Seed germination rate was the highest at 66% when seed were pre-chilled(30 days) and soaked in distilled water and then sown in bedsoil under 50% shading condition. Also we examined the effects of seedlings size, type of soil and shading condition for pot cultivation. Seedlings size as a samll (plant height 2.5 cm, number of leaves 4, leaf width 1.2 cm, leaf length 1.7 cm) was the best and efficiency quality in bedsoil under non-shading condition with the growth characteristics of plant height(39 cm), number of leaves(29), leaf width(9.1 cm), leaf length(5.9 cm), root length(14.2 cm) and fresh weight(above ground/below ground; 23/5.3 g) and dry weight(above ground/below ground; 4.6/1.3 g). Futhermore there was 94% or more flowering and fruiting in this condition. In this study, we have established the cultivation and ex situ conservation of S. takesimensis. Therefore propagated plants(over 3,000 individual) could be used for not only various research but also ex situ conservation. 2 134 Journal of Agriculture & Life Science 48(4) Ⅰ. 서론 멸종위기및희귀특산식물의현지외보전은온전한식물체를획득하는단계서부터시작된다. 온전한식물체를획득하는방법은자생지에서모수를채취하는방법과종자나식물체조직일부로부터유 무성번식하여획득하는방법이있다. 자생지내에서모수를채취하는방법은멸종위기및희귀특산식물의보전을위해서적절하지않아종자나식물체조직으로부터식물체를증식하는방법이가장이상적인현지내외보전방안으로제시된바 (Oldfield, 2009) 있다. 야생상태의멸종위기및희귀특산식물로부터온전한식물체를획득하고이를온전히보전하기위해서는종자채종및식물체조직확보에서부터종자발아, 기관유도, 유묘획득, 생육 ( 재배 ), 개화결실, 종자생산등의식물생활사의모든단계가규명되는종생물학적연구가기반되어야한다 (Korea Forest Service, 2013). 하지만국내에서의현지외보전연구는주로분포 (Park et al., 2012; Lee & Kang, 2011) 와발아력검정 (You & Kim, 2010; Cho et al., 2013) 등의기초연구가대부분으로종생물학적의전반적인정보가제공되는종은매우제한적이다. 특히현지외보전을위한멸종위기및희귀특산식물의재배는언제어느때이고지속생산이가능한의미로야생에서의희귀성때문에진행되기어려운분류, 표본, 형질, 분자생물, 증식, 생리, 생태, 복원등의멸종위기및희귀특산식물과관련된종생물학적연구가가능하다 (Theobald, 1989). 따라서중국에서는이미현지외보전의일환으로현지외재배라는용어를사용하고있으며지금까지알려진 33,000 여종의중국고등식물중약 70% 인 23,000 여종이재배되고있는것으로보고 (Huang, 2011) 된바있다. 