1 Journal of the Korea Academia-Industrial cooperation Society Vol. 17, No. 11 pp , ISSN / eissn 박서현, 이승호 * 상명대학교의생명공학과 Comparative studies of various transfection processes for the optimal luminescence signal analysis Seohyun Park, Sunghou Lee * Department of Biomedical Technology, Sangmyung University 요약형광간섭현상을최소화시켜상대적으로민감한측정이가능한 aequorin기반 luminescence기술은 G α16 단백질도입을통해세포내부의칼슘이동신호를감지하여 G 단백질결합수용체 (G protein-coupled receptor, GPCR) 의기능분석을가능하게하는세포기반분석기술로수용체및 G 단백질유전자전달의최적화과정이필수적이다. 본연구를위해 corticotropin releasing factor receptor subtype 2(CRF2) 수용체를모델시스템으로 CRF2와 G α16 단백질이구축된세가지안정화세포주를제작하였고, 이들을이용한서로다른세가지조건의임시발현세포주에서작용제 (sauvagine) 와길항제 (K41498) 의반응성을분석하여최적의유전자전달방법을도출하고자하였다. 그결과 sauvagine 및 K41498의농도에따른반응에서 CRF2-G α16 안정화세포주가임시발현세포주보다 10배이상의유효신호비율을나타내었고 (z =0.77) 임시발현세포주의경우 G α16 의안정화발현이후에 CRF2를전달하는경우가다른임시발현조건보다 2배이상높은효율을보였다 (z =0.84). 따라서임시유전자전달기술을 GPCR 세포기능분석시스템에활용할경우 G α16 단백질에대한안정화세포주를우선적으로구축하고, 목표하는다양한수용체들을단계적으로발현시키는것이최선의방법이라판단된다. Abstract By minimizing fluorescence interference phenomena, aequorin-based luminescence technology can provide a relatively sensitive detection platform with integration of Gα16 protein in order to track internal calcium mobilization by G protein-coupled receptors (GPCR). In this type of cell-based functional assay format, it is essential to optimize the transfection process of a receptor and Gα16 protein. For this study, corticotropin releasing factor receptor subtype 2(CRF2) was set as a model system to generate three stable cells with CRF2 and Gα16 in addition to transiently transfected cells under three different conditions. Agonist (sauvagine) and antagonist (K41498) responses in those cells were analyzed to develop the optimum transfection process. As a result, the effective signal ratio in the dose response experiments of sauvagine and K41498 were at least 10-fold higher (z'=0.77) in CRF2-Gα16 stable cells. For the transient transfection cells, stable expression of Gα16 prior to the CRF2 represented a two-fold higher signal (z'=0.84) than the other cases of transient transfection. In conclusion, for the utilization of transient transfection processes to develop a cell-based GPCR functional assay system, it is suggested to introduce various target receptors after stable expression of Gα16 protein. 