Therefore, it is not permitted for food materials in Korea. Traditionally, the classification of cyanobacteria has been based on morphological characters such as trichome width, cell size, division planes, shape, and the presence of character such as gas vacuole. But, some diagnostic features, such as gas vacuole or akinetes, can show variation with different environmental or growth conditions and even be lost during cultivation. Therefore, we developed detection method for functional foods containing Aph. flos-aquae by PCR. To design the primer, 16S rrna region of Aph. flos-aquae, Spirulina laxissima, and Spirulina spp. registered in the GeneBank ( have been used and for comparative analysis, BioEdit ver was used. As a result, we was design AFA-F1/AFA- R1 (363 bp) primer for the differentiation Aph. flos-aquae from chlorella, spirulina, green tea, and spinach. Also, it could be distinguished chlorella and spirulina products those are made to contain 1% Aph. flos-aquae. Key words: Aphanizomenon flos-aquae, 16S rrna, PCR 서론 시아노박테리아 (Cyanobacteria 문 ) 에속하는조류는전세계적으로수천년동안식품원료및가벼운질병을치료하기위하여사용되어졌다 1.2). 극동지역의해안가지방에서는기원전 6,000년전부터식품원료로해초등의대형조류 (macroalgae) 를사용하였으며, 서기 900년부터의약품으로써사용하였다는근거가있다 3). 대부분의미세조류생산업체는아시아태평양지역에밀집되어있고매년약 500톤의생산능력을보유하고있으며생산되는미세조류 *Correspondence to: Yong-Chjun Park, New Hazardous Substance Team, Food Safety Evaluation Department, National Institute of Food & Drug Safety Evaluation, Ministry of Food and Drug Safety, 187 Osongsaengmyeong2-ro, Osong-eup, Cheongwon-gun, Chungcheongbuk-do , Korea Tel: , Fax: yongchjun@korea.kr 는 Aphanizomenon flos-aquae, 클로렐라, 스피루리나, 두날리엘라등이주종을이룬다 4). 스피루리나의경우지난 30 년간사람또는동물의식품공급원및식품중색깔을내기위하여상업적으로생산되고있으며 5,6), 아프리카, 캘리포니아, 하와이, 태국, 중국, 타이완, 인디아에서는설비시설을갖춘야외연못에서배양하고있다 7). 그리고 1995년기준전세계적으로약 2,000톤이생산되고있다 5). 설비시설을갖춘야외연못에서생산하는스피루리나와다르게 Aph. flos-aquae는주로야외의대형호수에서수확하며 1980 년대초부터미국오레곤주의클래머스호 (Upper Klamath Lake) 에서가장큰규모로수확되며식품및건강식품보충제로판매되고있으며건조중량으로약 1,000톤이상이생산되고있는것으로보고되고있다 8). 기능성식품의측면에서 Aph. flos-aquae의지질중약 50%( 전체건조량의 5-9%) 는불포화지방산으로구성되어있으며동물실험결과식이보충제뿐아니라혈액의콜레스테롤수치를저 188

2 Development and Application of Detection Method for Aphanizomenon flos-aquae not Usable as a Food Materials in Korea 189 감화하는효과가있다고보고하고있다 9). 그러나클래머스호는독성물질을생산하는시아노박테리아의일종인 Microcystis aeruginosa가항상생존하고있어수확중에오염가능성이높다 10). Carmichael 등 8) 에따르면클래머스호에서수확한 Aph. flos-aquae의 80% 이상에서마이크로시스틴독소가검출되었다고보고하였다. 또한 Rapala 등 11), Ferreira 등 12) 및 Mahmood 등 13) 에의하면 Aph. flos-aquae 는 anatoxin-a, saxitoxin, neosaxitoxin 등의독소를생산할수있음을보고하였다. 