In vivo studies have suggested that, of these components, submucosal glands are thought to play an important role in nasal mucus secretion. These studies, however, are limited in dealing with pure secretory activity of submucosal glands. Therefore, in vitro study using cultured nasal submucosal glands is required to evaluate the pathophysiological mechanisms of nasal mucus secretion. The present study draws on experience obtained from the culture of human submucosal glands isolated from nasal mucosa. Materials and methodsto obtain isolated submucosal gland cells, the uncinate process mucosa was stripped of surface epithelum by stroking the luminal surface with cotton tip applicator and minced into small fragments. By enzymatic treatment, thereafter, the submucosal gland cells were isolated and plated on culture flasks containing media. Using millipore inserts and cover slide, culuted cell was stained AB- PAS to identify that confluent monolayer cells were submucosal gland cell. ResultsThe uncinate process mucosa were appropriate for the isolation of submucosal gland cell. An average of 1.210 6 cells were obtained in every preparation of cells. The exclusion ratio of trypan blue was 95%. The isolated cells were strongly stained in AB-PAS, suggesting that cells are of glandular origin. Plated gland cell formed colony on the 3rd day and became confluent after 57 days. The proportion of the stained cells to the total cells decreased depending the duration of cell culture. ConclusionWe succeeded in isolation and culture of submucosal gland cells which can provide model systems for studying the mechanisms of nasal glandular cell secretion in vivo. 분비선세포 배양 배양된 분비선세포 고 Collagen으로 도말한 배양용기에서 배양된 세포는 배양 안 2 일 후 용기에 달라붙었고, 3일에 군집을 이루었고, 배양 5일에 아융합, 7일에 융합을 이루었다(Fig. 2. A, B and C). 최근 각각의 세포의 고유한 특성을 알기 위해 세포배양법 세포의 모양은 난원형에서 다각형의 모양으로 뚜렷한 핵을 이 발달하면서 다양한 종류의 세포들의 증식이나 분화, 냉동 관찰할 수 있었다. AB-PAS 염색에서 청색, 자주색 등으로 보관을 통한 장기간의 저장법 등이 알려져 왔다.3)4)10)11) 그 염색되는 과립을 함유하는 세포가 관찰되었고, 이 세포들은 러나 선세포는 조직으로부터 분리하기가 어려워 단일세포 2일, 4일, 6일째 시간이 경과함에 따라 염색되는 세포의 비 를 배양하는데 문제점이 있으며,4) 배양과정에서 그 반응양 율이 줄어듬을 관찰할 수 있었다(Fig. 3. C 정이다. 본 연구에서는 인체 정상비점막에서도 선세포의 분리 많아 세포분리에 적당하지 않다고 생각되었다. 그러나 정상 가 가능하며 분리된 선세포를 배양하는 것이 가능하다는 것 인의 경우 사골동 점막은 채취하는 것이 어려운 반면 구상 을 나타내었다. 돌기의 점막은 외부로 노출되어 있어 비중격교정술을 하면 선세포의 분리는 토끼의 위점막에서 선세포의 분리가 시 12) 10) 13) 서 비교적 용이하게 얻을 수 있어 본 연구에서는 구상돌기 등의 동물의 기관점막에서 에 위치한 점막을 이용하였다. 이 부위의 선세포는 점막표 분리되어 실험에 이용되어 왔다. 특히 여러 실험동물의 기관 층과 가까이 위치하여 상피층을 물리적으로 제거하면 섬유 점막에는 선세포가 풍부하며 점막이 얇기 때문에 상피층을 아세포를 포함한 다른 세포를 비교적 적게 함유하게 되고, 제거하면 상피층 직하부에 분비선세포가 분포되어 있어 효 선세포의 밀도도 충분하였다. 분리된 대부분의 세포는 PAS 소처리를 하여 분리된 여러가지 세포 중 분비선세포가 차지 에 강하게 반응하는 분비과립을 함유하고 있는 세포질과 뚜 하는 비율이 높아 주로 기관지 점막에서 채취한 분비선세포 렷한 핵을 가지고 있어 선세포임을 알 수 있었고 비교적 쉽 를 배양에 이용하여왔다.10)14) 비점막에서 시도된 분비선세 고 성공적으로 분비선세포를 분리할 수 있었다. 도되었고, 소, 고양이, 개 포의 분리는 흰쥐와 기니픽 등의 실험동물에서 주로 시도되 15) 세포분리시에는 여러가지 다른 세포가 혼합될 수 있다. 주로 비중격점막을 채취함으로써 시도되어왔다. 세포의 분리와 배양에서 오염된 세포의 제거를 위해서는 이는 비중격점막의 상피층 직하부에 분비선이 고밀도로 존 세포분리시에 보다 순수하게 단일세포로 분리하는 방법 었는데, 15) 분비선간질이 적어 다른 세포가 혼합되지 않고 분 과 배양시에 오염세포를 제거하는 방법이 이용된다. 효소, 리하기가 쉽기 때문이었다. 인체의 정상비점막에서 분비선 EDTA16)17) 등으로 세포질간의 결합을 약화시키고, 세포간 세포를 분리하여 배양을 시도한 연구는 거의 미흡한 실정이 의 무게차이를 이용한 원심분리,10)15) 세포크기의 차이를 이 었으나, 최근 인체의 비점막에서 채취한 분비선세포의 일차 용하여 망상체로 세포를 거르는 방법,10)15) 분비선세포와 섬 배양이 시도되었다.3)9) 그러나 비점막 분비선세포배양은 비 유아세포, 상피세포가 평판되는 시간의 차이를 이용3)11)하여 재하며 3) 용조직에서 시도되거나, 9) 이 이루어졌었다. 조직편유출배양을 통한 세포배양 저자들은 세포분리를 위하여 적합한 비 순수도를 높일 수 있다. 본 실험에서는 효소, 세포무게를 이 용한 원심분리를 이용하여 세포의 순수도를 높였다. 점막조직을 찾기 위해 하비갑개와 구상돌기에서 비점막을 분리하여 배양된 선세포는 일반적인 다른 단일세포의 증 얻어 세포분리를 시도했다. 하비갑개의 점막은 비교적 두껍 식력과 같은 속도로 배양되었는데, 배양 2일에 세포가 소 고 선세포층과 상피세포층과의 간격이 클 뿐만 아니라 또한 군락을 이루었고 3 4일에 군집을 이루었으나 세포의 크 분비선세포 사이의 간격도 크고 섬유아세포가 상대적으로 기와 모양이 다소 불규칙하였다. 5일째 다각형의 세포로 모 585
