PB10001 Green Growth - Kenturky Lexington LANE'S END Stallions(USA) 조사연구박준형 2, 권태준 2, 배재호 2, 김대수 1, 박소영 1, 김대경 1, 정혜나 1, 강민수 1,2,3 1 제주대학교생명자원과학대학생명공학부, 2 제주대학교대학원동물생명공학전공 3 제주대학교마필산업연구센터미국 Kenturky 주 Laxington은세계경주마의수도라일컬어질정도로세계적인우수경주마를생산공급하고잇으며, 특히고능력종모마를많이사육하여원하는마필농가에게종부료를받아서비스시켜주는종모마목장들이여기저기산재해있다. 2007년여름에우리나라최초로제주도에마산업클러스트사업을정부로부터유치하기위한준비단계로선진국마필산업을시찰하기위하여 Kenturky 주 Lexington을방문하여이곳의 `대표적인종모마목장을시찰하하였다. 이때방문한목장이 LANE'S END Stallions으로서종모마가한마리씩방목된방목장에는목초가파랗게잘자라고있었으며, 이목장의종모마보유두수는약 20두정도되고있었고, 유럽풍의마사와청결한마방, 그리고넓은종부사를보유하고있어서마필농가가원할때언제라도선택된종모마와의교배가이뤄질수있게하고있다. 이목장의종모마중에가장최고의종부료를지불하는말은 A.P.Indy라는마명의말로무려 $ 300,000으로한화약 3억원이넘는금액이다. 한마리의망아지를임신시키기위해지불하는종부료가무려 3억원이라니그혈통의우수성과그에따른거액의종부료에감짝놀라지않을수없었다. 물론어떤종모마는고작 $ 5,000d에불과한것도없지는않았다. 우리나라에서근래도입된최고의종모마가격이 40억원정도라는점을염두에두고미국의경우를비교해보면경주마세계의자연교배원칙을다시금생각해보면서우리경주마의경주능력수준과높기만한세계의수준의벽을가히짐작케한다. Key words : Green Growth, Kenturky Lexington, LANE'S END Stallions(USA) PB10002 흑염소분변및혈액에서분리되는질병원인체분석최창용, 손동수, 정병열 *, 곽동미 **, 최순호, 류일선, 허태영, 정영훈, 강석진, 조상래, 손준규, 최선호, 김성재, 정연섭농촌진흥청국립축산과학원, * 국립수의과학검역원, ** 경북대학교수의과대학우리나라에서오랜기간동안약용으로이용되어온흑염소는최근육용으로도그수요가늘어나면서사육규모가다두화, 집단화되고있고, 질병의발생도복잡 다양해지고있는실정인데, 소, 돼지, 닭등의주요가축에비해발생되는질병의원인분석이상대적으로미비한실정이다. 따라서본연구에서는흑염소에서질병발생시분변및혈액에서세균및바이러스를분리하여질병발생의원인을분석하고자하였다. 설사증세를보이는흑염소분변을채취하여원인체를분석한결과소바이러스성설사병 (Bovine Viral Diarrhea, BVD) 이 34%(16점 /47점), 소전염성비기관염 (Infectious Bovine Rhinotracheitis, IBR) 이 2.1%(1점 /47점), 클로스트리듐 (Clostridium perfrigens) 30.6%(15점 /49점), 대장균증중용혈성대장균 2.0%(1점 /49점), 비용혈성대장균 8.2%(4점 /49점) 이분리되었다. 7개농가 98점의혈청을이용하여원인체분리한결과네오스포라 (Neopora canium) 와요네병 (Johnes Disease) 원인체는분리되지않았고, 렙토스피라 (Leptospira spp) 는 6.1%(6점 /98점) 분리되었다. 본연구를통해흑염소의설사병등질병발생도소, 돼지등과마찬가지로다양한세균과바이러스가원인으로확인되었으며, 이를바탕으로좀더다양한세균, 바이러스, 원충등의분리동정이연구되어져야할것으로사료된다. Key words : 흑염소, 분변, 혈액, 질병원인체, BVD, IBR PB10003 Reference ranges for biochemical and hematological values of dairy cows three week before and three week after parturition Tai-Young Hur, Young-Hun Jung, Seog-Jin Kang, Chang-Yong Choi, Ui-Hyung Kim, Dong-Su Son, Il-Sun Ryu, Sung-Jai Park, Hyun-Sup Kim National Institute of Animal Science, Rural Development Administration, Korea During the transition period, the dairy cows have critical endocrine and metabolic changes and it is necessary the reference ranges of laboratory results in normal transition dairy cows. The cows suffering mastitis, retained fetal membrane and metabolic diseases within the 3 weeks after calving were excluded. All biochemical analytes(β-hydroxybutyrate, glucose, cholesterol, calcium, and bilirubin) were statistically different precalving and postcalving groups. The hematological analytes were not significantly different execpt for hemoglobin. Different reference ranges for precalving and postcalving dairy cows should be determined to help the interpretation of the biochemical results in transition period. PB10004 암소품종에따른포유기한우송아지의행동특성김종선, 박연수 1, Lishaogang, 송영한강원대학교동물생명과학대학, 1 강원도축산기술연구센터본실험은암소품종에따른한우송아지에서의증체량및행동특성을알아보기위해 2009년 4월부터 6월까지강원도축산기술연구센터에서한우암소에인공수정 (Artificial Insemination; AI) 으로태어난한우송아지 5두및홀스타인암소에수정란이식 (Embryo Transfer; ET) 으로태어난한우송아지 5두를공시하였다. 행동에대한측정은육안및 CCTV camera(ir LED Camera; APD-7070V, Sony) 로비디오녹화후 08시부터 18시까지관찰하였다. 행동발현시간은 2분간격으로 Sampling하였으며행동발현빈도는행동발현시마다측정하였다. 송아지의일당증체량을비교한결과 ET에의해태어난한우송아지에서 AI보다높은경향을보였다. 포유시간및빈도에서도 ET에서유의적 (p<0.05) 인차이를나타냈으며, 홀스타인암소의비유량이많은이유로송아지의증체로이어졌고, AI에서유의적 (p<0.05) 으로높게나타난기립행동은비유량의부족에따라나타나는행동으로판단된다. ET의걷기행동에서도유의적 (p<0.05) 인차이가나타나풍부한모유섭취로개체의활동성이강한것을확인할수있었으며, 그에따른음수빈도에서도유의적 (p<0.05) 인차이를나타냈다. AI의핥아주기행동이유의적 (p<0.05) 으로높게나타난것은포유량의부족에따른구벽 (Inter-sucking) 등의이상행동이증가한것으로분석된다. 따라서, 본연구를통해암소의품종에따른송아지의행동특성파악할수있었으며추후, 송아지관리증진을위한요인별추가적인연구가지속되어야한다고판단된다. Key words : 한우, 송아지, 행동특성, 수정란이식, 인공수정 PB10005 희소한우정액의동결방법과평형시간에따른생존성및운동성비교손준규, 조상래, 최창용, 연성흠, 최선호, 손동수, 조창연, 이풍연, 고응규농촌진흥청국립축산과학원한우의품종중현재흑우와칡소는멸실위험에직면한희소한우로분류되어있다. 