스피루리나의경우독소를생산한다는보고는없으며 14), 클로렐라제품의경우 Aph. flosaquae에오염된제품이종종유통되기도한다 10). Aph. flos-aquae는남조류강 (Cyanophyceae Class) 연쇄체목 (Nostocales Order) 염주말과 (Nostocaceae Family) 에속하는남조류의일종이며담수에서자생하며녹조현상을일으키는주요미세조류의일종이다 15,16). Yamamoto 17) 에따르면비교적높은위도지역에서녹조발생시최초로발견된다는보고가증가하고있다고밝혔다. 국내의경우낙동강에서시아노박테리아에의하여여름과초가을에녹조현상이발생하며 18) 주요원인균중 Aph. flos-aquae, Microcystis, Anabaena 등은담수환경에나쁜영향을나타내는것으로알려져있다 19). 형태학적특징으로는수십에서수백 µm 의사상체로구성되어있으며사상체는대부분영양세포로구성되어있고중간에간헐적으로이형세포 (heterocyst) 가나타나기도하며기온이낮아지는등의생장환경의변화가발생하면무성포자를형성하여생존한다 20). 그리고환경조건중수온과 ph에의하여영향을많이받으며수온 15.1 o C 이상또는 ph9 이상에서는매우증식속도가높은것으로측정되었으나일조량에대하여는크게영향을나타내지않는다 20). 시아노박테리아의분류는사상체의넓이, 세포크기, 분열형태, 배열형태, 색소, 가스주머니의존재등형태학적특징에기본을두고있다 21,22). 그러나 Komarek 등은 22) 배양조건에따른환경변화로인하여 50% 이상은동정결과에차이가있을수도있다고보고하였다. 즉, 가스주머니등은생장조건또는서로다른환경에따라다양성을보여주며심지어배양중소실되기도한다 23). 이러한형태학적분류체계의한계를극복하기위하여최 근에는유전자염기구성비율, 유전자염기서열결정등에대한분자생물학적접근이시도되고있다 24). 상기에서기술하였듯이 Aph. flos-aquae는식품원료로활용가치가있지만독소를생성하기때문에소비자의건강을위하여현재국내에서는식품원료로사용할수없도록규정하고있다. 따라서시중에유통중인건강기능식품중클로렐라또는스피루리나제품에 Aph. flos-aquae가혼합되어있는지를확인하기위하여형태학적분석법은적용에어려움이있어종특이프라이머를이용한 PCR 법을개발하였다. 본연구에서개발한방법은소비자및선의의식품제조 유통업체를보호하는등식품안전관리에활용할수있을것으로기대된다. 재료및방법 시약및검체구입유전자증폭 (Polymerase Chain Reaction, PCR) 을위한프라이머및 Taq DNA polymerase는바이오니아 (Bioneer, Korea) 에의뢰하여합성또는구매하였다. PCR 장비는 C1000 Touch TM Thermal Cycler (Bio-RAD Laboratories, Inc. USA) 을사용하였으며, 유전자추출은 DNeasy Plant mini 키트 (Qiagen GmbH, Hilden, Germany) 를사용하였다. 표준시료인 Aph. flos-aquae, 클로렐라, 스피루리나는식품의약품안전처 ( 건강기능식품기준과 ) 로부터분양받았으며, 대조군으로사용된녹차, 시금치, 클로렐라제품 2종, 스피루리나제품 2종은일반마트에서직접구입하였다. 그리고표준시료를이용하여클로렐라및스피루리나에 Aph. flos-aquae가각각 10% 함유하도록실험실에서제조하였으며, 사용된제품에대하여는 Table 1에명기하였다. 유전자추출 DNeasy Plant mini 키트를사용하여유전자를추출하였다. 추출방법은제조사에서제공하는추출법에의거하여추출하였으며, 단최종용출단계에서는 AE 완충용액대신멸균증류수를사용하여 DNeasy Mini spin column으로부터용출하였다 Table 1. Polymerase Chain Reaction (PCR) 반응액조성및반응조건 PCR 을위한반응액의조성은주형 DNA 50 ng, dntps 200 µm, MgCl mm, 프라이머각 0.5 µm 이되게혼 Fig. 1. Aph. flos-aquae 판별을위한 PCR 반응조건은 95 o C에서 5분예비가열후, 95 o C 30초변성, 60 o C 15초재결합, 72 o C 30초중합반응등의과정을 35회반복하고마지막에 72 o C에서 3분간처리하였다. 결과확인최종산물의확인은반응액 2µl를취하여 2% 아가로즈젤로 100 V, 30분간전기영동하고 EtBr로염색 (1 µg/ml) 한후 UV투영기를이용하여확인하였다. 결과및고찰 종특이프라이머설계 Aph. flos-aquae의검출을위한종특이프라이머를설계하기위하여유전자은행 ( 에등록되어있는 Ahp. flos-aquae (Accession No. FJ 및 EU157995), Spirulina laxissima (Accession No. DQ393278) 및 Spirulina spp. (Accession No. HQ008228) 의 16S rrna 염기서열을이용하였다. Aph. flos-aquae의경우등록된염기서열의오류범위를줄이기위하여 2 종을, 스피루리나의경우 Spirulina laxissima 및 Spirulina spp. 를선정하였다. 