However, the precise colorectal carcinogenic mechanisms remain to be elucidated. he protein products of oncogenes and cancer suppressor genes play important roles in the carcinogenesis. he effects of the proteins are influenced by post-translational modifications as phosphorylation, acetylation, methylation, and ubiquitination. he aberrant sumoylation plays some roles in carcinogenesis. However, the expression pattern of small ubiquitin-related modifier (SUMO)-2/3 in the colorectal cancer has not been reported. We assessed the expression of SUMO-2/3 and evaluated the expression pattern in colorectal cancer. Methods: he SUMO-2/3 expression was tested in one normal colon mucosal cell line and 5 colorectal cancer cell lines by Western blot. We collected 322 cases of colorectal cancer operated from January 2000 to December 2010 at Soonchunhyang University Cheonan Hospital. We fabricated the tissue microarray and the expression of SUMO-2/3 was evaluated by immunohistochemistry. he results were analyzed with clinicopathologic parameters. Results: he SUMO-2/3 was not expressed in the normal colon mucosal cell line. However, it was expressed highly in all the 5 colorectal cancer cell lines as the beta-actin. he SUMO-2/3 was expressed in 68.3% of the colorectal cancers and its expression was correlated with the pathological tumor stage stage (odds ratio, 2.89; 95% confidence interval, 1.10 to 7.55; P= 0.031). Conclusion: he SUMO-2/3 plays some roles in carcinogenesis and progression of the colorectal cancer. Keywords: Colorectal neoplasms; Small ubiquitin-related modifier; Immunohistochemistry; issue array analysis 서론대장암은우리나라에서식생활습관이서구화됨에따라증가되는경향을보이는악성종양으로아직이의명확한발생기전이밝혀지지않고있는실정이다. 따라서이에대한다각적인연구가필요하다. 암발생에있어서암유전자산물과암억제유전자산물의평형이중요한역할을한다. 이들유전자산물인단백의기능은다양한전사후수정 (post-transcriptional modification) 에의하여그기능이조절된다. 단백질의전사후수정은인산화 (phosphorylation), 아세틸화 (acetylation), 메틸화 (methylation) 및유비퀴틴화 (ubiquitination) 등다양한형태로이루어지며이러한수정에의하여단백질기능의다양성을나타낸다. 이들중유비퀴틴화는단백질의분해에매우중요한역할을하고있다 [1]. Expression of SUMO-2/3 in Colorectal Cancer 46% 에서같은반면 SUMO-2 와 -3은 96% 의아미노산서열이같고기능도유사하기때문에 SUMO-2 와 -3을 SUMO-2/3 로기술하기도한다 [2,3]. 