Rotifer 섭식단계실험에서, 넙치자어의생존과성장은부화 12일째 rotifer만공급했을때보다 P. nn (nuplii) 단독으로공급했을때더높게나타났다. Arteim 섭식단계실험에서, 부화 3일째넙치자어의성장은 P. nn (C4-dult) 단독공급실험구와혼합공급 (P. nn+artemi) 실험구가다른실험구에비해높게나타났으나넙치자어의생존은모든실험구가유의적인차이를보이지않았다. P. nn nuplius와성체 (C4-dult) 의 n-3 HUFA 함량은각각 4.% 와 5.4% 였다. 결과적으로, 기수산요각류 P. nn는해수어종묘생산을위한초기생먹이로효과적인것으로판단된다. Astrct This study investigted the food vlue of Prcyclopin nn for flounder Prlichthys olivceus lrve in two feeding stges, rotifer (for 12 dys) nd Artemi (for 16 dys). In the rotifer feeding stge, survivl nd growth of flounder lrve in the only P. nn (nuplii) feeding experiment were higher thn in the only rotifer feeding experiment on 12 DAH (dys fter htching). In the Artemi feeding stge, the growth of flounder lrve in the only P. nn (C4-dult) feeding experiment nd mixture feeding experiment (P. nn+artemi) were higher thn in other experiments on 3 DAH, ut the survivl of flounder lrve did not differ significntly mong experiments. The n-3 HUFA contents of nuplius nd C4-dult were 4.% nd 5.4%, respectively. Overll the results of this study indicte tht the rckish wter cyclopoid copepod, P. nn, is n effective live food orgnism for lrvl seedling production of mrine fish. Keywords : Copepod, Prcyclopin nn, Prlichthys olivceus, food vlue, live food 1. 서론 해산어류나갑각류양식에서자어나유생의초기먹이로사용되는미소생먹이는주로 rotifer와 Artemi이다. 이들은영양학적으로해산어류에적합하지않다는치명적인단점을지니고있지만확보가용이하다는점에서아직까지이들을완벽히대체할수있는먹이가개발되 지못하고있다. 대부분의해수산어류는자어기에담수어류와달리정상적인성장과발달을위해필수지방산으로 EPA (eicospentenoic cid, 2:5n-3) 와 DHA (docoshexenoic cid, 22:6n-3) 같은 n-3 HUFA (highly unsturted ftty cids) 를요구하며해산자어의성공적인생산을위해서는이들지방산을충분히공급하는것이중요하다 [1-2]. 본논문은한국해양과학기술원연구과제 (PE99623) 로수행되었음. * Corresponding Author : Kyun-Woo Lee(Kore Institute of Ocen Science & Technology) Tel: emil: kyunu@kiost.c.kr Received Octoer 4, 218 Revised (1st Octoer 24, 218, 2nd Octoer 29, 218) Accepted Novemer 2, 218 Pulished Novemer 3,

