In esterase group, acid phosphatase showed high activity from both of A. simplex (s.s.) and A. pegreffii. Esterase (C4) showed activity only from SE and A. simplex (s.s.) showed higher activity than A. pegreffii. Alkaline phosphatase, acid phosphatase and naphthol-as-bi-phosphohydrolase showed higher activity in 3rd stage larvae than in 4th stage larvae of both species. In aminopeptidase group, only leucine arylamidase showed remarkable activity in SE of both anisakid species, and A. simplex (s.s.) SE showed higher activity than A. pegreffii SE. In glycosidase group, N-acetyl-β-glucosaminidase, α-mannosidase, α-fucosidase showed higher activity in A. simplex (s.s.) than A. pegreffii, and 4th larvae showed higher activity than 3rd larvae. These differences in hydrolase activity of anisakid nematodes larvae are thought to be due to different metabolism such as growth, moulting, digestion and feeding. Key words: Anisakis simplex sensu stricto, Anisakis pegreffii, Hydrolase activities, API ZYM 해양생물을매개로하여인간에감염되는인수 공통질병중기생충을원인으로하는질병으로는 조충류에속하는 Diphyllobothrium sp., 흡충류에속 하는 Clonorchis sp. 와 Opisthorchis sp., 선충류에속 하는 Anisakis spp. 와 Gnathostoma sp. 감염에의한 질병등이알려져있다 (Chai, 21). 이중아니사키 Corresponding author: Jeong-Ho Kim Tel: , Fax: jhkim7@gwnu.ac.kr 스속 (Genus Anisakis) 선충의유충에감염된어류나두족류를생식하거나조리가덜된생태에서섭취하였을때, 사람에게아니사키스증이일어날수있으며구토, 설사, 복통등의증세를수반하는것으로알려져있다 (Smith and Wootten, 1978). 이러한감염증은형태학적으로 Anisakis Type Ⅰ(Anisakis simplex sensu stricto, A. pegreffii, A. simplex C, A. typica, A. physeteris) 에속하는기생충에의해발생한다고알려져있다 (Audicana and Kennedy, 28).

2 26 전찬혁 위성 김정호 Anisakis Type Ⅰ에속하는각각의유충은형태적으로매우유사하여각각을동정하는데어려움이있어이전의대부분의연구들은 A. simplex (s.s.) 를대표종으로표기하거나 A. simplex (s.s.) 를대상으로하여연구가진행되어왔다. 하지만 singlestrand conformation polymorphism(sscp), multiplex PCR, PCR-RFLP, direct sequencing 등의분자생물학적방법이도입되면서유충을보다신속정확하게동정할수있게되었고, 특히 PCR-RFLP 방법을이용하여몇개의제한효소만으로도아니사키스속선충유충을대부분종수준에서동정할수있게되었다 (D Amelio et al., 2; Nadler et al., 2; Umehara et al., 28a; Zhu et al., 27). 최근일본에서는분자생물학적방법을사용하여아니사키스증환자에서분리한유충을동정한결과 A. simplex (s.s.) 인것으로확인되어, 아니사키스증을유발하는주요원인종이 A. simplex (s.s.) 인것으로보고하였다 (Arizono et al., 212). 그러나, 우리나라와일본에서아니사키스속선충유충을동정한몇몇연구결과에따르면, 연근해산어류및오징어 (Todarodes pacificus) 에서 A. simplex (s.s.) 