현재희소품종으로분류되어있는흑우는산업적인이용은불가능하나국내가축유전자원의다양성의확보차원에서소중히다루어지는국가적인중요한자원이다. 흑우및칡소정액의채취와동결보존에대한연구는보고되지않았으며, 각지자체연구기관에서연구목적으로제한적으로이용되고있는실정이다. 따라서본연구는유전적다양성을유지하고유전자원의안정적인보존을위해희소한우정액을동결보존코자실시하였다. 정액의희석제재는 AndroMed를사용하였으며, 흑우정액을채취한후 2시간정도평형을시켰을때는 75% 의생존율을보였다. 그러나동결융해후생존성은 50% 수준으로낮아진결과를보였으며, 채취한후 6시간, 18시간그리고 30시간에서의평형시킨후생존성은채정후와마찬가지로 75% 의생존율을보여각시간대별생존율은차이가없었으나, 54시간째에는 50% 로떨어지는결과를보였다. 흑우정액을동결시키기위해서냉각처리하는방법에따른정액의생존성과운동성조사에서는액체질소상단 10cm, 10분처리한후액체질소에동결하는것이가장효과적으로동결보존후생존성을높일수있었다. 정액동결시평형시간에따른생존성과운동성은 30시간까지차이가없었으며, 정액동결방법에서액체질소상단의위치와동결시간에따른생존성과운동성은 5cm 5분간동결을실시하였을때는생존성 45% 운동성 70% 의결과를보였으며, 5cm 10분간에서는 40% 생존성과 70% 운동성을나타냈다. 또한 5cm, 10분간처리하였을때는 50% 생존성, 70% 운동성을보여다른두처리구보다더효율적인결과를보였다. 현재흑우나칡소의경우멸실위험축종으로서분리되어있다. 이러한희소한우의증식을위한수단으로서정액의생산과보급이지속적으로이루어져야할것으로사료된다. Key words : 희소한우, 정액동결, 생존율, 운동성, 평형시간 PB10006 소의무인발정알림이개발및현장적용시험결과백광수, 박성재, 전병순, 임현주, 김현섭, 손준규, 박수봉농촌진흥청국립축산과학원본연구는소의발정발견효율성을제고하기위하여무인상태로발정을탐색할수있는승가검출기를개발하여그정확도를분석하였고, 전문경영체농가 3개소에서현장적용시험을실시하여발정발견정확도및수태율을조사하였다. 승가검출기는발정이예측되는개체의후지발목에센서를부착하도록고안되었다. 발정이발현되어승가또는승가허용행위를하게되면수직충격에의하여무게추와접점이접지되어신호가발생하게된다. 발생된신호는알림판으로전송되어승가또는승가허용횟수와최초및최종승가또는승가허용시점이표시되도록고안되었다. 승가행위에따른정확도를보면, 승가시도시 87.5%(7/8두 ), 미끄러졌을경우 100%(4/4두 ) 였고, 1초이상안정적으로승가를하였을경우승가허용우가허용시 80.0%(12/15두 ), 회피시 90.0%(10/11두 ) 로, 무인발정알림이를부착한소가승가행위를할때의알림판에표시되는평균정확도는 86.8%(33/38) 였다. 승가허용행위에따른정확도를보면, 허용을거부하여회피시 100%(2/2두 ), 1초이상안정적으로승가허용을할경우 100%(7/7두 ), 승가허용을하면서회피할경우 100%(1/1두 ) 로, 무인발정알림이를부착한소가승가허용행위를할때의알림판에표시되는평균정확도는 100%(10/10) 였다. 무인발정알림이를부착한소가승가및승가허용행위를할때의알림판에표시되는평균정확도는 90%(43/48) 였다. 현장적용시험에의한발정발견정확도는 85.7%(12/14) 였고, 발정이발견된개체들에대한수태율은 75.0%(9/12) 였다. PB10007 과배란처리된한우공란우로부터생산된수정란이식수태율조상래, 최선호, 최창용, 손준규, 최수호 1, 김영주 2, 박찬섭 2, 손동수농촌진흥청국립축산과학원, 1 경북축산기술연구소, 2 영주시농업기술센터본시험은우수한한우송아지를생산하기위해과배란프로그램에따라서처리된공란우에서과배란을유도하여인공수정을실시한다음수정란을회수하였다. 회수된수정란은수란우에이식하기위해서수정란을 0.25ml 스트로우에장착하여수정란수송기 (FHK, Japan) 를이용하여수송하였다. 수정란이식을위한수란우의발정동기화는발정주기와무관하게 day 0에 CIDR device를삽입과동시에 estradiol benzoate 1 mg을주사하였으며, day 7에 CIDR의제거와 25 mg PGF 2α를주사하였고 day 9에 100 ug gonadorelin주사하고 day 17에수정란이식을실시하였다. 수정란이식하루전에 1,500 IU의 hcg(chorulon, Intervet, Holland) 를주사하였으며, 수정란이식 1시간전에는 Flunixin meglumine(fluxinine R, BOMAC Lab. LTD, New Zealand) 350 mg을근육주사하였다. 시험의결과를다음과같다. 수정란이식을위한채란은총 4회를실시하였으며, 농가는 29농가 122두에이식을실시하였다. 1차수정란이식은 5농가 29두에이식하여 51.7%, 2차는 10농가 49두에이식하여 61.2%, 3차는 10농가 25두에이식하여 64%, 4차는 4 농가 19두에이식하여 58% 의수태율을나타내었다. 소수정란이식으로수태율을향상시키기위해서는공란우와수란우의사양관리와수정란이식에대한연구가필요할것으로사료된다. Key words : 한우, 공란우, 수란우, 수정란이식, 수태율 PB10008 유전자원보존을위한한우수정란의동결에따른생존효율성조사조상래, 최선호, 최창용, 손준규, 연성흠, 이풍연, 고응규, 조창연, 손동수농촌진흥청국립축산과학원본시험은희소한우의멸실위험대비및재래가축생식세포의안전한동결보존기술개발을위해서한우체내 외수정란동결융해후생존율을조사를위해서실시하였다. 체내수정란생산을위해한우공란우를사용하였으며공란우는 CIDR R (InterAg, New Zealand) 를삽입하고 estradiol benzoate(esrone, Samyang, Seoul, Korea), progesterone(ovaron, Samyang, Seoul, Korea), FSH제제인 Antorin R R 10(Kawasaki Pharm, Japan), dinoprost(lutalyse TM, Pharmacia & Upjohn, Belgium) 를각각처리하여과배란을유기한다음, 인공수정후수정란을채란하였다. 체외수정란은도축난소를이용하여수정란을생산하였다. 수정란의동결을위해완만동결보호제는 1.8M Ethylene Glycol(0.1M Sucrose+0.3% BSA) 을사용하였으며, 초자화동결은 Capillary Glass pipettes을사용하여 Open Pulled Straw(OPS) 방법을이용하였으며, 동결보호제는 VS1(TCM 199+20% FBS, 1분 ), VS2(TCM199+20% FBS+7.5% DMSO, 1분 ), VS3(TCM199+20% FBS+17% DMSO+20% EG, 20초 ) 를사용하여동결하였다. 수정란의생존율은융해후배양액에서 12시간이상배양하여수정란이재확장또는부화되는수정란을생존한것으로평가하였다. 실험의결과는초자화방법에서체내수정란은 85.1%(40/47), 체외수정란은 75.0%(141/188). 그리고완만동결방법에서는체내수정란 79.8%(67/84), 체외수정란 73.8%(116/157) 의생존율을보였다. 