In this study, we tried to isolate microorganisms from orchard soils, which can decompose the triazole fungicides, tebuconazole, fluquinconazole, and difenoconazole. Only difenoconazole was completely degraded in the enrichment culture, from which several difenoconazole-degrading bacteria were isolated. They showed the same rep-pcr pattern thus only one strain, C8-2, was further studied. The strain was identified as Sphingomonas sp. C8-2 based on its 16S rrna gene sequence and decomposed 100 mg/l of difenoconazole in a minimum medium to an unknown metabolite with a molecular weight of 296 within 24 hours. The inhibition effect of the metabolite against representative soil microorganisms significantly decreased compared to that of difenoconazole thus the bacterial strain is expected to be used for the detoxification of difenoconazole in soil and crop. Key words Bacteria, degradation, difenoconazole, triazole fungicide 서 론 환경에서농약의분해는주로미생물, 특히세균에의해일어난다는것이일반적인통설이다 (Fenner et al., 2013). 농약은적용대상에따라살충제, 제초제, 살균제로나눌수있으며이중살균제를분해하는미생물에대한연구는살충제나제초제에비해매우적은데이는살균제의급성독성이상대적으로낮기때문으로생각된다. 살균제중트리아졸 (triazole) 계살균제는 3개의질소원자를갖는트리아졸기를함유하는침투이행성의살균제로예방과치료효과를모두가지며곰팡이세포막형성에필수적인에르고스테롤의생합성을저해하여살균효과를나타낸다 (Bromilow et al., *Corresponding author yj7915@korea.kr 1999a; Sehnem et al., 2010). 각종원예및밭작물의흰가루병, 잿빛곰팡이병, 낙엽병등의방제에사용되며세계적으로가장소비량이많은살균제계통중하나로 2020년까지꾸준히사용될것으로예측되고있다 (Research and markets, 2014). 최근연구에서트리아졸계살균제는수중생물에대한독성 (Mu et al., 2016) 뿐만아니라내분비계장애물질로서인간을비롯한포유류의성적발달에영향을미칠수있음이보고되었다 (Taxvig et al., 2007; Zhou et al., 2016). 국내에서생산및수입되고있는트리아졸계살균제로는테부코나졸 (Tebuconaozle), 디페노코나졸 (Difenoconazole), 헥사코나졸 (Hexaconazole), 플루퀸코나졸 (Fluquinconazole) 등 17종이있으며 (KCPA, 2016) 트리아졸계살균제는일반적으로토양에서잔류기간이상대적으로긴것으로보고되어 (Bromilow et al., 1999a,b; Kim et al., 2003; Badawi et al., 2016) 토양및농작물에오랜기간남아환경에영향을미 349

350 안재형 노유미 이관형 박인철 김완규 한병학 유재홍 칠가능성이크다. 따라서이를무독화할수있는경제적이고안전한방법이요구된다. 본연구에서는전국에서채취한과수원토양으로부터트리아졸계살균제를분해할수있는미생물을순수분리하고그분해특성을구명하며그분해산물이토양미생물에미치는영향을분석하고자하였다. 를취하여동일배지 5mL에접종하고동일조건에서배양하면서수시로살균제의감소를확인하였다.

