주름의깊이를측정한결과 2주후에 mm로사용전에대비하여 9.58% 유의적으로감소하였고, 4주후에는 mm로 13.34% 유의 Figure 4. The clinical effect of sericin and alpha-mangostin on depth of crow s feet. 적으로감소하여 Sericin과 alpha-mangostin의함유한마스크가피부의주름을감소시킬수있음을확인하였다. 본실험에서는누에고치의실크성분인 sericin과열대과일인망고스틴주성분인 alpha-mangostin에의한항노화효과를확인한논문이다. Sericin은 collagen과 MMP1의발현을조절하는것으로알려져있고, 세포외기질로써세포배양에사용되는 collagen의대체물질로사용되어서세포의성장을돕는것으로알려져있다. 이에망고스틴의주요성분인 alpha-mangostin 을처리하였을때 Collagen의발현에상승작용이발생되는지를확인하였다. 각각이 collagen type 1a1의발현에미치는영향을확인하기위해서 sericin과 alpha-mangostin 각각단독으로처리하였을때 sericin 의경우농도의존적으로가장대표적인 collagen유전자인 collagen type 1a1의 mrna가증가하는것으로확인하였다 (Figure 2A). Alpha-mangostin을처리한결과세포의독성이없는것으로확인된 alpha-mangostin 200 μg/ml 이하의농도구간에서 collagen type 1a1의발현이증감하지않았다 (Figure 2B). 하지만 Figure 3에서와같이 sericin 을처리해주고 alpha-mangostin처리한결과 mangostin 의존적으로 collagen type 1a1의 mrna발현이증가하는것을확인하였고, 동량의 sericin만단독으로처리하였을때보다더많은발현을보여주었다. 때문에 sericin과 alpha-mangostin 간의상호작용을통해서 collagen의발현을증가시길수있음을확인하였다. 이러한세포생물학적인결과를통해서주름개선효능이있다고판단되었기때문에임상적용을통해서인체피부에서의주름개선효과를확인하였다. 임상실험은 sericin 과 alpha-mangostin이포함된마스크팩을이용하여서실험전과후의주름개선효과를확인하였다.
