The majority of them were designated as persistent organic pollutants (POPs) by the international community at the Stockholm Convention on Persistent Organic Pollutant. Extraction and clean-up method were developed using the QuEChERS method for residual organochlorine pesticides (OCPs) in soil and oriental melon, green pepper and lettuce. Recovery of OCPs in greenhouse soil and oriental melon, green pepper, and lettuce ranged from %. Limit of detection (LOD) of OCPs in soil and 3 crops were and μg/kg. The residues of OCPs in oriental melon, green pepper and lettuce greenhouse soil were analyzed by the developed method, and dieldrin, β- endosulfan and endosulfan sulfate were detected at , and μg/kg, respectively. The detection frequency of 3 compounds in soils were 52 (29.7%), 34 (19.4%) and 57 (32.6%) among 175 samples, respectively. However, these compounds were not detected in all crop samples. The residue level in 3 crops were lower than 1/58.8 of maximum residue level of them. These results showed that the OCPs residue in oriental melon, green pepper, and lettuce greenhouse soil were not as high as crop safety threatening. Key words Greenhouse, green pepper, lettuce, oriental melon, residual organochlorine pesticides 서 론 농림축산식품부 (MAFRA, 2015) 에따르면우리나라의채소류생산을위한노지재배면적은감소하는반면시설재배면적이꾸준히증가하여2014년에는 2013년에비해시설재배면적이6% 증가한 63,815 ha에서 2,714,519 톤의채소류가생산되었고, 이중참외, 풋고추및상추의재배면적과생산량은각각 5,358, 4,619 및 3,241 ha와 155,397, 185,915 및 89,673 톤으로전체시설재배지의 20.7% 와채소류생산량의 15.9% 를차지하고있다. *Corresponding author bjpark@korea.kr These authors contributed equally to this work. 참외 (Cucumis melo var. makuwa) 는박과에속하는 1년생덩굴성포복식물로우리나라의여름철대표적인내수지향적생식과일 (Kim et al., 2011) 로서, 시설재배기술의발달과소비패턴의변화에따라작형이다양하게분화되어이른봄부터연중생산되고있다. 고추 (Capsicum annuum L.) 는가지과에속하는 1년초단일작목으로일년연중재배되고, 주로음식의맛을내는향신료로사용되었으나최근에는웰빙에대한관심이높아지면서암유전자발현조절작용, 살균작용, 지방생성억제효과등다양하게연구되고있다 (Son et al., 2012; Kim et al., 2016). 상추 (Lactuca sativa) 는국화과에속하는여러해살이엽채류로우리나라에서는봄재배, 가을재배, 겨울재배및고랭지재배가이루어지고있으며, 전체쌈채소중재배면적과생산량에있어가장많은 319

