METHODS: Sprgue-Dwley rts (n=4) were rndomly divided into 4 groups (ge=7 weeks, n=1 in eh group): ontrol group (CON), sopolmine-indued ontrol group (SCO-CON), sopolmine+low intensity exerise group (SCO-LEX), nd sopolmine+high intensity exerise group (SCO-HEX). The exerise groups were trined progressively on tredmill, five dys per week for eight weeks following tredmill running progrms: i) low intensity (1-4 weeks: 1 m/min for 3 min, 5-8 weeks: 12 m/min for 3 min), ii) high intensity (1-4 weeks: 15 m/min for 3 min, 5-8 weeks: 16 m/min for 3 min). After 8 weeks-exerise progrmming, detrining ws onduted for the sme period. RESULTS: After detrining for 8 weeks, oth exerise groups signifintly inresed Ah onentrtion ompred with SCO-CON. Exerise groups signifintly redued AChE tivity, β-myloid nd ApoE ε4 protein volumes ompred to the SCO-CON. Both exerise groups showed signifint derese of espe lteny ompred to the ontrol group. CONCLUSIONS: In spite of the effets of detrining, regulr exerise is still onsidered to hve positive influene on the hnges of memory impirment relted ftors. Key words: Detrining, Sopolmine, Cognitive funtion, Memory impirment, Cholinergi 서론 알츠하이머 (Alzheimer s disese, AD) 는기억력과일상적활동을수행하는데필요한인지기능의점진적인저하를나타내며, 치매 (Dementi) 의가장주요한유형의하나로알려져있다 [1]. 현재까지 AD에대한기전은명확히밝혀지고있진않지만대다수 의선행연구들은산화스트레스와 holinergi dmge를 AD의주요요인으로간주하고있으며 [2,3], 이와더불어 myloid plques, neurofirillry tngles는 AD에관여하는단백질로보고되고있다 [4]. 중추콜린성신경계의감퇴는인지기능과기억력저하를초래하고 Aetylholin (Ah) 은신경전달물질로대뇌피질과해마의발달에중요한역할을하는것으로이들의순환농도는 Aetylholinesterse (AhE) 에의해 Corresponding uthor: Jin-Hwn Yoon Tel Fx E-mil yoonjh@hnu.kr * 이논문은 213 년정부 ( 교육부 ) 의재원으로한국연구재단의지원을받아수행된연구임 (NRF-213S1A5A2A119566). 2 조절되는데, AhE는 Ah를빠르게가수분해시킴으로써인지능력을저하시키는것으로보고되고있다 [5]. 이와함께지난 1년동안가장널리수용되고있는패러다임으로써 myloid 이론은세포외 myloid plques가뇌세포사멸에관여하여결과적으로치매증상이나타나는것으로인식되고있으며이밖에도과인산화 tu 단백질로구성된 neurofirillry tngles는 AD와연관된또다른위험요인이다 [6-1]. Apolipoprotein E (ApoE) 는뇌주요아포지단백의하나로지질과 Aβ를수송하는역할을한다. ApoE는세가지 vrints (ApoE-ε2, ApoE-ε3, ApoE-ε4) 가존재하고 ε2는 AD에방어적인반면, ε4 대립형질은 AD 발생에가장큰위험인자로특히 AD 환자들에서 ε4가높은빈도를보이며증가하는것으로보고된바있다 [11,12]. 