뿐만아니라근연종인 A. pegreffii가높은비율로발견되어 (Kim et al., 212; Setyobudi et al., 213, Umehara et al., 28b), 두근연종간의지리적분포와실제두근연종의감염에의한아니사키스증의유병율과는차이가있다. 아니사키스증은아니사키스선충유충이사람의소화관을뚫고들어갈때나타나는질병이다 (Oishi and Hiraoki, 1973). Audicana and Kennedy (28) 는 A. simplex 3기유충이사람의위장점막으로침투하기위해서는세포외기질 (extracellular matrix) 을분해할수있는강력한단백질분해효소들을분비하여조직을붕괴시키는메커니즘을가질것이라고기술하였다. 또한, 기생충의단백질분해효소들은숙주조직을붕괴시키는병원성뿐만아니라숙주면역반응에대한회피작용과도관련된것으로보고되고있다 (Sakanari et al., 1989). 기생충의가수분해효소는 esterases, peptidases, proteases 그리고 glycosidases 등으로구분되는데, 주로숙주조직을뚫고들어갈때분비되는단백질분해효소와숙주조직내로이동및생장할때사용되 는대사작용과관련된효소들을포함하고있어 (Ondrovics et al., 213), 아니사키스선충유충의병원성및대사작용과관련된메커니즘을이해하기위해서는단백질분해효소뿐만아니라이를포함하는가수분해효소활성에대한연구또한필요하다. 아니사키스속선충의병원성및대사작용에관련된연구로는 Suzuki et al.(21) 이 A. simplex (s.s.) 와 A. pegreffii 를대상으로한천배지위에서물리적침투력을비교하였을때, A. pegreffii 보다 A. simplex (s.s.) 에서더높은침투력을확인할수있어 A. pegreffii 보다 A. simplex (s.s.) 가사람에게아니사키스증을유발시킬확률이더높을것이라추정하였다. 아니사키스과선충의단백질분해효소에관한연구는 A. simplex와 Hysterthylacium aduncum 을대상으로이루어졌는데 A. simplex 의경우 serine proteases가, H. aduncum의경우 metallo proteases가숙주조직을분해하고숙주로침투하는과정에주로관여한다고보고하였다 (Malagón et al., 211; Morris et al., 1994). 한편, 단백질분해효소를포함한가수분해효소활성에관한연구는 A. simplex, Contracaecum rudolphii에서보고되었다. A. simplex 는섭이특성의차이에의해 3기유충과 4기유충에서가수분해효소활성차이가나타난다고보고되었으며 (Dziekońska et al., 23), C. rudolphii 성충의분비배설산물 (excretory-secretory products) 은숙주의상피조직에손상을줄수있으며, 대부분의선충에서나타나는몸체추출물 (somatic extracts) 의 glucosidases 활성은탄수화물유래에너지원으로작용한다고알려져있다 (Dziekońska et al., 2). 따라서, 본연구에서는아니사키스증을유발하는주요원인종인 A. simplex (s.s.) 와아니사키스증을유발하는증례는극히드물지만다양한해산어류와오징어에서 A. simplex (s.s.) 와함께흔하게발견되는 A. pegreffii 를대상으로각각의유충을배양하여 3기유충과 4기유충의분비배설산물 (excretory-secretory products) 과몸체추출물 (somatic extracts) 을얻은후, 아니사키스속선충의대사작용과관련된가수분해효소의활성을측정, 두종사이에서어떤가수분해효소가 A. simplex (s.s.) 와 A. pegreffii에서활성차이를나타내는지비교하였다.

3 어류기생성선충 Anisakis simplex sensu stricto 와 Anisakis pegreffii 유충의 excretory-secretory products 및 somatic extracts 의가수분해효소활성비교 27 재료및방법 Anisakis속선충의수집 A. simplex (s.s.) 3기유충은 211년 1월양양남대천으로회귀하는연어 (Oncorhynchus keta) ( 평균체장 : 69.3 cm, 평균체중 : 3.4 kg ) 의근육 (belly flaps) 으로부터, A. pegreffii 3기유충은 212년 1월부산자갈치시장에서구입한고등어 (Scomber japonicus) ( 평균체장 : 28.8 cm, 평균체장 : g) 의복강에서각각분리하였다. Excretory-secretory products의회수연어의근육에서분리된아니사키스속선충 1 마리와고등어의복강에서분리된아니사키스속선충 1마리를 3기유충과 4기유충의 ES products 회수를위해 마리씩나눈후, Iglesias et al. (22) 의방법에따라배양하였다. 