가축유전자원의안전한보존과증식을위해서수정란및생식세포동결보존에대한연구는지속적으로수행되어져야할것으로사료된다. Key words : 한우, 체내수정란, 체외수정란, 초자화동결, 완만동결 PB10009 Effect of Nandrolone Decanoate on The Expression of Steroidogenic Enzymes in The Rat Testis Ki-Ho Lee, Hee Jung Seo, Chan Wok Son, Seung Kyu Lee, Haing Woon Baik Department of Biochemistry and Molecular Biology, Eulji University, Daejeon Nandrolone decanoate (ND) is an anabolic-androgenic synthetic hormone which is frequently used for treatment of clinical diseases. An increase of illegal use of ND by young and adult athletics and people has been reported. Uncontrolled and prolonged use of ND results in occurrence of a number of side effects, including a decrease of serum level of testosterone and reduction of male fertility. However, the effect of ND on steroidogenesis in the testis has not been determined in detail. Thus, the present study was focused to reveal changes of gene expression of steroidgenic enzymes in the rat testis after ND treatment at different doses (2 mg and 10 mg ND/Kg BW/week) and for different periods (2 and 12 weeks). Subcutaneous injections of ND into male Sprague-Dawley rats were begun at 30 days of age, Molecules tested in the present study were steroidogenic acute regulator (Star), P450 cytochrome side chain cleavage (Cyp11a1), 17alpha-hydroxylase (Cyp17), Hsd3b1, P450 cytochrome aromatase (Cyp19), 17beta-HSD4 (Hsd17b4), aldose reductase (Akr1b1) and carbonyl reductase 1 (Cbr1). Results are summarized asfollowing: 1) significant decreases of Star mrna levels with all ND treatment; 2) significant decreases of Cyp11a1 and Cyp17 mrna levels at 10 mg ND treatment for 2 weeks and significant decreases of Cyp11a1 and Cyp17 mrna levels at both treatment doses for 12 weeks; 3) a significant decrease of Cyp19 mrna level with ND treatment for 12 weeks; 4) significant decreases of Hsd3b1 mrna levels by 12-week ND treatment; 5) no change of Hsd17b4 and Akr1b1 expression with ND treatment; and 6) significant decreasesof Cbr1 mrna expression with ND treatment for 12 weeks. In addition, significant decreases of serum testosterone levels were observed with prolonged ND treatment. Data obtained from the present study indicate that ND would affect syntheses of steroid hormones in the testis by modulating gene expression of steroidogenic enzymes andsteroid hormone metabolism-related molecules. These findings are important to understand a mechanism in which ND commonly used for clinical treatment and improvement of physical ability influences the male reproduction. PB10010 Oral Consumption Effect of Nandrolone on The Testis of The Male Reproductive Tract Ki-Ho Lee, Hee Jung Seo, Chan Wok Son, Inho Choi, Yong-Pil Cheon, Tae Hoon Cheon Department of Biochemistry and Molecular Biology, Eulji University, Daejeon Nandrolone is frequently used for improvement of physical ability. However, intramuscular administration of synthetic nandrolone often results in infertility in male. Boar meat contains non-negligible amount of naturally synthesized nandrolone. Thus, the present study was attempted to examine the effect of nandrolone fed orally with diet on male rat reproduction. Changes of fertility rate, testicular histology, and caudal sperm number were recorded. Gene or protein expression of steroid-metabolism relating enzymes in the testis was detected by real-time PCR or western blotting and immunohistochemistry, respectively. Results showed no significant change in fertility and body, fat, testis, and epididymis weight after nandrolone treatment for 6 weeks. However, a significant increase of caudal sperm number was found in rat treated with a high dose ofnandrolone. Immunohistochemical analyses revealed that most of molecules tested were localized in Leydig cells of the testis and there was no visible change in intensity of immuno-reactivity. However, treatment with nandrolone resulted in differential gene expression of molecules tested in the rat testes, even though protein levels and localization of these molecules were not significantly modulated. These findings indicate that oral administration of nandrolone doesn t affect male reproduction, presumably due to a quick transformation of nandrolone into inactive metabolites in the liver. Key words : Nandrolone, Testis, Steroidogenesis, Gene expression, Immunohistochemistry PB10011 개체식별한우체외수정란농가이식최선호, 조상래, 최창용, 손준규, 김양식 1, 박재영 2, 정연길 3 국립축산과학원, 1 임실군농업기술센터, 2 고성군농업기술센터, 3 이티바이오텍본시험은개체식별한우체외수정란을농가의번식우에이식함으로서농가실용화를위한기초자료를얻고자실시한실증시험에의해나타난결과이다. 