A challenge test was conducted using 1,708 olive flounder fingerlings obtained through 4 4 factorial cross from parents selected based on resistant degree. Regarding the means of resistant index, survival rate and days of survival by each mating group, the group(rr) produced by mating among resistant parents were 17.98, 13.77%, and days, respectively; the group(ss) produced by mating among susceptible parents were 12.46, 2.71%, and days, respectively. This study gives an indication that challenge test results may be successfully used as selection criteria for disease resistance to E. tarda in olive flounder, P. olivaceus. Key words : Olive flounder, Paralichthys olivaceus, Edwardsiella tarda, Selective breeding, Disease Resistance, Challenge test 우리나라의넙치양식은 1990년대초반부터동해및남해안과제주도연안에서발달하기시작하였다. 뿐만아니라고도의양식기술이개발되고있으며시설의규모화가진전되어지금까지주요양식산업으로자리잡고있다. 이와같은넙치양식산업의급속한발전으로양식넙치의생산량은 2008년을기준으로약 4만 7천톤으로전체어류양식생산량의 3.4% 에불과하나, 생산금액은 4,083억원으로전체양식생산금액의 26.8% 를차지하게되었다 ( 통계청 어업생산통계, ). 그러나최근에이르러잦은질병의발생과이 Corresponding Author : Jeong Ho Lee, Tel : Fax : , jhlee@nfrdi.go.kr 로인한집단폐사, 치료를위한항생제의과다한사용등은그자체비용뿐아니라궁극적으로환경오염과소비자들의건강에위해를가할수있는요소로언급되는등양식어류의불신으로이어져양식산업경쟁력약화의주요원인이되고있다. 이에따라넙치양식의안정적인생산을위한대안으로우수한형질을지닌개체를선발하여이들의교배를거듭하는전통적인선발육종방식이연구되어지고있다. 이는양질의어미확보및관리와더불어우수한품종을개발하여장기적이고체계적인육종연 275

2 276 김현철 손새봄 정종근 이정호 최혜승 노재구 박철지 민병화 김종현 김경길 명정인 구로써오랫동안이에대한연구가이루어지고있으며, 이를활용하여질병에대한저항성이향상된개체의생산가능성을제시하였다 (Cipriano and Heartwell, 1986; Ehlinger 1964, 1977; Embody and Hayford, 1925; Gjedrem and Aulstad, 1974; Gjedrem, 1983; Standal and Gjerde, 1987). 최근대서양연어 (Gjedrem and Gj en, 1995; deg rd et al., 2006), 무지개송어 (Henryon et al., 2002, 2005), 잉어 (Das Mahapatra et al., 2008), 대서양대구 (Kettunen and Fjalestad, 2006) 에대한연구는질병내성의향상에대한실현가능성을한층더높여주었다. Edwardsiella tarda는많은담수어나해산어에질병을일으키는에드워드증 (edwardsiellosis) 의원인균으로장내세균과에속하는그람음성세균이다 (Thune et al., 1993). 우리나라의주요양식산업대상어종인넙치에서보고된바있으며 (Nakatsugawa, 1983), 주요증상으로복부팽만과탈장, 간과신장의농양형성등전신적증상을나타내며많은폐사와함께양식산업에큰경제적손실을발생시키고있다. Ewing et al. (1965) 에의해E. tarda가어류병원체로보고된이후병원체의감염기작, siderophores, cell adhesion factors, cell invasion activity, two types of haemolysin와같은잠재적병원성인자, 항생제, 백신에대한연구가활발히진행되고있다 (Mohanty and Sahoo, 2007). 그러나명확한메카니즘이나치료대책이마련되고있지않아이에대한다른방향으로의연구가시급한실정이다. 본연구는넙치양식에있어질병에내성을 지니는우수한품종을개발하기위한품종개량의기초연구로넙치질병내성검증가계를대상으로 E. tarda를인위감염시켜내성지수, 생존율및생존일수를조사함으로써넙치의질병내성에대한가계별내성변이를검증하여양식되고있는넙치의유전적개량을위한기초자료를제공하기위해실시하였다. 시험균주 재료및방법 시험균주는경남지역의양식생물질병역학조사에서간충혈, 신장비대, 복수증상을나타내는넙치에서분리된 E. tarda를사용하였다. 