대한임상병리사 - 회지 : 제 2 권 1 호 1992 室內室中훌훌園義에대한數種消毒齊 의最低 fj1j 制 i 農度實驗 全南大學校醫科大學皮 I 賣科學敎室 金守哲 元永鎬 田寅 j 其 Key words : Air-born fungus, Disinfectants, Minial inhibitary concentration 1. 績 最近違業化의過程에서國內에도漸次室內動 務者가날로增加되고있다. 또한醫學의發達과 함께 A 間의平均壽命의延長으로老 AA 口가增 加히 - 고있을뿐아니라, 醫藥品의 i 藍誤用으로因한 副作用풍으로, 個體免授低下로非病原性微生物 에因한機會感梁 (opportunistic infection) 1) 發生이 增加되고있다. 호氣中에自然的으로存在하는호 中활園鍵 (air borne fungi) 은病院의各種實驗室 및病室에서흔히 containation 을일으키고있으 며, 또한抗原性을가지고上氣道릎通하여천식 (astha), 알레르기壘껏, 담마진 ( urticaria) 동의 틀.6. 늄빼 東洋化學 I 業 Co.), povidon(r)<povidon iodine, (C 6 H 9NO)n xr, 0 l 중 70 g; 현대약품공업 Co.>, zephanon (R) (benzalkoniu chioride W /V % solution; 同仁堂製藥 Co.), wydex (R) < glutaraldehyde 2g (W/V)% ; 同仁堂製藥 Co.> 의 種을使用하였다. 2. 方法 1) 空中휠蘭의採集 園種은病院內皮 l 賣科外來릅今擔室, 률園檢훌室, 手術室, 그리고入院室동의室內호氣中에서 SDA 杯板햄地를 0 分間露出 ( 午後 時 ) 하여自然 落下하는 air-born fungi 를採集하였다. 알러지성 f 동愚 의原 因이될뿐아니라, 皮庸에感 i 월 f 눔을를좋發할수있다는것도또한잘알려져있 는事實이다 2-) 따라서著者들은本病院의室內 에서數種의호中힐 E 힘難을 Sabour aud s dextrose agar (Dif Co CO.) 햄地에 t 꼼養後이들의分布를觀 察하였으며, 그들중자주觀察되는 種의품출園을 選擇하여比較的病院內에서자주使用되는 種의 消毒倒에의한이들률園의 trp 制했果를빠究하여 興味있는結果를얻었기에報告하는바이다. 2) 훨蘭의培養및同定 園採集한 SDA t 홉地를 2-C 에서 -7 일培養한 後에各園集落 (colony) 을計算하였다. 園의同定 (identification) 을위하여各各의集落을다시 SDA 試驗管힘面培地에移植하여圍集落의肉眼的인 成長模樣과形態를觀察하였다. 또한必 要에따라 서 corn eal agar edida 에再培養하거나, slide 培養法을施行하여 lactophenol cotton blue 梁色後 1. 材料 n. 實驗材料및方法 이들을모두肉眼的檢훌와아울러顯微鏡檢훌를 施行하여園을同定하였다. 그러나肉眼的및顯微 鏡的檢훌에서훌園이라하여도未同定된률園과, 微生物中同定된圍이라하여도이들은除外시켰다. Sabouraud dextrose agar(sda), corn eal agar (Difco CO) 를高壓薰氣滅園한후直쩔 9c의 Petri dishes 에 1l 씩분주하여훌園 t홉地로서주로 利用하였다. 消毒簡 j 로는國內에서製造된 cresol(r), (0 l 중 cres 이 0 l, oil 0 l, ethanol l, 수산화칼륨 6. g ; 안미약품 Co.), oxyful(r) (H 2 0 2, 함량 % ; ) 훨萬浮遊 lrl(fungus suspension) 의홈빼 및接種 各園珠의集落을 SaboUI aud dextrose broth 1l 가들어있는삼각플라스크에接種하여 2 0 C 에서 7 일간培養하였다. 培養中뼈子生成을防止 하기위하여每日數차례흔들어주었다. 園培養 波을 9,000 rp 으로 1 분간遠心하여沈澈된園絲 - 187-
덩어리를 ground glass tissue grinder(fischer scien- 그러고이들댐珠를이용한消毒簡 j 의 MIC 는표 tific Co.) 를移用하여園絲를짧게分節시켜서 2 에표시한바와같이 wydex는 0-0% 이상의 spectrophotoeter(spectronic 20, Bausch & Lob) 柳制效果를보였다. 그리고 oxiful은 0.-%, 를利用하여園淑灌度를波長 67 n 吸 -Jt 度 1.0 povidon 은 - %, zephanon 은 0.07 - %, cre- 로測定한후 μe 를各灌度의消毒培地에接 sol 은 0.07% 이하의柳制처果를各各나타냈으며, 種하였다. 표 7 에서표시한바와같이 air-borne fungus 에대 ) 消훌훌훌때包含휠簡培地의調훌빼 前述한 cresol, oxyf 비, povidon, zephanon, wydex 種의原波을試驗管에消毒勳 i 農度別로愈加한 후, 120 0 C 1 Lbs 1 분高壓薰氣滅園된 SDA (final ph.6) 를 0-0 o C 로하여混合히 - 였 A 며, 各消毒 倒의最終灌度를 0%, 20%, %, %, 2.