2009년경기양평, 전북임실, 경남고성의 3지역의한우번식우 50두이상의농가를대상으로수행하였으며, 이식전에직장검사를통해내부생식기를검사하여생식기이상과황체의유무를확인하였고, 황체가확인된번식우는 PGF2α 5ml를근육주사하였고, 10일후주사하였던대리모의황체의유무에따라대리모로선발하였다. 체외수정란은도축암소의난소를개체별로채취하였으며난포란은체외성숙을실시하였고, 다음날개체확인및육질등급을확인한후 1등급이상의개체만을체외수정을실시하여생산된배반포를수정란이식에공시하였다. 농가수란우발정동기화율은경기양평, 전북임실, 경남고성의순으로각각 74.5%(35/47), 66.0%(35/53), 71.2%(37/52) 로나타났으며, 수란우전체의평균 progesterone 농도는처리전 1.3~1.7 ng/ml가처리후 3.3~3.4 ng/ml로나타났으며, E2의경우는처리전 21~23 pg/ml가처리후 18~20 pg/ml로나타났다. 수정란이식수태율은각각 54.3%(19/35), 59.0%(23/39), 59.5%(22/37) 를나타냈으며, 지역간의유의적인차이는나타나지않았다. 이상의결과로개체식별한우체외수정란이식은농가의번식우의능력을효율적으로이용할수있어농가소득향상에기여할것이며, 도축으로인하여소실되는우수한능력의등록암소혹은고능력우의유전자원재활용적측면에유용하게쓰일수있을것으로사료된다. Choi 1 1 Department of Agricultural Biotechnology, Seoul National University, Seoul, Republic of Korea 2 Department of Animal Science, Pusan National University, Miryang, Korea Inappropriate feeding system may cause nutrition or health problems in Hanwoo cows. MPT (metabolic profiles test) is a useful tool for assessing metabolic status and helping in the diagnosis of metabolic disorders in domestic animals. MPT is based on an analysis of serum metabolites. In this study, total of 2205 Hanwoo cows were divided into 4 groups according to physiological stage. We analyzed the 12 indicators for MPT in each groups by using parametric or non-parametric method and established a reference range of those. Reference range of MPT showed the different levels at each physiological stage in all the test indicators. The efficacy of reference range was evaluated by using the feeding trial. In feeding experiments, incidence of deviation (%) was calculated in three different feeding groups. Our results indicate that the reference ranges is sensitive to the nutrition level, including the type of feeding in Honwoo cows. PB10013 Telomerase Activity In Rat Seminal Vesicle During Postnatal Development Hee Jung Seo, Chan Wok Son, Seong Kyu Lee, Haing Woon Baik, Ki Ho Lee Eulji university Telomerase is the protein-rna complexe present at the terminai of eukaryotic chromosomes and is required for several important cellular functions. The present study was to determine expression of telomerase and its associated proteins in the seminal vesicle of the male Sprague Daweley rat during postnatal development at 1week, 15days, 25days, 45days, 5months, 1year and 2years of ages. The seminal vesicle were collected for total RNA isolation and cdnas were generated a semi-quantitative real-time PCR analysis was employed for the present study. The molecules tested in the present study were telomerase, TERC(Telomerase RNA component), TEP1(Telomerase-associated protein1) and TERF1(TRF1)-interacting nuclearfactor2(tinf2). The expression of telomerase, TEP1, TERF1 and TEP1 genes were extremely low, except at 5months of age at which mrna levels were significantly increased. These results indicate that expression of these genes in the seminal vesicle of the male reproductive tract is differentially regulated in age-dependent. Telomere, male, seminal vesicle PB10014 Cloning and expression profiling of chicken neuron specific gene family member 1 during brain and gonadal development Deivendran Rengaraj, Jin Won Choi, Bo Ram Lee, Sang In Lee, Kyung Soo Kang, Hong Jo Lee, Young Sun Hwang, Gwonhwa Song, Jae Yong Han WCU Biomodulation Major, Department of Agricultural Biotechnology and Research Institute for Agriculture and Life Sciences, Seoul National University, 599 Gwanak-ro, Gwanak-gu, Seoul 151-921, Korea The Neuron specific gene (NSG) family consists of three members: all these members are implicated in synaptic