연구방법 1. 실험동물및식이방법실험동물은 7주령의 SD계수컷흰쥐 (ody weight: 187.8± 5.6 g) 4마리를통제그룹 (CON: n =1), sopolmine 투여그룹 (SCO-CON: n =1), sopolmine 투여 + 저강도트레드밀운동그룹 (SCO-LEX: n =1), sopolmine 투여 + 고강도트레드밀운동그룹 (SCO-HEX: n =1) 으로구분하였다. 식이는전실험기간동안고형사료 ( 단백질 22.5%, 지방 3.5%, 저섬유 7.5%, 회분 9.%, 칼슘.7%, 인.5%) 와물을충분히섭취하도록하였으며, 실험기간에동물실험윤리규정에따라동물의고통과불편을최소화하도록노력하였다. AD 개선과치료를위한가장효율적인방법중하나로약리적접근 은방출된 Ah 분해를억제하기위하여 AhE 억제제를투여함으로써 holinergi 신경전달을강화시키는것으로알려져있다 [13]. 이와더불어최근식이요법과운동및신체활동등의다양한라이프스타일중재는 AD 초기치료와예방을위한뇌형태및기능조절에긍정적결과를가져올수있는것으로보고되고있다. 이와관련하여 Mliszewsk- Cyn et l. [14] 은신체활동이 AD와관련된인지장애및병태를지연또는경감시켜전반적인뇌건강을향상시키는잠재력을지니고있으며, 다양한형태의운동은해마의신경발달을증가시키고기억력개선에긍정적영향을미치는것으로보고되고있다 [15,16]. 하지만선행연구의결과들은 AD의개선에대한운동의효과성에초점을두고있어 2. 연구절차 1) 운동처치운동강도는선행연구 [17,18] 를참고하여 sopolmine 투여 + 저강도트레드밀운동그룹 (SCO-LEX) 은 1-4주에는주당 5일간 1 m/min, 5-8 주에는주당 5일간, 12 m/min의속도로 1회당 3분운동을실시하였다. Sopolmine 투여고강도 + 트레드밀운동그룹 (SCO-HEX) 은 1-4주에는주당 5일간 15 m/min, 5-8주에는주당 5일간, 16 m/min의속도로운동을실시하였다. 8주간의운동이종료된후 8주동안동일한식이와실험실환경조건을유지한상태에서 detrining을실시하였다. 본연구에서운동강도에따른운동방법은 Tle 1과같다. 규칙적인운동의효과가어느정도지속되는지에대한연구는희소한 2) 기억력손상유도실험동물은 3일간의실험실환경의적응기간을거쳐노인성치매의특징인아세틸콜린계의효소를감소시키기위해무스카린수용체길항제 (musrini reeptor ntgonist) 로알려진 sopolmine (Sigm-Aldrih Co, USA) 1 mg/kg을.9% 식염수 1 ml에용해하여 7일동안 1일 1회복강내에주사하여기억력손상을유발하였다 [19]. 실정이다. 운동의지속적실천은다양한외부적환경에의해제약을받을수있으며, 운동중지 (detrining) 가 AD와관련된위험요인의변화에미치는영향을보고한선행연구는다소제한적이며미흡한실정이다. 따라서본연구에서는기억력장애흰쥐를대상으로 8주간의운동강도별트레드밀운동효과에대한후속연구로서뇌조직에서 detrining 에의한기억력장애요인들의변화를살펴보고자하였다. 3) 인지능력평가 인지능력사전훈련은 sopolmine 투여전에 1 회실시하였으며, 트 Tle 1. 6초동안수중미로내부를자유롭게이동한실험동물이 6초후에도 pltform에도달하지못할경우실험동물을자연스럽게 pltform으로유도한후 3초동안머물러있게함으로서시각적단서에익숙해지도록하였다. 최종테스트는 8주후실험동물을희생시키기전에 6초간자유수영검사를실시하여 pltform에도달하는시간을영상추적프로그램 (Pnl, S.L.

3 레드밀운동 8주후에운동이끝나고 24시간이지난다음실시하였다. 모리스수중미로는원형풀 ( 지름 12 m, 높이 6 m) 이며, 여기에수심 35 m로물 (25 C ± 2 C) 을채웠다. 그후수중미로를 4개의동일한 4 분원으로나누어구분하고, 남서 (SW) 사분원의중심부에원형 pltform ( 지름 1 m, 높이 3 m) 을설치한후실험동물이육안으로 pltform을볼수없도록오징어먹물가루로물을불투명하게하였다. pltform의표면은거칠게처리해실험동물이쉽게기어오를수있도록하였다 (Fig. 1). 6초동안수중미로내부를자유롭게이동한실험동물이 6초후에도 pltform에도달하지못할경우실험동물을자연스럽게 pltform으로유도한후 3초동안머물러있게함으로서시각적단서에익숙해지도록하였다. 