심포지움의막을열었다. 성체의뇌는새로운세포를만들어내지않을것이라는기존의생각과달리, 최근연구들에따르면포유류의 hippocampus 혹은 olfactory bulb에서평생동안새로운뉴런을만들어내고, 새롭게생성된뉴런은어느정도의시간동안독특한 neural processing을거친다. 또성체쥐의뇌에서새롭게생성된 oligodendrocyte는신경가소성 (Neural Plasticity) 에중요한역할을한다. Karolinska 연구팀은성체의뇌에서뉴런과 oligodendrocyte가만들어지는지는양을확인하였고탄소동위원소를이용하여포유류가진화하면서 cell generation이어떻게변화되어왔으며또인간에서는얼마나변화되어왔는지추적하는연구를진행하였다. 본연구팀은인간과쥐모두해마에서평생동안해마에서뉴런이생성되는반면, 성체의 olfactory bulb에서는다른포유류와달리인간에서는 neurogenesis가일어나지않으며, 대부분의포유류성체에서는뇌의선조체 (striatum) 에서 neurogenesis가거의일어나지않는반면, 인간에서는 interneuron이지속적으로생성된다고발표하였다. 또헌팅턴환자의경우, 성인이되어만들어진 striatal neuron은선택적으로고갈된다고주장했다. 한편, 쥐와달리성장기가끝난인간의 oligodendrocyte는굉장히 stable하기때문에인간과쥐의신경가소성이다르다고주장하였다. 결론적으로같은포유류라할지라도인간과쥐의뉴런생성은각각다른양상을띤다는점을알수있었고, 본연구팀은인간의뇌에서일어나는여러질병메커니즘과관련하여연구해보고자한다고밝혔다. ii. Session 1 Pluripotency, Differentiation & Reprogramming (Chair: Christa Buecker) 첫번째세션은캠브리지대학의 Thorsten Boroviak의 How to build a primate: Towards a synthetic model of primate embryogenesis 라는제목의발표로막을열었다. 포유류의 Preimplantation 발생동안 zygote는 pluripotent epiblast와 hypoblast, trophoblast로이루어진 blastocyst를형성하며, Implantation은 embryo의 reorganization이일어나는중요한과정으로, 쥐의 pluripotent epiblast는컵모양의 epithelium과 egg-cylinder를형성하지만, 영장류의 epiblast는 flat disc를형성하고 implantation 후에양막 (amnion) 으로분화한다. 또한영장류는, gastrulation에앞서 extraembryonic mesenchyme을만든다는점과 trophoblast의발생과침투면에서쥐와상당히다르다. 인간의 embryo implantation에관련한메커니즘은윤리적인이유로연구가용이하지않아명확하게밝혀져있지않으나, Early marmoset 발생은 rhesus macaque와인간에서거의동일하기때문에본연구팀은 marmoset embryo을이용하여 spatial methylome과 Transcriptome profiling (SHOT sequence) 진행함으로써영장류의 post-implantation의발생과정에대해연구하고자하였다. Laser capture microdissection approach를이용하여 embryo 각샘플의 original location을기록하였고, sample은 single-cell joint profiling을이용하여 methylome과 transcriptome간의 regulatory association을분석하였으며. 영장류배아발생을분석하기위해 SHOT-seq 및 immunofluorescence labelling을이용하여 digital 3D reconstruction을진행하였다. 또 Carnegie stage 6 배아에서 amnion, embryonic disc, secondary yolk sac, cytotrophoblast의 preliminary signature를만들었고, 이를바탕으로본연구팀은 amnion과 embryonic disc specification 연구를위해 in vitro 모델을만들고자하고있다. Decommissioning and Plasticity in Early Differentiation 을제목으로연구내용을발표하였다. FGF-Erk signaling은 primitive endoderm (PrE) 에서 embryonic stem cell의분화와 pre-implantation에서의분화를유도한다. 반면에 Erk를 inhibition시키면 ESC의 self-renewal이유도된다. Standard ESC 배양에서의 paracrine FGF-Erk 신호전달은 heterogeneity를유도하면세포는 pluripotency에서 PrE로역으로진행하는데, 여전히다시 self-renewal하는능력이있다. 본연구팀은이러한 ESC분화의역진행방향초기단계에서일어나는기전을알아보기위해, Erk 활성화를유도하는 cell intrinsic system을개발하여 Erk가 pluripotency를유지하는전사인자들을역으로억제함으로써 ESC분화를유도한다는것을밝혔다. 그러나이러한유전자들의전사가억제되어도전사인자단백질이여전히일정기간동안안정적으로발현되고있으며, 전사의초기단계에서 Erk-dependent repression에도불구하고전사인자 (TF) 가발현될뿐만아니라, enhancer에결합되어있다고주장하였다. 본연구팀은 TF가 enhancer에영구적으로부착되어 Erk가제거되었을때, pluripotency의 network가활성화되어분화단계의초기에서 plasticity를유지할수있다고주장했다. Institute Pasteur의 Pablo Navarro는 Mitotic bookmarking by TFs 라는제목으로연구내용을발표하였다. 