Min Sun Park Pil Son Choi Received: 17 November 2015 / Revised: 16 December 2015 / Accepted: 19 December 2015 c Korean Society for Plant Biotechnology Abstract To investigate the optimal conditions for shoot organogenesis in Astragalus sinicus L., hypocotyl explants were cultured in Murashige & Skoog s (MS) medium supplemented with 0.1, 1.0, 2.0, or 4.0 mg/l 2,4-dichlorophenoxy acetic acid (2,4-D) for 6 weeks. 2,4-D concentration significantly effected morphogenesis: some produced calli with adventitious shoots and roots, some produced calli with adventitious roots, some produced only calli, and some produced deep-brownish calli with roots. The formation of calli with shoots and/or roots was observed at lower levels of 2,4-D, whereas calli without shoots or with deep-brownish roots were formed after treatment with higher levels of 2,4-D. Also, a shoot organogenesis ability of callus clones was observed after treatment with medium with 0.1 or 1.0 mg/l 2,4-D grown in MS medium with combinations of benzyl adenine (BA) and 2,4-D for 4 weeks. Medium with a combination of BA and 2,4-D was effective for shoot formation, whereas root organogenesis from calli decreased. The greatest amount of shoot formation was obtained when calli were cultured in MS medium containing 1.0 mg/l 2,4-D and 0.5 mg/l BA. Upon shoot transfer into ½ MS basal medium, plantlets developed, and the plantlets grew well in soil in a greenhouse. M. S. Choi ( ) 남부대학교 한방제약개발학과, 약용식물형질전환연연구소 (Department of Oriental Pharmaceutical Development, Medicinal Plant Transformation Center, Nambu University, Gwangju , Korea) cps6546@hanmail.net Keywords Astragalus sinicus, Hypocotyl explants, Plantlets, Shoot organogenesis 서 론 자운영은 일본, 중국, 한국 등 아시아를 중심으로 서식하 는 2년생 초본 콩과식물로 알려져 있으며, 우리나라에서 는 답에서 벼 재배기술 향상을 위하여 자운영 종자를 파 종하는 연구가 진행되어 왔다(Kim et al. 2010). 가을에 발 아 된 후 동절기 생육기간을 거쳐 5월 하순경에 종자가 성숙하게 되는 동절기 초본식물이며(Kim et al. 2008), 생 육기간 동안 토양박테리아와 공생하면서 질소를 고정함 으로써 토양을 비옥하게 만드는 대표적인 콩과 식물이기 도 하다(Murooka et al. 1993). 그 뿐 아니라 이 식물은 동물 사료나 꿀벌의 먹이로서 이용되기도 하고(Yasue 1985), 약 리학적으로 해소, 인후염, 해열 등에 효과가 있는 것으로 알려지면서 3 ~4월경 연한 싹이나 꽃을 이용한 효소식품 제조 또는 기능성 식품의 원료로 이용되고 있고, 최근에 는 유전자원 보존을 위한 식물체 대량생산 연구(Erisen et al. 2010)와 새로운 유전자 도입을 위한 형질전환 연구 (Cho and Widholm 2002) 등이 진행되고 있다. 