Vol. 21, No. 1, 81~86 (6) Embryotoxicity of Bisphenol A in Daphnia magna Gab Soo Hwang School of Civil and Environmental Engineering, Kunsan National University ABSTRACT Embryotoxicity tests were performed in Daphnia magna to assess aquatic ecotoxicity of bisphenol A, a well known industrial compound showing estrogen-like activity in vivo, and to examine their effectiveness in the toxicological assessment. The whole embryonic developmental period was classified into 6 stages and developmental abnormality was checked to evaluate the embryotoxicity. In the present study, bisphenol A showed the ability to interfere with embryonic development, suggesting its antiecdysteroidal activity. The rates of mortality, delayed development, deformity and immobility all showed good concentration-response relationship, demonstrating their possibility as useful toxicological indices in daphnid embryotoxicity tests that have been rarely performed so far. It seemed favorable to the test sensitivity that embryos are removed from maternal daphnids around 7 hr after deposition from the ovaries to the brood chamber. These results suggest that daphnid embryotoxicity tests can be one of useful tools available for the assessment of ecotoxicity of various chemicals in the aquatic environment. Key words : Embryotoxicity, Daphnia magna, bisphenol A, aquatic ecotoxicity, developmental abnormality 서 론 현대산업사회에서우리인간은수많은화학물질들의홍수속에살고있다. 다양한환경중으로유출된각종유해화학물질들은그환경매체중에서식하는수많은생물종들에위해를가하여생태계전반의안정성을교란시키게되고그종국적폐해는결국인간에게미치게된다. 그러나그간의유해화학물질관리와관련된국제적연구노력은주로인간건강에대한위해성평가에보다편중되 To whom correspondence should be addressed. Tel: , Fax: gshwang@kunsan.ac.kr 어왔으며상대적으로환경중의생물체를대상으로하는생태위해성평가에대한관심은미흡했던것으로평가된다. 따라서근래생태위해성평가의중요성을인식하여하천, 해양, 대기, 토양, sediment 등의환경매체들에대한오염평가에있어물리화학적기법에의한오염물질의단순정량분석을지양하고다양한생물학적재료들을이용한생물검정시험법들의적용을통해생태계에대한실제적인위해성평가를수행하고자하는연구노력들이국내외에서활발히시도 ( 한국화학연구소안전성연구센터, 1997; West et al., 1997; Wernersson et al., 1999; Hisao et al., ; 이철우등, 5) 되어오고있으며향후효율적평가기술의개발과표준화를위한국제적노력은갈수록한층가속화될전망이 81

Bisphenol A 는식품캔등을가열시용출되는등우리의실제적인일상생활과밀접한관련을가지는내분비계장애물질인만큼그간많은연구노력이집중되어왔다. 그러나그독성학적중요성에도불구하고국내에서의 bisphenol A 에대한연구는미미하였고또한국외의관련연구결과들도대부분이내분비계장애물질로서척추동물에있어건강위해성평가에집중되어왔으며 (Bond et al., 19; Krishnan et al., 1993) 무척추동물등을이용한생태위해성평가와관련된연구는매우미미한실정이다. 산업폐수, 도시폐수등에의한수질오염이인간의건강과수중생물에심각한위해를초래함은주지의사실이며이들수중유해독성물질들을감시, 제어하기위한실제적인유용한접근법으로서다수의생물학적기법들이제시되어왔는데이중소형어류나갑각류의 embryo 를이용한독성실험은실험동물의배양및관리가비교적간편하고실험동물들이비교적짧은세대기간과생식주기를갖고있어필요한때에충분한실험재료 (embryo) 를얻기가용이하며짧은성장기간내에서우수한감도로서 developmental toxicity 에대한관찰이가능하다는장점때문에근래생태독성학자들을중심으로많은관심을받아왔다 (Dave et al., 1987; LeBlanc et al., ; Wirth et al., 1). 