Material extract from P. cornucopiae fruiting body showed the highest α-glucosidase inhibitory activity among several edible mushrooms. The α-glucosidase inhibitor was maximally extracted when P. cornucopiae fruiting body was treated with distilled water at A С for B hrs. The α-glucosidase inhibitor was purified by Sephadex G-100, G-50 gel permeation chromatography, ultrafiltration, C18 solid phase extraction and reverse-phase HPLC, and obtained the purified α-glucosidase inhibitor with inhibitory activity of mg/ml of IC50. The purified α-glucosidase inhibitor was tripeptides with sequence of treonin- isolucin alanin - phenylalanine isolucin - aspalatic acid and its molecular weight was 1, Dalton. The α-glucosidase inhibitor from P. cornucopiae was showed clear anti-diabetic action dosage-dependantly in streptozotocin-treated diabetic induced rat. Key words : Anti-diabetic, Pleurotus cornucopiae, α-glucosidase 1. 연구목표 느타리버섯은 우리나라의 주요 식용버섯으로 면역조절효과, 항암활성, 항바이러스, 항염, 항생제, 콜레스테롤저감활성 등이 알려져 있다(Wang et al., 2000; Regina et al., 2008). 만성질환중의 하나인 당뇨병은 2000년 이후에는 성인 발병률이 10명중 1명으로 10%수준 으로 증가하고 있으며, 전세계적으로는 3번째로 위험한 질병으로 분류되고 있으나, 우리 나라 에서는 사망원인 5위의 성인병에 해당되고 있다(Baek, 2007). 특히, 당뇨병은 뇌하 년도 시험연구보고서

4 라. 분자량 및 아미노산 서열 동정 펩타이드 동정은 gel digestion 으로 처리하여 Q-TOF2 mass spectrometer (AB Sciex Instruments, CA USA)와 결합된 Nano-ESI로 분석하였다. 분석조건은 상 온에서 규산으로 코팅한 nanoelectrospray needles (EconoTipTM, New Objective, USA)에 1 kv전력으로 조절하였고, 안정적인 flow rate (10-30 nl/min)를 유지하기 위 해 0 5 psi으로 조절한 nitrogen back-pressure와 연결시켰으며 cone 전압은 40 V으로 조절하였다. 4중극 분석기(quadrupole analyzer)는 헥사폴 충돌세포(hexapole collision cell)에서 분열에 적합한 전구체 이온(precursor ions)을 택하기 위해 수행되었다. 충돌가 스(collision gas)는 Ar을 압력 6 7x10-5mbar으로 조절하였으며, 충돌에너지(collision energy)는 25 40V였다. 생산된 이온은 반사장치(reflector)와 micro-channel plate detector, a time-to digital converter를 적합한 조건으로 맞춘 orthogonal TOFanalyze 로 분석하였다. 마. 효능검증 수컷 실험용 쥐는(Sprague-Dawley male rats) 체중 g이고 5 7주 생육한 것을 Orientbio Co.(Korea)에서 구입하였으며, 12시간 낮, 12시간 밤시간을 유지하고 온 도와 습도는 실험 1주전에 조절하였다. 모든 실험용 쥐는 5마리씩 무작위로 선발하여 처리하였으며 α-glucosidase 억제제와 당뇨를 streptozotocin 투여 방법은 Stavric et al.(1975)와 Hall et al.(1990) 방법에 준하여 처리하였다. 실험용 쥐의 혈청내 혈당수준을 높이기 위해 최후 약제처리 1시간전에 streptozotocin 를 280 mg/kg 복강내 주입하였다. 노랑느타리버섯추출물은 300 mg/kg, 100 mg/kg, 50 mg/kg 3수준 농도로 급여하였고, soluble starch 는 3 g/kg 농도로 처리별 혼합하여 경구 투여하였다. 혈액은 심장에서 채취 하였으며, 상온에서 1시간 정도 응고한 후 3,000rpm으로 10분간 원심분리하여 혈 청을 분리 20 C에 저장하여 실험에 사용하였다. creatinine, alkaline phosphatase, triglycerides 등은 Sigma-Aldrich(St. Louis, Mo, USA), α-glucosidase assay kit는 Abnova Corporation제품을 사용하였다. 3. 결과 및 고찰 <시험1> 느타리버섯의 항당뇨성 물질 분리 및 동정 느타리버섯을 포함한 4종의 식용버섯 추출물의 α-glucosidase 억제활성을 분석한 결과(표 1), 노랑 느타리버섯에서 48.5%로 가장 높았고, 분홍 느타리버섯은 (Pleurotus salmone ostramineus) 39.1%의 효과를 나타냈다. 일반 느타리버섯과 큰 느타리버섯은 활성도가 없었다.
