In the present study, we attempted to induce keratinizing squamous epithelium from human cholesteatoma epithelial (HCE) cells using an air-liquid interface (ALI) technique. We also examined the effect of RA on the phenotype of keratinizing HCE cells. Materials and Method:HCE cells were cultured in RA-free defined media at an ALI or in a submerged state. We examined the morphological differences between ALI and the submerged cultures, and histologically investigated changes of the phenotype after RA treatment. We also determined the effect of RA on the mrna expressions of the cornifin-α and mucin genes as indicators of squamous and mucous differentiation, respectively. Results:Using an ALI technique, we were able to diffe-rentiate HCE cells into a keratinizing squamous epithelium. When we treated the keratinizing HCE cells with RA, the morphological phenotype progressively changed to mucociliary epithelium. In addition, the expression of cornifin-α mrna was suppressed, and the expressions of mucin gene 5AC (MUC5AC) and MUC5B mrna were increased progressively with RA treatment. Conclusion:We successfully developed a culture system for keratinizing differentiation of HCE cells using the ALI technique in a defined medium. 합 에 레티노익산이 어떠한 영향을 미치는지 관찰해 보았다. 류 7일 후 진주종 상피 세포는 각질 중층 편평 상피 세포 로 분화되었으며(Fig. 1A), 이는 stratum basale, stratum spinosium, stratum granulosum, stratum cornium으로 구성되어 있어 생체내의 진주종 조직과 유사하였다.

PCR에 의한 산물을 작하였고(Fig. 2A), 4일째 되는 날은 거의 대부분의 각질 ethidium bromide이 포함된 2% agarose gels(fmc By- 층이 벗겨졌다(Fig. 2B). 처리 10일 후 1~2개의 기저층 products, Rockland, ME)을 이용하여 분리하였고, CSC Che- 을 제외한 편평 세포층이 사라졌고(Fig. 2C), 21일 후에는 miluminescence s Detection Module (Retest, Strau- 원주 섬모 상피로 분화하였다(Fig. 2D). 처리 21일 후 주 benhardt Germany)를 이용하여 bands를 분석하였다. 사전자현미경(SEM)을 통하여 관찰된 상피 세포는 많은 섬모로 덮여 있음을 확인할 수 있었다(Fig. 3). 결 과 각질화된 진주종 상피 세포에서 cornifin-α mrna 발 진주종 상피 세포 배양에서 각질 편평 상피로의 유도 현에 미치는 레티노익산 처리의 효과 배양된 세포는 증식하여, 5일이 지나면 합류(confluence) 편평 세포 분화의 표식자인 cornifin-α 유전자의 발현 를 이루었으며, 이후 여러 층의 상피세포로 분화하였다. 합 에 레티노익산이 어떠한 영향을 미치는지 관찰해 보았다. 류 7일 후 진주종 상피 세포는 각질 중층 편평 상피 세포 로 분화되었으며(Fig. 1A), 이는 stratum basale, stratum spinosium, stratum granulosum, stratum cornium으로 구성되어 있어 생체내의 진주종 조직과 유사하였다. 특히 분화된 상피 세포는 유핵(有核)의 stratum cornium를 형성 A 하는 parakeratosis현상을 보였고, 이는 생체내 진주종 조직 과 유사한 특징이다. 그러나 배양액에 잠긴 상태(submerged) 로 배양된 진주종 상피 세포는 각질층을 형성하지 않았다 (Fig. 1B). 이는 공기중으로의 노출이 진주종 상피 세포의 각질화에 필수적이라는 사실을 말해주고 있다. Cross-sections of the intact cultures were stained with H & E. The HCE cells were cultured in RA-free medium for 7 days after confluence and then treated with 10-7M RA for up to a further 21 days. The keratin layer began to detach on the 1st day following treatment (Fig. 2A), and progressively exfoliated on the 4th day (Fig. 2B). On the 10th day, only 1- or 2 layers of basal cells remained (Fig. 2C). The epithelium eventually changed into columnar epithelium with cilia (arrow heads) on the 21st day following RA treatment (Fig. 2D). 729

The cornifin-α mrna levels were measured by Northern blotting from total RNA isolated on the 1st, 4th, 10th and 21st day following the RA treatment.

Note that the apical surface of the epithelium is covered with abundant cilia. 세포에 레티노익산을 처리 1일 후, cornifin-α mrna는 4day 10 day 21 day RA MUC5AC MUC5B 급격히 감소하였고, 4일 후에는 cornifin-α mrna가 거 의 발현되지 않았다. 그러나 레티노익산이 없는 배양액에서 β-2m cornifin-α mrnas는 배양 기간과 관계없이 강하게 발현 되었다. 대조 유전자인 β-2m의 발현은 큰 변화를 보이지 않았다(Fig. 4). 이는 레티노익산이 진주종 상피 세포가 편평 세포로 분화하는 것을 억제시킨다는 사실을 말해준다. 각질화된 진주종 상피 세포에서 Mucin mrna의 발현 에 미치는 레티노익산의 효과 Fig. 5.