즉, 외관상으로손상을입지않은유충을 PBS로 2~3회세척한후, 2% FBS(Lonza, Switzerland), antibiotic solution (Sigma, USA), 1% commercial pepsin(sigma, USA) 이포함된 RPMI-164(Sigma, USA) 배지를 ph 4. 으로조절한후 12 well plate 에각각의유충을옮긴후 37, % CO 2 조건에서각각 1일과 6일배양하였으며, 배지는매일교환하였다. 1일간배양한유충의배양액은 3기유충의 ES products으로, 6일간배양한유충의배양액은 4기유충의 ES products 로사용하였으며, 4기유충으로의탈피는꼬리부분의 mucron 이없어진것을현미경으로확인하여결정하였다. 각각의분리된배양액은 8 에보관하였으며, 사용시종류별로나눠배양액을 pooling 한후, Bradford(1976) 의단백질정량법에따라 1 ug/ml로적정하여가수분해효소활성측정을위한시료로사용하였다. Somatic extracts의회수 3기유충과 4기유충의배양에사용된배양액을제거하고, PBS로 2~3회 washing 한후, 남은유충을따로회수하여각각의충체들을절반으로잘라절반은 -8 에보관하였다가 A. simplex (s.s.) somatic extracts과 A. pegreffii somatic extracts으로나눠충체를 pooling 하였다. 각각의 pooling된시료 들은 on ice 상태에서 PBS 1mL을첨가한후, homogenizer(pt 12, Switzerland) 와 sonicator(vcx 13, USA; % amplitude, 1min) 를사용하여마쇄한후, 18, g에서 1분간원심분리하였다. 회수된상등액은 Bradford(1976) 의단백질정량법에따라 1 ug/ml로적정하여가수분해효소활성측정을위한시료로사용하였다 Anisakis속선충의동정 3기유충과 4기유충의 somatic extracts 으로사용하고남은 2 마리의아니사키스속선충유충은 Wasko et al.(23) 의방법에따라핵산을추출한후, D Amelio et al.(2) 이 rdna의 ITS region (ITS1-.8S-ITS2) 을타겟으로제작한 A ( -GTC GAA TTC GTA GGT GAA CCT GCG GAA GGA TCA-3 ) 와 B ( -GCC GGA TCC GAA TCC TGG TTA GTT TCT TTT CCT-3 ) primer를사용하여 PCR을실시하였으며, 증폭된산물은 HhaⅠ(NEB, UK) 과 HinfⅠ (NEB, UK) 제한효소로처리한후, 2% agarose gel을사용하여결과물을전기영동하고 UV transilluminator 상에서분절패턴을확인하였다. API ZYM 연어에서분리된 A. simplex (s.s.) 와고등어에서분리된 A. pegreffii 의 3기와 4기유충의 ES products 및 somatic extracts를 19종의가수분해효소기질을포함하고있는 API ZYM kit (Biomerieux, France) 를사용하여각각의 strips에 6 μl씩분주한후, 37 에서 4시간반응시켜각각의가수분해효소활성을반복구를두어비교하였다. 활성치는판독표를기준으로 ~4 nm 까지순차적으로기록하였으며, somatic extract의경우 PBS를, ES products의경우배양에사용된 RPMI-164 배지를분주한실험구를대조군으로사용하였다. 결과 PCR-RFLP 증폭된 PCR product를제한효소 HhaⅠ과반응시켰을때, 연어에서분리한아니사키스선충유충

28 전찬혁 위성 김정호 과고등어에서분리된아니사키스선충유충모두, 43 bp의분절을확인할수있었다. HinfⅠ제한효소를이용하여각각의패턴을비교해본결과에서는연어에서분리한아니사키스속선충유충의경우 62, 2 bp의분절을확인할수있었으며, 고등어에서분리한아니사키스속선충유충의경우 37, 3, 2 bp의분절을확인할수있어, 각각 A. simplex (s.s.) 와 A.

pegreffii 에서 종의가수분해효소에서활성이나타났다. N-acetyl-β-glucosaminidase와 α-mannosidase에서는 3 nmol과 2 nmol로비교적높은활성을나타냈지만 esterase lipase (C 8), naphthol- AS-BI-phosphohydrolase에서는두종모두에서 1 nmol 이하의비교적낮은활성이나타났다.