하지만국내에서는멸종위기및희귀특산식물중몇 % 가현지외에서보전되고재배되는지또한생물학적정보가얼마나축적되어있는지에대한정보가부족한실정이다. 국내멸종위기및희귀특산식물중현지외보전의국내한계와문제점을가장잘나타내는대표적인 예가섬현삼 (Scrophularia takesimensis) 으로지금까지섬현삼관련연구는분포 (Ahn et al., 2006; Kim et al., 2013), 발아력검정 (Ahn et al., 2006; Lee, 2007, Kong, 2009; Kang et al., 2009) 및자생지생태 (Han et al., 2010) 등의연구가진행되었으나유묘출현및확보, 환경에따른재배 ( 생육 ), 개화결실, 종자생산등의종생물학적기초연구는진행된바없다. 울릉도해안에서제한적으로자라는섬현삼은다년생초본으로높이 1 m에달하고줄기에날개가있다. 꽃은 6-7월에원추화서로많은꽃이달리며, 과실은삭과로 9-10월에익는다 (Lee, 1980). 섬현삼은울릉도에만제한적으로자생하기때문에산림청국립수목원의희귀식물목록집 (Korea National Arboretum, 2008) 에국제단위수준의멸종위기종 (CR: Critically Endangered) 으로지정되어있고환경부에서는멸종위기야생동식물 Ⅱ급으로지정되어있다 (Ministry of Environment, 2005). 현재섬현삼개체수는 2,000 여개체로보고되어있으나성숙개체보다 1 2년생의미성숙개체가많고, 환경변화에의한급격한개체수감소가우려되어적극적인보호가필요하며지금까지제시된대량증식법의체계적인정리의시급함이제시 (Kim et al., 2013) 된바있다. 따라서본연구는실험실수준에서규명된섬현삼의발아조건을야외온실에서적용하여실제유묘확보가가능한발아율 ( 유묘출현율 ) 을살펴보고, 획득된유묘를다양한조건의분화재배를시도하여개화및결실까지식물생활사의전반적인단계를구명함으로써희귀특산식물섬현삼의현지외보전의기초자료를확보하고지속적인생산을위해수행되었다. Ⅱ. 재료및방법 2.1 공시재료본연구에사용된섬현삼종자는 2012년 8월중

3 Lee et al : Ex situ Conservation of Scrophularia takesimensis through Seed Germination and Pot Cultivation 135 순울릉도도동등대일대 (Fig. 1 (A); N E ) 에자생하는섬현삼꼬투리를채종하여지퍼백에담아실험실에가져온후 1.00 mm 표준체 (Sie , Chung-gye Sang gong sa, Korea) 에걸러이물질과불량종자를걸러내었다. 선별된섬현삼종자는사용전까지모두 2 ( 건조 ) 냉장보관하였다. 2.2 종자저온습윤처리, 전처리및차광처리가섬현삼발아에미치는영향지금까지보고된섬현삼발아율중가장높은발아율을보고한조건인음건후 4 저장종자를 48 시간 GA 3 용액 (800 ppm) 침지처리방식 (Kong, 2009) 과저온 (5 ) 30일저장 (Lee, 2007) 방식을단용및혼합하여적용하였다. 저온저장은건조및습윤처리를달리하였으며습윤처리는페트리디쉬 (90 15 mm, SPL, Korea) 에여과지 (Whatman No. 1, USA) 2매를깔고증류수 5 ml를공급한후파라필름 (Sigma-Aldrich, USA) 으로밀봉하여 2 에서 30일간저장하였고, 증류수를공급하지않은저온건조저장대조구를두었다. 전처리는 GA 3 용액 (Gibberellic acid 3, Duchefa, Netherlands, 800 ppm) 침지 (Kong, 2009) 및증류수 48시간침지처리한처리구와전처리하지않은대조구로달리하였다. 처리된종자는클라스만상토 (potgrond H, Klasmann-Deilmann, Germany) 를 2 cm 높이로채운육묘용묘판 (60 cm 35 cm 3 cm) 에 30립씩 3반복으로 3월 29일파종하였으며복토는하지않고 1일 1회미스트관수하였다. 