쥐의배아줄기세포는 blastocyst의 inner cell mass에서유래하며, pluripotent하고무한정으로 self-renewal할수있다. Ex-vivo에서는 pluripotency가무한하게분할하는동안유지되는반면, embryo에서는 pluripotency가일시적으로만존재한다. 따라서본연구팀은이러한특별한상태를유지하게하는메커니즘을알아보기위해 mitotic bookmarking이라고알려진과정에서 mitosis 동안특정전사인자들이 ES cell 발생과어떤관련이있는지알아보고자하였다. IMBA의 Ulrich Elling은 The Genetics of Cell Identity Change 라는제목으로연구내용을발표하였다. Cell identity는핵치환기술이나 Yamanaka cocktail과같은전사인자들에의해바뀔수있다. 현재까지 Lineage를정의하는전사인자가작동하는메커니즘에대해많이알려져있지만, lineage를역으로진행시키는전사인자와기전에대해서는많이알려져있지않다. 본연구팀은새롭게개발한 CRISPR 기술을적용하여 cell identity conversion의 genetics를확인하고, Ascl1과 Ngn2 발현에의해유도되는 neurogenesis의경로를밝히려한연구를소개하였다. University of Padua의 Marco Pellegrini는 Direct generation of human naïve induced pluripotent stem cells from somatic cells in microfluidics 라는제목으로연구내용을소개하였다. ipsc (Induced pluripotent stem cell) 는 OCT4, SOX2, KLF4와 cmyc의전사인자를발현시켜만들어지는데, murine naïve ipsc와달리인간 ipsc는 primed pluripotency 라일컬어지는조금더 advanced 한 state에서만들어진다. 본연구팀은 genetic manipulation의과정대신에, microfluid와변형시킨 mrna를 delivery하여 1000개미만의 primary human somatic cell로부터직접 human naïve ipsc (nipsc) 를만들었다. NANOG와 OSKM를 12일동안발현시켜만든 nipsc는 transgene이없고, 정상의핵형을가지며, transcriptional, epigenetic, metabolic한측면에서 naïve state를보이며, three germ layer로분화될수있다고밝혔다. 본연구팀이만든 microfluid는 patient specific nipsc를안정적이면서도저렴하게만들수있어, 질병모델링이나약물스크리닝, 재생의학연구에적용할수있다고주장했다. B. 3 월 22 일주요내용 i. Session 2 Germ Lines & Totipotency (Chair: Kikue Tachibana) 두번째세션은 Institute Curie 소속 Déborah Bourc his의 Maternal epigenetic contribution to zygotic genome activation 라는제목의연구내용발표로시작하였다. 포유류의정자와난자의 genome은 DNA methylation의양과분포면에서명확히다르며, 수정되어만들어진 embryo는비대칭적인 methylome을가진염색체를갖게된다. 모계의 Dnmt3L의돌연변이를이용하여난자유래의 DNA methylation을제거하면 mid-gestation에서 lethality가유발되는데, 이는배아발생과정에서 maternal imprinting가중요함을함의한다. 본연구팀은모계유래의 DNA methylation이수정직후의 transcriptome의조절에미치는역할을밝혀내기위해, 난자유래의 methylation을제거한 model 연구를통해 preimplantation embryo의발생에대해분석하였고, High throughput single-cell sequencing을이용하여 transcriptome의조절에관여하는난자유래의 DNA methylation의역할을분석하였으며, Dnmt3L mutant를이용하여난자 DNA methylation의제거가특정분할단계 (Zygote genome activation의초기단계 ) 에미치는영향에대해분석하였다. MRC London Institute of Medical Sciences의 Petra Hajkova는 Molecular principles of epigenetic reprogramming and gametogenesis 라는제목으로연구내용을발표했다. 쥐의초기발생후접합자와발생중인 primordial germ cell (PGCs) 는크로마틴구조나히스톤변형, genome 차원에서의 DNA demethylation과같은 global한 epigenetic reprogramming을거치지만이에대한 mechanism은잘밝혀져있지않다. 따라서본연구팀은쥐의 gonadal primordial germ cell (PGCs) 에서 epigenetic reprogramming 동안일어나는 DNA modification의변화와 chromatin configuration의독특한양상, 그리고 global DNA의디메틸화와관련한 germline developmental program의기전에대해서연구하였다. Kyushu University의 Katsuhiko는 A stem cell-based approach to understanding of mammalian oogenesis 라는제목으로강연하였다. Female germ line은 totipotency를부여하기위해여러독특한분화단계를거치기때문에, Pluripotent stem cell을이용하여난자를다시만들어보는것은 totipotency에대한이해를넓힐수있다. 