Principal component analysis and partial least squares were applied to the H- NMR profile to reduce its complexity and establish class-related clusters. Relative lipid regions were distinguished in vaccinated and non-vaccinated serum. Then, the lipids were extracted from the serum and analyzed. Triolein was identified. Key words: Metabolomics, PCA, PLS, Triolein, Edwardsiella tarda, NMR 서 론 최근동식물의대사물질변화분석을통해대사경로의조절과생화학적대사경로를이해하고자 metabolic profiling 의연구가활발하게이루어지고있다 (Kim et al., 26). 대사물질의변화를분석하기위해 HPLC, GC, MS 등여러장비들이활용되고있지만복잡한전처리과정과많은시간이소요된다. 이에비해핵자기공명분석 ( H-nuclear magnetic resonance: H-NMR) 은전처리과정이간단하며정성및정량분석이가능할뿐아니라재현성이뛰어나다. 또한여러화합물질로부터특정물질을분석하고다양한조직과혈청으로부터대사체의정보를얻을수있는유용한분석법으로 (Ala-Korpela, 995; Griffin & Kauppinen, 27), 다변량통계분석기법을적용해데이터베이스및라이브러리를통하여복잡한화학정보로부터유용한정보를유추및해석할수있다 (Brindle et al., 23). 이러한방법을 metabolomics 로정의하고있으며주요대사체군의변화및규명을가능하게한다 (German et al., 25). 지금까지의 metabolomics 연구들은대부분이인체대사물질변화에관한연구와식물의표현형차이에대한연구들로국한 되어있고어류에관한연구는보고된바가없다. 따라서본연구는 H-NMR spectroscopy 와다변량통계분석을통해넙치의대사물질변화를분석하기위해고안되었다. 우리나라의대표적인양식어종인넙치 (Paralichthys olivaceus) 는고밀도사육으로인한 Edwardsiella tarda, Vibrio anguilarum 및 Streptococcus iniae 등과같은세균성질병이보고되고있다. 그중 E. tarda 는지느러미및복부발적및충혈, 복부팽만탈장등을나타내는에드워드감염증 (edwardsiellosis) 의원인균으로성체의집단폐사를야기시켜양식에큰문제가되고있다 (Austin & Austin, 999; Castro et al., 26; Mohanty & Sahoo, 27). E. tarda 의감염을예방하기위해서는백신접종이가장효과적이고합리적인방법으로여겨지고있으며최근에백신개발에관한연구가활발하게이루어지고있다 (Kawai et al., 24; Liu et al., 27; Kim et al., 29). 본연구는넙치의백신접종에따른대사물질의변화를분석하기위한실험으로백신접종전후의대사물질을비교분석하고백신에반응하는물질 (vaccine-response substance; VRS) 을분석하여 indicator 로서선정하고자하는것이그목적이다. 대사물질의변화분석을위해서는수소핵자기공명분 Article history; Received 8 March 22; Revised 9 June 22; Accepted 3 August 22 *Corresponding author: Tel: Fax: address: jycho@sch.ac.kr Kor J Fish Aquat Sci 45(4) , August 22 pissn:374-8 c The Korean Society of Fishereis and Aquatic Science. 2 368 조지영 석 ( H-nuclear magnetic resonance: H-NMR) 과주성분분석 (principal components analysis: PCA) 및부분최소제곱분석 (partial least square: PLS) 을실시하였다. 실험어사육 재료및방법 실험어는양식장 ( 극동수산, 경북포항 ) 에서생산된넙치 ( 평균 5±7 g) 를사용하였고 2 리터원형수조에동일한사육밀도로 3 마리씩수용을하였다. 사료는배합사료 ( 조단백질 52% 이상 ) 를사용하였으며사료공급량은어체중의.-3.5% 를 일 회공급하였으며급이횟수, 수온, 환수량등모든양식조건을동일하게사육하였다. 실험기간중의사육수온은 22± 를유지하였다. 