receptor endocytosis and membrane protein trafficking in the somatodendritic and axonal compartments of nerve cells. However, characterization and functional analysis of any NSG family genes in avian species is not known. The aim of the present study was cloning and characterization of chicken NSG1 (cnsg1) as a genetic resource to study the relationship between brain and gonads. Protein sequence comparisons revealed that the amino acid sequences of cnsg1 is highly conserved with zebrafinch (99%) and various mammalian NSG1 proteins (88-98%). RT-PCR and real-time PCR analysis on limited series of tissues at E6.5 and during sexual maturation revealed that cnsg1 was highly involved in brain, central nervous system and gonadal development. We further examined the mrna localization of cnsg1 during brain and gonadal development by in situ hybridization analysis. cnsg1 expression was strongly detected in all parts of the brain and spinal cord. On the other hand, cnsg1 was differentially expressed during gonadal development. cnsg1 was initially detected in the primordial germ cells, and continued expression in the germ cells until hatch. After hatch, cnsg1 expression was detected at a low level in males and at a high level in females. In summary, we characterized chicken NSG1 which might play a role in endocytosis and involve in the signaling dialog between brain and germ cells. The epididymis is divided into 4 different segments, initial segment and caput, corpus, and caudal epididymis, depending upon functional and morphological features. Cell-cell interaction is an important to create unique microenviroment in each segment of the epididymis. Among several forms of cell-cell junctional complexes, gap junctions are specialized channels that directly link the cytoplasm of neighboring cells and are consisted of a number of connexin isoforms. The present study was designed to examine mrna expression of connexin isoforms by semi-quantitative real time PCR analysis in the epididymis of male rats during postnatal development. Results showed that nine connexins of 13 isoforms were present in the entire epididymal regions including connexins 26, 30, 30.3, 31.1, 32, 37, 40, 43, and 45. In addition, the abundance of these connexin isoforms mrna levels was different within each epididymal regions at different postnatal ages. The current study demonstrates differential regulation on expression of connexin isoforms in the rat epididymis in segment-dependent and age-dependent manners. PB10016 Identification of ERBB receptor feedback inhibitor 1 gene expression in chicken Whasun Lim, Suzie E. Ahn, Jin Won Choi, Jae Yong Han and Gwonhwa Song WCU Biomodulation Major, Department of Agricultural Biotechnology, Seoul National University, 599 Gwanak-ro, Gwanak-gu, Seoul 151-921, Korea ERBB receptor feedback inhibitor 1 (ERRFI1) also known as mitogen-inducible gene 6 (MIG-6) is a negative regulator of epidermal growth factor (EGF) signaling. In mice, ablation of Errfi1 gene causes epithelial hyperplasia, adenoma, and adenocarcinoma in organs such as the lung, gallbladder, and bile duct. However, characterization and functional analysis of any NSG family genes in avian species is not known. The aim of the present study was cloning and characterization of chicken ERRFI1 gene to compare expression patterns and function in the development of reproductive tracts and hormonal regulation between chicken and mice. In the present study, we observed ERRFI1 gene expression in each embryonic stage and adult organs by RT-PCR, quantitative RT-PCR and in situ hybridization analyses. The ERRFI1 gene was expressed brain, kidney, small intestine, muscle in male chicken, and detected at a low level in female kidney, heart, liver and muscle. Of particular note, ERRFI1 gene