%,1.2 %, 2%, 0.%, 0.1%, 0.07% (volue in volue percent : v/v%) 가되도록調節하였다. ) 最小빼뼈 l 옳度 (inial inhibitory concetration) 의측정 各灌度別로消毒簡 j 가恩加된 SDA 平板 t 꼼地에 園을接種후 2 0 C 로培養하여 日째, 7 日째 2 回 觀察하여肉眼的으로園發育을볼수없는灌度 培地를消毒倒의最終판 n 制 i 農度 (MIC) 로定하였다. ill. 實驗成績 호氣중에存在하는 air-borne fungi 를培養하여 同定된園珠는표 1 에서표시한바와같이 Myceria(20.%), Alternaria(17.8%), Horodendru (1.2%), Rodotorura(1.%), Penicillu (11.6 % ), Aspergillus(8.9 %) 동의순서로分離培養되었다. Aspergillus Sp. ), 量的接種을위해培養한園絲를分節하였고, 灌度 別 t 곱地에接種할園浮遊波을만들어分尤光度計 67 n 에서吸 Jt 度 1. 0 으로測定하여 MIC 측정 배지에 μ e 을接種施行하였다 20). } 즙養溫度는 2 0 C 에서 1 주일培養하면서 MIC 를判讀하였으며, 接種후 일부터園集落을볼수있었고 7 일이후 는거의變化가없었다. 實驗에서各消 毒簡 j 의 MIC 는 oxyful(r) 은 0.- %, zephanon (R) 은 0.07-0.1%, cresol(r) 은 0.07 % 이하, povidon(r) 은 - %, wydex(r) 은 0% 이하였으며, 實驗한消毒劃중 cresol 이이가운데 가장낮은灌度에서호中훌園鍵이 f 印制되었으며, 各各의호中훨園鍵에대한 MIC 는비교적비슷한 結果를보였다. 이와같은實驗結果는實驗條件즉 n

ABSTRACT In these day, the people working in the roo and old aged persons are increasing by rapid advanceent of civilization in this country. Also the incidence of drug abuse or isuse are increasing. So there is a growing tendency at the outbreak of the diseases by the non-pathogenic organiss in the host with iune disturbance. Air born fungi like soil fungi can cause the opportunistic infection in iunedeficiency persons. They also ay have a role at inducing the allergic disease such as astha, allergic rhinitis, urticaria. We tried to figure out the antifungal effects of disinfectants, which use frequently in the hospital, against several air born fungi including Horodendru sp., Aspergillus sp., Penicilliu sp., Alternaria sp., which are isolated in this hospital. Air born fungi was collected by air exposure of Saubroud glucose edia. Mycelia sterilia sp.(species), Alternaria sp., Horodendru sp., Rodotrura sp., Penicilliu sp., Aspergillus sp., Nigrospora sp., Gliocladiu sp. and Paciloyces sp. Were sound in decreasing order. The suspension of fungi was ade by the growing at the nutrient broth and its concentration was easured by spectroeter. Each fungus was inoculated into Saubroud glucose agar plate and MIC (inial inhibitory concentration) was deterined after the growing of 7 days at 7 c. The MICs at the five disinfectants were less than 0.07% in Cresol, 0.-0.% Oxyful, - Povidon, 0.07-0. % Zephanon, ore than 0% Wydex. There is no difference of MIC according to kinds of fungi.