또한농업용차광망 ( 차광율 50%, 80%) 을설치하여온실내대조구 (46,000 53,000 lux, 920 1,150 µmol m -2 s -1, 실외 66,000 86,000 lux, 1,500 µmol m -2 s -1 ), 50% 차광 (19,000 23,000 lux, µmol m -2 s -1 ), 80% 차광 (10,000 14,000 lux, µmol m -2 s -1 ) 의 3조건으로달리하여차광에따른발아율을비교하였다. 발아조사는매주측정하였으며더이상발아가진행되지않는 60일 (8주차, 5월 27일 ) 차데이터를공시하였다. 2.3 유묘크기, 토양종류및차광조건이섬현삼분화재배에미치는영향유묘크기, 토양종류및차광조건이섬현삼분화재배에미치는영향을살펴보고자발아하여획득된유묘를임의의크기대 (Big), 중 (Middle), 소 (Small) 로나누어 (Table 1) 분화재배실험에사용하였다. 토양종류는클라스만, 클라스만 + 펄라이트 (2:1, v/v), 클라스만 + 마사토 (2:1, v/v), 원예용상토 ( 바로커, 서울바이오 ) 의 4 조건으로달리하여포트 ( 직경 13 cm, 높이 10 cm) 에채운후준비된유묘를 5월 28일 ( 파종 61일후 ) 이식하였다. 또한차광조건이분화재배에미치는영향을살펴보기위해발아율실험과동일한조건과장소의차광처리구 ( 대조구, 50% 및 80% 차광 ) 에서수행하였다. 각처리구당 6개체씩 3 반복하였으며, 각조건에따른생육특성은초장, 엽수 ( 매 ), 엽폭, 엽장, 근장 및생중량 (g, 지상 하 ) 을각조건당 9개체씩 9월 4일 ( 발아 161일, 이식 100일후 ) 에측정하였다. 엽폭과엽장은완전히전개된위에서 2 3번째의건실한잎 Table 1. 4 136 Journal of Agriculture & Life Science 48(4) 을대상으로 5반복측정하였고, 근장은길이가긴 5 개를측정하여평균값을구하였다. 건중량 (g, 지상 하 ) 은시료를 20일간음지에서충분히자연건조한후전자저울을이용하여 9월 26일에측정하였다. 또한각조건당생식생장단계인개화주를 9월 4일조사하였으며종자결실유무는 10월 24일 ( 발아 181 일, 이식 120일후 ) 에조사하였다. 모든실험은경기도양평군용문면에위치한국립수목원유용식물증식센터비닐온실에서수행하였다. 2.4 통계처리본실험에수집된데이터는평균 (means) ± 표준편차 (standard deviation) 로산출하였으며, 다변량분산분석 (MANOVA :Multivariate ANOVA) 을통해각각의요인이미치는영향을살펴보았으며유의성이있는경우 Duncan multiple range test(p=0.05) 로 2차검증하였다. 또한모든통계처리는 SPSS(ver Kor) 프로그램을사용하였다. Ⅲ. 결과및고찰 3.1 종자저온습윤처리, 전처리및차광처리가섬현삼발아에미치는영향저온습윤처리, 전처리및차광처리가섬현삼종자발아에미치는영향을살펴본결과는다음과같다 (Table 2). 2 저온건조및습윤처리등의종자저장방식에따른섬현삼의발아율은저온건조저장에서 0 58% 의발아율을보였고, 저온습윤처리에서는 43 66% 의발아율을보여, 저온건조저장보다최대 66% 의뚜렷한발아율차이 ( 유의적수준 P<0.001) 를나타냈다. 전처리또한섬현삼의발아율에유의적인수준 (P<0.001) 으로영향을미쳤다. 건조저장종자는증류수침지처리를통해발아율을향상시킬수없었고저온습윤처리종자는증류수침지처리에의해발아율이 3 11% 상승하였으나뚜렷한차이를보이지않았다. 수분 ( 증류수 ) 침지처리는종자내부로수분이침투되어세포를팽창시켜부피가커져종피가 파열되면서수분과산소투과성이증진되며광이투 과할수있는통로를제공하여배가발아하는것으 로알려져있으나 (Choi et al., 2011) 본실험결과에 서는뚜렷한발아율향상을보이지않았다. GA 3 는배휴면을타파하고배를신장시켜종피를뚫고나오는것을도움으로써발아를촉진 (Machado de Mello et al., 2009) 시키는것으로알려져있다. 