HinfⅠ제한효소를이용하여각각의패턴을비교해본결과에서는연어에서분리한아니사키스속선충유충의경우 62, 2 bp의분절을확인할수있었으며, 고등어에서분리한아니사키스속선충유충의경우 37, 3, 2 bp의분절을확인할수있어, 각각 A. simplex (s.s.) 와 A. pegreffii로동정하였다 (Fig. 1). API ZYM A. simplex (s.s.) 와 A. pegreffii 의 L3와 L4 ES products 및 somatic extracts의가수분해효소활성데이터는 Table 1에요약하였다. 3기유충 ES products에서는 A. simplex (s.s.) 에서 4종, A. pegreffii 에서 종의가수분해효소에서활성이나타났다. N-acetyl-β-glucosaminidase와 α-mannosidase에서는 3 nmol과 2 nmol로비교적높은활성을나타냈지만 esterase lipase (C 8), naphthol- AS-BI-phosphohydrolase에서는두종모두에서 1 nmol 이하의비교적낮은활성이나타났다. acid phosphatase의경우 A. simplex (s.s.) 에서는활성이나타나지않았지만, A. pegreffii 에서는 nmol의활성을확인할수있었다. 3기유충 somatic extracts의경우 A. simplex (s.s.) 와 A. pegreffii에서각각 7종의가수분해효소에서활성이확인되었다. A. simplex (s.s.) 는 esterase (C 4), leucine arylamidase 그리고 acid phosphatase에서 A. pegreffii 보다 1 nmol 높은활성을나타냈지만, A. pegreffii는 N-acetyl-β-glucosaminidase와 α-fucosidase에서 A. simplex (s.s.) 보다높은활성이나타났고, 특히 N-acetyl-β-glucosaminidase의경우 A. simplex (s.s.) 에서활성이나타나지않은데반해, A. pegreffii 에서는 2 nmol의비교적높은가수분해효소활성이나타났다. 4기유충 ES products에서는 A. simplex (s.s.) 와 A. pegreffii 모두 종의동일한가수분해효소에서활성이확인되었다. 두종모두 acid phosphatase와 N-acetyl-β-glucosaminidase에서 3 nmol 이상의높은활성을나타냈다. A. simplex (s.s.) 는 alkaline phosphatase 에서 A. pegreffii 보다높은활성을나타냈지만, A. pegreffii는 acid phosphatase와 α-mannosidase에서 A. simplex (s.s.) 보다높은활성을나타냈다. 4기유충 somatic extracts에서는 A. simplex (s.s.) 에서 7종, A. pegreffii 에서 6종의가수분해효소에서활성이확인되었다. A. simplex (s.s.) 는 alkaline phosphatase, esterase (C 4) 그리고 leucine arylamidase에서 A. pegreffii 보다높은활성이나타났고, A. pe- Fig. 1. Anisakis pegreffii L3 L4 L3 L4 ESP a SE b ESP SE ESP SE ESP SE greffii는 naphthol-as-bi-phosphohydrolase와 N-acetyl-β-glucosaminidase에서 A. simplex (s.s.) 보다높은활성을나타냈다. 고찰 우리나라의회충, 편충, 구충등의장내기생충감염율은집단검진과투약으로크게감소하고있으나, 어류를생식하는식습관이보편화되면서어류매개성질환은크게증가하고있는추세이다. 어류매개성질환은크게선충류, 조충류, 흡충류등에의한감염증을들수있는데, 조충류와흡충류의경우 Praziquantel 로구제가가능한반면, 선충류의경우 tiabendazole과 albendazole 등에서약제활성이있는것으로알려져있으나유효한치료제는 아직개발되어있지않다 (Chai, 21; D Amelio et al., 21). 하지만, 치료법개발에앞서아니사키스선충유충이사람에게감염되는메커니즘과이들유충의대사작용과관련된화학적특징에대해이해하는것도중요하다. 일반적으로기생충의단백질분해효소는숙주의혈액응고를억제하고, 숙주면역반응에서기생충을보호하며, 유충의부화와탈피능력을증가시킬뿐만아니라섭이작용에중요한역할을하는등여러기생충들의생활사와숙주관계에있어중요한역할을하며, 특히유충의숙주침투력과도관련되어있다고알려져있다 (Dziekońska et al., 23; Malagón et al., 211). 6 3 전찬혁 위성 김정호 정한결과 A. pegreffii가우점하는지역에서도 A. simplex (s.s.) 에의한감염이더높은것으로확인되었고, 한천배지위에 A. simplex (s.s.) 와 A. pegreffii 를올려놓고배지내로침투하는침투력을확인해보았을때 A. simplex (s.s.) 