본연구팀은쥐의만능줄기세포에서난자발생의과정을구현할수있는배양 system을개발하여, ESCs/iPSCs로부터 primordial germ cell을유도하였고, 배아의난소에있는체세포와결합시켜 Reconstituted ovary 를만들어약 5주의시간동안몇단계의배양과정을거쳐실험실에서도다수의성숙한난자를생산하였다. Institute of Epigenetics and Stem Cells의 Eva Hoermanseder는 Epigenetic Barriers to Cell Fate Reprogramming 라는제목으로연구내용을발표하였다. Vertebrate egg는체세포의역분화과정을유도하는데이용되는데, 난자에서몇몇유전자는체세포상태와같이유전자발현 memory를가지고있기때문에, 분화된핵을 reprogramming하기에는효율적이지못한부분이있다. 이와같은같은유전자들은전사과정없이세포분할시기이후강력하게발현되며이를 ON-memory 유전자라한다. 되고전사의재프로그래밍화와 NT-embryo의발생이촉진된다. 따라서 H3K4-메틸화는 NT-embryo 에서의후성유전학적인 ON-memory를제공하여핵의효율적인재프로그래밍을방해하는역할을하기때문에 H3K4 메틸화를제거하는것은재프로그래밍전략을보다더향상시킬수있는방법이라고설명하였다. Broad Institute of MIT and Harvard의 Jian Shu는 Reconstruction of cellular reprogramming landscapes and trajectories from large-scale single-cell profiles 이라는제목의발표에서 ancestordescendant fate를분석하기위한방법으로 Waddington-OT를도입하여 large-scale의 single-cell RNA-seq와 single-cell ATAC-seq data를설명할수있는프로그램을모델링하였고 fibroblast를 Yamanaka factor를이용하여 pluripotency 또는 totipotency를가진세포로재프로그래밍되는과정에서추출한 50만개의 scrna-seq와 scatac-seq profile 연구에 WADDINGTON-OT를적용하였을때 powerful한검정능력이있었다고발표하였다. 또본연구팀이개발한고해상도 Map은 pluripotency 나 totipotency로의다양한 reprogramming 과정을보여줄수있어매우유용할것이라고전망하였다. < 학회 Break time > ii. Session 3 Organoids (Chair: Sasha Mendjan) 세번째세션은 MRC Laboratory of Molecular Biology 소속의 Madeline Lancaster의 Advances in brain organoid technologies and their application to human biology 라는제목의발표로막을열었다. 본연구팀은초기인간배아뇌의전형적인 organization의자발적인 selforganization과형성을자극하는 3D 미세환경을이용하여직접분화시켜 cerebral organoid라불리는 in vitro human model system을만들었다. 본연구팀은 neuronal migration과 positioning, axon guidance와 neuron maturation 등의신경발생후기단계를모델링할수있는조직을만드는데이방법을적용하였으며, 이방법을통해신경회로의생성및 CNS tract의형성, 더나아가운동회로를통한근육조절과수축까지만들어낼수있을것으로내다보았다. Cambridge Stem Cell Institute의이주현 (Joo-Hyeon Lee) 선생님은 Stem Cells and Their Dynamic Niche in Lung Repair and Regeneration 라는제목으로연구내용을발표하였다. 본연구팀은 genetic lineage tracing과 3D organoid co-culture 방식을결합하여폐포손상회복과정동안일어나는 AT2 세포의증식과 AT1 세포로의분화를유도하는역동적인염증반응의환경에대해실험하고있다고밝혔다. IMBA 소속의 Reiner Wimmer는 Human blood vessel organoids model diabetic vasculopathy 라는제목으로연구내용을발표하였다. 당뇨의발병은전세계적으로증가하는추세에있으며, 당뇨는 basement membrane의팽창과 vascular cell의소실로정의되는혈관변화를일으키기때문에실명이나신부전, 심장질환, 뇌졸중, 하지절단을유발하는주요한원인이다. 당뇨병은내피세포의기능에악영향을미치며 EC-pericyte communication을방해하지만, 아직까지 Endothelia/pericyte dysfunction이어떻게 diabetic vasculopathy에영향을미치는지에대해서는많은부분이명확하지않다. 따라서본연구팀은 pluripotent stem cell로부터 self-organizing 3D human blood vessel organoids를만들어, endothelial cell과 pericyte를포함하는 human blood vessel organoid가 basement membrane으로둘러싸인 capillary network로자가조립되며, 쥐에이식된 Human blood vessel organoid는관류하는혈관분지들즉, artery, arteriole, venule이안정적으로형성됨을확인할수있었다. Blood vessel organoid가 in vitro에서 hyperglycemia나 inflammatory cytokine에노출되면 vascular basement membrane이두꺼워지며, Human blood vessel이쥐의 in vivo에서당뇨환경에노출되면당뇨환자와같은 microvascular change를일으킴을확인할수있었다. 