2 Flow Cytometry란 무엇인가? A technology that simultaneously measures multiple characteristics of single cells at a rapid rate 즉 유액( 流液 )상태의 입자(Particle) 나 세포(Cell) 가 일정 감지지역 (sensing area)를 통과할 때 각각의 입자나 세포가 갖는 여러 특징을 동시에 측정하는 방법 relative Size : FSC relative Granularity or Internal Complexity : SSC relative Fluorescence Intensity : FL1, FL2, FL3...

3 Flow cytometry (FC) is an emerging technique which holds great promise for the separation, classification and quantitation of cells.

4 Flow Cytometry 발전 역사 1930년대 스웨덴 Karolinska Institute의 Caspersson이 세포내 핵산과 단백질 양을 측정하기위해 microspectrophotometer를 개발한 것이 시초. 1947년 Northwestern대학(미국)의 Gucker등에 의해 공기를 빠 른 속도로 주입하면서 검체를 실려보내고 입자가 통과하면 빛이 산란되어 photodetector에서 전기 신호로 검출해낼 수 있는 최 초의 Flow Cytometer가 개발. 1950년대 형광 항체 개발, hematology counter 도입 1960년대-형광 측정이 cytometry에 도입, DNA content 분석 시도 1970년대에는 Herzenberg 들에 의해 fluorescence flow cytometry 와 cell sorting법이 개발되어 살아있는 세포를 분리 하여 연구에 사용할 수 있게 됨.

5 Flow Cytometry 발전 역사(계속) 현재는 초당 10만개의 세포를 분석할수있으며, 100만개 중에 하 나 섞여있는 드문 세포를 찾아낼 수있고, 1%이내의 정밀도로 형 광을 측정하고, 세포 표면에 붙어있는 수백개 분자의 형광 항체 를 검출해 낼 수 있다. Slow flow system에서는 관찰 시간을 microsecond에서 millisecond로 늘리므로서 한 분자의 형광 분자의 검출이 가능 해짐으로서 세균, 바이러스, macromolecule등의 연구에 이용될 수 있다. 또한 이들에 microfluidic technology가 도입됨으로써 flow를 정지시키거나 뒤로 돌릴 수도 있게 되었다.

48 결론 Flow Cytomery는 세포를 이용한 연구에서는 아 주흔히이용될수있는기본술기이다. 세포를 이용한 작업을 할때는 한번쯤 Flow Cytometry를 이용해서 할수 있는 방법은 없나를 생각한다. 다수의 세포를 분석하고 객관적이면서 정량적인 정보도 얻을 수 있다. 기본 setting을 자유롭게 변화시키면서 분석을 시도해 볼수 있다.
