Ltd., Kimje , Korea (Received on August 8, 2011; Revised on January 27, 2012; Accepted on Feruary 28, 2012) This study was conducted to examine the effects of the seeding rate on the disease incidence of sharp eyespot (Rhizoctonia cerealis) on three different varieties and to select effective chemicals to control the disease. When the seeds were sown twice as many as the recommendation, the disease incidence increased y approximately 13%. However, the susceptile variety Jopummil alone showed the significantly enhanced disease incidence at a two-fold seeding rate. Two chemicals such as Hexaconazole EC and Teuconazole EC highly inhiited the fungal growth on agar medium. However, two stroilurin fungicides such as Pyraclostroin EC and Trifloxystroin SC were relatively weak. The fungicides tested displayed the similar in vivo antifungal activities as in vitro activities. Hexaconazole EC and Teuconazole EC showed the strongest oth protective and curative activities and the protective activities of the chemicals were generally higher than the curative activities. Hexaconazole EC and Teuconazole EC controlled the disease y 64% and 73%, respectively, and the two chemicals reduced the disease y 45% and 39%, respectively, when they were applied one day after pathogen inoculation. These results indicate that oth Hexaconazole EC and Teuconazole EC could e used to control sharp eyespot on wheat. Keywords : Chemical control, Seeding rate, Sharp eyespot, Wheat 서론 밀은 Gramineae(Poaceae) 과 Triticum속으로, 전세계적으로분포하고있으며대부분의유럽과아프리카의매우주요한식량작물이다. 국내에서도밀은쌀과더불어영양과에너지의주요공급원으로이용되어져왔으며, 최근밀의이용량증가에따라자급율제고를위한정책적목표와더불어재배면적과생산량이급격히증가하고있는추세이다. 우리나라에서밀에발생하는곰팡이성병해로는흰가루병, 붉은곰팡이병, 녹병류, 잎집눈무늬병, 깜부 *Corresponding author Phone) , Fax) ) pacc43@korea.kr 기병류등을비롯하여현재 17종이보고 (The Korean Society of Plant Pathology, 2009) 되어있으나, 발생과방제연구는미흡한실정이다. 이중잎집눈무늬병 (Sharp eyespot, Rhizoctonia cerealis) 은 Kim 등 (1991) 에의해보고된이후국내에서뿐만아니라 (Kim 등, 1992). 세계적으로보리, 밀등화본과작물에피해를일으키는것으로알려져있다 (Daamen 등, 1990). 온난한지역에서주로발생하며 (Lipps 등, 1982), 병원균은포자를형성하지않고작물의잔재물이나토양에서균사체또는저항성을갖는구조체로살아가다기주에침입하여병을일으키게된다. 또한차갑고습한조건에서발병이잘일어나며광과양호한배수토양상태에서병반의진전이확대된다 (Cromey 등, 2002). 2 밀잎집눈무늬병의발생에파종량이미치는영향과방제약제선발 11 저해로잎이황화되고, 도복발생과줄기고사와종실의등숙에도영향을주게되어수량감소피해를일으키게된다 (Cromey 등, 2005). 