백신제작 E. tarda 를 % NaCl 이첨가된 Brain Heart Infusion broth (BHIB) 에접종하여 25 에서 24 시간동안배양하였으며세균배양액에포르말린 (37% formaldehyde) 을최종적으로.3% 농도가되도록처리하여실온에서 시간정치한후 4 에서 24 시간동안두어불활화시켰다. 불활화된세균배양액은 6,52 rcf 에서 2 분동안원심분리하여수확하였고.2% NaN3 가첨가된멸균생리식염수에습중량 mg/ml 의농도로희석하여 4 에보관하였다. 백신투여 포르말린으로불활화시킨 E. tarda 를멸균생리식염수로희석한후 MS-222 로마취된넙치의복강내에. ml 를주사하였다. 백신효능검증의어체크기는직경 5 cm, 무게 5 g 으로주사가가능한가장작은크기의어체로선택하여실험을진행하였으며백신의농도는 mg/kg bodyweight 로실험을진행하였다. 응집가반응 (Kim et al., 29) 을통해항체가형성여부를확인하였다. 혈청분리 넙치를 MS-222 로마취한후미부혈관에서채혈하여상온에서 시간, 4 에서 24 시간방치한후 5,9 rcf 에서 분간원심분리하여상등액을분리하였다. 분리된혈청은 -8 에나누어보관하며분석에사용하였다. H-NMR 분석 혈청은진공농축기 (JEIO Tech, Korea) 를이용하여농축하였고용매 D2O 에녹여 2 µl 의샘플을 5 mm Shigemi NMR tube 에주입하였다. H-NMR spectra 의측정은 Jeol JNM- ECP 5 NMR spectrometer 를사용하여 5 MHz 에서측정하였다. NMR 분석은 nt 32, 63, 28 scan 으로실시하여최적의분석조건을확인하였으며온도에따른 peak pattern 의변화를알아보기위하여 28 scan 을기준으로하여온도 2, 25, 3 로설정하여실행하였으며 splitting 이가장많은 ppm 영역의 peak 들을비교하였다. 모든실험은 5 반복구로실행하였다. Pulse sequence 는 RD-9 -t-9 -tm- 9 -FID 로하였으며.5 s 의 relaxation delay 로실시하였다. NMR spectra 는 -2 ppm 까지 scan 하였고 X-WINNMR (version 2., Bruker Gmbh, Germany) 을이용하여 calibration 을실시하였다. 다변량통계분석 각각의혈청으로부터얻어진 H-NMR 스펙트럼은 peak 의 chemical shift 와 split pattern 그리고 integral 값을이용하여정리하였고 data 간격은 ppm 부터 2 ppm 으로조정하였다. H-NMR 스펙트럼데이터는 Matlab (version 6.5) 에도입하였고 Unscramber (version X.) 이용하여 total metabolite 에대한주성분분석 (PCA) 과부분최소제곱법 (PLS) 을실시하였다. PCA 는자료의변수들이가진정보를최대한확보할수있는적은수의새로운변수를생성하는것으로새로운변수를주성분 (principal component) 이라하며자료가보여주는변이를최대한설명할수있는주성분을선택한다 (Massart and Vandeginste 988; Joo and Cho 25). PC 는 scores 와 loading 값으로나타내며 PCA 분석을통해서대조구대비실험구의차별성및신뢰성을확인하였다 (Brindle et al. 23). PLS 는작은변이를설명하는변수를제외시킴으로써차원축소를통해안정적인결과를얻을수있는방법으로 PLS 분석을통하여 score plot 에서 clusting 을확인하고 loading plot 에서 indicator 로추측되는인자를탐색하며 VIP value 를이용하여인자의중요도를확인하였다. PCA 와 PLS-DA 분석에서 component 는 7 로설정하였으며 non-linear iterative partial square algorithm (NIPALS) 으로실행하였다. VRS 성분분리및분석 H-NMR 분석과다변량통계분석을통해백신을첨가한경우와그렇지않은경우에지질함량이차이가나는것을확인하였고혈청으로부터 VRS 성분을분리하기위해우선 hexane 을이용하여지질층 (total lipid) 을분리하였다. Hexane 추출층에메탄올을첨가하여극성성분을완전히제거하였고, 비극성층을 TLC (thin layer chromatography) 를이용하여전개용매 hexane/ether (6:) 로분리하였다. 대조군에비해함량에차이를보이는 band (R f.7) 를 H-NMR 과 3 C-NMR, FAB-MS 를이용하여구조분석을실시하였다.