was highly expressed in adult testis as well as female brain and oviduct. In situ hybridization analyses determined the location of ERRFI1 mrna in the gland area of oviduct and seminiferous tubules of testis. Collectively, our current data indicate unknown hormone-mediated transcriptional activation of ERRFI1 expression increases in the testis and oviduct in a cell type-specific manner, and provide new insights into the reciprocal communication between the germ cell development and the formation of reproductive organ in chicken. Key words : ERBB receptor feedback inhibitor 1, testis, ovary, chicken, gene expression PB10017 사료내생균제와생약제첨가가이유자돈의장내미생물변화에미치는영향조광근 1,*, 정미애 1, 김병욱 1, 박화춘 2, 류연선 3, 강승하 4, 강상기 5, 김삼웅 6 1 국립진주산업대학교동물소재공학과, 2 다산종돈, 3 ( 주 ) 조이바이오텍, 4 CSIRO Livestock Industries, 5 서울대학교농생명공학부, 6 국립진주산업대학교양돈과학기술센터 본연구에서는시판중인대표적생균제 2종과생약제를사료에첨가, 급여하였을때이유자돈의장내미생물에미치는영향을규명하고자하였다. 사양시험은이유자돈총 100두를공시하고항생제가첨가되지않은기초사료처리구 ( 대조구 ), 항생제처리구 ( 기초사료 + 티아무린, 엔라마이신각 0.1%), 생균제 A 처리구 ( 기초사료 + 생균제 A 0.04%), 생균제 B 처리구 ( 기초사료 + 생균제 B 0.1%), 생약제처리구 ( 기초사료 + 생약제 0.2%) 로구분하여실시하였다. 장내미생물분석은 CombiMatrix사의 4x2K format DNA chip을사용하였으며 negative control 9개를포함하여총 747개의 probes를디자인하였다. DNA chip 분석결과, 문 (phylum) 수준에서는모든처리구에서 Firmicutes( 전체 hybridizaion signal의약 50~52%) 와 Proteobacteria(18~24%) 가우점하는것으로나타났다. Species( 종 ) 수준에서는대조구 429종, 항생제처리구 462종, 생균제 A 처리구 466종, 생균제 B 처리구 452종, 생약제처리구에서 453종의미생물이확인되어대조구의미생물다양성이가장낮은것으로나타났다. 생약제처리구를제외한전처리구에서가장많이확인된 phylospecies는 Listeria monocytogenes( 전체 hybridization signal의 6~7%) 였으며생약제처리구에서는 Facalibacterium prausnitzii(6~7%) 가가장많이존재하였다. 본실험에서사용된항생제, 생균제 A, 생균제 B, 생약제처리는돼지장내미생물다양성을증가시키는것으로확인되었다. 또한생약제처리구의경우항생제처리구와유사한미생물변화를보이는것으로나타나생약제와항생제가장내미생물에대해유사하게작용할것으로사료된다. Ahn, Jin Won Choi, Hee Won Seo Hyun Jun Jang, Young Min Kim, Hee Jung Choi, Gwonhwa Song, Jae Yong Han WCU Biomodulation Major, Department of Agricultural Biotechnology, Seoul National University Proteases are involved in the degradation and remodeling of the extracellular matrix and are associated with cell transformation, differentiation, motility, and adhesion. These functions are also related to cancer cell invasion and metastasis. Chickens spontaneously develop epithelial ovarian cancer and are therefore a good animal model for human ovarian cancer. However, no studies have investigated the expression of Proteases in chickens with ovarian cancer. We chose two protease family, cystein cathepsins (CTSs) and matrix metalloproteinases (MMPs), and investigated its expression in chicken ovarian cancer. In the present study, CTSB, CTSC, CTSS, and MMP3 genes were highly expressed in cancerous chicken ovaries. Messenger RNAs for the three CTSs were localized to a gland-like area, a major characteristic of cancerous ovaries, but the three CTSs showed no specific localization in normal ovaries. MMP3 mrna was predominantly localized to stroma, which is very similar to MMP3 expression in human cancers. Immunoreactive CTSB protein was present in the gland-like area of cancerous ovaries. Collectively, our current data suggest that CTSB, CTSC, CTSS and MMP3 have important functions in the development of epithelial ovarian cancer and provide the basis for understanding the mechanism(s) responsible for the development of ovarian cancer in chickens. PB10019 수송이젖소의생리변화및생산성에미치는영향이인형, 이성은, 김나라, 안중현, 이상락건국대학교동물생명과학대학동물생산 환경학전공본연구는수송에따른스트레스가착유우및임신한육성우의생리적변화와착유우의유생산성에미치는영향을조사하고, 수송전안정제및영양제처리의스트레스완화및유생산성에대한효과를판단하기위하여실시하였다. 실험 1에서는평균 41월령의착유우 10두를공시하여대조구와처리구로각각 5두씩나누어대조구가축사의스탄촌에보정을한상태로대기하는동안처리구는트럭으로 4시간의수송을하였고, 실험 2에서는착유우 20두및임신한육성우 10두를공시하여대조구와처리구로각각착유우 10두및육성우 5두로나누어처리구에수송전안정제및영양제를처리한후대조구및처리구모두 2 시간 40분의수송을하였다. 실험 1에서생리적변화는수송중 cortisol 및 NEFA는수송중 (P<0.001) 증가, WBC, neutrophil, glucose는수송후증가 (p<0.001), N/L비율의증가 (p<0.05) 하였고, 유조성분의변화는 fat 및 MUN이수송후유의한감소, protein, lactose, SNF는유의하게증가하였다. SCC는수송시작후급격히증가하였다가수송중점차감소하는경향이나타났다. 