최종테스트는 8주후실험동물을희생시키기전에 6초간자유수영검사를실시하여 pltform에도달하는시간을영상추적프로그램 (Pnl, S.L.U, Spin) 을이용하여인지능력을평가하였다 [2,21]. 4) 조직시료의준비모든동물은실험이종료된 24시간경과후 Zoletil 5 R (1 mg/kg, i.p.; Vi Lortories, Crros, Frne) 을이용하여마취한후 Mthis et l. [22] 의방법을참고하여대뇌피질과해마조직을추출하였다. 조직을추출한후증류수로혈액을완전히제거하고 4 C.25 M surose 를포함하는 5 mm potssium phosphte uffer (ph 6.5) 용액을각시료조직의 3배 (w/v) 가한후 utohomogenizer (GTR-1, Tokyo Rikk- Miki Co., Ltd., Tohyo, Jpn) 로파쇄하여 25, rpm에서 3분간원심분리하고상층액을취하여시료로사용하였다. 5) Ah 함량측정대뇌피질의 Ah은 Lee et l. [23] 의방법으로측정하였다. 대뇌피질부위를적출한균질액 (12 mm sodium phosphte uffer ph mm NCl) 5 ml를취하여 1% hydroxylmine 5 ml를첨가하여혼합한후 HCl을이용하여 ph를 1.2 ±.2로조절한후 Fel3 (1% in.1 N HCl) 을 5 ml를첨가하여분광광도계 (UV-16A, Shimdzu Co., Kyoto, Jpn) 를이용하여 54 nm에서흡광도를측정하였다. 6) AhE 활성측정대뇌피질의 AChE 활성의측정은선행연구 [24,25] 방법을참고하여측정하였다. 각 miroplte well에.1 M Tris uffer, ph 8. (Trizm HCl+ Trizm se) 을 3 μl,.1 M dithionitroenzoi id (DTNB; Sigm Aldrih Corportion, St. Louis, MO, USA) 2 μl, enzyme suspension 1 μl을연속적으로첨가하고흡광도측정직전에기질인.1 M etylthioholine hlo ride (Sigm, Aldrih Corportion ) 1 μl을첨가한후, 96 well miroplte reder (ELISA reder) 를이용하여 45 nm에서흡광도변화를 5분동안관찰하여 AChE의활성 (unit/min/mg protein) 을측정하였다. 7) Western lotting을이용한 Aβ, polipoprotein 분석해마의 Aβ, polipoprotein 발현을분석하기위하여 Gel상의단백질을 15 mm Tris-glyine uffer (19 mm glyine, 5 mm Tris-se,.5% SDS, 2% methnol) 를사용하여 1 V에서 1시간동안 PVDF memrne (Pll Corportion, USA) 에 trnsfer 하였다. 그후 memrne을 TBS 시약으로 1분씩 3차례 wshing하고, loking uffer (TBS 34 ml, BSA 1.5 g) 에서 1시간동안 shking시킨후, primry ntiody (Aβ: snttuz, ApoE: sntruz) 를 loking uffer에 1:1,의비율로희석하여 old room에서 overnight하였다. Memrne을씻어내고 seondry ntiody (nti-rit IgR) 를 1:2,의비율로희석하여상온에서 1시간동안처리하고 wshing 후, 마지막으로 memrne에부착된단백질을 western lotting detetion system을이용하여측정하였으며, density Fig. 1. 4 meter (Shrp jx-33) 를이용하여스캔한후, 단백질의정량분석은이미지분석프로그램 (Imge Mster ver. 3., Phrmi Biotehnology, Pistwy., NJ, USA) 을이용하여산출하였다. 3. 자료처리방법통계적자료분석을위해서 SPSS version 2을이용하여각집단의평균및표준편차를산출하였다. 운동중지에따른뇌조직 Ah, AhE, 인지기능, Aβ, polipoprotein 발현차이를알아보기위하여일원변량분석 (One-wy etween mesures ANOVA) 실시하였다. 일원변량분석후통계적차이가있을경우, 사후검증으로 Dunn을이용하여그차이를알아보았다. 