2 1. 서론 G 단백질결합수용체 (G protein coupled receptor, GPCR) 의구조와신호전달기전이밝혀진후로생체내다양한생리학적인반응을유발시키는원인을알게되면서현재개발중인신약의 40% 이상이 GPCR에초점을맞춰개발되고있다 [1]. 국외연구선두그룹에의해다수의 GPCR구조를밝히는등 [2] 연구가활발히진행되고있는반면국내에서구조연구는투자및연구진행방향과기술력이부족하여성장이더디고있다. 향후 GPCR을타겟으로하는생리활성물질의개발 [3-5] 및신약개발연구를위하여구조및기능에관한활발한연구가필요하다고판단된다. 분자유전학의발전에따라약물로개발될수있는가능성을지닌타겟의수가매우증가하였고타겟에대한새로운후보물질을탐색하기위해많은물질들을효율적으로빠른시간내에검색이가능한고효율약효검색스크리닝 (High-throughput screening, HTS) 기법이접목된다양한분석실험들이개발되어활용되고있다 [6]. 이러한실험들중다양한 GPCR를타겟으로하여개발되는기능분석시스템은주로세포를기반으로분석되기때문에유전자전달과정이필수적이다. 유전자전달방법은원형형태의벡터를이용하여임시로유전자가발현되도록하는임시유전자전달방법과선형화시킨벡터를세포로전달하여발현된유전자가환경적인요인에의해서사라지지않고안정적으로발현이되게하는안정적유전자전달방법으로나뉜다 [7]. 안정적유전자전달방법은제작기간이길지만양질의실험결과를얻을수있는가능성이높은반면임시유전자전달방법은안정화단계를거치지않기때문에바로이용할수있지만실험결과의변동성이크게나타나는단점이있다. 300여종이상의서로다른 GPCR 수용체들에대한기능분석시스템이요구되는현재의연구환경을고려한다면빠른시간내에최적의실험조건을검색가능한임시유전자전달방법의체계적인활용이필수적이다. 이에본연구에서는 GPCR 타겟들에대한세포기능분석을위해필요한 G 단백질과수용체유전자들의전달방법에따른실험결과를모델시스템을통하여상호비교하였다. 이를위하여중추신경계부터피부질환까지다양한영향 [8-10] 에관계한다고알려져있는부신피 질자극호르몬수용체 (Corticotropin-releasing factor receptor subtype 2, CRF2) 를모델시스템으로 G 단백질 (G α16) 과수용체 (CRF2) 유전자들의전달방법에따른실험결과의비교를통하여다양한 GPCR 기능분석시스템에효율적으로활용가능한기초자료를제시하고자하였다. 2. 재료및방법 2.1 재료및기기수용체활성화에따른세포내칼슘이동신호를 luminescence 신호로감지하기위하여개발된세포주인 A30(ES-000-A30) 은 HEK293 세포에칼슘과반응하는광단백질인 aequorin[11, 12] 유전자가안정적으로구축된세포주 [13, 14] 로서 Perkinelmer사 ( 미국 ) 로부터라이센스를구입하여사용하였다. CRF2(GenBank Acc#, AY449734)[15] 와칼슘신호전달에필요한 G 단백질인 G α16(genbank Acc#, M63904) cdna는 Missouri S&T cdna Resource center( 미국 ) 로부터구입하였으며 CRF2 수용체에대한작용제인 Sauvagine은 American peptide company(cat# , 미국 ) 사의제품을사용하였다. CRF2에대한길항제인 K41498은 TOCRIS(cat# 2070, 영국 ) 사의제품을사용하였다. 세포내부의칼슘이동의변화를측정하기위하여 proaequorin의 cofactor로이용되는 Coelenterazine h는 Discoverx(cat# , 미국 ) 사의제품을사용하였다. 아울러측정에이용되는 96-well black polystyrene plate는 Costar(cat# 3915, 미국 ) 사의제품을사용하였으며수용체활성화에따른 luminescence 측정은 Mithras LB 940(Berthold, 독일 ) 을사용하였다. 2.2 세포배양 A30 세포는 MEM 배지 (cat# LM007-08, WELGENE, 한국 ) 에 10% fetal bovine serum(gibco, 미국 ), 100IU/ml penicillin, 100μg/ml streptomycin(hyclone, 미국 ) 과 10 μg/ml zeocin(invitrogen, 미국 ) 이첨가된배양액을사용하였으며세포는 100mm tissue culture dish(cat# , Corning, 미국 ) 에서 5% CO 2 가공급되는 37 의온도조건에서배양하였다. 세포밀집도가 80% 에도달했을때 Phosphate buffered saline(hyclone, 미국 ) 5ml로세척하고 0.05% Trypsin-EDTA(GIBCO, 미국 ) 641