실험2에서는착유우의수송후처리구의 cortisol 농도가대조구보다유의하게낮았고 N/L 비율이처리구가낮았으며, 유생산은대조구는수송후 3일에 15.5% 가감소하였고, 처리구는수송후 1일에 19.3% 감소하였으나수송후 3일이후부터처리구의유량이대조구보다더높게나타났다. 육성우는처리구의 total protein, albumin, glucose 농도가대조구보다유의하게감소하였다. 이상의결과로수송전안정제및영양제의처리는수송스트레스의완화에도움이될것으로판단된다. Key words : 수송스트레스, 안정제, 생리적변화, 유조성분, 유생산, 착유우, 임신한육성우 PB10020 마우스를이용한 SNCA 유전자와아프리카수면병과의관련성정덕원, 조창연, 이풍연, 고응규, 연성흠, 손준규, 김상우, 최순호, 유용희, 윤종택 * 농촌진흥청국립축산과학원가축유전자원시험장, * 한경대학교 Synuclein Alpha(SNCA) 유전자는주로비장과초기적혈구세포에서높게발현되며헤모글로빈합성에관여한다고보고되어있다. 한편포유동물이원충인 Trypanosoma Congolense에게감염되면중추신경계손상과적혈구파괴를통해빈혈을유발시키고가축에서는생산성의저하와폐사를유발시키는것으로보고되어있다. 본연구에서는 SNCA 유전자발현과포유동물의 T.Congolense 감수성및저항성의관계를규명하고자실시하였다. SNCA 유전자가발현하는 C57BL/6-SJL(+) 계통과 SNCA가 knock-out된 C57BL/6-OlaHSD(-) 계통의마우스에게 T.Congolense 인공감염하여헤모글로빈수치를측정하였다. 두계통은인공감염후 2주동안급격한헤모글로빈수치의감소가관찰되었다. 그러나 + 계통은 2-4주사이에증가를보인이후, 4주차이후에는다시감소하는경향을보였다. 한편 -계통은 2주이후지속적로완만한헤모글로빈감소를보였다. 인공감염이후의생존성을조사한결과 + 계통은 33% 이었고 -계통은 20% 로관찰되어 SNCA 유전자발현군이높은생존율을보였다. 체중변화를조사한결과두계통모두감염이후대조군보다체중이증가하는경향을보였으며, 특히양계통모두인공감염이후체중의증가를보이지않는개체는폐사하는것으로관찰되었다. 이상의결과로마우스에서 SNCA 유전자가발현의유무는헤모글로빈수치및체중증가의차이를보인것으로밝혀졌다. SNCA 유전자의발현을조사로수면병에저항성을가지는계통의조성이가능할것이시사되었다. Key words : 아프리카수면병, Trypanosoma Congolense parasite, SNCA PB10021 천연한방제재의급여가한우송아지설사예방에미치는영향최창본 *, 백경훈 *, 김병기 **, 김수정 ***, 이상욱 **** 영남대학교 *, 경상북도축산기술연구소 **, 대구한의대학교효능검증원 ***, 농업회사법인목원 ( 주 ) **** 한우송아지설사는포유기발생비율이 43.1% 나되는전염성이높은급성질병으로폐사율이매우높으며 ( 최고 25%), 송아지의발육및성장에도심각한장애를나타내어초기증체저하및출하체중의감소등실제축산농가에막대한경제적손실을초래하고있다. 송아지설사병을치료하기위한방법들중현재시중에서판매. 유통되고있는약물들은일시적인효과는있으나치료를중단하게되면병원균이잠복하였다가재발하거나만성화될우려가있으며, 특히장기간반복치료시내성문제가심각하게대두될수있다. 한방에서는오래전부터한약재를이용해서가축질병을치료해왔으며, 이러한한약재는질병을미연에예방하는예방약적인부분에특히강점을가지고있으며, 무엇보다도독성이적거나무독하고가축의체내에잔류하지않으며, 약물의내약성을일으키지않는다는장점을가지고있다. 따라서본연구는천연한방제재를이용한송아지설사예방및치료첨가제를개발하여송아지설사병으로인한농가의어려움과송아지폐사에따른경제적손실을감소시키고, 안전한축산물생산으로축산물의소비를증가시키고자실시되었다. 천연한방제재의급여는농가관행구에비해이유시까지의한우송아지총증체량및일당증체량을 11.8% 개선시켰으며, 사료요구율을 21.4% 개선시키는결과를나타내었다. 또한농가관행구의설사발생빈도수는천연한방제재급여구보다 29.6% 높은것으로나타났으며, 경제성분석결과, 1kg 증체에소요되는비용이천연한방제재급여구 (C-4구) 가 1,258원으로농가관행구 (C-1구, 1,592원 ) 에비해매우유리한것으로조사되었다. PB10022 한우번식우백신및사양관리카렌다를활용한번식효율제고백경훈 *, 정슬기 *, 김성일 **, 이상욱 ***, 정근기 ****, 최창본 * 영남대학교 *, 군위축협 **, 농업회사법인목원 ( 주 ) ***, ( 사 ) 목산한우연구소 **** 한우번식우사육농가의경우, 두수증가에따른개체관리가용이하지않은까닭에적절한사양관리 ( 임신감정, 백신주사등 ) 시기를놓치는수가많았으며, 이는결국번식우뿐만아니라송아지폐사율의증가로이어지기때문에시급히해결해야하는축산업의큰과제로남겨져있다. 이러한현장애로사항들은농장주및번식우사양농가의사육의지와는무관하게현재까지많은두수를일괄적으로관리할수있는기술즉, 체계적인관리시스템의개발이미진한것이가장큰이유라할수있을것이다. 따라서본연구팀은농촌진흥청의지원을통해번식우사양농가들의현장애로를해결하기위한 한우번식우백신및사양관리카렌다 를제작하게되었으며, 실제약 2년간의현장활용시험을통해주요번식지표의개선효과를확인할수있었다 ( 공태일 : 최대 46% 감소, 첫수정월령 : 최대 7% 단축, 분만간격 : 최대 28% 단축 ). 또한주간분만유도법, 아까바네예방법등과같이실제한우번식농가에매우유용한기술적정보들이카렌다에제공되어있기때문에한우번식농가를위한지침서역할도겸비하고있어본카렌다를번식우사육농가에서적극활용한다면한우의대외경쟁력제고등한우산업전반에걸쳐기여하는바가매우클것으로기대한다. Key words : 한우, 번식우, 카렌다, 번식효율 PB10023 한우호흡기질환예방을위한최적의백신조건탐색백경훈 *, 임연수 **, 정슬기 *, 정근기 ***, 최창본 * 영남대학교 *, 농협중앙회한우개량사업소 **, ( 사 ) 목산한우연구소 *** 한우의호흡기질환은바이러스가비말에의하여접촉또는공기전파되므로전염력이광범위하기때문에사육농가의피해가심각한실정이다. 현재국내에서가장문제시되는호흡기질환유발바이러스로는전염성비기관염바이러스 (IBRV), 소바이러스성설사병바이러스 (BVDV), 소파라인플루엔자바이러스 (PI3V), 소합포체폐렴바이러스 (BRSV) 등을꼽을수있는데, 이들은소의집단사육에의한환기불량및과다습도등이주요인이며분뇨에서발생되는암모니아가스, 소의호흡으로발생되는탄산가스등에의해호흡기점막을자극하여발병하거나증세를악화시키기때문에, 사육환경의개선과함께적절한예방백신을활용한사전예방이매우중요한것으로판단된다. 따라서본연구는한우호흡기질환의예방을위한최적의백신조건탐색연구로항생제, 호흡기질환주요바이러스 4종에대한백신, P.hemolitica 백신및구충제들을복합처리한뒤각각의중화항체생성도, 세균분리검사및실제임상진료자료분석등을실시하였다. 백신주사에의한바이러스간항체가는 BVDV가 3.86±0.202로가장높았고, IBRV가 1.89±0.202로가장낮았다 (P<0.05). 시험군들간중화항체가분석결과, 시험 4군이 4.13±0.226 으로대조군 (1.97±0.226) 과비교해가장높았으며 (P<0.05), 시험 1군, 시험 2군및시험 3군간에는유의적인차이가나타나지않았다. 또한약 5개월에걸친호흡기질병관련발생두수및치료내역을분석해본결과, 호흡기질환에대한예방이발병일기준으로보면시험 3군에서가장효과가있었으며, 치료횟수를기준으로보면시험 2군과시험 4군에서가장효과적인것으로나타났다. [ 방법 ] 시험축사내사육환경온도는상온환경 (20± 2 ) 과고온환경 (30± 2 ) 및초고온환경 (34± 2 ) 으로나누어배치하였으며 (5~8월), 각기경정맥 catheter를장착한거세한우송아지 4두를공시축으로하여조농비 3:7비율의사료를급여하고매일오전식전에채혈한후원심분리후혈장분리후정성분석시까지초저온냉동고에보관하였다. 