수량감소는병발생정도와차이가있으나심한경우한개체에서 18%(Cromey 등, 2002), 영국과웨일즈에서는평균 26% 가보고 (Clarkson과 Cook, 1983) 되었다. 중국에서는 2005년에서 2008년까지밀재배지의 6백만 ha 이상이피해를입어십조위엔 ( ) 이상의경제적피해가보고 (Chen 등, 2008) 되었다. 최근월동후저온, 강우등잦은이상기상과함께밀면적의확대, 지속적인재배에따라국내에서도병의발생과피해증가가예상된다. 또한, 잡초방제를위한친환경재배목적과수량성제고를위해권장파종량에비해높은밀식재배를함으로이에따른병발생의증가우려도나타나고있다. 그러나국내에는아직병의방제법과관련한연구는보고되어있지않으며방제약제도등록되어있지않은실정이다. 따라서본시험에서는이병의피해방제기술개발을위해파종량에따른병발생정도와화학적방제를위한적정약제선발, 처리방법등에대한시험을수행하였다. 재료및방법 파종량에따른발병정도. 재배법에따른발병정도를확인하기위해파종기와파종량에따른발병정도를조사하였다. 시험재료는품종별저항성차이를보이는 (Lee 등, 2011) 안백밀 ( 중도저항성 ), 탑동밀 ( 중도감수성 ), 조품밀 ( 감수성 ) 을이용하였다. 파종량은농가권장량 160 kg/ ha를기준으로, 기준량대비 1.5배, 2배, 3배량수준으로처리하였다. 파종은일반포장에 10월 20일경파종한후병원균접종및관리등을위해 3월상순경에포트 ( cm 3 ) 로이식하여온실내에서재배하였다. 접종은뿌리활착을위해이식 2주후에실시하였으며, 접종원은 Lee 등 (1995) 이보고한 oat-meal 모래배지 (Lee 등, 1995) 의양과조성을변형하여만든밀기울모래배지 ( 밀기울 : 모래 : 물 =3:10:6) 를이전보고 (2011) 와같은방법으로제조하여균사가자란밀기울을잘게부수어토양중에고르게접종하였다. 접종량은 40 g의접종원을처리하고 3일간비닐을덮어습도조절을통한발병을유도하였다. 발병정도는접종한달후에전체줄기에대한발병줄기수의비율로조사하였다. 배지내약제반응. Rhizoctonia spp. 에의한병가운데벼잎집무늬마름병, 인삼잘록병, 잔디라이족토니아마름병등의방제약제로국내등록되어있는 20종을 1차선발하였다 (Tale 1). 배지내병원균억제반응을검토하여 ( 데 Tale 1. Selection: According to the in vitro antifungal activities on agar medium in the preliminary experiment, the 7 fungicides were chosen for this study. 이터미포함 ) 약효및농도별시험에는 Tale 1의비고에표시한 Hexaconazole 등 7종의약제를선발하여배지내균의생육억제능을시험하였다. 7종의약제중우수한효과를보인두약제를선발하여적정농도처리선발을위해각각의약제별기준농도대비 1/2배, 1배, 2배등 3처리로시험하였다. 접종균주는 Lee 등의보고 (2011) 에서와같은국내밀재배포장분리된 JS0901, JS1007, JS1022 등 3 균주를이용하였다. 배지내균생육의억제반응조사를위해 Potato Dextrose Agar(PDA) 배지를이용하였다. 배지중앙에 paper disc를부착시키고각각의약제를 10 µl씩흡수시킨뒤배양한분리균주의균사선단을 5 mm cork orer로떼어중앙으로부터상하좌우 4cm 정도간격을두어치상하였다. 배지는 25 o C에서 5일간배양후균사생장저지대길이를통해배지내약제별효

3 12 박종철 이은숙 조광민 이미자 강천식 최재성 과를확인하였다. 식물체약효검정. 밀재배상태에서의약제효과를검정하기위한식물체처리를통한시험을수행하였다. 시험재료는 Lee 등 (2011) 의결과에서중도저항성을보인알찬밀과감수성을보인조품밀과신미찰밀 -1 등 3품종을이용하였다. 품종별로 70% ethyl alcohol로 1분 30초간종자소독을한후건조시켜 Petri-dish에서발아시켰다. 발아된종자는각 20립씩포트 ( cm 3 ) 에심어약 10 cm 가량생육시킨후시험을수행하였다. 처리약제는배지내약효검정에서우수한것으로선발된 Hexaconazole유제, Teuconazole유제, Pyraclostroin유제, Pencycuron액상수화제등 4종을이용하였다. 약제별약효검정은병원균접종전약제처리와균접종후약제를처리하는접종후처리두가지처리로수행하였다. 접종원조제는파종량과동일하게수행하였다. 접종전처리시험은포트에각약제를 10a 당사용기준희석농도에따라처리하고, 24시간후접종원을약 2g씩토양에접종하고, 3일간비닐을덮어습도를유지시켰으며일주일후발병줄기수를조사하였다. 