통계적유의수준 (α) 은.5로설정하였다. 연구결과 1. 8주간의운동중지후대뇌피질의 Ah 농도 8주간의운동중지후대뇌피질의 Ah 농도변화에대해분석한결과모든그룹간유의한차이가나타났으며 (p<.5), 사후검증결과 Ah 농도는운동그룹이 sopolmine 투여통제그룹에비하여통계적으로유의하게높게나타났으나 (p<.5) (Fig. 2), 운동강도별유의한차이는없었다. 2. 8주간운동중지후대뇌피질의 AhE 활성 8주간의운동중지후대뇌피질의 AhE 활성을비교한결과, 통제그룹 (CON: n =1), Sopolmine 투여통제그룹 (SCO-CON: n =1), Sopolmine 투여 + 저강도트레드밀운동그룹 (SCO-LEX: n =1), Sopolmine 투여고강도 + 트레드밀운동그룹 (SCO-HEX: n =1) 간유의한차이가나타났으며 (p <.5) 사후검증결과, AhE 활성은트레드밀운동그룹이 Sopolmine 투여통제그룹보다유의하게감소되었다 (p <.5) (Fig. 3). 운동강도에따른집단간차이는고강도운동그룹이저강도운동그룹에비해증가하는경향을나타내었지만유의한차이는나타나지않았다 주간운동중지후도피잠재기 8 주간의운동중지후도피잠재기를비교한결과 Fig. 4 와같이통제 그룹 (CON: n =1), Sopolmine 투여통제그룹 (SCO-CON: n =1), Sopolmine 투여 + 저강도트레드밀운동그룹 (SCO-LEX: n =1), Sopolmine 투여 + 고강도트레드밀운동그룹 (SCO-HEX: n =1) 간유의한 차이가나타났으며 (p <.5) 사후검증결과, 도피잠재기는트레드밀운동 집단이 Sopolmine 투여통제집단보다유의하게감소하였다 (p <.5) (Fig. 4) 주간운동중지후해마 β-myloid 발현 8 주간운동중지후해마 β-myloid 발현을비교한결과 Fig. 5 와같 이통제그룹 (CON: n =1), Sopolmine 투여통제그룹 (SCO+CON: n =1), Sopolmine 투여 + 저강도트레드밀운동그룹 (SCO-LEX: n =1), Sopolmine 투여 + 고강도트레드밀운동그룹 (SCO-HEX: n =1) 간유의한차이가나타났으며 (p <.5). 사후검증결과, β-myloid 발현은트레드밀운동집단이 Sopolmine 투여통제집단보다유의하 AhE tivity Fig. 3. Comprison of β-myloid expression on detrining in sopolmineindued memory impirment rts.,, Different letters in the sme olumn represent signifintly different mong the groups t p<.5. 기억력감퇴및인지기능의저하는신경퇴행성질환을앓고있는환 자에게서흔히발생할수있는증상으로중추 holinergi system 은무 스카린및니코틴수용체를통해학습과기억력에밀접한관련이있 고, AhE 활성증가에따른 Ah 감소및독성 Aβ, ApoE ε4 발현증가는 AD 와같은신경퇴행성질환을가속화시킬수있다. 본연구는후속 연구로써규칙적인강도별트레드밀운동후, 8 주간 detrining 이 sopolmine 으로유도된기억력장애쥐뇌조직에서 Ah, AhE, β-myloid, ApoE ε4 발현및인지기능에미치는영향을관찰함으로써운동 의트레이닝지속효과에대해규명하고자하였다. Sopolmine 은비선 택적무스카린수용체의길항제로중추콜린성신경활성을억제시킬 뿐아니라, 노화동안발생되는퇴행성신경, 신경정신병병리및외상 성 (trumti) 뇌손상을가정한기억장애의약리학적모델에자주사, d ApoE ε4 expression (volume) ApoE ε4 β-tin 25, 2, 15, 1, 5, 용되고있다 [26-28]. Fig. 6. Comprison of ApoE ε4 expression on detrining in sopolmineindued memory impirment rts.,, Different letters in the sme olumn represent signifintly different mong the groups t p<.5. 