Hepatic ratio of glutathione disulfide to glutathione (GSH) and levels of malondialdehyde, oxidative stress markers were not changed in rats treated with TBBPA. Hepatic expression of antioxidant enzymes including GSH peroxdiase-1 (GPX-1)/ GSH reductase (GR), alpha-, mu- and pi-class glutathione-s-transferase (GST) and gamma-glutamylcysteine ligase catalytic subunit was determined using immunoblot analysis. Alpha-class GSTs, GPX-1 and GR levels were significantly decreased in rats treated with TBBPA at the dose of 5 or 1, mg/kg. These results show that TBBPA results in down-regulation of hepatic expression of antioxidant enzymes related with GSH, suggesting the liver in TBBPA-treated rats may be more sensitive to oxidants. Tel: , Fax: sangkim@cnu.ac.kr 퓨터, TV 와같은가전제품, 건축용자재, 실내장식재등플라스틱이나섬유를원료로한각종가연성제품에첨가되고있으며, 저렴한가격과높은난연효과로전세계난연제시장의약 % 정도를차지하고있다. PBBs (polybrominated biphenyls), PBDEs (polybrominated diphenyl ethers), TBBPA 33

Vol. 24, No. 4 (tetrabromobisphenol-a) 등의세종류가대표적인브롬화난연제로사용되었으나 PBBs 는이미 197 년대부터발암성과간독성이확인되어사용이중지되었다. 최근보고들에의하면 PBDEs 의경우에도간독성과생식이상을일으킴이밝혀졌으며, 생물축적성이있다는이유로잠재적인 POPs (persistent organic pollutants) 로주목받고있다 (Kemmleina et al., 3). 유럽연합은 4 년 8 월 15 일부터 PBDEs 중에서 penta-bde, octa-bde 2 종에대해사용및판매금지를시켰고, 국내의경우에도환경표지제도로간접규제를시행하고있다. 따라서, 현재안전하게이용될수있는브롬화난연제로는 TBBPA 를들수있다. 국내에서 TBBPA 는가전제품, 조리기구, 건축재료및내염성플라스틱소재에활용되며, 연간수요량이 3 년기준으로 3, 톤이나된다. 전체브롬화난연제수요량이 46, 톤임에비추어국내브롬화난연제의 65% 정도가 TBBPA 임을알수있다. 북유럽인을대상으로하는임상시험에서인간의혈액이나모유에서 TBBPA 가검출됨이보고되었으며 (Thomsen et al., 2a, b), 그잠재적위해성때문에 International Programme on Chemical Safety 와 World Health Organization 에서는 TBBPA 의독성연구를수행한바있다. 그결과, 혈액독성, 행동독성, 발암원성등의기초연구에서는 TBBPA 가무해한것으로보고되었으나최근일련의연구결과들은 TBBPA 의잠재적간, 신장및면역독성의가능성을제시하고있다 (Canesi et al., 5). 따라서, TBBPA 의주요장기에대한독성영향평가및작용기전규명은환경독성학측면에서상당히큰의미가있음을알수있다. 특히, 화학물질에의한독성발현에중요한인자인항산화방어체계와 TBBPA 독성과관련한연구는매우미흡하다. 항산화방어체계 (antioxidant defense system) 는저분자의항산화물질과항산화효소로구성되며산화성물질을무독화시킨다. 저분자의항산화물질로는 glutathione (GSH) 를비롯하여 vitamin C 와 E, bilirubin, hypotaurine 등이있으며항산화효소계는 superoxide dismutase, GSH peroxdiase-1 (GPX-1)/GSH reductase (GR), glutathione-s-transferase (GST), catalase 등으로구성된다. 특히, GSH 와 GSH 를기질로이용하는 GPX/GR, GST 등은항산화방어체계의핵심을구성한다 (Kim and Novak, 7). 또한 GSH 합성의속도결정단계를촉매하는 gamma-glutamatecysteine ligase 의 catalytic subunit (GCLC) 은산화적손상에의해유도되며 GSH 의항상성을유지하는효소이다 (Kim et al., 4). 