2 320 임성진 오영탁 양지연 노진호 최근형 류송희 문병철 박병준 점유율을나타내고있다 (RDA, 2007). 잔류성유기오염물질 (persistent organic pollutants, POPs) 은독성, 생물농축성, 잔류성및장거리이동성을가지는물질로이들의감소를목적으로제조, 사용및수 출입을제한또는금지하는스톡홀름협약이 2001년 5월과 2004년 5 월에채택 발효되었고, 우리나라도 2007년에이를비준하여 잔류성유기오염물질관리법 으로 POPs 물질을관리하고있다 (Shin et al., 2010; Lim et al., 2016a). POPs 물질중염소를함유하고, 농약으로사용된물질인잔류성유기염소계농약류 (residual organochlorine pesticides, ROCPs) 는 aldrin, endrin, dieldrin, toxaphene, chlordane, heptachlor, mirex, hexachlorobenzene (HCB), dichloro diphenyl trichloro ethane (DDT), α-hexachlorocyclohexane (HCH), β-hch, pentachlorobenzene (PeCB) 및 endosulfan 13종으로이들중 chlordane과 mirex를제외한 11종이우리나라에서살충제로등록되어사용된바있다 (Lim et al., 2016a and 2016b). 식품의약품안전처 (Korea Food Code, 2016) 는농산물중잔류성유기염소계농약의단일성분또는대사체가독성을가지는경우이들의합으로서잔류허용기준을설정하여관리하고있다. Aldrin과 dieldrin의잔류허용기준은이들의합으로서 , endirn (endrin과 δ- ketoendrin의합 ) 은 , endosulfan (α-endosulfan, β- endosuflan 및 endosulfan sulfate의합 ) 은 , heptachlor (heptachlor와 heptachlor epoxide의합 ) 는 mg/kg 으로설정하여농작물별로관리하고있다. 잔류성유기염소계농약류중 dieldrin, endrin, DDT 및 heptachlor는 1970년대, endosulfan은 2011년에우리나라에서제조 사용이금지되었음에도불구하고농업환경및농작물에서꾸준히검출되고있다 (Park and Ma, 1982; Suh et al., 1984; Park et al., 2011; 1984; Lim et al., 2016). 또한농업환경중이들에대한모니터링조사에 gas chromatography (GC)-electron capture detector (ECD) 가많이사용되고있으므로 (Chen et al., 2005; Gonzalez et al., 2005; Mikes et al., 2009; Lim et al., 2016a, 2016b) 본연구에서는꾸준하게재배면적이증가하고있는참외, 풋고추및상추시설재배지토양과작물중잔류성유기염소계농약류에대한 GC-ECD를활용한동시분석법을확립하고, 참외시설재배지 61지점, 풋고추시설재배지 62지점및상추시설재배지 52지점의토양및작물중잔류성유기염소계농약류의잔류양상을파악하고자하였다. 재료및방법 시약및표준품본연구의시설재배지토양및농작물중잔류성유기염소계농약류의잔류량을조사하기위한 aldrin (99.0%), 2,4- dichloro diphenyl dichloro ethane (DDD, 99.5%), 4,4-DDD (99%), 2,4-dichloro diphenyl dichloro ethylene (DDE, 97.0%), 4,4-DDE (98.5%), 2,4-dichloro diphenyl trichloro ethane (DDT, 98.0%), 4,4-DDT (98.0%), dieldrin 98.3%), endrin (99.0%), α-endosulfan (97.0%), β-endosulfan (99.5%), endosulfan sulfate (98.5%), heptachlor (98.5%), heptachlor epoxide (98.5%), hexachlorobenzene (99.5%), α-hexachlorocyclohexane (α-hch, 98%), β-hch (97.7%), γ-hch (99.0%) 및 δ-hch (98.5%) 표준품은 Dr. Ehrenstorfer GmbH (Ausburg, Germany) 사에서구입하여사용하였다. 