본시험에서는각시험사육환경온도대별로대사판정검사 (metabolic profile test : MPT) 주요항목 (glucose, urea-n, FFA, TG) 농도와 insulin 농도를측정함과동시에 glucose부하시험을실시하여 glucose 처리계수 (K-value) 를측정하였다.[ 결과 ] 고온스트레스에서는혈중 urea-n, FFA, glucose 및 insulin농도는감소하였으며 TG농도는상승하였다. glucose 부하시험결과 K-value는상온, 고온, 초고온환경에서각기 5.2, 3.1, 5.5를나타냄으로서고온환경에서 glucose 처리능력이약 40% 저하되는것으로나타났으며초고온환경에서는저insulin혈증이지속되었다. 본연구결과는간장의 glucose 처리능력검사등을기초로하여사계절능력이우수한반추대동물을조기선발하는검사기술 (bovine early selection test : BEST) 을체계적으로개발하는데기여할것으로사료된다. PB10025 제주馬씨수말의견월악放牧地性行動조사연구권태준, 배재호, 김대수, 김소영, 김대경, 정혜나, 강민수제주대학교생명자원과학대학생명공학부, 제주대학교대학원동물생명공학전공제주대학교마필산언엽구센터본연구는재래가축중유일하게문화재로지정된제주재래마에방목습성을규명함으로서재래마의동물학적인특징을기록하며재래마의방목지에서의합리적인관리방안을제시하게되고아울러종모마의성행동에관한조사를통해효율적인번식생리에필요한기초자료를제공할목적으로수행하였다. 제주마씨수말의행동양식에대해성행동즉 flehmen, 교배시도, 교배행동등을조사하였고, 번식기인 3~8월까지제주특별자치도축산진흥원에서관리하고있는견월악방목지에서 A.M 7~ A.M 10, P.M 4~ P.M 7시까지선선한시간때를이용하여조사가진행되었다. 행동양식에따라행동유형별빈도와지속시간등을정밀측정하였다. Flehmen은발정이시작되는 3~5월에는빈도가 15~22회정도이루뤄지나, 7월 ~ 8월에는 10회이내로감소하는경향을보였다. 씨수말의교배시도행동의경우 3.4월에는평균 13회정도, 5월에는 16회정도로증가하며, 7~8월에는평균 6.5 회로감소하는양상을나타냈다. 교배 ( 삽입 ~ 하마 ) 의경우는하루에평균빈도는 3번정도이뤄지며, 교배지속시간은평균 17.51 초정도로이뤄졌다. 교배중음경왕복운동 ( 피스톤운동 ) 를 30회조사하여측정한결과평균 9회로나타났다. 이상의연구결과는장차제주마종모마의번식률향상및번식관리기술개선에활용할수있는기초자료가될것으로여겨진다. Thoroughbred 씨수말의성행동양식에대해성행동즉 flehmen, 교배전준비운동시간 ( 음경발기까지시간 ), 교배시도, 교배지속시간, 음경왕복운동 ( 피스톤 )) 등을조사하였고, 한국마사회제주육성마목장내종부소에서총 30회의성행동에대해유형별빈도와지속시간등을정밀측정기록하였다. Flehmen의평균빈도는 4~5회정도이뤄졌고, flehmen 지속시간은짧게는 5초내외에서길게는 30초까지다양한시간대분포를보여주었으며, 교배전준비운동시간 ( 음경발기까지시간 ) 은길게는 15분짧게는 1분정도로개체간에큰차이를나타났다. 교배시도행동의평균빈도는약 4.5회로나타났으며, 교배지속시간 ( 삽입 ~ 하마 ) 은 24.56초로개체간에큰차이가없었다. 음경의왕복운동 ( 피스톤 ) 평균빈도는약 9.2회로나타났다. 이상의연구결과는 Thoroughbred 씨수말의번식관리의제고를위한과학적인기초자료를제공할수있을것으로여겨진다. Key words : Thoroughbred, Behaviour, Flehmen, Stallion PB10027 한우에있어서성호르몬처리에의한난소내 apoptosis의가속화정학재, 김재희, 유동민, 최혜영, 남상일, 류일선, 최연호, 이명식, 전기준, 홍성구농촌진흥청국립축산과학원한우시험장연구배경 : 포유류의번식과정에있어서성스테로이드호르몬의처리는발정및배란을유기하는수단으로사용되어져왔다. 최근난소를비롯한난포에서의낭종발생빈도는점점증가하는추세를보이고있다. 따라서본연구의목적은인공수정및수정란이식전처리과정에서투여하는성스테로이드호르몬의난소에어떠한영향을미치는지에대하여조사해보았다. 실험방법 : 발정동기화및배란동기화유도를위하여처리하는 GnRH, PGF2, HCG CIDR 등을각각 1, 2, 3차에걸쳐처리하여난소를적출, 면역조직화학방법에의해난소내의 apoptosis( 자연세포사 ) 를관찰하였다. 결과및결론 : GnRH, PGF2를 1차로처리한난소에서는무처리군 ( 대조군 ) 에비하여자연세포사발생이크게차이는관찰되지않았으나, 2, 3차처리된난소에서는대조군에비하여약 3배이상의자연세포사가발생되는것이관찰되었다. 또한, HCG와 CIDR 처리에의한난소상의자연세포사관찰에서는원시난모세포주변의과립막세포에서의자연세포사의발생율이높았다. 특히, HCG 및 CIDR을처리된난소내의자연세포사는대조군에비하여약 5배이상으로발생되는것이관찰되었다. 이러한결과들이제시하고있는것은외인성호르몬들의투여는난소주기에있어서난포및그주변세포들을자극하여생리적활성에효과를미치고있으나, 이러한자극들은난포성장에관여하는세포들의자연세포사를유도할수있다는것을의미한다. PB10028 제주馬씨수말의종부소性行動에관한조사연구배재호 2, 권태준 2, 박준형 2, 김대수 1, 박소영 1, 김대경 1, 정혜나 1, 강민수 1,2,3 1 제주대학교생명자원과학대학생명공학부, 2 제주대학교대학원동물생명공학전공 3 제주대학교마필산업연구센터본연구는제주마씨수말의번식관리제고를위한목적으로종모마의성행동에관한조사연구를실시하였다. 제주특별자치도축산진흥원내에소재한종부소에서씨수말을대상으로총 30회의종부행동을조사하여유형별빈도와지속시간등을정밀측정기록하였다. 즉제주마씨수말의 flehmen, 교배전준비시간 ( 음경이발기되기까지의시간 ), 교배지속시간 ( 삽입 ~ 하마 ), 음경왕복운동 ( 음경의피스톤운동 ) 등을조사하였다. 씨수말의 flehmen은평균빈도가 5~6회정도이뤄졌고, 종부지속시간은짧게는 3초내외, 길게는 24초까지다양한시간대를나타냈다. 교배전준비시간 ( 음경발기되기까지의시간 ) 은 Thoroughbred의경우개체간에큰차이가나타난데반해제주마씨수말의경우에는평균 2분 30초정도로개체간에별차이를보이지않았다. 교배시도행동의평균빈도는약 2.7회로나타났으며, 교배지속시간 ( 삽입 ~ 하마 ) 은 18.20초로제주마씨수말이 Thoroughbred 종종모마보다약 6초정도짧은것으로나타났다. 이상의연구결과는제주마씨수말이종부소에서이뤄지는성행동에대한실태에대해과학적인기초자료를제공하므로서제주마씨수말의합리적인번식관리에기여할수있을것으로여겨진다. Key words : Jeju Horse, Behaviour, Flehmen, Stallion PB10029 제주馬仔馬를분만포유중인어미말의放牧地行動조사연구권태준 2, 배재호 2, 박준형 2, 김대수 1, 박소영 1, 김대경 1, 정혜나 1, 강민수 1,2,3 1 제주대학교생명자원과학대학생명공학부, 2 제주대학교대학원동물생명공학전공 3 제주대학교마필산업연구센터본조사연구는제주마의방목지행동양식을연구할뿐만아니라제주마를둘러싼환경과의관계속에서어떤방목지행동을발현하는가를현장에서연구하며, 아울러그행동발현의메카니즘을구명코자수행되었다. 제주마종빈마가자마분만시부터이유시까지방목지에서이뤄지는방목습성에대해조사기록한내용을분석하였다. 본조사연구는자마가분만하고나서 7개월령까지매월 2회씩방목지에서조사기록하고분석하였다. 방목지에서의관찰시간은 A.M 8:00~P.M 5:00 까지점심시간한시간을제외한총 8시간동안조사원 1명이어미말 1두씩담당하였다. 조사결과채식시간은자마가어렸을때는어미말이채식에소요되는시간이비교적짧았으나자마가성장함에따라채식시간이점차길어졌다. 자마의 1개월령시에는전체관찰시간의 50% 를채식에소비하였으나 6개월령시에는약 80% 의시간을채식에소비하였다. 다음으로는자마에게수유하는시간은 3 개월령까지는점차길어지다가 4~5개월령부터는다시짧아지는경향을나타냈으며, 수유횟수는 4개월령까지는관찰시간중 7~11회정도였고, 그이후에는조금씩단축되었다. 이상에서얻어진연구결과는제주마의방목지에서의번식기술제고및제주마의관리기술의향상에기초자료로활용될것으로여겨진다.