대부분의 약초식물은 여러 가지 함유되어 있는 이차대 사산물과 페놀화합물에 의해 조직 또는 세포배양을 통해 식물체 재생과정이 어려운 것으로 알려있는데, 이는 식 물체 자체에 함유되어 있는 유효성분이 세포분열을 억제 하거나 약초부위로 이용되는 뿌리를 배양절편으로 이용 할 경우 오염문제 등이 중요한 원인이 되고 있다. 그러한 문제점은 세포분열능이 높은 어린 무균식물체 배양절편 을 이용하거나(Cho and Soh 1995; Cho et al. 1998), 가장 효 과적인 호르몬의 선택과 최적 농도를 조사함으로써 해결 될 수 있다(Erisen et al. 2010; Turgut-Kara and Ari 2008). 예

2 J Plant Biotechnol (2015) 42: 를 들면 시호와 당귀와 같은 약초식물에서 성숙한 뿌리 를 사용하는 것보다 무균 발아된 어린 식물체의 배축 절 편을 배양함으로써 기내에서 대량으로 부정근을 생산하 거나, 기관 발생을 통한 식물체 재분화에 성공할 수가 있 고, 체세포배발생과 같은 형태발생은 2,4-D가 효과적으 로 알려져 있지만(Soh et al. 1996), 박과 작물이나 자운영 속(genus) 식물은 2,4-D보다는 IAA를 고농도 처리하거나 BA, Tidiazuron과 같은 cytokinin을 NAA와 혼합처리 하였을 때 효과적인 것으로 보고하고 있다(Luo et al. 1999; Turgut-Kara and Ari 2008). 또한 같은 속(genus)으로 알려진 Astragalus nezaketae의 경우 호르몬의 종류와 조합처리는 캘러스 형 성과 형태발생이 다양하게 나타날 수 있으며, 특히 BA와 NAA조합처리는 캘러스로부터 신초발생에 매우 효과적 인 것으로 보고된 바 있다(Erisen et al. 2010). 이와 같이 식물의 종에 따라 배양재료, 배지에 첨가하는 호르몬의 종류 및 농도는 형태발생에 매우 중요하다 할 수 있으며, 같은 속이나(genus) 종에서도 다른 반응을 보일 수 있기 때문에 신초발생과 같은 형태발생을 통한 식물체 대량생 산 연구를 위해서는 호르몬의 선택과 적정농도의 선택이 필 수 불가결하다 할 수 있다. 그러나 최근 Astragalus속(genus) Astragalus adsurgens (Luo and Jia 1998), Astragalus cicer (Basalma et al. 2008), Astragalus melilotoides (Hou and Jia 2004), Astragalus chrysochlorus (Turgut-Kara and Ari 2008), Astragalus Canadensis (Hung and Xie 2008) 등 각각의 식물에서 유전자원 보존 을 위한 식물체 재분화 연구가 진행되어 왔으나 자운영 (Astragalus sinicus L.)에서는 모상근 절편으로부터 신초발 생 연구(Cho and Widholm 2002)와 트립토판 합성효소 유 전자가 발현되는 자운영 형질전환체 개발 연구(Cho et al. 2000)외에는 거의 찾아 볼 수 없어 체계적인 식물체 재분 화 시스템 구축이 필요한 실정이다. 따라서 본 연구에서 는 자운영 배축 절편으로부터 기관발생과 캘러스로부터 식물체 재생을 위한 체계적인 최적의 배양조건을 규명하 기 위하여 시도 하였다. 상 암 배양 하였다. 배양 6주 후 자운영 배축 절편으로부 터 형성된 부정근을 갖는 캘러스, 신초와 캘러스를 갖는 캘러스, 캘러스, 캘러스의 특징 등을 조사하였으며, 처리 구 당 1회 20개씩, 3회 이상 반복하여 수행하였다. 신초발생능 캘러스 형성에 대한 BA의 효과 자운영 배축 절편으로부터 배양 6주 후 0.1또는 1.0 mg/l 2,4-D배지에서 자란 연한 노란색의 캘러스를 선발하였 고, 대량증식을 위하여 1.0 mg/l 2,4-D배지에 계대배양 하 였다. 이러한 캘러스로부터 신초 형성에 대한 BA효과를 조사하기 위하여 1.0 mg/l 2,4-D와 0.1, 0.5, 1.0, 2.0 mg/l BA를 각각 조합 첨가한 MS배지에 옮겨 24 C, 16시간 광 주기(46 μmol m -2 s -1 )조건으로 약 4주 동안 배양하였다. 배 양 4주째에 캘러스부터 신초를 형성하는 캘러스 수를 기 록하였으며, 처리구 당 1회 20개의 캘러스 클론을 3회 반 복 수행하였다. 