규칙적인운동이 AD 와관련된인자들에긍정적개선효과가있음 이밝혀지고있는데, 이와관련한선행연구에서, sopolmine 의주입 은해마 CA1 영역의 AChE 발현을증가시켜 ACh 의분해를가속화시 킬수있고, 이에대한방어적형태로써트레드밀운동은 sopolmine 에의해유도된 AChE 발현을억제시킬수있는것으로보고되었다 [29]. Wng et l. [3] 은 C57l/6 mie 를대상으로한연구에서 12 일동 안의자발적달리기운동이 Aβ25-35 로발생된기억손실을감소시키 는것으로보고하였으며이와유사한연구결과로써, 우리의이전연구 에서도 8 주간의저강도및고강도의트레드밀운동이기억력장애흰 쥐의대뇌피질과해마에서 Ah, AhE, β-amyloid, ApoE ε4 및 Morris wter mze test 의수행에긍정적변화를확인할수있었다 [31]. 이를통 해규칙적인운동이단기및공간학습및인지기능과같은뇌기능을 개선시키고, 신경발생및신경영양인자의발현에긍정적영향을미칠 수있음을유추해볼수있다. 반면, 기억력과관련된운동의긍정적효 과와달리운동중지후이전의운동효과가어느정도지속되는지에 대한연구는제한적이며잘알려지지않았다. 본연구에서 8 주동안트 레드밀운동후동일한기간에따른운동중지효과에대해분석한결 과, 해마의 ACh 농도차이는운동강도와는독립적으로운동집단 (SCO- LEX, SCO-HEX) 이 sopolmine 투여집단 (SCO-CON) 보다유의한차 이를보이며높게나타났으며, ACh 농도에직접적영향을미치는 AChE 활성을살펴본결과, 운동강도별유의한차이는나타나지않았으나, 운동집단이 sopolmine 투여집단에비하여유의한차이를보이며 낮은활성을나타내었다. 또한, 인지기능과기억력에중추역할을하는, d 1 Chun-Seop Jung, et l. 6 해마의 Aβ 및 ApoE ε4의발현에대한결과, 도피잠재기는모든집단간유의한차이가나타났으며운동집단이 sopolmine 투여집단에비하여유의한차이를보이며감소하였고, Aβ 및 ApoE ε4 발현에서도두요인모두운동집단이 sopolmine 투여집단과비교하여유의한차이를보이며감소됨을확인할수있었다. 우리의결과는규칙적인운동의효과에대해보고한이전의결과와는직접적인비교는어려우나, 8주간의운동중지에도불구하고 ACh, AChE 활성및 Aβ, ApoE ε4 발현이통제집단과비교하여비교적긍정적지속효과를확인할수있었고, 이는일반적으로저항성운동을통한근비대현상이운동중지와동시에이전의상태로급격히돌아가지않는것과유사한것으로판단된다. 아직까지 AD rt 모델을대상으로운동중지가콜린성신경계와해마 Aβ 및 ApoE ε4 발현에미치는영향은보고되지않고있어이에대한다각적인논의는제한될수있으나, 우리의결과는운동이장기증강 (long-term potentition, LTP) 강화와 BDNF mrna 발현의상위조절에의해 AD 모델 APP/PS1 trnsgeni mouse의학습과기억능력을개선시킬수있음을보고한연구 [32] 와, 운동이뇌기능과신경계에긍정적영향을가져올수있으며특히, 기억의주요영역인해마에서운동의효과는 Wnt 신호전달체계와관련되고, 시냅스가소성과장기증강 (longterm potentition, LTP) 을개선시킬수있다고 [33,34] 보고한선행연구결과와밀접한관련이있는것으로생각된다. 또한, Kim et l. [35] 은 STZ로유도된당뇨쥐에서 12주간의트레드밀운동은 AD와관련된기억손실을완화시키는연구결과를보고하였는데, 이는 AD와같은신경퇴행성질환에서 Wnt 신호전달체계활성을통한신경생성의증가와관련이있음을제안하였다. 이러한관점에서지속적인운동은운동중지기간에상관없이 Wnt 신호전달체계의활성과 LTP 강화에긍정적영향을미칠수있으며이는 Aβ 독성에대비한신경보호역할 [36] 을하고, 해마의기억력과인지기능개선에기여할수있을것으로생각된다. 이와같이과거운동경험은 detrining 기간에도불구하고긍정적효과가보존되었지만, 본연구에서는기억력감퇴개선과관련된적정한운동강도를명확히제시하지는못하였다. 이는아마도 detrining 기간동안실험동물의활동성또는환경적요인등과같은외적요인이작용한것으로판단된다. 또한, 아직까지 detrining과관련된선행연구가다소제한적임으로이를뒷받침할과학적근거를제시하지못하는것은본연구가지니는제한점이라할수있다. 따라서추후이와관련된다양한접근을통한연구가꾸준히실시되어야할것으로생각된다. 결론 이상의결과를종합해볼때, 이전의규칙적인운동실천에따른운동경험및효과는 detrining 영향에도불구하고기억력감퇴와관련 된인자들의변화에있어여전히긍정적효과를가져올수있는것으로생각된다.