2 350 안재형 노유미 이관형 박인철 김완규 한병학 유재홍 칠가능성이크다. 따라서이를무독화할수있는경제적이고안전한방법이요구된다. 본연구에서는전국에서채취한과수원토양으로부터트리아졸계살균제를분해할수있는미생물을순수분리하고그분해특성을구명하며그분해산물이토양미생물에미치는영향을분석하고자하였다. 를취하여동일배지 5mL에접종하고동일조건에서배양하면서수시로살균제의감소를확인하였다. 살균제의감소가확인되면이배양액을 R2A agar (Difco, USA) 배지에도말하고 28 o C에서배양하였다. 형성된콜로니를살균제 100 mg/l를포함하는최소배지에접종후배양하여살균제분해여부를판정하였다. 재료및방법 수입량및생산량이많고상대적으로토양반감기가긴테부코나졸 (597일 ), 플루퀸코나졸 (350일 ), 디페노코나졸 (318 일 ) ( 을선발하여실험에사용하였는데, 테부코나졸은바이엘크롭사이언스로부터, 플루퀸코나졸과디페노코나졸은아그로텍으로부터각각원제를제공받아사용하였다. 경기, 강원, 충청, 경상, 전라지역의 22개과수원에서채취한토양을 2mm 체로거른후 100 g을취하였다. 각각의살균제원제를 100 mg/ml의농도로아세톤에용해시킨후, 100 mg/kg soil의농도가되도록토양에처리하였다. 질소와인을황산암모늄과인산염의형태로각각 5 mg N/kg soil과 0.5 mg P/kg soil의농도로되도록토양에첨가한후주기적으로혼합하면서 1개월동안실온에서관리하였다. 토양 1g 을취하여살균제 100 mg/l를포함하는최소배지 5mL에첨가후 28 o C에서 150 rpm으로진탕배양하면서 1개월간격으로살균제농도를측정하였다. 최소배지의조성은다음과같다 (1 L 당첨가량 ): Na 2 HPO 4 12H 2 O 3.58 g, KH 2 PO g, (NH 4 ) 2 SO g, MgSO 4 7H 2 O 0.05 g, CaCl 2 H 2 O g, yeast extract 0.01 g, trace minerals 10 ml, vitamin solution 10 ml. Trace mineral과 vitamin solution은 DSMZ medium 141에따라조제하였다 ( 살균제농도는 Shimadzu SCL-10Avp HPLC system (Shimadzu, Japan) 을이용하여조사하였는데컬럼은 YMC-Triart C 18, 이동상은아세토니트릴 : 물 =7:3, 유량은 0.7 ml/min이었으며 photodiode array detector를이용하여 200 nm에서분석하였다. 시료는아세토니트릴로 1/20로희석하고 0.2 µm 필터로여과후분석에사용하였다. 살균제의감소가확인되면 0.5 ml 결과및고찰 세종의살균제중디페노코나졸만 100% 분해되었으며다른살균제는분해가관찰되지않았다. 이는알려진토양반감기와상관이있는것으로판단된다. 디페노코나졸분해가확인된 5개의서로다른배양액으로부터 ( 충북충주, 전남보성, 경기이천, 경남창원, 강원춘천에서채취한토양시료이용 ) 디페노코나졸을분해할수있는 5균주를순수분리하였으며 (GTG) 5 프라이머를이용한 rep-pcr (Jarocki et al., 2016) 분석을통해이들이모두동일균주임을확인하였다 (Fig. 1). 이중 C8-2 균주만을선택하여이후연구에사용하였으며 16S rrna 유전자염기서열분석을통해 Sphingomonas 속으로동정하였다 (Fig. 2). 살균제디페노코나졸분해세균분리및특성분석 351 C8-2 균주의디페노코나졸분해특성을구명하기위하여 nutrient agar (Oxoid, USA) 배지에 28 o C로 5일간배양후디페노코나졸 100 mg/l가포함된최소배지에흡광도 (OD 600 ) 가 0.1이되도록접종하였다.