또한 캘러스로부터 신초를 조심스럽게 분리하여 호르몬이 첨가되지 않은 1 / 2 MS기본배지에 옮 겨 6주 동안 배양하여 식물체를 유도하였으며, 유식물체 는 토양으로 옮겨 온실에서 순화하였다. 결과 및 고찰 자운영 배축 절편을 2,4-D가 첨가된 각 처리군에서 배양 하면서 기간별로 관찰 하였다. 배양 2주째부터 절단부위 로부터 세포분열이 시작되면서 캘러스가 형성되기 시작 하였고, 연한 노란색의 캘러스가 왕성하게 자라기 시작 하였다. 배양 4주째부터 연한 노란색의 캘러스 표면에 뿌 리가 왕성하게 형성하였으며, 일부에서 신초가 관찰되기 시작하였다. 배양 6주째에는 흰색의 뿌리와 녹색을 띄는 신초가 자라기 시작하였다(Fig. 1A, B). 2,4-D단독배지에 서는 신초발생 보다는 캘러스와 뿌리 발생이 이루어졌 고, 고농도배지에서는 오히려 캘러스 생장이 억제되었다 재료 및 방법 배축절편 배양 자운영(Astragalus sinicus L.) 종자를 국립종자원으로부터 분양 받아 70% 알코올에 1분, 0.4% 클로락스용액에 15분 간 침적하여 표면 살균한 후 멸균수로 3-5회 수세하였다. MS기본배지(Murashige and Skoog 1962)에 치상 하여 4주 후 유식물체를 얻었다. 유식물체의 배축으로부터 기관발 생능을 갖는 캘러스 형성을 조사하기 위하여 2 x 3 mm 크기의 절편을 만들어 0.1, 1.0, 2.0, 4.0 mg/l 2,4-D가 각각 첨가된 MS배지(pH 5.8)에 치상 하였고, 24 C에서 6주 이 Fig. 1 Morphogenesis of cultures of hypocotyls explants of Astragalus sinicus grown in MS medium containing various concentrations of 2,4-D for 6 weeks. 또한 같은 속(genus)으로 알려진 Astragalus nezaketae의 경우 호르몬의 종류와 조합처리는 캘러스 형 성과 형태발생이 다양하게 나타날 수 있으며, 특히 BA와 NAA조합처리는 캘러스로부터 신초발생에 매우 효과적 인 것으로 보고된 바 있다(Erisen et al. 2010).

3 398 J Plant Biotechnol (2015) 42: Table 1 Morphogenesis of Astragalus sinicus hypocotyl explants grown in MS medium containing various concentrations of 2,4-D for 6 weeks 2,4-D (mg/l) Callus with adventitious shoots and roots a No. of explants with various responses Callus with adventitious roots Callus Deep-brownish callus with roots ± ± ± ± ± ± ± ± ± ± ± ±3.58 a Each value represents the mean±standard error of at least three replicates (Table 1). 2,4-D 농도별 차이를 보면, 0.1 mg/l에서는 신초 와 뿌리가 동시에 발생하거나 뿌리만 발생하는 경우가 각각 1.9개와 6.5개로 조사되었으며, 농도가 높아질수록 감소하는 경향을 보였다. 캘러스 형성은 1.0 mg/l농도까 지는 증가하다가 고농도로 갈수록 감소하였고, 특히 4.0 mg/l 2,4-D가 첨가된 배지에서는 신초와 뿌리를 갖는 캘러스 형성은 거의 없었으며, 오히려 캘러스가 생장하지 않고 점차 진한갈색으로 변화되면서 괴사하였다(Fig. 1C). 많 은 식물로부터 캘러스 형성이나 기관 발생과 같은 형태 발생은 최적의 호르몬 종류와 적정농도를 선발하여 사용 하는 것이 중요하다(Choi et al. 2002). 예를 들면 자운영 (Astragalus sinicus L.) 모상근으로부터 체세포배 발생능 캘러스 형성은 7.5 ~ 10.0 mg/l 2,4-D의 높은 농도가 요구 되어(Cho and Widholm 2002) 대두의 경우(Choi et al. 2002) 와 유사한 경향을 보였지만, 보통의 식물에서 요구하는 농도(1.0 ~ 2.0 mg/l 2,4-D)와는 다른 반응을 보였고, 같은 속인 Astragalus chrysochlorus 식물에서는 2,4-D보다는 3.0 mg/l IAA가 효과적인 것으로 보고되어(Turgut-Kara and Ari 2008) 식물 종에 따라 그리고 같은 속(genus)이라 할 지라도 확연히 다를 수 있음을 보여준 좋은 예라 할 수 있다. 