The total aflatoxins were quantified by the immunoaffinity column clean-up method followed by high performance liquid chromatography (HPLC)-fluorescence detector (FLD). Aflatoxins were found in 45(8.5%) samples including 34 nuts and nut products, 7 soybean pastes, meju, bean product and corn snacks with a range of 0.0 ~ 3.96 µg/kg. These results show that the contamination level of aflatoxin in foods consumed in Korea is low compared with the standard in Korea Food Code(0 µg/kg as aflatoxin B ). Aflatoxin B content was increased in peanuts and corn snacks during storage but it was decreased in doenjang (soybean paste). Key words : aflatoxin, immunoaffinity column, HPLC-FLD 곰팡이독소 (Mycotoxin) 는 Aspergillus속, Penicillium속및 Fusarium속에의해서생성되는독소이며, 아플라톡신 (aflatoxin) 은 Aspergillus flavus, Asp. parasiticus, Asp. nomious 등의 Aspergillus속곰팡이에의해생성되는 차대사산물이다. 지금까지 0여종의아플라톡신이알려,) 져있으나일반적으로발견되는아플라톡신은 B, G 로서주로곡류, 두류, 견과류등농산물과그가공품에서광범위하게발견되고있다. 이중에서도아플라톡신 B 은발암성, 기형유발, 간장독성등을유발하는가장강력한독성물질로, 국제암연구회 (IARC, International Agency for Research on Cancer) 에서 group (carcinogenic to humans) 으로분류하고있다 3). 미국, 영국, 일본등선진국에서는아플라톡신을비롯한많은곰팡이독소에대하여활발한연구가이루어지고있으며특히아플라톡신에대하여는농산물, 가공식품및사료등에 5~50 ppb 수준으로허용기준을설정하여관리하고있다. 아플라톡신에대한각국의관리현황을살펴보면유럽연합 (EU) 의경우땅콩, 견과류, 건조과일류, 곡류, 가공 *Correspondence to: Min-Jung Parkyeonggi-do Institute of Health and Environment, North branch, #66, Je Chengsa -ro, Uijeongbu-cityyeonggi-do, , Korea Tel: , Fax: enrogel@gg.go.kr 식품등에대하여아플라톡신 B 으로서 ~8 µg/kg, 총아플라톡신으로서 4~0 µg/kg을최대기준으로정하여규제하고있으며, 미국식품의약국 (FDA, Food and Drug Administration) 에서는브라질넛, 식품, 땅콩, 땅콩가공품, 피스타치오에대하여총아플라톡신으로서 0 µg/kg의최대기준치를정하고있고, 국제규격위원회 (CODEX) 에서는아몬드, 헤이즐넛, 피스타치오, 비가공땅콩에대하여총아플라톡신으로서 5 µg/kg의기준치를설정하고있다. 우 4-6) 리나라에서도곡류, 두류, 견과류및그단순가공품에대하여아플라톡신 B 으로서 0 µg/kg의기준을설정하여관리하다가최근에된장, 고추장, 고춧가루와땅콩및견과류를함유한과자류까지확대하여규제를강화하고있다 7,8). 곡류를주식으로하고있고많은식품과원료농산물을수입에의존하고있는우리나라의경우아플라톡신에노출될가능성이높을것으로예상되므로유통되는식품에대한아플라톡신오염도조사는식품안전성측면에서매우중요하다. 따라서본연구에서는아플라톡신오염이우려되는견과류, 장류, 곡류및그가공품을대상으로 HPLC-FLD 분석법을이용하여아플라톡신에대한오염실태를조사하였다. 또한식품의저장 유통과정에서의아플라톡신생성을알아보고자식품개봉후저장기간에따라변화하는아플라톡신오염도를조사하였다. 08