3 한국산학기술학회논문지제 17 권제 11 호, ml을첨가하여세포를떼어낸뒤항생제가없는배양액 5ml을첨가하여 1,000xg에서 5분동안원심분리를진행하였다. 침전된세포에항생제가없는배양액을 10ml 첨가하여혼합후 개의세포를 100mm tissue culture dish 로옮긴뒤 10μg/ml zeocin(invitrogen, 미국 ) 을주입하여 5일간격으로계대배양하였다. 2.3 안정화세포주구축및 luminescence 신호분석을위한임시유전자전달제조사의프로토콜에따라 4D-Nucleofector system (LONZA, 스위스 ) 을사용하여 A30 세포주에각각선형의 CRF2 수용체유전자 3μg과 G α16 유전자 3μg을전달한후적정항생제농도와세포수로 96-well culture plate(bd Falcon, 미국 ) 에 5% CO 2 가공급되는 37 의온도조건에서배양하였다. 약 2-3개월에걸쳐만들어진클론들을칼슘과 camp에대한반응성실험을통하여최대반응성을갖는클론을선택하였으며각각 A30CRF2와 A30G16로명명하였다. 실제 CRF2 수용체연구를위해구축된안정화세포주인 A30G16CRF2는 luminescence 신호분석을위한비교세포주로사용하였다. 임시발현세포는 A30CRF2 세포주에 G α16 유전자를전달한 A30CRF2/G16, A30G16 세포주에 CRF2 유전자를전달한 A30G16/CRF2, A30 안정화세포주에 G α16 과 CRF2 유전자를동시에전달한 A30/G16CRF2 세포들로서로상호비교하여최적의 luminescence 신호분석을위한임시발현조건을검색하고자하였다. 2.4 세포내칼슘측정을위한 aequorin 기반 luminescence 신호분석 aequorin기반수용체기능분석시스템을이용하여수용체활성화에따른세포내부의칼슘이동을 luminescence 신호로측정하였다 [6, 16]. 임시유전자전달세포는 48시간동안배양후 Phosphate buffered saline(hyclone, 미국 ) 10ml로세척하고 Accutase(PAA, 영국 ) 1ml을사용하여세포를떼어낸뒤 Phosphate buffered saline(hyclone, 미국 ) 9ml을첨가하여 1,000xg 에서 5분간원심분리하였다. 침전된세포에 cells/ml이되도록 DMEM/HAM s F12 배지 (cat# LM002-05, WELGENE, 한국 ) 와 0.1% Albumin bovine Fraction V(SERVA, 독일 ) 의조성을갖는완충액을첨가한뒤, 500μM의 Coelenterazine h를첨가하였다. Rotator(FINEPCR, 한국 ) 를사용하여암실에서 의온도조건하에약 18시간동안반응시킨후완충액으로세포수를 cells/ml로맞추고 암실에 1 시간동안방치하였다. 작용제활성신호를측정하기위해 96-well black plate(costar, 미국 ) 에준비해둔세포 50μ l/well(5,000cells/well) 에추가로 Sauvagine을 50μ l/well(10μm 4.57nM) 로분주후측정하였다. 길항제의활성신호는 K41498을 50μl/well(A30G16CRF2: 3μ M 1.37nM, A30CRF2/G16: 1μM 457pM, A30G16/CRF2: 300nM 137pM, A30/G16CRF2: 1μ M 457pM) 로준비해둔세포 50μl/well에첨가한뒤 15분동안빛을차단시키고측정기계와동일한온도조건에서반응시킨후 50μl/well(500nM) 의작용제를분주하여측정하였다. Mithras LB 940(Berthold, 독일 ) 장비를사용하여반응을통해생성되는 luminescence 신호를실시간으로 20초 /well( 작용제활성 ) 또는 18초 /well( 길항제활성 ) 로측정하였으며측정된 Relative light units(rlu) 값을시간에대한 Area under the curve(auc) 로변환시킨수치인 RLU(AUC) 로사용하였다. 2.5 자료처리및통계분석농도에따라얻어진데이터수치는 PRISM version 5.0(GraphPad Software Inc., 미국 ) 의 Non-linear curve fitting 기능으로분석하였으며데이터의유의관계를비교하기위해 One way ANOVA 및 Bonferroni s multiple comparison test로분석하였다. 작용제와길항제의투여농도에따른반응성에서최대와최소수치, EC 50, IC 50, Hillslope의변수값을통하여그래프를나타내었다. 데이터의평균, 표준편차, 변동계수는 Microsoft Excel(Microsoft Corporation, 미국 ) 을사용하여분석하였다. 3. 실험결과 3.1 안정화세포주에서반응성확인 G α16 유전자와 CRF2 유전자를안정적으로발현시킨안정화세포주 (A30G16CRF2) 에서투여농도에따른작용제와길항제의반응성을확인하였다 [Fig 1]. A30 세포내에존재하는아세틸콜린수용체에대한아세틸콜린의 642