3 살균제디페노코나졸분해세균분리및특성분석 351 C8-2 균주의디페노코나졸분해특성을구명하기위하여 nutrient agar (Oxoid, USA) 배지에 28 o C로 5일간배양후디페노코나졸 100 mg/l가포함된최소배지에흡광도 (OD 600 ) 가 0.1이되도록접종하였다. 접종한배지를 28 o C, 150 rpm 에서진탕배양하면서시간에따른디페노코나졸농도를조사하였다. 비교균주로는 C8-2 균주에대한 16S rrna 유전자염기서열유사도가 99.5% 이면서다양한유기합성화합물을분해하는것으로알려진 Sphingomonas wittichii RW1 (=KCTC 42168) (Miller et al., 2010) 을비교균주로사용하였다. S. wittichii의경우배양 160시간동안디페노코나졸이유의하게감소하지않은반면 C8-2 균주는 24시간이내에검출한계미만으로감소하였다 (Fig. 3). C8-2 균주배양액의크로마토그램에서디페노코나졸외에머무름시간 3분대에새로운피크가생성된것을확인할수있었다 (Fig. 4). 이피크의면적은디페노코나졸농도와역의상관관계를보였으며 (Fig. 5) 이를통해디페노코나졸의분해산물인것으로추정되었다. 이피크는배양 160시간까지잔존하여 C8-2 균주가이물질을이용하지못하는 것으로판단된다. 이 3분대의피크는알려진디페노코나졸분해산물인 1,2,4-트리아졸이나 CGA (1-[2-chloro-4- (4-chloro-phenoxy)-phenyl]-2-(1,2,4-triazol)-1-yl-ethanol) 의피크와는일치하지않았다 (Fig. 4). 이물질의분자량을텐덤질량분석기를이용하여분석하였다. 분석은전북대학교공동실험실습관에서수행하였으며 Agilent 1200 series HPLC 와 6410 triple-quadrupole mass spectrometer를이용하였다. 이동상은 0.1% 포름산과 0.1% 포름산을포함한아세토니트릴용액을사용하여 ESI positive 모드에서분석하였다. 컬럼은 Pheomenex Kinetex C18 ( mm, 2.6 µm I.D.) 을사용하였으며컬럼온도는 30도, 흐름속도는 0.5 ml/min, 주입부피는 5µL였다. 분석결과 3분대피크를형성한물질은분자량 296임을확인하였다 (Fig. 6). 토양세균인 Pseudomonas putida KACC 10266, Streptomyces graminisoli KACC 16472, Bacillus subtilis KACC 10854와토양곰팡이인 Penicillium roqueforti KACC 47196를농촌진흥청농업미생물자원센터 (KACC) 에서분양받아디페노코나졸과 C8-2 균주에의한디페노코나졸분해산물이각각의토양미생물의생장에미치는영향을조사하였다. 5 살균제디페노코나졸분해세균분리및특성분석 353 1, 5, 10 mg/l가첨가된 Potato dextrose agar 배지 (Oxoid, USA) 에접종한후 28 o C에서배양하면서균사의성장을육안으로관찰하였다. C8-2 균주에의한디페노코나졸분해산물이토양미생물에미치는영향분석을위해먼저 C8-2 균주를디페노코나졸 100 mg/l가포함된최소배지에접종하고 28 o C에서 20일간진탕배양후 0.45 µm 공극의필터로여과한액을 1배 (1 ), 0.5배 (0.5 ), 0.1배 (0.1 ), 0.05배 (0.05 ), 0.01배 (0.01 x), 0배 (0 ) 희석한액에세균의경우 R2A를, 곰팡이의경우 Potato dextrose agar를정량대로첨가하였다. 이배지에세균또는곰팡이균사를접종하고세균의경우 28 o C, 150 rpm으로진탕배양하면서시간에따른흡광도 (OD 600 ) 를측정했고곰팡이의경우균사의성장을육안으로관찰하였다. 결론 실험결과토양세균인 B. subtilis와 S. graminisoli의성장은디페노코나졸농도가 1mg/L에서 10 mg/l로증가함에따라점차저해되었고 (Fig. 7A) 토양곰팡이인 P. roqueforti 의성장은디페노코나졸농도 1 mg/l 이상에서완전히억제되었다 (Fig. 8A). 반면 C8-2 균주의분해산물은토양세균인 B. subtilis와 S. graminisoli, 토양곰팡이인 P. roqueforti의성장에대한저해효과가현저히감소함 (Fig. 7B, 8B) 을확인할수있었다. 이상의결과로부터 C8-2 균주는디페노코나졸을빠르게분자량 296의물질로전환하였으며이물질은원물질에비해토양미생물에대한영향이현저히감소하였다. 따라서향후이균주를디페노코나졸오염토양및농산물의정화에이용할수있을것으로기대한다. 감사의글 본논문은농촌진흥청국립농업과학원농업과학기술연구개발사업 ( 과제번호 : PJ ) 의지원으로수행된결과이며연구비지원에감사드립니다.