배양은 1% mannitol 이첨가된 Salmonella-Shigella (SS, Difco) 배지에서 25, 48시간배양후흑색 colony를 1.5% NaCl 첨가한 Tryptic soy agar (TSA, Difco) 에도말하여 27, 24시간배양후인위감염실험에사용하였다. 시험어 Table 1. Number of offspring identified for each of the families generated in the full factorial cross Dam D1 D2 D3 D4 Observed total per sire Sire S S S S Observed total per dam ,705 수정가능정도에따라각각 2 가계를최종선정 하여 4 4 팩토리얼디자인에의해 16 가계를 생산하였다 (Table 2). 자치어는동일한환경에서사육하기위해각 가계당 3 ml수정란을 FRP 5 톤사육수조에혼합 하여사육하였다. 먹이로는부화직후소량의클로렐라를사육수에혼합하였고, 부화후 3일령부터는로티퍼를공급하였으며, 성장함에따라알테미아와배합사료를공급하였다. 사육수의관리는배합사료공급이전에는지수식으로, 그이후부터는일부환수와유수식으로관리하였다. 사육기간동안의수온은 17.6~20.4, 염분은 30.7~34.8 psu, DO는 6.0~7.9 mg /l 범위였다. 인위감염시험 시험균주에대한넙치의폐사율을조사하기위하여부화후 118일령까지사육한넙치종묘 를이용하였으며, 인위감염시체장은 10.5~15.2 cm범위였다. 인위감염시험은총 1,708마리의넙치종묘를동일수조에서 cfu/ ml농도의시험균주로 2.5시간동안침지하여실시하였다. 수온은 24 ± 0.5 로유지하였으며, 폐사율은 1일에 4회씩 34일간기록하였다. 폐사어는폐사원인을확인하기위해폐사즉시해부하고간, 신장과비장조직을무균적으로절취하여 Salmonella-Shigella (SS, Difco) 배지에도말한후 E. tarda를재분리하였다. 인위감염후친자확인된개체수는 1,705마리였으며, 가계별개체수는 Table 3과같다. 친자확인 친자확인의시료는계측형질측정시각개체의넙치가슴지느러미조직을절취한다음 phenol extraction 방법으로 DNA를추출하여 multi-

4 278 김현철 손새봄 정종근 이정호 최혜승 노재구 박철지 민병화 김종현 김경길 명정인 plex PCR 반응에사용하였다. 유전자서열분석기 (ABI 3100) 를이용하여 8개유전좌위에대한각개체의유전자형을결정하였으며, 친자확인은멘델의유전법칙에위배되는친어를배제시키는방법 (Exclusion method) 으로친어및자손의유전자형을분석하여추정된부모조합을교배지침과비교하는방법으로실시하였다. 하였다. 조사형질을대상으로교배그룹, 교배암컷, 교배수컷의효과를분석하기위해 SAS Package (Ver. 8.2) 를이용하여 ANOVA-test를실시하였으며, 평균치간의유의성검정은 Duncan's multiple range test를실시하여귀무가설을유의수준 5% 로각각검정하였다. 통계학적분석 교배그룹별, 교배암컷별, 교배수컷별내성지수 (Resistant index), 생존율 (Survival rate), 생존일수 (Days of survival) 를조사하였으며, 내성지수, 생존율및생존일수는아래의공식을이용 Fig. 1. 5 E. tarda 인위감염에의한넙치의가계별내성변이 279 타나내성친어간의교배에의해생산된그룹이다른그룹에비해유의적으로내성지수가높은것으로나타났으며, 민감친어간의교배에의해생산된그룹이유의적으로내성지수가낮은것으로나타났다 (p<0.05). 생존율의경우, RR, RS, SR, SS가각각13.77 ±5.85%, 3.66±1.40%, 7.32±3.46%, 2.71± 0.74% 로나타나 RR 그룹이 RS, SR, SS보다각각 10.1%, 6.5%, 11.1% 생존율이높은것으로나타났다 (p<0.05). 생존일수의경우, RR, RS, SR, SS가각각18.36 ±0.51일, 15.33±0.42 일, 14.56±0.35일, ±0.35일로나타나동일인위감염조건하에서 RR 그룹이 RS, SR, SS보다각각 3.0 일, 3.8 일, 6.0 일을더생존한것으로나타났다 (p<0.05). 교배암컷의효과 교배에사용된암컷에대한내성지수를조사한결과 (Table 5, Fig. 2), 내성암컷 D1, D2가각각 15.66±0.66, 17.67±1.86으로나타났으며, 민감암컷 D3, D4는각각 14.13±0.87, 13.15± 0.81로나타나내성암컷 D2가민감수컷D3, D4보다유의적으로내성지수가높은것으로나타났다 (p<0.05). 생존율의경우, D1, D2, D3, D4가각각6.43± 2.52%, 11.00±6.58%, 7.78±3.22%, 2.25±0.81% 로나타나내성암컷 D2의생존율이가장높은것으로나타났으나유의적인차이는보이지않았다. 생존일수의경우, D1, D2, D3, D4가각각 15.63±0.44일, 18.28±0.50일, 13.71±0.50일, 13.38±0.35일로나타나동일인위감염조건하에서내성암컷 D2이 D3, D4보다각각 4.6일, 4.9일을더생존한것으로나타났다 (p<0.05). 