본연구에서는대표적인브롬화난연제인 TBBPA 를모델화합물로이용하여 TBBPA 가표적장기인간에서항산화방어체계에미치는영향을평가하였다. 이를위하여랫드에 4 주간 TBBPA 를반복투여하고간을절취한후간에서 GSH 를매개로한항산화효소의발현변화를 immunoblot analysis 를통하여평가하였으며또한산화적스트레스의지표로 GSH disulfide (GSSG)/GSH 의비율과 malondialdehyde (MDA) 의변화를측정하였다. 1. 실험동물 재료및방법 특정병원체부재의 Sprague-Dawley 계랫드를사용한다. 입수된동물은무작위로폴리카보네이트 cage 에분리수용하여최소한 1 주이상의순화과정을거쳐사용하였다. 동물실의사육환경은실내온도 23±3 C, 상대습도 ~% 를유지하고 15~ 3 Lux 의조도로 12 시간씩명암을조절하였다. 2. 시약 TBBPA, GSH, NADPH, GR, Tris-HCl, thiobarbituric acid, O-phthalaldehyde 와기타시약은 Sigma- Aldrich (St. Louis, MO) 에서구입하였다. Immunoblot analysis 를위한항체중 GCLC, GR, glyceraldehyde 3-phosphate dehydrogenase (GAPDH) 그리고 GPX-1 의항체는 Lab frontier (Seoul, Korea) 에서구입하였다. GST alpha, mu 와 pi class 항체는 Detroit R & D (Detroit, MI) 에서구입하였다. 3. 동물투여및처리 Sprague-Dawley 계랫드 ( 암, 수 ) 를구입하여교배시킨후그차산자에서생후 18 일부터 28 일간 1 일 1 회반복투여한후동물을부검한다. 기존의연구보고에의해 TBBPA 의 NOAEL 값이 25 mg/kg/day 로평가되었다 (Darnerud, 3). 따라서 NOAEL 값을저용량군으로설정하였으며, 중용량군은 5

3 December 9 Lee et al. : 브롬화난연제에의한항산화활성변화 35 mg/kg/day, 고용량군은 1, mg/kg/day 로투여하였다. 랫드를 diethyl ether 로마취시키고복강을절개하여적출된간에 3 배용량의 homogenizing buffer (1 mm 의 EDTA 와 5 mm 의 Tris-HCl 을포함하는.154 M KCl 용액, ph 7.4) 를가하여분쇄한후 4 C, 1, g 에서 high speed centrifuge (Model J2,-MC, Beckman Instruments Inc., California, USA) 로 분간원심분리하고상등액을취하였다. 상등액을 4 C, 14, g 에서 1 시간동안 ultracentrifuge (Model L, Beckman Instruments Inc., California, USA) 로초원심분리하여상등액을얻었다. 상등액을 cytosol sample 로이용하며, cytosol sample 은실험시까지 -7 C 이하로유지되는 deep freezer (Model ULT- 149, Revco., Asheville, NC) 에보관하였다. 단백질의농도는 BCA TM Protein Assay Kit 를통해측정하였다. 4. GSH 와 MDA 측정 GSH 와 MDA 를정량하기위해약 1g 의간을각각 ice-cold 1 M perchloric acid 또는 1.15% KCl 용액에서분쇄하였다. Total GSH 와 GSSG 의함량은 GSH reductase 를이용한 enzyme recycling 방법을사용하였다. Eppendorf tubes 에.3 mm NADPH 용액.7 ml, 6 mm 5,5-dithio-bis-(2-nitrobenzoic acid) (DTNB) 용액.1 ml, 검체또는 GSH 표준액.2 ml 을가하여잘섞은후상온에서 4 분간방치하였다. 반응액에 12 units/ml 농도의 GSH reductase 를가하고잘섞은후 412 nm 에서약 2 분간흡광도의변화를측정하여 linear 한 1 분간의기울기변화를구하고검량선으로부터 GSH 의농도를계산하였다. MDA 는 thiobarbituric acid 로유도체화하고형광검출기와 C18 컬럼을장착한 HPLC 를사용하여분리및정량하였다 (Kim et al., 5). 