시설재배지토양및농작물시료중잔류성유기염소계농약류분석을위한전처리과정에사용된 activated carbon, magnesium sulfate, sodium chloride 및 sodium citrate는 Sigma-Aldrich (Saint Louis, USA), Q-sep quick, easy, cheap, effective, rugged, and safe (QuEChERS) dispersive solid phase extraction (dspe) tube (150 mg magnesium sulfate, 50 mg primary secondary amine, 50 mg C 18, 2 ml) 는 Restek (Pennsylvania, USA) 사에서구입하였다. 시료채취및시료조제본연구에서는참외, 풋고추및상추주산지의시설재배지토양및작물을조사대상으로하였다. 참외는경상남도창원군, 경상북도성주군및경기도여주시시설재배지에서토양및작물을각각 22, 24 및 15점 ( 총 61점 ) 을채취하였고, 풋고추는전라북도임실군, 전라남도나주시및경상남도밀양시에서토양및작물을각각 11, 31 및 20점 ( 총 62점 ) 을채취하였고, 상추는경기도고양시와경남밀양시에서토양및작물을각각 20및 32점 ( 총 52점 ) 을채취하였다. 참외, 풋고추및상추시설재배지토양및작물시료는 Lim 등 (2016a, 2016b) 의방법에따라채취하였다. 토양은토양시료채취기를이용하여 10개지점에서 10 cm 깊이로각각 g 채취하여혼화하고, 이로부터약 500 g을취하여음건한다음 2mm 토양체를통과시켜분석시료로사용하였다. 작물은 10개지점에서각각 g 채취한다음약 1kg 을믹서기로분쇄 혼화하여분석시료로사용하였다. 분석법의유효성검증조사대상시설재배지토양및작물시료중 aldirin등 19종잔류성유기염소계농약의잔류량분석을위한분석법의유효성은검량선의직선성, 회수율시험, 검출한계 (limits of detection, LOD) 및상대표준편차 (relative standard deviation, RSD) 로검증하였다 (Lim et al, 2016a, 2016b). 검량선의직선성은분석의효율성을높이기위하여 Group I (α-hch, β-hch, γ-hch, δ-hch, α-endosulfan, β-endosulfan, endosulfan sulfate, endrin, 2,4-DDD, 2,4-DDT) 과 Group II (aldrin, dieldrin, HCB, heptachlor, heptachlor epoxide, 2,4-

( o C) Hold time (min) Initial Ramp Ramp Ramp DDE, 4,4-DDE, 4,4-DDD, 4,4-DDT) 로나누어 μg/l 수준에서확인하였고, 검출한계 (LOD) 는 S/N (signal to noise ratio) 이 3.3이되는농도로하여아래의식으로부터산출하였다. 최소검출량 (ng) 최종재용해량 (ml) LOD (mg/kg) = 기기주입량 (µg) 시료량 (g) 잔류성유기염소계농약의회수율시험은 Group I과 Group II 혼합표준용액을토양과작물에각각 4와 20, 10과 20 μg/kg 수준으로처리한다음 Fig. 1의시험방법에따라 3 반복으로실시하였고, 잔류성염소계농약 19종의회수율은각검량선에해당성분의피크면적을대입하여산출한농도와첨가농도의비로부터구하였다. 또한분석기기의실험실내정밀성시험은 5회반복하여실시하였고그결과를 RSD (%) 로나타냈다. 잔류성유기염소계농약잔류량분석참외, 풋고추및상추시설재배지토양및작물중잔류성유기염소계농약의잔류량분석은 QuEChERS 법을다소변형한 Lim et al. (2016a, 2016b) 의방법에따라수행하였다 (Fig. 1). 시설재배지토양 50 g을삼각플라스크에칭량하고, magnesium sulfate 20 g, sodium chloride 5 g 및 sodium citrate 5 g을첨가한다음 acetone 150 (100+50) ml로 2시간동안 2회진탕추출하였다. Acetone추출액을감압여과하고, 40 o C에서 rotary evaporator (IKA RV 10 Digital, Staufen, Germany) 를이용하여감압농축한다음 acetonitrile 4 ml로재용해하고, 이중 1.5 ml를취하여 QuEChERS dspe tube에넣고 5분간진탕하였다. 이를 3,000 rpm 속도에서 10분간원심분리 (Combi 514R, Hanil, Incheon, Korea) 한다음상등액을 0.22 μm syringe filter로여과하고 gas chromatography (GC, Agilent Technologies, Santa Clara, USA) - micro ECD (μecd) 를이용하여 Table 1의기기조건에서분석하였다.