본연구에서건조저장종자를 GA 3 용액에침지처리하여발아율을향상시킬수있었으나저온습윤처리는 GA 3 침지처리에의해발아율이향상되지않거나오히려감소하였다. 이러한이유는저온습윤처리는발아억제제인 ABA를불활성화시키고 GA 3 를활성화시켜발아를촉진 (Khan, 1977) 시켰기때문인것으로판단된다. 하지만건조저장종자를 GA 3 용액 800 ppm 침지처리시최적의발아율 78% 를보였다는보고 (Kong, 2009) 와본연구결과의건조저장종자의최적발아율 58% 와 20% 이상의차이를보였으나이는실험실 (Kong, 2009) 과온실내 ( 본실험 ) 의미기후등의미세환경및울릉도내채종지역과채종개체차이에서기인된것으로생각된다. 차광처리또한섬현삼의종자발아에유의적수준 (P<0.001) 으로영향을미쳤으며, 50% 차광처리조건 ( 습윤, 증류수침지 ) 에서가장우수한 66% 의발아율을보였으나저온습윤처리종자를 GA 3 침지처리시대조구나증류수침지처리에비해발아율이 20 23% 감소하였다. GA 3 침지처리된종자는유묘출현후과다한길이생장으로묘가연약해져결국고사하게되므로유묘출현율과상이할수있다고보고 (Kang et al., 2007) 하였으나본연구에서는 GA 3 침지처리로유묘가연약해지거나고사현상이관찰되지않았다. 따라서종자저온습윤처리와 GA 3 침지처리및 50% 차광처리 ( 광질 ) 와의관계분석을통해종자생리특성이나발아율이감소한원인을구명하는연구가필요한것으로생각된다. 차광처리는노지보다수분증발을억제하여보다습한환경과낮은온도를제공함으로써종자발아와생육에긍정적인영향을미치는것으로알려져있으며 (McLaren & McDonald, 2003) 이러한이유로 50% 차광처리조건 ( 습윤, 증류수침지 ) 에서섬현삼 의발아율이향상된것으로생각된다. 지금까지진행된섬현삼발아검정실험은 Ahn et al. (2006) 이 5 저장기간 (0 15, 30, 45, 60) 중 60일저온처리종자를 20 에서발아시켰을때 67% 발아율을보고하였으며, Lee (2007) 는 5 저장기간 (0 15, 30, 45, 60) 중 30일저온처리종자를 20 에서발아시켰을때 69% 발아율을획득했다고보고하였다. 하지만위두실험은종자저온저장방식 ( 습도, 건도및습윤 ) 에대한정보가명확히제시되지않았다. Kong (2009) 은 8주간음건한후 4 에서저장한종자를 GA 3 용액 (800 ppm) 에 48시간침지처리시 78% 발아율을보고하였다. Kang et al. (2009) 은채종된종자를종이백에담아실험실실온 (18-22, 상대습도 40-60%) 및저온 (5 ) 저장하여실험하였으며, 상온저장종자를 GA 3 용액 (200 ppm) 에 24시간침지처리하여 20 에서발아시 80% 이상의발아율을획득하였다고보고하였으나저온건조저장및습윤처리 (60일) 는전혀발아하지않았다고보고하여본연구결과와상이하였다. 이는저장방법과실험실조건의차이에서기인된것으로판단된다. 이상과같이이전의연구들은선행연구를바탕으로실험을수행하였으나정보가제대로전달되지않아다른요인을적용 (Kang et al., 2009) 하여기존연구와의연계성보다차별성만부각시켜연구방법들과의연계성이떨어졌고명확한정보가제공되지않는등의한계를드러냈다. 따라서명확한정보가제공된부분을본연구에적용하여 2 건조저장종자를 30일간저온습윤처리하고증류수에 48시간침지처리하여클라스만상토가채워진육묘용묘판에파종후 50% 차광처리하면 66% 의최대발아율과유묘획득율을보였으며, 본실험중최적조건으로나타났다. 3.2 유묘크기, 토양종류및차광조건이섬현삼분화재배에미치는영향유묘크기, 토양종류및차광조건이섬현삼분화재배에미치는영향을살펴본결과유묘크기가대인