4 반응성은최대농도신호값 188,850 ± 4,233과최저농도신호값 30,920 ± 15,906으로 S/B 비율 (Signal to Background ratio) 이 7.10이었고 EC 50 는 1.16μM로확인되었다. CRF2에대한작용제인 sauvagine의반응성은최대농도신호값 194,400 ± 3,201, 최저농도신호값 8,981 ± 6,494로 S/B 비율이 21.65이었으며 EC 50 는 56.85nM으로확인되었다. CRF2에대한길항제인 K41498에대한반응분석에서 negative control 수치가 36,359 ± 2,689, positive control 수치가 437,095 ± 7,981로 S/B 비율이 12.02이었으며실험데이터의품질을평가하는통계파라미터인 z 수치 [17] 는 0.77로나타났다. 아울러투여농도에따른반응성실험에서최대농도신호값 426,739 ± 16,044, 최저농도신호값 16,252 ± 12,658로 S/B 비율이 26.26로나타났으며 IC 50 은 45.14nM으로확인되었다. A30CRF2/G16과 A30G16/CRF2, 그리고 G α16 단백질및 CRF2 수용체를동시에전달시킨 A30/G16CRF2, 세가지세포조건에서내재된아세틸콜린수용체에대한작용제의최대농도신호값은각각 86,901 ± 4,395, 141,130 ± 5,746, 333,637 ± 6,959로 A30/G16CRF2 세포가가장높게나타났으며최저농도신호값은 4,313 ± 13,395, 15,282 ± 13,814, 15,613 ± 14,410으로 A30CRF2/G16 세포가가장낮게나타났다. S/B 비율은 A30/G16CRF2 세포가 21.37로가장높았고 S/B 비율이 9.24로가장낮은수치값을갖는 A30G16/CRF2 세포에비해 2.31배높았다. 그리고 A30CRF2/G16과 A30/G16CRF2 세포는유사한 S/B 비율을보였다. CRF2에대한 sauvagine의최대농도신호값은각각 49,301 ± 1,898, 105,826 ± 2,846, 27,262 ± 2,858이었으며최저농도신호값은 4,211 ± 1,154, 12,778 ± 3,075, 12,022 ± 1,681로 A30G16/CRF2 세포에서최대농도신호값이가장높은것으로확인되었다. 이에따라 S/B 비율은 A30CRF2/G16 세포에서 11.71로가장높았으며 A30/G16CRF2 세포에서 2.27로가장낮은수치를보였고 EC 50 는각각 491nM, 175nM, 945nM로확인되었다. Presented data were representative data set from at least three separate experiments performed in triplicate, and each data points were expressed as mean ± SEM(Standard error of mean). 3.2 임시유전자전달에서작용제반응성비교임시로 G α16 과 CRF2 유전자를발현시킨세포에서투여농도에따른작용제반응성을비교분석하였다 [Fig. 2]. 5 한국산학기술학회논문지제 17 권제 11 호, 임시유전자전달에서길항제반응성비교 CRF2 길항제 K41498의투여농도에따른반응성을비교하고실험데이터의품질을평가하는통계파라미터인 z 수치를비교분석하였다. A30CRF2/G16, A30G16/CRF2, A30/G16CRF2 세포조건의반응성실험에서 negative control 값이각각 6,503 ± 311, 10,932 ± 972, 10,239 ± 1,626이었으며 positive control 값이 64,366 ± 2,253, 256,307 ± 3,663, 21,253 ± 1,757로 S/B 비율은 9.90, 23.45, 2.08로나타났다. z 수치는 0.62, 0.84, -1.61로확인되었다. 투여농도에따른반응성에서최대농도신호값은각각 61,275 ± 2,140, 260,277 ± 9,318, 34,195 ± 8,626이었으며최저농도신호값은 7,891 ± 950, 14,959 ± 14,540, 13,425 ± 1,540으로확인되었다. 이에따라 S/B 비율이 7.77, 17.40, 2.55로 A30G16/CRF2 세포에서가장높은수치를보였으며 IC 50 는 3.65nM, 13.5nM, 1.75nM로현저한차이가확인되지않았다 [Fig. 3]. Presented data were representative data set from at least three separate experiments performed in triplicate, and each data points were expressed as mean ± SEM(Standard error of mean). 