4 322 임성진 오영탁 양지연 노진호 최근형 류송희 문병철 박병준 잔류성유기염소계농약은 POPs로지정되어우리나라에서도사용및제조가금지되어사용되지않는약제이므로참외, 풋고추및상추중이들의잔류량은시설재배지토양을분석한후검출된농약만을분석대상으로하였고, 참외, 풋고추및상추 10 g을칭량하여삼각플라스크에넣고, sodium chloride 1 g, sodium citrate 1 g 및 activated carbon 1g을첨가한다음 acetone 150 (100+50) ml로 2시간동안 2회진탕추출하였다. Acetone 추출액을감압여과하고, 40 o C에서 rotary evaporator를이용하여감압농축한다음 acetonitrile 4 ml로재용해하고, 이중 1.5 ml를취하여 QuEChERS dspe tube에넣고 5분간진탕하였다. 이를 3,000 rpm 속도에서 10분간원심분리한다음상등액을 0.22 μm syringe filter로여과하고 GC-μECD를이용하여 Table 1의기기조건에서분석하였다. 또한 GC 분석에서잔류성유기염소계농약이검출된시설재배지토양및작물시료에대해서는 GC-mass spectrometry (MS, Agilent Technologies, Santa Clara, USA) 를사용하여재확인하였다. 결과및고찰 분석법의유효성토양및작물중잔류성유기염소계농약류잔류분석법의적합성을검증하기위하여 Group I (α-hch 등 10종 ) 과 Group II (Aldrin 등 9종 ) 의크로마토그램, 회수율및상대표준편차를 Fig. 2, Table 2 및 Table 3에나타냈다. 잔류성유기염소계농약류 19종은 Table 1의기기분석조건에서 분의머무름시간을나타냈고 (Fig. 2), μg/l 범위에서작성한검량선의직선성은결정계수 (R 2 ) 가 으로양호하였다. 국내에서의회수율시험기준은분석성분의처리수준을검출한계의 5-10배 ( 저농도 ) 및 배 ( 고농도 ) 농도의 2 개수준에서수행할것을요구하고있다. 잔류성유기염소계 Fig. 2. Recovery of residual organochlorine pesticides in crops Crops Pesticides Recovery (%) LOD RSD (%) 10 μg/kg 20 μg/kg (μg/kg) 10 μg/kg 20 μg/kg Oriental melon Dieldrin 94.2 ± ± Dieldirn 84.2 ± ± Green pepper β-endosulfan 92.4 ± ± Endosulfan sulfate 83.2 ± ± Dieldirn 92.6 ± ± Lettuce β-endosulfan 77.7 ± ± Endosulfan sulfate 81.5 ± ± 농약은국내에서사용이금지되어있고, 토양으로부터농약의작물로의흡수 이행율또한매우낮으므로작물에서의회수율시험을저농도범위의 2개수준에서수행하였다. 잔류성유기염소계농약 19종의토양중회수율, RSD 및 LOD 는각각 %, % 및 μg/kg 범위이었고 (Table 2), 3 작물 ( 참외, 풋고추, 상추 ) 중회수율, RSD 및 LOD는각각 %, % 및 μg/kg 범위로나타났다 (Table 3). 이상의토양및 3 작물에서의회수율및 RSD는회수율 %, RSD 10% 이하로잔류분석법기준에적합하였다. 시설재배지토양중잔류성유기염소계농약의잔류량참외, 풋고추및상추주산지시설재배지토양의잔류성유기염소계농약의잔류수준, 검출빈도및분포도를 Table 4에나타냈다. GC (μecd) 크로마토그램상에서검출된 dieldrin, β-endosulfan 및 endosulfan sulfate 성분에대해서는 GC-MS 질량스펙트럼의대표적이온분자량 (m/z) dieldrin 79, 81, 82, β-endosulfan 195, 237, 241 및 endosulfan sulfate 272, 274, 387을확인하여동일성분임을확인하였다 (Fig. 3). 참외주산지 3개시군의시설재배지토양 (61지점) 에서는조사대상잔류성유기염소계농약 19종중 dieldrin만검출되