교배수컷의효과 교배에사용된수컷에대한내성지수를조사한결과 (Table 6, Fig. 3), 내성수컷 S1, S2가각각 17.58±1.68, 15.21±0.99로나타났으며, 민감수컷 D3, D4는각각14.97±1.12, 12.85±0.60으로나타나내성수컷 S1이민감수컷S3, S4보다유의적으로내성지수가높은것으로나타났 Table 5. Survival rates of olive flounder challenged with E. tarda by mating dam (D1: Resistant dam 1, D2: Resistant dam 2, D3: Susceptible dam 1, D4: Susceptible dam 2) 다 (p<0.05). 생존율의경우, S1, S2, S3, S4 가각각 16.38± 5.40%, 4.71±1.13%, 3.58±1.19%, 2.79±1.07% 로나타나내성수컷 S1 의생존율이민감수컷 S3, S4 보다각각 12.8%, 13.6% 높은것으로나 타났다 (p<0.05). 생존일수의경우, S1, S2, S3, S4 가각각 ±0.51 일, 14.86±0.35 일, 15.18±0.48 일, 12.73± 0.32 일로나타나동일인위감염조건하에서내 성수컷 S1 이 S3, S4 보다각각 2.2 일, 4.7 일을 더생존한것으로나타났다 (p<0.05). 고 찰 선발육종은수산생물의유전적개량을위한 가장일반적이고효과적인방법으로이용되고있으며, 특히어류및패류에서선발육종및육종효율예측을목적으로중요경제형질들에대한양적유전학적평가가적용되어지고있다 (Argue et al., 2002; Gjerde et al., 2004; Zheng et al., 2006). 본연구는넙치양식에있어질병에내성을지니는우수한품종을개발하기위한품종개량의기초연구로실시되었다. 에드워드질병내 Fig. 3. Survival rates of olive flounder challenged with E. tarda by mating sire (S1: Resistant sire 1, S2: Resistant sire 2, S3: Susceptible sire 1, S4: Susceptible sire 2) 성검증을위하여 2007년에실시한육종넙치핵집단 2세대의인위감염결과와친자확인결과를기초로가계별내성지수를계산하였으며, 이를근거로내성가계와민감가계를선정하였다. 질병내성가계와민감가계각각 2가계, 암 수각각 4마리를선정하여 4 4 팩토리얼디자인에의해 16가계를생산하였으며, 동일수조에서부화후 118일까지사육한후 E. tarda에인위적으로감염시켜매일발생하는폐사개체에대하여친자확인을실시하였다. 친자확인결과를기초로내성지수, 생존율, 생존일수에영향을미치는교배그룹, 교배암컷, 교배수컷의효과를분석하였다. 내성암컷 2마리와내성수컷 2마리간의교배에의해생산된교배그룹이내성지수, 생존율및생존일수모든조사형질에서다른교배그룹보다유의적으로내성이높은것으로나타났으며, 민감친어간의교배를통해생산된교배그룹은유의적으로질병내성이낮은것으로나타났다. 이와같은결과는 2007년에실시한핵집단의인위감염실험결과를재현하는것으로에드워드내성관련형질도일반적인성장형질과마찬가지로선발대상형질로이용이가능할것으로판단된다.

7 E. tarda 인위감염에의한넙치의가계별내성변이 281 교배에사용된친어개체중내성암컷 D2 를 이용하여생산된후대의모든에드워드내성관련형질에서다른교배암컷에비해내성이높은것으로나타났으며, 반대로교배암컷 D4를이용하여생산된후대의에드워드내성은낮은것으로나타났다. 내성수컷인 S1을이용하여생산된후대의모든에드워드내성관련형질에서다른교배수컷에비해내성이높은것으로나타났으며, 반대로교배수컷 S4를이용하여생산된후대의에드워드내성은낮은것으로나타났다. 따라서 2007년도핵집단의인위감염실험을통해선발된 D1, D2, S1, S2 친어가운데서도각친어의개체별내성정도에서차이가나타나는결과는가계별질병내성정도를분석하여가계선발하는것도중요하지만, 팩토리얼교배로후대생산을통한개체선발도병행하는것이바람직하다는것을나타낸다. 이상의결과에서동일한환경조건에서에드워드질병감염실험에의한질병내성정도를확인한결과, 가계간, 개체간내성정도의차이가존재하는것을확인하였다. 이는질병내성관련형질도유전적인변이가존재하여선발대상형질로적합하다고판단되며, 보다정확한선발을위해각형질별로유전력을기초로한육종가를추정하여이를기준으로선발하는것이바람직할것으로판단된다. 요 약 본연구는에드워드내성정도를기준으로선발된넙치친어를이용하여 4 4 펙토리얼교배를통해얻어진 16가계넙치의에드워드인위감염결과를이용하여교배그룹, 교배암컷, 교배수컷의효과를분석하였다. 내성친어간의교배에의해생산된교배구 (RR) 의내성지수, 생존율, 생존일수는각각 17.98, 13.77%, 일로나타났으나, 민감친어간의교배에의해생산된교배구 (SS) 는각각12.46, 2.71%, 12.40일로나타났다. 이상의결과에서, 에드워드질병 내성형질에대한가계간, 개체간내성정도의차이가존재하는것을확인하였고, 인위감염결과를이용하여질병저항성에대한선발이가능할것으로판단된다. 감사의글 이연구는국립수산과학원 ( 육종기술개발, RP AQ-091) 의지원에의하여연구되었습니다. 참고문헌 Argue, B.J., Arce, S.M., Lotz, J.M. and Moss, S.M.: Selective breeding of Pacific white shrimp (Litopenaeus vannamei) for growth and resistance to Taura Syndrome Virus.