또한 DII4-Notch3는 human blood vessel에서 diabetic vasculopathy를일으키는중요한인자임을확인하였다. 따라서본연구팀은 human stem cell로부터만들어진 organoid를통해 human blood vessel의구조와기능을보여주고, 또한 diabetic vasculopathy를모델화하고이과정을조절하는인자를찾는데매우적절하다고설명하였다. C. 2019 년 3 월 23 일주요내용 i. Session 4 Adult Stem Cells (Chair: Noelia Urban) 네번째세션은 University of Lausanne의 Marlen Knobloch가 Metabolic regulation of neural stem cells 이라는제목의발표로막을열었다. 포유류뇌의특정부분은평생에걸쳐 Neural stem/progenitor cell (NSPC) 로부터새로운뉴런을생성한다. Adult neurogenesis를 tissue homeostasis와 physiological brain function에중요하며, neurogenesis의손상은 major depression과 epilepsy 등의정신과적질환과관련이있다고알려져있다. NSPC quiescence와 proliferation의엄격한조절은평생에걸친 neurogenesis에중요하며 stem cell pool의 exhaustion과 uncontrolled growth를막는다. 그러나이러한평형을어떻게미세하게조율할수있는지에대해서는알려진바가거의없다. 본연구팀은 cellular metabolism이 NSPC activity에미치는영향과, metabolic program 이 quiescent 및 proliferating NSPC 사이에서변화하는양상, lipid의 build-up (de novo lipogenesis), break down (fatty acid oxidation) 이 NSPC의 proliferation과 quiescence에미치는영향에대해소개 SY-Stem (Symposium for the next generation of stem cell researchers) 참석후기임현경 Page 7 / 10
하였다. 또, single metabolite의 manipulation이 NSPC의 behavior를변화시키며다른 stem cell system에도적용될수있다고설명하였다. Center of Regenerative Medicine in Barcelona의 Carolina Florian은 Aging alters the epigenetic asymmetry of HSC division 이라는제목으로연구내용을소개하였다. Stem cell은 symmetric과 asymmetric division의과정을거치는데, Stem cell division을조절하는기전에대해서는아직명확히확립되지않았다. 본연구팀은대응방법 (paired fashion) 으로젊은쥐의 hematopoietic stem cell (HSC) 과나이든쥐의 HSC의 first division에서기원하는 2개의 daughter cell의 epigenetic signature, transcriptome, function을비교하였다. Young HSC는주로비대칭적으로분할되는반면, aged HSC는주로대칭적으로분할된다는사실을밝혀냈고, 이러한분할방식은 stem cell polarity 와밀접하게관련되어있으며, small RhoGTPase Cdc42의활성화에의해조절된다고발표했다. 또한 Daughter cell의 potential은 epigenetic mark인 H4K16ac의양과, daughter cell에할당된 open chromatin, Cdc42 자체와밀접하게연관되어있지만, transcriptome과는관련이없는것으로확인되었다. 종합하면, epigenetic mechanism은 stem cell division의 functional outcome을결정하며, Cdc42 활성화를통해 HSC polarity의 division 방식이결정되고, 결국 HSC polarity를조절하는분자들은 daughter cell의재생능력과 stem cell division을조절하는역할을한다고설명하였다. IMBA의구본경 (Bon-Kyoung Koo) 선생님은 Defining the identity and dynamics of adult gastric isthmus stem cells 이라는제목으로연구내용을발표하였다. Gastric corpus epithelium은 gastro-intestinal tract에서가장두꺼운부분으로 rapid tissue turnover의특징을가진다. 여러 marker들은 Isthmus와 base region에서 gastric corpus stem cell에중요한역할을미치나, isthmus stem cell의 identity와 distinct stem cell population 사이의상호작용에대해아직뚜렷하게밝혀진바가없다. 본연구팀은 unbiased genetic labeling과 biophysical modeling을통해, corpus gland가 2 개의 independent zone으로구획화되며, actively-cycling stem cell은 pit-isthmus-neck region를, slowcycling reserve stem cell은 base를차지함을관찰할수있었다. 