4 01 Technology PyroHotStart Technology Enzyme-mediated hotstart Primer dimer 형성과같이 PCR 반응시작전에발생하는비특이적반응제거 매 cycle 발생하는 PPi를제거하여 PCR 효율증가 PyroHotStart 기술은 pyrophosphate ( 이하 PPi ) 와 pyrophosphatase ( 이하 PPase ) 효소를이용하여, PCR 반응을시작하기전 (zero cycle) 에발생하는비특이적반응을줄여특이도를높인바이오니아고유의특허기술입니다. 또한매증폭 cycle시발생하여 PCR 저해물질로작용하는 PPi를제거함으로써반응효율을높입니다. 따라서특이도가요구되는 multiplex PCR 반응에유용하며, 비특이적반응물로인해겪게되는결과분석의어려움을해소시킵니다. DNA polymerase가활성을갖기위해서는 Mg 2+ 이온이필요한데, mixture에첨가되어있는 PPi는 Mg 2+ 이온과높은친화력을갖기때문에 Mg 2+ 이온을붙잡아둠으로써 PCR 반응을억제합니다. Mixture에첨가되어있는 PPase는변성 (denaturation) 단계에서반응온도가올라가면활성화되어 PPi를 Pi로분해하여 Mg 2+ 이온을해리시킵니다. 해리된 Mg 2+ 이온은 DNA polymerase에결합하여 PCR 반응을진행할수있도록 enzyme을활성화합니다. 활성화된 DNA polymerase는 PCR cycle이진행됨에따라 target DNA를증폭합니다. 16 05 Instrument for Real-Time PCR Exicycler 96 Real-Time PCR Quantitative Thermal Block Superior 5-color Real-Time Quantitative PCR System 제품 개요 Exicycler 96은 96 well 동시 검출 방식의 Real-Time qpcr장비입니다. 바이오니아의 특허인 Light Tunnel (이하 LT ) 기술과 편광을 이용한 형광 검출 기술로 광학적 민감도 와 정확도를 높였습니다. 또한, 96 well 간의 Ct 편차를 최소화하여 reference dye에 의한 normalization 없이 각 well당 5채널의 형광값을 정밀하게 분석할 수 있습니다. Exicycler 96은 10~109 copies의 넓은 detection range로 저농도 및 고농도의 시료에서 DNA 검출이 가능합니다. 분석 소프트웨어는 Absolute Quantification, Relative Quantification, SNP genotyping, Existence/Nonexistence 모드를 제공 하여, 간편하게 viral load analysis, 유전자 발현분석, SNP genotyping 등 다양한 분야 의 실험 분석이 가능합니다. 뛰어난 민감도와 정확도 편광을 이용한 형광 검출 특허기술 (Figure 1)을 적용하여 반사광에 의한 노이즈를 최소화하여 민감도와 정확도 를 향상 시켰습니다. 또한 LT 특허기술 (Figure 2)을 적용하여 조사하는 광원을 모든 well에 균일하게 변환하여, 어떤 well에서 PCR 하여도 Ct 값 편차가 0.3 cycle 이내로 유지됩니다. 특장점 Ramp Rate 향상으로 반응시간 단축 Thermal block의 온도 제어 기술 개발로 ramp rate가 향상되어, PCR 반응에 걸리는 시간을 당사 기존 모델 대비 40% 이상 줄였습니다. 정확한 온도와 균일성 Well간 온도 편차가 ±0.3 이내로 thermal block의 정확성 및 균일성이 우수한 제품입니다. 5-multiplex PCR Well 간 광학 편차를 기구적으로 해결하는 바이오니아의 LT 기술을 이용하여, 일반적으로 편차 보정을 위해 사용 되는 reference dye가 필요하지 않으므로 5채널의 형광을 모두 시료에 활용할 수 있어 real 5 multiplex PCR이 가능합니다.