이는 본 연구에서도 호르몬 농도에 따라 신초와 뿌 리가 동시에 발생하거나, 뿌리만 발생하는 경우 그리고 신초와 뿌리 발생이 없는 캘러스만 형성되는 경우 등으로 Fig. 2 Shoot organogenesis and plant regeneration from the cultures of Astragalus sinicus callus clones grown in MS media with combinations of BA and 2,4-D for 4 weeks. MS배지에 2,4-D가 0.1 또는 1.0 mg/l로 첨가된 배지에 서 6주 이상 배양하여 선발한 연한 노란색의 캘러스를 동일배지에서 증식하였다. 이러한 캘러스를 1.0 mg/l 2,4-D 와 BA (0.1, 0.5, 1.0, 2.0)를 각각 조합 첨가한 배지에 옮겨 4주 동안 배양하였다. 배양 4주 후 신초가 발생된 캘러스 수(Fig. 2A)를 조사한 결과 1.0 mg/l 2,4-D mg/l BA 배지에서는 10.2개, 1.0 mg/l 2,4-D mg/l BA배지에 서는 15.6개, 1.0 mg/l 2,4-D mg/l BA배지에서는 14.7개, 1.0 mg/l 2,4-D mg/l BA배지에서는 7.5개로 조사되었다. 반면 뿌리를 형성하는 캘러스는 1.0 mg/l 2,4-D mg/l BA배지에서 3.7개 조사되었을 뿐 나머 지 조합에서는 거의 뿌리 발생이 이루어지지 않았고, 일 부 캘러스는 신초나 뿌리가 발생되지 않았다(Table 2). 신 초는 1 / 2 MS기본배지에 옮겼을 때 정상적인 식물체로 자 랐으며, 토양에서도 잘 적응하여 완전한 식물체로 생육 되었다(Fig. 2B, C). BA는 조직배양으로부터 신초발생이 나 신초발생능 캘러스 형성에 매우 효과적인 것으로 알 려져 있을 뿐 아니라(Erisen et al. 2010), 일부 박과 작물이 나 Astragalus속 식물에서는 캘러스로부터 체세포배 발생 에도 중요하게 작용하는 것으로 알려져 있다(Luo et al. 1999). 그 뿐 아니라 자운영과 동일 속(genus)인 Astragalus nezaketae의 경우 잎과 엽병조직 절편으로부터 오옥신으로서 2-4-D와 NAA를, 사이토키닌으로서 BA, Kinetin, Tidiazuron을 단독 혹은 조합첨가 하였을 때 연하고 부드러운 녹색의 캘러스 형성은 오직 BA와 NAA로 조합첨가 하였을 때 이루어 졌고, 그러한 캘러스를 신초발생능 캘러스로 보 고한 바 있다(Erisen et al. 2010; Mirici 2004). 이는 본 연구 결과와 매우 유사함을 보여 주는 결과라 생각되며, 많은 작물의 신초발생 연구에서 BA가 매우 효과적 이다는 결 과와 일치하고 있다(Malik and Saxena 1992; Gill and Saxena 1992; Barwale et al. 1986; Wright et al. 1986). 반면 자운영 과 동일 속(genus)일지라도 Astragalus melilotoides의 경우 배출절편으로부터 신초 발생능 캘러스는 BA에 비하여 2,4-D 와 kinetin이 조합되었을 때 가장 효과적인 것으로 보고하

MS배지에 2,4-D가 0.1 또는 1.0 mg/l로 첨가된 배지에서 형성된 캘 러스로부터 신초발생능 캘러스 형성을 조사하기 위하여 2,4-D와 BA 를 혼합 첨가한 배지에 옮겨 4주 동안 배양하 면서 조사하였다. 먼저 2,4-D 농도에 따라 신초와 뿌리를 형성하는 캘러스, 뿌리를 형성하는 캘러스, 캘러스, 진한 갈색으로 변하는 캘러스 등 여러가지 형태발생을 관찰 할 수 있었다. 일반적으로 신초와 뿌리를 갖는 캘러스는 2,4-D가 낮은 농도로 첨가 되었을 때 형성되었으며, 반면 신초가 없는 캘러스, 갈변 되는 캘러스는 고농도로 처리 되었을 때 형성되었다. 또한 BA와 2,4-D가 조합첨가된 배지에서는 신초발생에 효과적이었으며, 뿌리 발생은 감 소하였다. 가장 효과적인 신초발생은 1.0 mg/l 2,4-D 와 0.5 mg/l BA가 조합 첨가되었을 때 얻어졌으며, 이러한 신초들은 1 / 2 MS기본배지에서 대부분 유식물체로 생육되 었으며, 토양에서 정상적으로 적응하여 생장하였다. 사 사 이 논문은 2015학년도 남부대학교 학술연구비의 지원을 받아 연구되었음.