3 다변량통계분석을통한넙치의혈청성분분석 369 결과및고찰 넙치혈청성분의변화를분석하고 health indicator 를찾기위해 E. tarda 불활화백신을넙치의복강내주사하였다. 백신접종 4 일후혈청을분리하여응집가반응을통해불활화백신에대한항체가형성되었음 (2 antibody titer) 을확인하였 ppm Fig.. Arrows indicate distinguished peak regions from non vaccinated fish serum. 고 H-NMR 분석에사용하였다. 온도에따른 H-NMR peak 의 pattern 을확인하기위해온도조건을 2, 25, 3 로설정하여 splitting 이가장많은 ppm 영역의 peak 들을비교하였다. Peak pattern 은같았으며온도변화에따른 peak pattern 에변화가없는것으로판단되어분석온도는 25 로설정하였다. 백신을처리한경우의혈청과백신을접종하지않고사육한넙치의혈청의 H-NMR 은 Fig. 에나타내었고.65 부터 5.8 ppm 에서 peak 를확인할수있었다. 대부분의 chemical shift 에서처리군과대조군의차이를나타나지않았으며.65 ppm 과 ppm 에서차이를나타내었다. 각 peak 의 integral 값과 split pattern 및 relative compounds (metabo-types) 는 Table 에나타내었다..65 ppm 은 methlyl 기를나타내며가장많은 integral 값을가지는 δ.53 은지질성분의반복되는 methylene [(2)n] 을나타낸다..53 ppm 부터 3.2 ppm 은 lipid 성분의 methlylene 의 peak 이며 3.4 ppm 부터 4.38 ppm 은아미노산과 glycerol 의 methine 으로확인된다. 두실험구의 NMR spectra 는 methine 부분은유사한형태로나타나고있으며 methylene 부분의 peak 값의변화를확인할수있다. H-NMR 을통하여얻은 metabolite profiling 의다변량분석을위해 PCA 와 PLS-DA 분석을실시하였다. Table. 4 37 조지영 PC-2 PC-2 PCA 분석의 loading plot 과 scores plot 을나타냈으며 sample -5 는대조구의 sample 이며 sample 6- 은실험구 ( 백신접종후얻은혈청 ) 의 sample 로나타내었다. Scores plot 에서실험구와대조구 sample 의확연한차이를보이며 (Fig. 2-a) loading plot 의 correlation 결과.65,.53,.64, 2.5, 2.22, 5.2 ppm 의 peak 가중앙값에서가장멀리나타나다른 peak 들과구분되는것을확인할수있다 (Fig. 2-b). PLS 분석에서는 PCA 에서보다대조구와실험구의샘플이더욱명확하게구분되는것을확인할수있어 regression coefficients 의 VIP value 를확인한결과.65,.53, 2.5, 2.22 ppm 의 peak 가가장두드러지게구분되었으며이 peak 들은지질성분의나타내는 methylene 과 methyl 기로확인되었다 (Fig. 3). PLS regression coefficients based on data from H- NMR spectra. 수있었고 TLC 를통해대조군과구별되는 band 를분리하여구조분석을실시하였다. 물질은 FAB-MS 분석결과분자량이 로나타났으며 NMR ( H, 3 C) 과 MS 를통해분자식은 C57H4O6 로확인되었다. 3 C-NMR 을통해 4. 과 4. ppm 사이에서 3 개의지방산 methyl carbon (C-8, 8 8 ), ppm 사이의 9 개의 methylene carbon (C-(3,4,7 ) 2, C-3, 4, 7 ), ppm 사이의 8 개의 methylene carbon (C-(5, 6,7,2-5 ) 2, C-5, 6,7,2-5 ), ppm 사이의 2 개의 methylene carbon (C-(2,8,, ) 2, C-2,8,, ) 을확인하였고, 3.5 ppm 에 6 개의 methine carbon (C-(9, ) 2, C-9, ) 을확인하였다. 또한 65. ppm 에 2 개의 glycerol methylene carbon (C-, C-3), 69.5 ppm 에 glycerol methine carbon (C-2) 을확인하였고 73.5 ppm 에서 3 개의 carbonyl carbon (C- 2), C- ) 을확인하였다. H-NMR 에서는 3 C-NMR 에상보되는 3 개의지방산 methyl proton 과 42 개의 methylene proton, 6 개의 methine proton 을나타냈으며 2 개의 glycerol methylene proton 과 개의 glycerol methane proton 을확인하였다. NMR 과 MS 의 data 분석결과 glycerolipid 로확인되었으며 Ramsewak et al. (2) 가보고한문헌의자료와비교한결과 triolein 으로확인되었다 (Fig. 4). 몇몇문헌에서특정노출에서어류의혈청성분중 triglyceride 의변화를보고한바있다. Tilapia Nilotica (Oreochromis niloticus) 의사육에서항산화효과는증대를위해마늘을섭이시킨경우혈청내 triglyceride 의함량이감소하였으며 (Metwally, 29), African catfish (Clarias gariepinus) 의양식에서 potassium permanganate (KMnO4) 으로 chemotherapeutant 를시킨경우, 역시혈청내 triglyceride 의함량은감소하였다 (Kori-Siakpere et al. 2). 어류의혈청에서 triglyceride 는내재성의에너지원으로 (Jeon et al., 995) 혈청내에서의변화는이화작용의결과로나타난것으로볼수있다. 본연구를통해넙치에백신을접종한경우항체가는증가하였고 triglyc-