Residues of residual organochlorine pesticides in greenhouse soil Greenhouse Pesticides Detection range (μg/kg) Detection number (Ratio of detection, %) Oriental melon Dieldrin (44.2) Dieldrin (17.7) Green pepper β-endosulfan (17.7) Endosulfan sulfate (51.6) Dieldrin (26.9) Lettuce β-endosulfan (44.2) Endosulfan sulfate (48.1) 었으며, 검출범위와검출빈도는각각 μg/kg과 44.2% 이었다. 풋고추주산지 3개시군의시설재배지토양 (62지점) 에서는 dieldrin, β-endosulfan 및 endosulfan sulfate 3성분의잔류성유기염소계농약이검출되었으며, 검출범위는각각 , 및 μg/kg, 검출빈도는각각 17.7, 17.7 및 51.6% 이었다. 상추주산지 2개시군의시설재배지토양 (52지점) 에서도 dieldrin, β-endosulfan 및 endosulfan sulfate 3성분의잔류성유기염소계농약이검출되었으며, 검출범위는각각 , 및 μg/kg, 검출빈도는각각 26.9, 44.2 및 48.1% 이었다. 이상의결과는최근 Lim et al. (2016a, 2016b) 의딸기및들깨시설재배지토양에서검출되는성분및검출수준과유사하였다. 또한일부토양시료에서 aldrin, DDT, dieldrin, endrin 등의사용금지시기가유사함에도불구하고 dieldrin 만이검출되었다. Lim et al. (2016b) 은이원인으로반감기가 aldrin 365일에비해 diedrin 1,000일로더길고, DDT (2,000일 ) 와 endrin (4,300일) 에비해반감기는짧지만토양에노출된 aldrin이 dieldrin으로분해되어잔류되므로사용된aldrin양만큼토양중 dieldrin의잔류량이증가되고, 시험대상토양에상대적으로 aldrin 또는 dieldrin의사용량이많았기때문인것으로판단하고있다. 참외, 고추및상추중잔류성유기염소계농약의잔류량우리나라식품의약품안전처는농약의잔류허용기준을작 물별로설정 관리하고있으며, 이중본연구에서검출된 dieldrin과 endosulfan류에대해서는잔류허용기준을각각 과 mg/kg으로정하고있다 (Korea Food Code, 2016). 또한, 국립농산물품질관리원은 농산물등안전관리결과보고서 (2015) 에서 2014년조사대상농산물 47827건중 455회 ( 부적합 27회 ) 에걸쳐 endosulfan이검출됨을보고하였다. Endosulfan은 POPs로지정 (2011) 되어사용되지않음에도불구하고농산물에서꾸준히검출되는것은우리나라농경지에 endosulfan이잔류하고있고, 작물에흡수 이행됨을나타낸다. 본연구에서의시설재배참외, 풋고추및상추중잔류성유기염소계농약의잔류수준은 3개작물의시설재배지토양에서 dieldrin, β-endosulfan 및 endosulfan sulfate가각각최대 72.5, 78.7 및 μg/kg 수준으로검출되었음에도불구하고모든참외, 풋고추및상추시료에서식품의약품안전처의잔류허용기준보다 1/58.8 보다낮은수준인검출한계미만으로검출되어 Lim et al. (2016a, 2016b) 의시설재배딸기및들깨에서검출한계미만으로검출된결과와동일하였다. 이상의결과는우리나라참외, 풋고추및상추시설재배지조사대상토양 175개시료중 dieldrin (52개시료, 29.7%), β-endosulfan (34개시료, 19.4%) 및 endosulfan sulfate (57개시료, 32.6%) 3성분의잔류성유기염소계농약이 μg/kg 수준으로잔류 (Table 4) 하고있으나작물에서는이들성분이검출되지않아식품의약품안전처의잔류허용기준보다현저하게낮은

Agric. 3: 국내에사용되어금지된유기염소계농약류의다중분석법과모니터링 임성진 오영탁 양지연 노진호 최근형 류송희 문병철 박병준 * 농촌진흥청국립농업과학원화학물질안전과 요약본연구에서는참외, 풋고추및상추시설재배지토양및작물중스톡홀름협약에서대부분잔류성유기오염물질 (POPs) 로지정된잔류성유기염소계농약의잔류량을조사하였다. 토양및작물중잔류성유기염소계농약분석을위한추출및정제는 QuEChERS 방법을다소개선한방법으로수행되었다. 시설재배지토양, 참외, 풋고추및상추중잔류성유기염소계농약의회수율은각각 , , 및 % 이었으며, 토양과 3 개작물에서의검출한계는각각 과 μg/kg 수준이었고, 상대표준편차는토양및 3 개작물에서각각 , , 및 % 로 20% 미만으로적합한범위이었다. 참외, 풋고추및상추시설재배지토양에서 dieldirn, β-endosulfan 및 endosulfan sulfate 3 성분이각각 , 및 μg/kg 수준으로검출되었으나참외, 풋고추및참외의모든시료에서는검출되지않았다. 이러한결과는참외, 풋고추및상추시설재배지토양이농산물안전성을위협할정도로잔류성유기염소계농약으로오염되지않았음을나타냈다. 색인어 시설하우스, 잔류성유기염소계농약, 참외, 풋고추, 상추