3.4 안정화세포주와임시유전자전달세포의반응성비교안정화세포주인 A30G16CRF2와임시로유전자를전달시킨 A30CRF2/G16, A30G16/CRF2, A30/G16CRF2 세포들의실험결과를일원분산분석과함께 Bonferroni 사후검증을실시하고그결과를표에나타내었다 [Table 1]. 6 투여농도에따른작용제와길항제의반응성결과는최대농도신호값과 S/B 비율에서세포간의유의성이현저하게나타났다. CRF2 작용제인 sauvagine의농도에따른반응성결과에따라 A30G16CRF2 세포는최대농도신호값과 S/B 비율에서 A30/G16CRF2, A30CRF2/G16 세포와유의성을보였고 A30CRF2/G16 세포는최대농도신호값에서 A30G16/CRF2 세포와유의성을보였으며 A30/G16CRF2 세포는 EC 50 수치를기준으로 A30G16CRF2, A30CRF2/G16, A30G16/CRF2 세포와유의성을보였다. 길항제인 K41498의농도에따른반응성결과에따라 A30/G16CRF2 세포는최저농도신호값에서 A30CRF2/G16 세포와유의성을보였고최대농도신호값에서 A30G16CRF2 세포는 A30/G16CRF2, A30CRF2/G16 세포와유의성을보였다. 또한 A30CRF2/G16 세포는 A30G16/CRF2 세포와유의성을보였으며 A30/G16CRF2 세포는 A30G16/CRF2 세포와유의성을보였다. 그리고 A30CRF2/G16 세포는 S/B 비율에서 A30G16/CRF2 세포와유의성을보였다. 4. 고찰및결론세포를기반으로한 GPCR의기능분석은주로 G단백질을매개로생성되는이차전달자인 camp 또는 IP 3 의변화를측정할수있도록개발되었고 [18] 특히, GPCR의활성화를통하여변화되는세포내부의칼슘을형광 (Fluorescence imaging plate reader, FLIPR) 또는 luminescence(aequorin) 로측정하는기법이광범위하게활용되고있다 [19]. 이경우세포내칼슘이동신호를전달하기위하여 Gq 단백질의연결이필요하며 Gi/s 단백질이연계된 GPCR들의경우칼슘신호를감지할수있게하기위하여추가로 promiscuous G 단백질인 G α 16[20, 21] 의도입이필요하여다중유전자도입에따른칼슘신호의최적화과정이필수적이다. 이러한과정을통하여형광간섭이최소화되어상대적으로민감하고넓은신호영역이제공되므로광범위하게이용되고있는 luminescence 기술 [11, 12, 22] 을사용한 CRF2 수용체기능분석시스템을개발하고자하였다. 실험에영향을미치는유전자전달방법의최적화과정을확인하기위하여안정화세포주 (A30G16CRF2) 와임시발현세포들 (A30CRF2/G16, A30G16/CRF2, A30/G16CRF2) 을비교분석한결과안정화세포주인 A30G16CRF2의작용제최대농도신호값이 230,000 RLU(AUC) 이상으로다른세조건의임시발현세포주들에비해최대 8.85배이상의높은수치를보였다. 또한길항제반응성에서최대농도신호값이 260,000 RLU(AUC) 이상으로다른세조건의임시발현세포주에비해최대 7.65배이상의높은수치를보여안정화세포주에서최상의반응성을보였다. 그리고세조건의임시발현세포주에서는 A30G16/CRF2 가 A30CRF2/G16, A30/G16CRF2에비해작용제와길항제반응성의최대농도신호값과 S/B 비율이 2배이상의높은수치를보였다. 이는 G α16 이먼저세포막에구축되어있을때외부로부터전달되는 CRF2와보다더효율적으로결합하여 CRF2 자체의신호를 Gq 신호경로로전달하게되어칼슘측정에높은반응성을나타낸것으로보인다. A30/G16CRF2 세포조건에서는측정시작용제반응성에서큰효율을보이지않았으며작용제로인한반응성이낮아역학반응초기에불안정한신호수치를나타내어실험이불가능하다고판단되었다. 이에대한원인은보고된바와같이외부로부터인위적으로여러유전자들을도입할때세포내에서발현정도를조절하는것이매우어려우며 [23]G α subunit에서 C말단끝의 5개아미노산 [24] 과 4α/6β loop, N말단 α-helix 지역등이수용체결합특이성을결정하여상호작용을한다고알려진내용 [25] 과관계가있다고판단된다. 또한비록수용체와 G α subunit이융합된단백질유전자를전달시킨경우가더향상된결과를주지만융합되지않는수용체와 G α subunit의유전자를동시전달시킬경우수용체가세포막에있는내생의 G α subunit에우선적으로선택성을가지고결합이되기때문에 [26] 이러한결과가초래된다고판단된다. 본논문에서 CRF2 수용체를모델로이용하여 G 단백질과수용체유전자들의전달방법을비교분석한결과 aequorin기반 luminescence 실험에서안정화세포주가가장반응성이높지만임시발현세포를사용할경우우선적으로 G 단백질을안정적으로발현시킨후목표하는수용체를 2차적으로발현시키는방법이가장성공가능성이크다고판단된다. 645