그리고클로렐라의경우활용할수있는 16S rrna 염기서열이없어이론적비교 분석어려워가능한여러종류의프라이머를설계하여실험을통한최적의프라이머를선정하였다. Aph. flos-aquae에대한특이프라이머를설계하기위하여정방향 3종 (AFA-F1, AFA-F2 및 AFA- F3) 과역방향 5종 (AFA-R1, AFA-R2, AFA-R3, AFA-R4 및 AFA-R5) 을설계하였다. 그리고가공식품에대하여적용이가능하도록하기위하여 PCR 산물의크기는가급적으로 300 bp 이내가되도록조합을구성하여총 5종류 (AFA-F1/ AFA-R1, AFA-F2/AFA-R2, AFA-F3/AFA-R3, AFA-F3/AFA- R4, AFA-F3/AFA-R5) 의조합이이루어지도록하였으며 (Fig. 1), 설계된프라이머에대한정보는 Table 2에기술하였다. Aph. flos-aquae 판별프라이머선정 Carmichael 등 13) 에따르면클래머스호에서수확한 Aph. flos-aquae의 80% 이상에서마이크로시스틴독소가검출되었다고보고하고있으며, 클로렐라, 스피루리나의생산단가가높아일부생산업체에서는 Aph. flos-aquae를혼합하여제품을생산하여도현재의분석법으로는혼합여부를확인할수없다. 그리고 Aph. flos-aquae, 클로렐라, 스피루리나는고유의형태학적특징이있으나배양조건등환경변화에의하여형태학적분류에오류가있을수있어 22) 유전자분석법을시도하였다. 유전자분석법에는염기서열결정등의방법이있으나분석시간, 분석기술, 분석비용등을고려하여신속 정확하고분석방법이간편한종특이프라이머를이용하였다. 또한대조군으로는녹차및시금치를추가하여수행하였다. 설계된총 5종류 (AFA-F1/ AFA-R1, AFA-F2/AFA-R2, AFA-F3/AFA-R3, AFA-F3/AFA- R4, AFA-F3/AFA-R5) 의프라이머를이용하여 PCR을수행한결과 AFA에서는 PCR 산물이뚜렷이생성되고클로렐라, 스피루리나, 녹차및시금치에서는특이적밴드가형성되지않는 AFA-F1/AFA-R1 (363 bp) 을최종선정하였다 (Fig. 2) 유통제품에대한적용성검토국내의 건강기능식품에관한법률 제 19 조에의거한 Fig. 2. 5 192 Yong-Chjun Park, Seung-Jung Shin, Ho-Yeon Lee, Yong-Sang Kim, Mi-Ra Kim, Sang-Jae Lee, and Hwa-Jung Lee 건강기능식품공전제3. 개별기준및규격 2. 기능성원료에따르면 25) 클로렐라및스피루리나는제조방법을 인공적으로배양하고건조하여제조하여야함 그리고기능성분 ( 또는지표성분 ) 의함량은 총엽록소를각각 10 mg/g, 5 mg/g 이상함유하고있어야함 으로명기하고있다. 또한일반적으로자연호수에서채취하며 8) 독성물질을생산하는특징 11,12,13) 이있는 Aph. flos-aquae에대하여식품의약품안전처에서는식품원료로사용으로할수없도록규정하고있다 26). 따라서국내에유통중인건강기능식품에대하여식용불가원료인 Aph. flos-aquae 함유여부를조사하였다. 대상시료로는클로렐라및스피루리나제품각 2건을구입하였으며, 분양받은 Aph. flos-aquae, 클로렐라, 스피루리나를이용하여 Aph. flos-aquae가 1% 함유되도록실험실조건에서제조하였다. 본연구에서설계한 AFA- F1/AFA-R1 프라이머를이용한 PCR 분석결과유통중인제품에서는 Aph. flos-aquae가검출되지않았으며 Aph. flosaquae가 1% 함유되도록제조된시료에서는모두 PCR 산물을확인하였다 (Fig. 3). 따라서본연구에서확립된시험방법은의도적으로혼합하거나비의도적으로혼입된가공식품에대하여 Aph. flos-aquae의확인이가능하여소비자의건강및건전한제조 유통업체를보호할수있는식품안전관리에활용이가능할것으로기대된다. 감사의말 이논문은 2013년도식품의약품안전처자체연구개발과제 (13161소비자102) 연구비로수행된결과의일부로서이에감사의글을전합니다. 요약 Aphanizomenon flos-aquae는시아노박테리아일종이며 anatoxin-a, saxitoxin, neosaxitoxin 등의독소를생산할수있어국내에서는식품원료로사용이금지되어있다. 전통적으로시아노박테리아는사상체넓이, 세포크기, 분열방법, 세포형태, 가스주머니의존재유무등의형태학적특징에의한분류가가능하다. 그러나가스주머니혹은무성포자와같은특징은주변환경또는생장조건에따라차이가있으며경우에따라소실되기도한다. 따라서 PCR 에의한 Aph. flos-aquae를함유하는기능식품을검출할수있는분석법을개발하였다. 프라이머를설계하기위하여유전자은행 ( 에등록되어있는 Aph. flos-aquae, 스피루리나의 16S rrna 염기서열을이용하였으며, 비교및분석에는 BioEdit ver 프로그램을사용하였다. 최종적으로클로렐라, 스피루리나, 녹차, 시금치로부터 Aph. flos-aquae를검출할수있는 AFA-F1/AFA- R1 (363 bp) 프라이머를최종선정하였다. 그리고상기프 라이머는 Aph. flos-aquae가각각 1% 함유되도록제조된클로렐라, 스피루리나제품에서모두혼입여부의확인이가능함을확인하였다. 참고문헌 1.