5. Immunonlot anlalysis 항산화효소의 immunoblot analysis 를위해, 간의 cytosol (loading 시 μg 의 protein) 1% sodium dodecyl sulfate-polyacrylamide gel electrophoresis 를이용하여 nitrocellulose membrane 에 transfer 하여.5% Tween 를포함한 phosphate buffered saline (PBST) 에 5% milk powder 를희석하여 blocking 하였다. Immunodetection 을위해 1 차항체로서 anti- GST alpha (1 :,, 5% milk 를녹인 PBST 로희석 ), anti-gst mu (1 :,, 5% milk 를녹인 PBST 로희석 ), anti-gst pi (1 : 1,, 5% milk 를녹인 PBST 로희석 ), anti-gclc (1 : 9,, 5% milk 를녹인 PBST 로희석 ), anti-gr (1 : 2,, 5% milk 를녹인 PBST 로희석 ), anti-gpx-1 (1 : 2,, 5% milk 를녹인 PBST 로희석 ) 그리고 anti-gapdh (1 : 1,, 5% milk 를녹인 PBST 로희석 ) 로반응시켰다. 반응후 PBST 로 washing 한뒤 2 차항체로서 horseradish peroxidase (1 : 1,, 5% milk 를녹인 PBST 로희석 ) 를실온에서 2 시간동안반응시키고 ECL 용액으로발광시켰다. 분석은 chemiluminescence 를이용한 ChemiDoc XRS digital imaging system 으로 detection 하였으며 densitometry 를이용한 Quantity One Software (Bio-Rad Laboratories) 으로정량하였다. 6. 통계처리 실험군사이의통계적차이는 analysis of variance (ANOVA) 후 Dunnett s test 로검사하였다. 모든실험결과는평균 ± 표준편차로표시하였다. 결과및고찰 1. TBBPA 가투여된랫드에서산화적손상지표의변화 TBBPA 가처리된랫드의간에서 GSH, GSSG, GSSG/GSH 와 DMA 의함량을측정하였다 (Table 1). GSH 는양적으로가장풍부한 non-protein thiol 로간세포에서 mm 농도로존재한다. GSH 는친전자성물질과비효소적으로반응하여무독화시킬뿐만아니라 GST 와 GPX 의기질로서효소적으로친전자성물질과활성산소종을제거한다 (Lu, 9). 특히, 간의 GSH 는혈중 GSH 의공급원으로기능하여신장등다른장기에혈액을통해 GSH 를공급한다 (Lu, 9). GPX 의기질로사용되어산화된 GSH 인 GSSG 는 GR 에의해환원되어 GSH 로재생되거나세포의축적을막기위해혈액또는담즙으로배설된다 (Lu, 9). 산화적손상은 GSH 의소모를증가시키고 GSSG 생성의증가를유발한다. 따라서 GSSG/GSH 는산화적손상의지표로사용

Data are mean±sd for three rats. significantly different from the animals treated with vehicle only at p.1 (ANOVA followed by Dunnett s multiple range test) 되고있다. 본실험에서 GSH 의함량은 5 mg/kg 용량의 TBBPA 에의해대조군에비해약 191% 로증가하였다 (Table 1). 반면 GSSG 의농도는동일용량의 TBBPA 에의해대조군대비약 131% 로증가하였으나유의성은관찰되지않았다. GSSG/ GSH 의비율은대조군에서 1.38±.17% 였으며 TBBPA 의처리에의해유의적인변화를보이지않았다. 지질의과산화는산화적손상과정에서흔히발생하며반응생성물로 MDA, 4-hydroxynonenal, organic peroxides 등이생성된다. 이들물질은강한반응성을가지며자발적으로다른물질과반응하여소거된다. 따라서지질과산화의생성물은산화적손상의지표로서광범위하여사용되나산화적스트레스에정량적으로반응하지못할가능성이있다. 특히 thiobarbituric acid 와반응시켜 spectrophotometer 를이용하여 MDA 를측정하는방법은 aldehydes 등을비롯한다른물질의간섭으로인해정확성에문제가있다 (Volpi and Tarugi, 1998). 본연구에서는 MDA 와 thiobarbituric acid 의반응생성물을 C18 컬럼이장착된 HPLC 로분리하고형광검출기로정량함으로써간섭을최소화하였다. 실험결과 TBBPA 의처리는간에서 MDA 의농도에변화를유발하지않았다. 본실험결과는산화적손상의지표인 GSSG/GSH 그리고 MDA 의농도가 4 주가반복투여된 TBBPA 에의해영향을받지않으며따라서산화적손상은 TBBPA 에의해발생하지않았음을시사한다. 2. TBBPA 가투여된랫드에서항산화효소의발현변화 TBBPA 를처리한랫드간의 cytosol 에서항산화효소의발현변화를관찰하였다 (Figs. 1, 2). Immunoblot analysis 에서단백질의 loading control 로사용한 GAPDH 는 TBBPA 의처리에영향을받지않았다 (Fig. 1A). GSTs 는주로간조직에발현하며친전자성물질과 GSH 의포합반응을매개할뿐만아니라과산화수소, organic peroxide 에대한 seleniumindependent peroxidase 활성을가지는항산화효소이다 (Sun et al., 1996; Ketterer, 1998; Townsend and Tew, 3; Casalino et al., 7). 