정상적인세포가다양한종류의 stress를받게되면 DA의불안정화로암화 (carcinogenesis) 가쉽게일어난다. 이러한암화과정에여러단백질들의 SUMO-2/3 SUMO화 (sumoylation) 가일어나기때문에 SUMO-2/3 단백증가는암발생을예측할수있다. 그러나지금까지대장암에서의 SUMO-2/3 단백발현변화에대한보고는없는실정이다. 이에본연구에서는 322예의대장암조직의 tissue microarray (MA) 에서 SUMO-2/3 의발현을면역조직화학적방법으로검사하고이들결과를병리조직학적요인들과비교분석하여 SUMO-2/3 단백발현과대장암과의관계를연구하였다. 대상및방법 1. 연구대상 1) 환자조직 2000년 1월부터 2010 년 12월까지순천향대학교천안병원에서병리학적으로암종으로확인되어외과적수술받은환자중 paraffin block의보존상태가양호한 322명의조직을연구대상으로하였다. 2) 세포주대장암세포주 (HC116, LoVo, SW480, H29, Colo205) 와정상대장세포주 (CCD-841-Cor) 를이용하였다. 2. 방법 1) 임상기록검토환자의임상기록을토대로성별, 연령및방사선학적검사기록을조사하였다. 2) 병리조직학적검색환자의조직슬라이드를재검색하여조직학적진단및분화도를정하고 American Joint Committee on Cancer (AJCC, 7th ed.) 의종양병기지침에근거하여 tumor () 병기를정하였다. 3) issue microarray 제작환자의조직 slide를검색후해당 paraffin block에서각예마다 2 개의 2 mm core를채취하여 recipient block에심어한 block당 30예의조직을심었다. 4) 면역조직화학적염색 MA block을 4-5 µm 두께로절편을만들어 xylene으로 5분간 3회씩탈파라핀시킨후 100%, 95%, 90%, 70% 및 50% 의 ethanol 에각각 3분씩처리하여함수시켰다. Antigen retrieval을위해 0.1 M sodium citrate buffer (ph 6.0) 에 15분간 microwave 에서가열시킨후실온에서 30분간식혔다. 그후 phosphate buffered saline (PBS, ph 7.4) 용액으로세척후조직내의내인성과산화효소의활성을억제하기위하여 0.3% hydrogen peroxide-methanol 에 15분간실온에서처리후증류수로세척하였다. 절편을 PBS로세척후비특이적항원을차단하기위하여 30분간염소혈청과반응시킨후여분의용액을제거하고일차항원 mouse monoclonal antibody SUMO-2/3 (AGen, Seoul, Korea) 1:200으로희석하여실온에서 2시간반응시켰다. 그후절편을 PBS로 5분간 3회수세후일차항체 enhancer (Biovision, Mountain View, CA, USA) 와 30분간실온에서반응시켰다. 그후 PBS로 5분간 3회수세후 polymer (Biovision) 와 30분간실온에서반응시킨후다시 PBS로 5분간 3회수세하였다. 발색은 diaminobenzidine을사용하였고 Mayer s hematoxylin으로약하게대조염색한후다시 graded ethanol로탈수후 xylene으로투명화시켰고그후 balsam으로봉입하여광학현미경으로관찰하였다. 면역조직화학적염색판독은핵과세포질에염색정도를전혀염색되지않는것 0, 경미한염색 1, 중등도의염색 2, 강한염색 3의 4등급으로구분하였다. 또한염색부위를 0-20% 염색 1, 20-60% 염색 2, 60% 이상염색 3으로등급구분하여염색정도와염색부위의등급합이 5 이상을양성으로판독하여통계처리하였다. 5) Western blotting HC116, LoVo, SW480, H29, Colo205, CCD-841-Cor 각각의세포를 37 C 5% CO 2 humidified incubator 를이용하여 RPMI1640 (+10% fetal bovine serum, +1% adult bovine serum) 에서배양한후 Western blot 을시행하였다. BCA protein assay (Pierce, Rockford, IL, USA) 를이용하여 protein의양을측정한뒤 30 µg/µl로정량한후 Laemmil Sample Buffer (Bio-Rad, Hercules, CA, USA) 와 1:1로섞은뒤 100 C에서 10 분간 heating 후 ice에서식혔다. Mini-PROEA 4-20% gradient GX gel (# ) 에 sampling한 protein 을 30 µl씩 loading 후 100V (400 ma), 60분간전기영동을진행하였다. 