이와같은장점에도불구하고 bisphenol A 의위해성평가와관련된그간의연구보고들을검토해보면소형어류나갑각류의 embryo 를이용한 embryotoxicity 시험의연구결과는극히찾아보기어렵다. 따라서본연구에서는 daphnia 의 embryo 를사용한 embryotoxicity 실험을통하여대표적인내분비계장애물질로알려져있는 bisphenol A 의생태위해성평가를수계환경을대상으로수행하고나아가 daphnia 의 embryo 를활용한효율적생태위해성평가기법의개발에기여하고자하였다. 재료및방법 1. 실험동물 (Daphnia magna) 의배양 금번연구의수행에있어물벼룩의배양및관리는 Olmstead 등 (Olmstead et al., 2) 의방법에준 하여수행하였다. 그내용을요약하면, 물벼룩의사육을위하여항상 ±2 C 의온도, 16L/8D 의광주기가유지되어지며매일오전중정기적으로먹이공급여부, sieving 여부, 사육수교체여부, 물벼룩의건강상태등이점검되어졌다. 사육수로서는증류수에 KCl 8 mg/l, MgSO 4 1 mg/l, NaHCO mg/l, CaSO 4 H 2O 1 mg/l 가첨가되는 Hard constituted water medium 이조제되었고여기에 selenium 1 mg/l 와 vitamin B 12 1 mg/l 가첨가되어사용되었으며물벼룩의먹이로는 tetrafin fish food 와 green algae (Selenastrum capricornutum) 의배합된적당량이매일 2 차례공급되어졌다. 물벼룩은 1 L 의배양액중 45 마리의밀도로유지배양되었으며배양액은 1 주일에 3 회씩교환되었다. 이와같은실험실적배양조건하에서단성생식 (parthenogenetic reproduction) 을통해물벼룩의 female 들이연속적으로배양되었다. 2. Bisphenol A 의 embryotoxicity 실험 본연구에서는 bisphenol A 가물벼룩에대한발육독성물질 (developmental toxicant) 인지의여부와함께물벼룩 embryo 들의 brood chamber 내출현후성숙시간별에따른독성실험을수행하여독성감수성의차이를확인하여가장효율적인 daphnia embryotoxicity 평가기법의수행조건들을확립하는데기여토록하였다. 물벼룩 embryo 의전성숙과정은 6 단계들로구분되어졌으며 (Kast-Hutcheson et al., 1) 실험의안정성과감수성이충족되도록 brood chamber 내 embryo 출현확인후 6~7.5 시간, 8~8.5 시간, ~.5 시간의연령대를대상으로관련 embryo 독성실험들이수행되었다. 실험수행을위해 6~7.5 시간연령대의경우각농도별로 ~ 45 개, 8~8.5 시간및 ~.5 시간연령대의경우 16~ 개의 embryo 들이가급적동일어미의 brood 들에서채취되어 ~ mg/l 농도범위에서 2 배수계열농도의 bisphenol A 함유배양액 µl 가주입 (.1% ethanol 함유 ) 된 96 well microtiter plate 의각 well 에개별적으로분리되었다. 분리된 embryo 들은 C, 16 h 광주기하에서배양되었고치사율 (mortality), 발육지연 (delayed development), 형태이상 (deformity), 비운동성 (immobility) 등을산출하기위해매 12 시간마다현미경을통하여발육상의이

3 March 6 Hwang : Bisphenol A 의 embryo 독성 83 상유무가면밀히관찰되어졌다. 결과및고찰 Embryo 독성시험의유용성을인식하고물벼룩 embryo 를이용하여 bisphenol 의생태발육독성을평가하고자수행된본연구에서초기 6~7.5 시간연령대의물벼룩 embryo 를사용하였을때 sigmoid 형의전형적인농도의존적치사반응곡선을얻을수있었으며 (Fig. 1) 이를위한최적관찰시간은폭로개시 72 시간후임을알수있었다. 반면 8 시간이후의 embryo 연령대에있어서는적용된 bisphenol A 의전농도범위에걸쳐폭로후 72 시간동안매우낮은치사감수성을나타냄으로서농도의존적치사반응곡선을제공하지못하였다. 물벼룩 embryo 의발육단계에있어 stage 1 단계는기존의보고 (Kast-Hutcheson et al., 1) 와는다소다르게본실험에있어서는 embryo 가 brood chamber 내에출현된후 18~19 시간정도까지지속되는것으로관찰되었는데초기연령대의 embryo 사이에있어서도외부환경에대한감수성의차이가매우뚜렷하였다. 구체적으로 6 시간연령이전의 embryo 들은실험수행중안정성을제공하지못하고거의파열됨을보여주었으며 8 시간이후연령대의 embryo 들은 bisphenol 에대해강한저항성을나타내어실험수행을위한높은감수성을제공하지못하였다. Fig. 2 는 bisphenol A 의폭로로부터생존한 embryo 들중에서운동성의유무에대해관찰한결과이다. 