특히, alpha-class GST 와 mu-class GST 는간에서발현하는대표적인 GST 로간에서발현하며항산화활성의중요한역할을담당한다. Cytosol 에존재하는 GSTs 는 homo- 또는 heterodimeric 효소로존재하며간에서 alphaclass GSTA1, A2, 그리고 A3 subunit 그리고 muclass GSTM1 과 M2 가주로발현한다 (Mannervik et al., 1985). 본실험에서 TBBPA 투여는 mu class GST 의발현에변화를유발하지못하였다 (Fig. 1B). 반면 alpha-class 인 GSTA1/2 와 GST3/5 의발현은 5 mg/kg 용량으로처리된 TBBPA 에의해유의적으로감소하였다 (Fig. 1C, 1D). 태자의간과간암에서발현하는 pi-class GST 는본실험에서검출한계이하였으며 TBBPA 처리에의해서도검출되지않았다 (data not shown). 이결과는 TBBPA 가 GST isoform 에선택적으로발현변화를유발함을시사한다. 또한간의주요 GST 는 alpha class GST 의감소는 TBBPA 가친전자성물질의포합반응및 peroxides 의무독화능력을저하시킬가능성을시사한다. Gamma-glutamate-cysteine ligase (GCLC) 는 GSH 신생합성의속도결정단계를촉매하는효소로활성을가지는 heavy subunit, 즉 GCLC 와조절기능을가지는 light subunit 으로구성된다 (Lu, 1999; Serviddio et al., 4). GCLC 의발현은산화적손상에의해유도되며이현상은산화적스트레스에적응하

Data are mean±sd for three rats.,significantly different from the animals treated with vehicle only at p.5 or p.1, respectively (ANOVA followed by Dunnett s multiple range test). 기위한중요한과정이다. 산화적스트레스에의한 GCLC 의발현증가는 NF-E2-related factor2, AP-1 등의전사인자에기인한다 (Kim et al., 8). 본연구에서 GCLC 의발현은 TBBPA 의영향을받지않았다 (Fig. 2A). 따라서 TBBPA 에의한 GSH 상승은신생합성을매개하는 GCLC 의변화로설명할수없다. GPX/GR 은과산화수소등 peroxide 의무독화를매개하는핵심효소로 peroxide 를환원시키는과정에서생성된 GSSG 는 NADPH 로부터환원력을제공받는 GR 에의해다시 GSH 로환원된다. 따라서 GPX 와 GR 의변화는산화적스트레스에대한세포의저항성을결정하는중요한요소이다 (Sun et al., 9). TBBPA 의처리는 5 mg/kg 용량에서 GR 의발현을대조군의약 79% 수준으로감소시켰으며용량을 1, mg/kg 으로증가시킬경우유의적으로 GR 의발현이감소하였다 (Fig. 2B). GPX 는 selenocysteine 을활성부위에포함하고있으며과산화수소외에다양한구조의 hydroperoxide 를무독화시킨다 (Ursini et al., 1995; Brigelius-Flohe, 1999). 현재까지 4 가지 isoform 이발견되었으며그중에서 GPX-1 은세포내에널리분포하고과산화수소

TBBPA 의처리는 5 mg/kg 용량에서유의적으로 GPX-1 의발현을감소시켰다 (Fig. 2C). 이결과에서 TBBPA 의처리가 GPX/GR 을통한 peroxide 의무독화능력을감소시킴을시사한다. 결 론 TBBPA 는독성으로사용이금지되었거나규제를받고있는 PBBs, PBDEs 와달리상대적으로독성이낮은것으로보고되고있다. 그러나최근일련의연구결과들은 TBBPA 의잠재적간, 신장및면역독성을제시하고있다. 본연구에서는브롬화난연제 (brominated flame retardants) 로광범위하게 TBBPA 를반복적으로 4 주간처리한랫드의간에서항산화방어체계의변화를관찰하였다. 실험결과 4 주간의반복처리는간에서 MDA, GSSG/GSH 의비율을증가시키지않았으며반면 total GSH 의농도는 5 mg/kg 용량에서유의적으로증가하였다. GSH 의합성과대사에관여하는효소의발현변화를평가한결과 GST 중에서 alpha class GST, GR/ GPX-1 의발현이유의적으로감소하였다. 이결과는 TBBPA 의처리가일부항산화효소의발현을감소시켜결과적으로산화성물질에대한간세포의반응성을증가시킬수있음을시사한다. 감사의글 본연구는 8 년도식품의약품안전청독성물질국가관리과제의지원으로이루어진것임. Fig. 2.
