SPE 제품의선택 SPE 제품의올바른선택방법 1. 시료의특성즉, 매트릭스 (Matrix) 와관련된극성도, 하전된작용기의존재유무, 시료의용해도, 분자량등과같은요소들을고려하여 SPE 충진베드 (Bed) 에얼마나강하게시료를머무르게할것인지를결정해야한다. 2. 시료를머무르게하는방법의선택. 두가지방법이가능하다. 방법 1. 원하는성분은충진물을통과시키고간섭물질을머무르게하는방법방법 2. 원하는성분은충진물에흡착시키고그이외의간섭물질은통과시키는방법으로이방법은또한시료가용리되는동안시료를농축시킬수있다. 3. 적합한충진형태와 Bed 크기의선택. 최고의회수율을가진순수한추출물을얻기위해선알맞은충진물을선택해야한다. 충진베드의크기가적합하지않을경우종종추출회수율이떨어진다. 즉, 충진베드가너무크면용리가불완전하고, 충진베드가너무작으면머무름 (Retention) 이불완전해진다. 많은시료중에포함된미지요소들로인해응용을위한최적의충진베드의크기를결정하기위하여때로는예비실험이요구되기도한다. 최적의충진베드의크기를결정하기위한방법으로는먼저 200mg, 또는 500mg과같은중간정도의충진베드로시작한다. 만약완전한머무름이얻어지면, 더작은크기의충진베드와용리부피 (Elution Volume) 를사용할수있다. 반면불완전한머무름을보인다면더큰크기의충진베드와용리부피를사용해야한다. 4. 적당한컨디셔닝, 세척그리고용리용매의선택. 충진물질과관련하여용매의세기를고려해야한다. 최종의컨디셔닝용매는용리용매로작용하지않도록그세기가약해야하며잠재적으로전하를띤화합물의이온화를조절하기위하여완충용액 (Buffer) 을사용해야한다. 세척용매는시료가용리되지않도록충진베드에머물러있는간섭물질을서서히제거해야한다. 용리용매는소량 (1~2ml) 의용매로시료를완전히용리시키기위해용리세기가강한용매를사용한다. IC-RP 는계면활성제, 유기산, 단백질그리고다른유기계면활성 제를제거하는데확실한방법을제공한다. 많은이온크로마토그래피응용에서가장까다로운부분은시료매트릭스로부터간섭하는성분을제거하는것이다. 이러한간섭물질은함께용리되거나또는중요한피크를간섭하며, 컬럼에과부하를주고, 컬럼충진물에결합되어세척할수없게하므로컬럼수명을급격히단축시킨다.