그뒤 polyvinylidene fluoride membrane 에 rans-blot urbo (Bio-Rad) 를이용하여 transfer하였다. Membrane 를 5% skim milk에서 blocking 후 SUMO-2/3 primary antibody (AGen) 를 1:1,500으로 ris-buffered saline/ween20 (BS) 를이용하여희석후 4 C에서 overnight 반응시켰다. Secondary antibody (peroxidase labeled anti mouse goat antibody) 를 1:1,000으로 BS를이용하여희석후실온에서 1시간반응시켰다. BS를이용하여 washing한뒤 Enhanced chemiluminescene 반응시켰고 chemiluminescent gel document로 detection 하였다. 96

Expression of SUMO-2/3 in Colorectal Cancer 종양조직 근처의 정상조직이 포함된 24예의 정상조직 모두에서 음 현이 높은 것으로 나타났다(P = 0.031). 단변량분석(able 2)에서 성 성을 보였고 암종 인접 정상 상피세포에서는 SUMO-2/3의 핵 내 발 별, pathological nodal status (p stage)와의 유의성이 없는 것으로 현이 관찰되었다. Adenoma에서 발생한 암종에서는 adenoma의 나타났지만, pathological tumor stage (p stage, P = 0.031)와 분화 핵 내 발현이 관찰되지 않은 반면 암종의 핵 내에서는 강한 발현을 도 moderate differentiation에서 SUM-O2/3 발현이 높은 것으로 보였다. 분화도에 따른 SUMO-2/3의 발현은 분화도가 좋은 종양에 나타났다(P = 0.017). 서는 발현되지 않은 반면 분화도가 낮은 암종에서는 세포질과 핵 내에 강한 발현이 관찰되었다(Figs. 2, 3). 고 찰 세포의 생명활동을 유지하기 위해서 단백질의 기능이 매우 중요 3. SUMO-4 의유전자 code 는밝혀졌으나세포내에서이의발 현및기능에대해서는잘알려져있지않다 [9]. SUMO-2/3 의표적 단백과결합은표적단백의 ΨKXE 일때 K (lysine) 에 SUMO 가결합 한다. Ψ 란큰 hydrophobic amino acid residue 를의미한다. SU- MO-1 과 SUMO-2/3 의차이점은 SUMO-1 은세포내에다른단백 에결합상태로존재하는반면 SUMO-2/3 은유리형태 (free form) 로 존재하다가세포가어떤 stress 를받으면바로표적단백에결합하 여 conjugation 이된다. 세포단백의 SUMOylation 은세포의정상 적생명현상에매우중요한역할을하지만비정상적인 SUMOylation 은악성종양발생뿐만아니라신경변성질환 (neurodegenerative disease) 및심장병들을일으킬수있다 [11]. SUMOylation 과연 관된물질들이암발생과관련있음이알려졌는데 [12,13] 예로 Ubc9 (SUMO conjugating enzyme, E2) 은사람의악성종양이발 현증가가관찰되었을뿐아니라이의발현증가는암세포성장을 촉진시킨다 [14,15]. 활성화된 SA3 를억제하는 SUMO E3 protein PIAS3 은다양한종양에서발현증가가관찰되고있으며 [16] SUMO E1 효소의발현증가는간세포암환자에서생존율감소와 관련있음이알려졌다 [17]. 또한 SUMOylation 은암억제유전자인 p53, prb, p63, p73 및 Mdm2 등의활성에관여함으로악성종양발 생에일조하기도한다 [18]. 또한 SUMOylation 은유방암과전립선 암발생에관여하기도한다. 유방암발생에관련있는 BRCA1 에 SUMO-2/3 이결합되면 BRCA1 이 androgen receptor 에직접적으 로작용하여 receptor 기능을항진시켜전립선암발생에기여한다 99