가 A. pegreffii 보다더높은침투력을가지는것으로확인되어, 근연종인 A. simplex (s.s.) 와 A. pegreffii 에서물리적침투력과관련된병원성에차이가있을것이라보고하였다. 한편, 단백질분해효소와가수분해효소의활성에관한연구는아니사키스과 (Family Anisakidae) 선충중 A. simplex, Contracaecum rudolphii 그리고 Hysterothylacium aduncum에서만보고되어있다 (Dziekońska et al., 23 and 2; Malagón et al., 211). 따라서, 본연구에서는아니사키스증을유발하는주요원인종인 A. simplex (s.s.) 와아니사키스증을유발하는증례는적지만다양한해산어류와오징어에서 A. simplex (s.s.) 와함께흔하게발견되는 A. pegreffii를대상으로사람에게감염될수있는유충단계인 3기유충과 4기유충의분비배설산물 (excretory-secretory products) 과몸체추출물 (somatic extracts) 을사용하여아니사키스속선충의대사작용과관련된가수분해효소의활성을측정, 두종사이에서어떤가수분해효소가 A. simplex (s.s.) 와 A. pegreffii에서활성차이를나타내는지비교해보았다. 본연구에서비교한총 19종의가수분해효소중 9종에서각기다른가수분해효소활성차이가확인되었다. Esterase 그룹의가수분해효소활성은 acid phosphatase, esterase (C 4), alkaline phosphatase 그리고 naphthol-as-bi-phosphohydrolase에서나타났다. acid phosphatase의경우 A. simplex (s.s.) 와 A. pegreffii 의 3기유충의 ES products를제외한모든그룹에서활성이높게나타났으며, alkaline phosphatase와 esterase (C 4) 의경우 A. pegreffii 보다 A. simplex (s.s.) 에서다소높은활성을나타내는것을확인할수있었지만, naphthol-as-bi-phosphohydrolase의경우 A. simplex (s.s.) 보다 A. pegreffii 에서같거나좀더높은활성을확인할수있었다. Ruitenberg and Loendersloot(1971) 는 acid phosphatase는 Anisakis sp. 의체벽과내장에서높은활성을나타내며, phosphatase는대사작용의제어에있어 중요하다고보고하였다. 본실험에사용된 A. simplex (s.s.) 와 A. pegreffii가성충이아닌 3기유충과 4기유충이므로 ES products과 somatic extracts에서각각높은활성이나타났다고생각되며, 체벽과내장에높은활성이나타난다고보고되었으므로 ES products 보다 somatic extracts에서다소높은가수분해효소활성을나타내는것으로생각된다. 또한, 3기유충보다 4기유충에서더높은가수분해효소활성을나타내는것은 3기유충에서 4기유충으로성장, 탈피하는것보다 4기유충에서성충으로성장, 탈피하는데더많은에너지원이필요하기때문이라고생각된다. naphthol-as-bi-phosphohydrolase 의활성은 A. simplex (s.s.), 돼지회충 (Ascaris suum), 그리고 sheep botfly (Oestrus ovis) 에서도보고된바있으나기능에대해서는보고된바없다 (Dziekońska et al., 23; Dziekońska, 26; Tabouret et al., 23). Liew et al.(212) 은그람음성균인 Burkholderia pseudomallei에서 naphthol-as-bi-phosphohydrolase의높은활성은세균의생장에있어필수적으로필요한 housekeeping enzyme의역할을할것으로추측하였는데, A. simplex (s.s.) 와 A. pegreffii에서도이와비슷한역할을할것으로추정된다. Aminopeptidase 그룹의가수분해효소활성은 leucine arylamidase와 valine arylamidase에서 A. pegreffii보다 A. simplex (s.s.) 의 somatic extracts에서활성이높게나타났으며, ES products에서는활성이나타나지않았다. Aminopeptidase는 A. suum이부화와탈피를하는동안호르몬과효소전구체의촉매제로매우중요한것으로알려져있으며 (Rhoads et al., 1997), 성숙한 A. suum의체벽, 난소, vulva, 내장그리고체강액에서발견되어소화와섭이작용과관련된것으로생각된다 (Lee, 1962; Rhodes et al., 1966). 본연구에서 A. simplex (s.s.) 에서 A. pegreffii 보다높은활성이나타난이유는 A. simplex (s.s.) 가 A. pegreffii 보다유충의체장과 ventriculus 의길이가길기때문에 (Quiazon et al., 28), 그에따른소화와섭이작용이더많이이루어지기때문이라고추정된다. N-acetyl-β-glucosaminidase 의경우 A. simplex (s.s.) 