또한 Stmn1과 Ki57 발현에기반한 independent lineage tracing을이용하여 rapidly-cycling IsthSC가 corpus gland의 pit-isthmus-neck에존재한다는것을확인하였다. 결론적으로 single cell RNA-seq analysis는 molecular identity와 single, cycling IsthSC population의 lineage relationship을밝혀내는데사용될수있으며, 이를통해 actively-cycling IsthSc single population의 identity와 functional behavior를밝혀낼수있다고발표하였다. University Medical Center of the University Mainz의 Sophie Peron은 Direct in vivo glia-toneuron conversion in the postnatal mouse cerebral cortex 라는제목으로연구내용을발표하였다. 뇌의 glial cell은 in vivo에서 neurogenic transcription factor들이발현될때뉴런으로바로 reprogram된다. 성체쥐의피질에서는이러한 reprogram이 local glial proliferation이발생할수있는 brain lesion 후에만일어나는것으로알려져있다. 따라서본연구팀은재프로그래밍화과정에영향을미치는 glia의 proliferative state를명확히하기위해, proliferative glia가 postnatal mouse cortex에서병변 (lesion) 없이도 in vivo에서 lineage convert가될수있는지실험해보았다. Glial cell 은초기 postnatal cortex에서부분적으로증식하는데, 본연구팀은이세포들의 lineage conversion 을알아보기위해 postnatal mice의 cortex에 neurogenic transcription factor를코딩하는 retrovirus를 SY-Stem (Symposium for the next generation of stem cell researchers) 참석후기임현경 Page 8 / 10
주입하였다. 그결과 Asc1, Neurog2 또는 NeuroD1가단독으로과발현되면 glia가 neuron으로전환하는데비효율적인것으로나타났다. 또본연구팀은다른 molecular pathway와의 synergy가 reprogramming을유도하는지실험해보았고, 흥미롭게도 Ascl1과 Sox2 또는 Ascl1과 Bcl2의동시발현시키면 glial cell이 neuron으로전환되고, 이들중일부는 inhibitory neurotransmitter인 GABA를발현하며, Bcl2는 Neurog2-mediated glial의 neuron으로의전환을촉진하는것을확인할수있었다. 특히, 이두인자가함께발현되면, conversion efficiency가매우증가했을뿐만아니라 dendritic spine이나 long-distance projection이가능한정교한뉴런이생성될수있다고밝혔다. 본연구팀은현재 reprogram된뉴런의통합적인기능과전기생리학적특징에대해서연구중이라고발표하였다. ii. Session 5 Regeneration & Ageing (Chair: Bon-Kyoung Koo) 다섯번째세션은 Institute Pasteur의 Elisa Gomez가 Ageing and inflammation drive the specific loss of HSC-independent tissue resident macrophages 라는제목의연구내용발표로막을열었다. 노화는선천면역세포의과도한생산, 적응면역반응의결함, 전신적인만성염증과밀접한관련이있다. 대부분의 adult tissue에서는 Embryo-derived Hematopoietic Stem cell (HSC)- independent resident macrophage와 HSC-derived monocytes/macrophage의 2-lineage의 macrophage가공존하는데, Adult tissue에서 resident macrophage가유지될수있는이유에대해아직까지밝혀진바가거의없다. 개체발생과정에서 tissue homeostasis를유지하기위해서는 resident macrophage가중요한역할을하는데, Resident macrophage는 HSC-derived cell에의해교체되며개체발생에서 tissue resident macrophage의교체는노화에영향을미치는것으로생각되어왔다. 본연구팀은대부분의조직에서 resident macrophage density가노화와함께감소하는데, HSCderived cell에의한 compensatory mechanism이존재하는다른 tissue myeloid population과달리 resident macrophage는 compensatory mechanism이존재하지않는다고밝혔다. 한편, aged mutant mice에서는 impaired inflammation sensing이 resident macrophage의손실을억제하는반면, 지속적인염증은 resident macrophage의손실을유도한다고설명했다. 본연구팀은 Ageing tissue에서의 Resident macrophage 변화양상에대한연구는 Aged tissue에서의 resident macrophage homeostasis mechanism을이해하는데에새로운시각을제공해줄것이라고기대했다. iii.