접종후처리시험은약 2g의접종원을토양위에먼저접종한후 3일간비닐로덮어습도를유지하면서발병유도하였다. 그뒤표준희석농도로희석한약제를위와같은양으로관주하고일주일후발병줄기수를통해약제효과를평가하였다. 결과및고찰 파종량에따른병발생정도. 파종량에따른병발생정도를조사한결과처리간차이를보였다. 기준파종량에비해 2배량까지증가할수록평균발병률은높아지는경향이었다 (Tale 2). 기준량의평균발병률은약 51% 였으나 2배파종량에서는 64.2% 로조사되었다. 그러나 3배량의파종에서는발병률이약 45% 로감소되는결과를보였다. 전체품종들의파종량별평균발병률에대한차이를검정한결과, 기준파종량에비해 2배량에서약 13% 발병률이높아졌으나유의적인차이는보이지않았다. 이는본시험에이용된 3 품종의저항성이각각다른원인인것으로생각된다. 품종별로차이를검정한결과에서는조품밀에서만이파종량이 2배량까지증가할수록발병률이높아지는유의적인차이를보였다. 안백밀의경우는 2배량파종처리에서도기준량에비해발병율이증가하지않았으며, 3배량에서는약 15% 감소하였다. 탑동밀과조품밀은 2배량까지발병률이증가를보였으며, 3배량에서는감소하는결과를보였다. 이결과는국내밀육성품종에대한검정결과안백밀은중도저항성, 탑동밀과조품밀은각각중도감수성과감수성이었던결과 (Lee 등, 2011) 와같았다. 본시험결과파종량은잎집눈무늬병의발생정도에영향을주는것으로나타났으며, 조품밀처럼감수성인품종의경우발병정도가더증가하는영 Tale 2. 4 밀 잎집눈무늬병의 발생에 파종량이 미치는 영향과 방제 약제 선발 향을 받는 것으로 나타났다. 잎집눈무늬병의 발생에는 기주식물체의 밀집정도와 분 얼정도가 관련이 있는 것으로 보고(Colach 등, 1997)되 어 있다. 기주와 병원균과의 거리가 짧을수록 균의 침입 확률이 높으며, 한편 분얼수가 많은 경우 침입되는 줄기 수의 비율이 낮아지기 때문에 상대적으로 발생률은 낮게 나타나게 된다. 또한 R. cerealis는 다른 균에 비해 생장 속도가 늦으며(Lipps와 Herr, 1982), 기주 침입 시 오직 Fig. 1. A: Hexaconazole, B: Teuconazole, C: Pencycuron+Teuconazole, D: Fenuconazole+ Thifluzamide, E: Pyraclostroin, F: Trifloxystroin, G: Pencycuron, H: control treatment with sterilized distilled water asored paper disc. 13 균사만으로 기주의 세포벽을 융해 시켜 침입하기 때문에 침입속도가 늦으며, 기주간의 간격이 아무리 좁은 환경조 건을 가져도 한정되어 있는 병원균이 침입할 수 있는 기 주의 적정량은 정해져 있을 것으로 생각된다. 본 시험에 서 3배량에서 병 발생이 낮은 이유는 접종원의 비율이 가 장 적은 이유로 생각된다. 한편 안백밀의 경우 2배량에서 도 발병률이 증가하지 않았는데, 이와 관련된 품종의 저 항성에 대한 연구와 포장상태에서 균의 밀도에 따른 병 원균의 침입, 병 발생 기작에 대한 정밀한 연구가 더욱 필요할 것으로 보인다. 또한 병 발생 억제를 위한 경종적 방법에 대한 연구도 필요할 것으로 보인다. 배지 내 약제 반응. 잎집눈무늬병의 방제 약제 선발 을 위해 1차 배지 상에서의 약제별 효과를 검정한 결과 는 Fig. 1과 Tale 3과 같다. 약제별 기준 처리 농도에 서 균의 생육 억제 정도를 조사한 결과 Hexaconazole과 Teuconazole 처리에서 16.5 mm와 16.2 mm의 저지대를 형성하여 7종의 약제 중 균사생장 억제에 우수한 효과를 보였다(Tale 3). 반면, Pencycuron 수화제의 경우에는 1.1 mm 억제대 형성으로 효과가 없는 것으로 조사되었 다. Pencycuron +Teuconazole도 11.4 mm의 저지대 형성 으로 효과를 보였으며 이는 Teuconazole 성분이 함유된 이유로 생각된다. 그 외 나머지 3종의 약제는 효과를 나 타내지 못한 것으로 조사되었다. 방제 효과를 보인 Hexaconazole, Teuconazole과 Fenuconazole은 모두 triazole계 약제이다. 이 트리아졸계 약제는 대표적인 에르고스테롤 생합성 저해제(ergosterol iosynthesis inhiitor, EBI)로써, 친유성이며 에르고스테롤 의 생합성 전구물질인 라노스테롤과 비슷한 형태를 가지 고 있어 정상적인 생합성에 장애를 일으키게 된다(Kataria Tale 3. Kataria 등 (1991) 은 R. cerealis의배지상억제능과밀유묘에서모잘록병의방제시험에서 triazole계인 Cyprocolazole 를가장우수한약제로보고하여본시험의결과와도같았다. 