와 A. pegreffii 의 ES products 에서 3 nmol의비교적높은가수분해효소활성이나타났으며, somatic extracts의경우 A. simplex (s.s.) 에서는가수분해효소활성이나타나지않았지만, A. pegreffii의 3기유충 (2 nmol) 과 4기유충 ( nmol) 에서가수분해효소활성이나타났다. α-mannosidase와 α-fucosidase는 A. simplex (s.s.) 보다 A. pegreffii에서같거나좀더높은활성이확인되었는데, α-mannosidase 는 ES products에서, α-fucosidase 는 somatic extracts 에서만각각의활성이나타났다. glycosidase 그룹의가수분해효소중가장높은활성을나타낸 N-acetyl-β-glucosaminidase는 hyaluronic acid 소화물의가수분해에서중요한역할을하며, 결합조직의주요성분중하나로써기생충이숙주로침투하는과정을돕는것으로알려져있다 (Hotez et al., 1994). Nisbet and Billingsley(2) 는닭진드기 (Dermanyssus gallinae) 와 flour mite (Acarus siro) 의추출물에서 3 nmol 이상의높은가수분해효소활성을확인하였고, 다른두종의진드기 (Psoroptes ovis, Tetranychus urticae) 에서도 nmol 이상의가수분해효소활성을확인하였는데, N-acetyl-β-glucosaminidase는모든무척추동물의탈피과정에있어중요한역할을한다고보고하였다. 또한 A. suum이돼지에감염되었을때근육, Perienteric fluid, 내장기관, 내장벽추출물에서동일하게높은활성이보고되었다 (Dziekońska, 26). N-acetyl-β-glucosaminidase는대부분 ES products를통하여소화기관에서분비되며유충의탈피와관련되어있을것이라판단되지만, A. pegreffii 의 3기유충의 somatic extracts에서나타난가수분해효소활성에대해서는좀더추가적인연구가필요할것이라판단된다. 본연구에서는 API ZYM kit를사용하여 A. simplex (s.s.) 와 A. pegreffii의가수분해효소의활성을비교해본결과 6종의 Esterase 그룹의가수분해효소중 종의가수분해효소에서활성이나타났는데, naphthol-as-bi-phosphohydrolase 를제외한 4가지의가수분해효소에서 A. pegreffii 보다 A. simplex (s.s.) 에서유사하거나더높은가수분해효소활성이확인되었고, Aminopeptidase 그룹의가수분해효 소에서는활성이나타나지않은 cystine arylamidase 를제외한 2 종류의가수분해효소에서 A. pegreffii 보다 A. simplex (s.s.) 에서높은가수분해효소활성이확인되었다. 그리고 Glycosidase 그룹의가수분해효소의경우대부분활성이확인되지않았지만, N-acetyl-β-glucosaminidase, α-mannosidase 그리고 α-fucosidase 에서 A. simplex (s.s.) 보다 A. pegreffii에서더높은활성이확인되었다. 본연구를수행한결과, A. simplex (s.s.) 와 A pegreffii간의성장, 탈피, 섭이및소화작용과관련된가수분해효소활성차이가확인이되었으나, 실제사람의아니사키스증의유병율과관련된두종간의가수분해효소활성차이의특별한경향은찾아볼수없었다. 차후다양한단백질분해효소의활성을비교함으로써, 두종간의병원성및대사작용과관련된매커니즘을이해하는데도움이될것으로생각된다. 요약 Anisakis simplex sensu stricto와 A. pegreffii의 3기유충과 4기유충에서얻은 excretory-secretory (ES) products 및 somatic extracts의가수분해효소활성을 API ZYM kit를이용하여비교하였다. Esterase 그룹의가수분해효소중 acid phosphatase는 A. simplex (s.s.) 와 A. pegreffii 모두에서높은활성을나타냈으며, esterase (C 4) 의경우 somatic extracts에서만가수분해효소활성이나타났는데 A. simplex (s.s.) 가 A. pegreffii와비교하여 3기와 4 기유충모두에서 2배가량높은활성을나타냈다. alkaline phosphatase, acid phosphatase 그리고 naphthol-as-bi-phosphohydrolase 의경우 A. simplex (s.s.) 와 A. pegreffii 모두 3기유충보다 4기유충에서더높은가수분해효소활성이확인되었다. Aminopeptidase 그룹의가수분해효소활성은 leucine arylamidase에서관찰되었는데, somatic extracts의경우 A. pegreffii 보다 A. simplex (s.s.) 에서가수분해효소활성이두배가량높게확인되었으며, 대부분의다른효소들에서는활성이거의나타나지않았다.