이때 초이상전혀운동성이보여지지않은경우비운동성 embryo 로 check 되었으며최적관찰시기는 bisphenol A 폭로후 ~62 시간사이로이시기에 6~7.5 시간및 8~8.5 시간연령대의모든생존 embryo 들은발육단계분류상 stage 5 의후반기에해당하였다. 실험결과 6~7.5 시간과 8~8.5 시간연령대의경우들이감수성의차이에따른일정한반응 gap 을나타내면서대체적으로농도의존적으로증가하는반응 pattern 을보여주었다. Fig. 3 은 bisphenol A 가물벼룩 embryo 들에대해발육저해작용 (developmental inhibition) 을나타냄을보여주고있다. 최적관찰시간은폭로후 72 시간전후인것으로확인되었으며이시기에정상적인 embryo 들은완전히성체로발육된발육단계분류상 stage 6 에해당하나 bisphenol A 의폭로에의해일부의 embryo 들이 stage 5 에해당되는발육저해를보이는것으로나타났으며특히 6~7.5 시간연령대의경우에있어 8~8.5 시간연령대의경우에비해전형적인농도의존적반응 pattern 을보여주었고이경우 bisphenol A 의폭로농도 mg/l 에서모든 embryo 들이 stage 5 단계에해당하는것으로나타났다. 이러한발육저해작용에대한실험결과는 Fig. 4 와관련된형태이상에대한관찰결과와함께 bisphenol A 가물벼룩에대해 antiecdysteroidal activity 를가지고있음을시사하는것으로볼수 Immobile embryos/survivals (%) 6~7.5 hr 8~8.5 hr.1 1 Fig. 2. 4 84 J. ENVIRON. TOXICOL. Vol. 21, No. 1 있으며이와관련하여 Mu 등 (5) 은 bisphenol A 가물벼룩에있어 molting 을지연시키고 embryo 의발육을저해시키는 antiecdysteroidal activity 를야기한다고보고하였다. Fig. 4 에서는 bisphenol A 에폭로된물벼룩 embryo 들에있어생존 embryo 들중형태이상 (deformity) 을가진 embryo 들의존재비율을도시하였다. 이때 embryo 발육상에있어형태이상에대한대표적인관찰대상내용은 shell spine 과 antennae 등의발육이상 (Fig. 5) 으로최적관찰시간은폭로후 72 시간전후였다. 본실험에있어 6~7.5 시간연령대의경우는 bisphenol A 의폭로농도.3 mg/l, 8~ 8.5 시간연령대의경우는 bisphenol A 의폭로농도 5 mg/l 에서부터형태이상이농도의존적으로유발 됨을보여주었으며 ~.5 시간연령대의경우생존 embryo 들중모두에서형태이상은관찰되지않았다. Fig. 6 에서는 6~7.5 시간연령대의 embryo 들에있어상기단일독성반응변수들에대한대표적인부가변수반응결과를산출, 도시하였는데제시된 2 개부가변수들 (dead+developmental inhibition, dead+deformity) 모두의경우에있어전형적인농도의존적 S 자반응곡선을제공함으로서물벼룩 embryo 독성시험이전반적인발육이상 (develop- Stage 5/survivals (%) 6~7.5 hr 8~8.5 hr.1 1 Exposure concentrations of bisphenol A (mg/l) Fig. 3. 5 March 6 Hwang : Bisphenol A 의 embryo 독성 85 mental abnormality) 을검정하는효율적시험법으로서화학물질의발육독성을평가하는데있어유용한평가수단이될수있음을확인시켜주었다. 아울러 8 시간이후연령대의 embryo 들에대해서는반응감수성의약화로인해폭로농도변화에따른유효한반응분별성을얻기어려운것으로판명된바발육독성평가를위한물벼룩의 embryo 독성시험을위해서는 7 시간전후연령대의 embryo 를사용함이가장적합함을알수있었다. 이상본연구에서수행된실험결과들로부터물벼룩 embryo 들을사용한독성실험은다양한독성 parameter 들의활용에대한합리성을부여해주며실험수행의경제성및발육독성관찰에대한효율성의제공을통해화학물질의생태독성평가를위한유용한평가수단으로서제시될수있음을알수있었다. 결 론 대표적인내분비교란물질로알려져있는 bisphenol A 에대해실험동물로서물벼룩 (Daphnia magna) 을사용하여 embryo 독성시험을수행한본연구결과 1. 관찰된독성반응들 (mortality, immobility, delayed development, deformity) 에대해전체적으로양호한농도상관성을보여주었다. 2. bisphenol A 는전반적으로물벼룩 embryo 의발육을저해하여무척추동물에있어 antiecdysteroidal activity 를가지고있음을시사하였다. 3. 물벼룩 embryo 를사용하여화학물질의발육독성평가를수행시 brood chamber 내출현후 7 시간전후연령대의 embryo 들을사용함이가장적합하며전반적인반응 parameter 들에대한최적관찰시간은폭로후 72 시간임을알수있었다. 4. 전체적으로물벼룩을이용한 embryo 독성시험이화학물질의생태독성평가에다양한유용성을제공할수있음을시사하였다. 감사의글 본연구는지역대학우수과학자연구지원사업 ( 과제번호, R ) 으로학술진흥재단의지원으로수행되었음에감사드립니다. 참고문헌 이철우, 박수영, 윤준헌, 최경희, 정영희, 김현미.