Expression of SUMO-2/3 in Colorectal Cancer [19,20]. 또한 desumoylation 효소인 SUMO protease SEP1 은전립선암과갑상선암에서발현증가가관찰되며이의발현증가는전립선암발생을촉진시키기도한다 [21]. 암발생의외적요인중유전물질의돌연변이를유도하는방사선, 다양한종류의화학적발암물질및체내에서생성되는활성산소등을들수있다. 그러나이들이외에유전자돌연변이가없어도유전자집합체인염색체의불안정화가암발생의한원인이되기도한다. 염색체의안정화를위해서는염색체에부착되어있는특수구조물인동원체, 텔로메어등의안정화가필수적인데이런유전자집합체인염색체는여러단백의결합에의하여외부의유해자극으로부터보호되지만이기능이없으면염색체가불안정해짐으로암을비롯한다양한질병이발생된다. SUMO는이러한염색체의이질염색체 (heterochromatin) 의조절에중요한역할을한다. 즉 SUMO는염색체안정화에중요한역할을하는여러가지단백질의기능을조절해주는데이러한 SU MO 의기능에이상이있을때악성종양이발생하기도한다 [22]. 이외에 SUMO-2/3 conjugating enzyme 인 E2 (Ubc9) 의발현증가는폐선암종, 흑색종, 유방암및난소암등에서관찰된다 [23]. p53는세포유전자의위해가가해졌을때위해로부터세포를보호함과아울러위해받은유전자의복구를도와줌으로암발생을억제해주는암억제유전자의대표적인물질이다. 따라서 p53의대사에관여하는모든물질들은암발생과직접적혹은간접적으로관련이있다. Mdm2는 p53 기능을조절하는물질중하나이다. Mdm2는 SU- MOylation 의표적물질로 E3 ligase 작용을한다 [24]. 그러나아직은 Mdm2의 SUMOylation 과 p53의안정성과의관계가학문적으로확실시된것은아니나최근 SUMOylation 과 desumoylation 에의한 Mdm2의기능조절기전이밝혀지고있다 [25]. SUMOylation 된 Mdm2는 Mdm2의자가유비퀴틴화를감소시켜정상적인상태에서 p53의양이감소되도록조절해준다. SUSP4 는 Mdm2의 de- SUMOylation 효소인데 UV에의하여 DA가손상받으면 SUSP4 가증가하여 Mdm2의 desumoylation 이일어나고 Mdm2의자가유비퀴틴화 (self-ubiquitination) 가촉진된다. 이러한결과로 p53가안정화되어 p53 의존적인세포사멸과세포성장억제등이이루어진다 [23]. 암전이는악성세포가혈관내로이동하며다른장기로퍼지는현상을말하며이러한현상으로암에의한사망률이높게된다. 이러한암전이는암연구의최종목표라할수있는어려운문제가되고있다. 최근에 SUMO가암전이에대한조절역할을한다는연구결과가발표되었다. KA11은 F-kB의표적유전자로암전이억제유전자기능을한다. 최근 KA11유전자의암전이억제가 ip60과 beta-catenin을통하여이루어진다는것이밝혀졌다. 비전이암세포에서는상대적발현량이많은 ip60의기능으로 KA11이발현되어암전이가억제되지만전이암세포에서는전사억제복합체로작용하는 beta-catenin 과 reptin의양이상대적으로증가되어 ip60 의급격한감소를초래하고따라서 KA11 발현이억제되어암전이 가유발된다 [26]. 최근에 SUMO 가전이억제유전자인 KA11 발현을 조절한다는사실이밝혀졌다 [23]. SUMO 의기능을다양한문헌고찰로검토해본결과 SUMO 는 암발생과전이에직접혹은간접적으로관여함을알수있었다. 그 러나대장암종발생과의관련에대해서는아직까지보고가없는실 정이다. 따라서본연구에서는 322 의대장암을대상으로 SUMO- 2/3 의발현을면역조직화학적으로조사하여이의결과를임상및 병리학적으로검토하였다. 본연구에서총 322 예의대장암중 220 예 (68.3%) 에서 SUMO-2/3 의강한발현이종양세포의핵과세포질 내에서관찰되었다. 이로미루어볼때 SUMO-2/3 의발현은대장 암발생과관련있음을알수있다. 또한 SUMO-2/3 의발현은성별, 연령, p, p, 혈관전이, 림프관전이등과의통계학적유의성은없 는반면 moderated differentiation 에서통계학적유의성을보여 SUMO-2/3 의발현과분화도가관련이있는것으로나타났다. 단변 량분석에서는 p stage 에서 stage 가높을수록 SUMO-2/3 의발현 이높은것으로나타났다. 따라서대장암종에서 SUMO-2/3 의발 현은암발생과진행에역할을한것으로생각된다.