2 A Survey of the Presence of Aflatoxins in Food 09 재료및방법 재료식품중아플라톡신오염도조사에사용된재료는 007년 월부터 008년 월까지경기지역의시중마트에서소포장과벌크형태로판매되는제품과, 국내온라인상에서판매하는제품을구입하였으며견과류및그가공식품 (79건), 장류 (9건), 곡류가공품등가공식품 (50건) 등총 58건을대상으로하였다. 58건중시중마트소포장제품은 87건, 벌크포장제품은 건, 온라인판매제품은 50건이었다. 개봉후저장기간에따른아플라톡신변화량조사에사용한재료는 차오염도조사에서아플라톡신 B 이검출된된장 건, 옥수수스낵 건, 땅콩가루 건, 땅콩 건등 6건을개봉한후다시밀봉하여상온 (5~5 ) 에서 6개월보관하면서시료로사용하였다. 표준품및시약실험에사용한아플라톡신 B 표준품은 Sigma 사에서구입하여사용하였으며추출과분석에사용된 acetonitrile, methanol은 HPLC용으로 (J. T. Baker, U.S.A.), trifluoroacetic acid (Fluka, Switzerland) 는특급을사용하였다. 정제용칼럼및분석장비시료정제용칼럼은 immunoaffinity column (AflaTest, Vicam Co., U.S.A.) 을사용하였고, 분석장비는 fluorescence detector가부착된 HPLC (Agilent 00 series, Agilent Technologies, U.S.A.) 를사용하였다. 표준용액조제아플라톡신 B 표준품각각 mg을 acetonitrile 0 ml에녹여표준원액으로하고아플라톡신혼합표준용액은각표준원액 ml씩을분취하여질소로완전히농축한다음 trifluoroacetic acid 00 µl로 0분간유도체화시킨후각각의최종농도가 0., 0.5,.0, 5.0, 0.0 ppb가되도록이동상 (30% acetonitrile) 으로희석하여사용하였다. 시료전처리방법시료의아플라톡신전처리과정은 005년식품의약품안전청연구결과보고서에서확립한방법을적용하였다. 균 9) 질화한시료 5 g에 % NaCl이포함된 70% methanol 용액 00 ml를첨가하여 0분간진탕한후여과하고이여액 0 ml에증류수 0 ml를혼합한용액 5 ml를취하여 immunoaffinity column에통과시킨후증류수 0 ml로 column을세척하고 acetonitrile 3 ml로용출하였다. 용출액을질소로완전히농축한다음 trifluoroacetic acid 00 ml로 0분간유도체화시킨후이동상을 800 ml 첨가하여시험용액으로하였다 (Fig. ). Instrument HPLC (Agilent 00 series) Column Capcell pak C 8 UG0 (50 mm 4.6 mm Shiseido) Detector Fluorescence detector Wavelength Ex 360 nm, Em 450 nm Mobile phase Water : Acetonitrile = 70 : 30 Flow rate 0.7 ml/min Injection volume 50 µl HPLC에의한정량아플라톡신정량분석을위하여 Table 과같이검출기는 fluorescence detector (FLD, Ex. 360 nm, Em. 450 nm), column 은 Capcell pak C 8 UG0(50 mm 4.6 mm, 5 µm, Shiseido), 이동상은 30% acetonitrile로유속 0.7 ml/min, 시료용액주입량은 50 µl로하여아플라톡신을분석하였다. 결과및고찰 시료중아플라톡신회수율아플라톡신혼합표준용액의 HPLC 크로마토그램은 Fig. 와같았으며아플라톡신 B 의검출한계 (limit of detection, LOD) 는최소검출량을구하는방법으로신호대잡음비 (S/N) > 3으로볼때 0.0 µg/kg이었다. 총아플라톡

4 A Survey of the Presence of Aflatoxins in Food 견과류및그가공식품 79건, 장류 9건, 곡류가공품등가공식품 50건을대상으로아플라톡신오염실태를조사한결과, Table 와같이총 58건중 45건 (8.5%) 에서아플라톡신오염이확인되었고, 오염수준은아플라톡신 B 으로서 0.0~3.3 µg/kg 로서 0.0~0.68 µg/kg 으로서 0.04~0.6 µg/kg 로서 0.03~0. µg/kg, 총아플라톡신으로서 0.0~3.96 µg/kg 범위로나타났다. 식품유형별로는견과류및그가공식품의경우 79건중 34건 (43.0%), 장류는 9건중 8건 (7.6%), 그외가공식품은 50건중 3건 (6.0%) 에서아플라톡신이검출되어견과류및그가공식품에서검출빈도가가장높은것으로나타났다. 견과류및그가공식품중땅콩버터의경우 건모두에서아플라톡신 4종 (B ) 이검출되었고검출량또한총아플라톡신으로서 0.7~3.96 µg/kg으로조사대상식품중가장높은오염도를보였다. Jang 등 ) 은곡류에비해견과류에서아플라톡신검출빈도가높았고특히땅콩및땅콩버터에서 30% 와 84.6% 로높은검출빈도를보였다고발표하였고, Park ) 의연구결과에서도땅콩및땅콩버터중 % 에서아플라톡신오염이확인되었다고하였다. 