반면효과가좋지않았던 Pyraclostroin과 Trifloxystroin은 stroilurin 계로서, 이계통은세포내의전자전달계교란을통해활성본체의호흡을억제시켜살균작용을나타내게되는데, 균사생장보다는포자발아억제능이높은것으로보고 (Karadimos 등, 2005) 되어있다. 이같은약제의살균기작차이가본실험에서의균사생장억제효과가나타나지않은원인으로생각된다. Pencycuron 은 urea 계통으로원형질막의삼투압과유동에변화를주어살균작용을하는것으로보고 (Kim 등, 1996) 되어있으나, 본시험에서효과를보이지않았다. 따라서 R. cerealis 는생육억제를통한방제에 triazole 계통이효과적인것으로나타났다. 1차검정에서유의성있는효과를보인 Hexaconazole과 Teuconazole 두약제에대해농도에따른효과를검정하기위해표준농도대비 1/2배, 2배로농도처리로균사생육억제정도를확인하였다. 그결과 Hexaconazole은표준 ( 유효성분 10%) 의 2배농도 ( 유효성분 20%) 에서가장좋은약효를보였다 (Tale 4). Teuconazole은표준농도 ( 유효성분 25%) 와 2배농도 ( 유효성분 50%) 에서효과적인균시생육억제차이를보였다. 따라서본시험결과약제방제시에약제별적정농도에맞게처리하는것이비용과효과면에서도효율적인것으로나타났다. 식물체약효검정. 밀을포트재배하면서배지시험결과선발된 4종의약제에대한효과를확인하였다. 약제처리는병원균접종전과접종후두처리를실시하였는데, 두처리에서모두배지내시험결과와같은약제별효과를나타내었다. 접종전처리에서는약제별로 % 로차이를보였다 (Tale 5). 배지상에서효과를보였던 Hexaconazole과 Teuconazole 두약제처리에서 18.9% 와 14.4% 의발병률로다른약제에비해유의성있는효과의차이를보였다. The same letter within columns means no significant differences etween the treatments y Duncans multiple range test (P<0.05). 리에서도무처리에비해약 14% 의발병률억제효과를보였으나, 유의성있는차이로나타나지는않았다. 접종후처리에서는접종전처리에비해약제처리시에도발병률이약간높게나타났으나접종전처리에서와같은약제별효과를보였다 (Tale 6). Hexaconazole과 Teuconazole 두약제처리에서 26.1% 와 28.9% 의발병률을보여다른약제에비해효과적인것으로나타났다. Pyraclostroin과 Pencycuron 처리에서는접종전에비해무처리의 47% 발병률과비슷한약 42% 의발병률을보여차이가없는것으로나타났다. 약제처리후 Teuconazole의경우발병되지않은유묘중일부에서약간의황화증상이나타나약해가의심되었으나다른처리에서는나타나지않았다. 본시험결과로밀에서발생하는잎집눈무늬병의약제방제에는 Hexaconazole과 Teuconazole이효과적이었으며, 방제시기는발병이예상되는시기에미리처리하는것이방제효과를높이는것으로조사되었다. 이를위해병의효율적이고정밀한발생예찰기술개발이필요할것으로보인다. 한편국내에는잎집눈무늬병의방제약제가등록되어있지않은실정으로본시험결과에서나타난우수약제에대한포장평가를통해방제약제의효과검정과개발을위한시험도필요할것으로생각된다. 요약 본연구는국내육성밀 3품종을이용하여파종량과밀잎집눈무늬병의발생정도를조사하고, 이식물병을효과적으로방제하기위한약제를선발하기하기위하여수행되었다. 파종량이기준량의 2배로증가할때발병률이약 13% 증가하였다. 그러나감수성인조품밀만이 2배량파종시에발병률이높아지는유의적인차이를보였다. 잎집눈무늬병의방제약제선발을위해배지상에서약제별효과를검정한결과, Triazole 계통인 Hexaconazole EC과 Teuconazole EC는균사생장을효과적으로억제하였다. 하지만 stroilurin 계통의 Pyraclostroin EC과 Trifloxystroin SC는균사억제효과가약하였다. 식물체에직접처리하여약효를검정한결과, in vivo 활성과 in vitro 활성은유사하게나타났고예방효과가치료효과보다높은것으로나타났다. 즉, Hexaconazole EC와 Teuconazole EC의예방효과는각각 64% 와 73% 였고, 치료효과는각각 45% 와 39% 였다. 본연구결과는 Hexaconazole EC와 Teuconazole EC가밀에발생하는입짚눈무늬병의방제제로이용될수있다는것을나타낸다.