이러한경향을볼때땅콩등견과류가아플라톡신생성곰팡이에가장오염되기쉬운식품중의하나임을알수있었다. 본연구결과아플라톡신오염수준은국내 ( 아플라톡신 B 으로서, 0 µg/kg 이하 ) 의기준규격을초과하는것은없는것으로나타났으나우리나라의경우곡류를주식으로하고있기때문에지속적인모니터링으로아플라톡신노출에주의를기울여야할필요가있다. 특히 45건의오염된시료중 0건에서아플라톡신 B 과함께 B 또는 G 가검출되어향후총아플라톡신에대한기준설정등안전성검토가이루어져야할것으로사료된다. Table 3은조사대상식품을구입한유통경로별로아플라톡신오염도를살펴본것으로시중마트에서소포장으로판매하는제품 (9.%) 에비해벌크형태로판매하는제품 (38.%) 이나온라인상에서구입한제품 (58.0%) 에서아플라톡신검출빈도가높은것으로나타났다. 특히땅콩등견과류에서의아플라톡신검출율은소포장판매품이 4.5% 인반면, 온라인판매제품은 44.7%, 벌크제품은 50% 로벌크제품과온라인판매제품에서높은검출율을보였고, 옥수수스낵의경우도소포장제품에서는아플라톡신이검출된제품은없었으나벌크제품에서는 5건중 건이검출되어 40% 의검출율을보였다. 또한온라인상에서구입한땅콩버터에서는 건모두아플라톡신이검출되어 00% 의검출율을보였다. 이는식품의포장형태및보관장소등저장조건또는유통경로에따라아플라톡신오염수준이달라질수있음을보여준다. 개봉후저장기간에따른아플라톡신변화량식품을개봉하여저장하는동안변화하는아플라톡신함 Table 3. Food Concentration of AFB (µg/kg) After opening After month After 6 months Doenjang Corn snack () Corn snack () Ground peanut () Ground peanut () Peanut 량을조사하기위하여유통식품중아플라톡신이검출된된장 건, 옥수수스낵 건, 땅콩가루 건, 땅콩 건등 6 건을선정하여상온에서보관하면서 개월후, 6개월후의아플라톡신 B 함량을분석하였다. 그결과 Table 4와같이된장을제외한 5건의식품은아플라톡신 B 이저장기간에따라증가하는경향을보였다. 아플라톡신은식물이수확후소비가되기까지전과정에서오염될수있으므로저장기간이길수록아플라톡신오염량은증가될것으로예상할수있다. 5 Min-Jung Park, Mi-Hye Yoon, Hae-Geum Hong, Tae-Suk Joe, In-Sook Lee, Jeong-Hwa Park and Hoan-Uk Ko 따라아플라톡신함량을측정한결과, 양은차이가있었으나저장기간이길수록아플라톡신생성이증가하였다고하였다. 한편본연구에서된장은다른식품과는달리저장기간에따라다소감소하는것으로나타났는데이는된장의여러성분과발효과정중에상재하는미생물에의해아플라톡신함량이저하된것으로사료된다. 실제로 Kang 등은 4,5) 아플라톡신생성균인 Aspergillus flavus, Asp. parasiticus에대해가장강력한길항력이있는균을분리하여 Bacillus sp. 인것으로보고하였고길항균과아플라톡신생성균을처리하여제조한된장에서아플라톡신 B 함량이 87% 이상감소하였다고하였다. Park 등도 Asp. parasiticus를 6) 오염시켜메주를전통방법으로발효했을때아플라톡신생성은일어났지만, 된장, 간장등을만들기위해발표된메주를소금과물을넣고 3개월정도숙성하였더니약 63% 의아플라톡신 B 이파괴되었다고보고하였다. 아플라톡신은자연발생적으로생성되는독소로일단생성이되면완벽한제거와불활성화는어려운것으로알려져있다. 세척및가열등일반적인식품가공으로는아플라톡신제거가어렵기때문에 Yeo 등은사전에오염을 7) 방지하거나오염된식품의섭취를예방하는것이최선의방법이라고제안하고있다. 본연구결과에서도보듯이아플라톡신은식품의저장기간이길수록오염이증가될수있으므로식품섭취에의한아플라톡신노출을피하기위해서는되도록빨리섭취하고, 곰팡이가발생하지않도록건조하고온도가낮은조건에서보관하여야한다. 요약 경기지역시중마트와국내온라인상에서유통되고있는견과류및그가공식품 79건, 장류 9건, 곡류가공품등가공식품 50건에대해 immunoaffinity column 정제방법을이용하여 HPLC-FLD로아플라톡신오염실태를조사한결과총 58건중 45건 (8.5%) 에서아플라톡신오염이확인되었고, 오염수준은아플라톡신 B 으로서 0.0~3.3 µg/kg, 총아플라톡신으로서 0.0~3.96 µg/kg 범위로국내기준치인 0 µg/kg을초과하는것은없었다. 아플라톡신이검출된식품은견과류및그가공식품에서 34건 (43.0%) 장류에서 8건 (7.6%), 그외가공식품에서 3건 (6.0%) 으로견과류및그가공식품에서검출빈도가가장높았다. 식품을개봉하여 개월, 6개월보관후의아플라톡신 B 함량은저장기간이길어질수록땅콩, 옥수수스낵에서는증가하는경향을보였으나된장의경우에는다소감소하였다. 참고문헌.