2 이들지방산은자어의성장과생존율향상은물론자어의망막발달과시력에중요한역할을하고어류의색소침착에도영향을미치는것으로보고되고있다 [3-4]. 이러한지방산은자연에서해산어류의먹이로주로소비되는동물플랑크톤인요각류에풍부하게존재하기때문에이들을해산자어어의먹이로사용하려는연구가현재까지계속이어지고있지만상업적배양의어려움으로실효를거두지못하고있는실정이다 [5-7]. 이러한상황에서, 최근기수산요각류인 Prcyclopin nn의대량배양시도가있었으며비교적고밀도의배양이가능한것으로보고된바있다 [8-9]. 따라서본연구는넙치자어의성장및생존에대한 P. nn의초기생먹이로써의영양학적가치를평가하기위해 rotifer 및 Arteim와비교조사하였다. 2. 재료및방법 2.1 넙치자어의 rotifer 공급단계실험은 nuplius만공급한실험구 (C), nuplius와 rotifer를 1:1로공급한실험구 (CR) 및대조구로 rotifer 만공급한실험구 (R) 로하여 3반복실시하였다. 5 L 수조 ( 사육수 4 L) 에넙치의발안란을 1,2개씩수용하였다. 2일후부화하였고부화율은 92.7±5.83% 였다. 이때초기부화자어의크기는전장 2.7±.15 mm, 체폭.73±.5 mm였고건중은.9±.3 mg이였다. 사육수는온풍기를사용하여 18±.5 로유지하였고매우약하게통기하였다. 부화 5일째부터사이폰을사용하여매일바닥청소후 3% 씩환수하였다. 실험에사용한 rotifer는 Brchionus rotundiformis로 15 에서 12시간동안 A1 Super Selco R (INVE Aquculture) 로영양강화하였고부화 2일째에 2 개체 /ml 공급하였으며점차공급량을증가시켜 2일째 1-15 개체 /ml를유지하였다. 먹이공급횟수는오전 9시와오후 6시로나누어 1일 2 회공급하였다. P. nn의먹이공급은 P. nn의건중량이 B. rotundiformis와비교하여 2.7배가적었기때문에 rotifer 공급량의 2.7배많게공급하였다. 부화일부터 3일마다넙치자어 3마리씩전장과체폭을측정하였고실험종료일인부화 12일째는넙치의전장, 체폭과함께전중도측정하였다. 넙치의지방산분석을위해 5 μm sieve를사용하여모든넙치를수확한다음증류수로헹 군후, -8 에보관하였다. 2.2 넙치자어의 Artemi nuplius 공급단계실험구는 P. nn만공급한구 (C), P. nn와 Artemi를 1:1로공급한실험구 (CA), P. nn를실험 1 일째부터실험 5일까지공급하다가 Artemi만실험구 (CA5) 및대조구로 Artemi만공급한실험구 (A) 로하여 3반복실시하였다. 부화 14일째되는넙치를사용하여 1 L 사육수가담긴 25 L 수조에 13마리씩수용하였다. 사육수는 를유지하였고약하게통기하였으며 2일마다 5% 씩환수하였다. 바닥청소는매일먹이공급직전에실시하였으며폐사개체를기록하여생존율계산에사용하였다. 먹이는 P. nn 및 Artemi를 5-1 개체 /ml를유지하였고 1일 1회공급하였다. 2일간격으로무작위로 5마리씩을취하여전장, 체폭및건중을측정하였다. 실험종료후모든넙치는증류수로헹군다음분석을위하여 -8 에보관하였다. 2.3 지방산분석지방산분석은시료를담수로깨끗이세척한후 -8 에서보관하였다. 이렇게냉동된시료를진공동결건조시킨후지질을추출하였다. 14% BF 3-methnol (Sigm, USA) 로지방산을 methyltion 시킨후, cpillry column (HP-INNOWx, 3m.32mm.5 μm, USA) 이장착된 gs chromtogrphy (Shimdzu, GC-17A, Jpn) 로지방산을분석하였다. Crrier gs는 helieum (3 ml/min) 을사용하였으며, oven 온도는 17 에서 225 까지 1 /min 증가시켰고, injector의온도는 25, detector (FID) 온도는 27 로설정하였다. 지방산은알려진표준지방산의 retention time과비교하여분석하였다 [1]. 2.4 데이터분석모든결과는 one-wy ANOVA test를실시하였으며, 처리평균간의유의성 (P<.5) 은 Turkey의다중검정법 (Turkey's multiple comprison test) 으로하였다. 모든통계분석은 SPSS Version 17.(SPSS Inc, Chicgo, IL, USA) 을사용하여실시하였다. 3. 실험결과 683

3 한국산학기술학회논문지제 19 권제 11 호, 넙치자어의 rotifer 공급단계 넙치자어의생존율은 P. nn만공급한실험구 (C, 33%) 가 rotifer만공급한실험구 (R, 2%) 보다높게나타났고 (P<.5) rotifer와 P. nn를 1:1로혼합공급한실험구 (CR) 는 rotifer만공급한실험구보다높은생존율을보였지만모든실험구와유의적인차이는보이지않았다 (P>.5; Fig. 1B). 넙치의크기는부화 12 일째, 전장은 P. nn 만공급한실험구 (6.3 mm), rotifer 와 P. nn 를 1:1로혼합공급한실험구 (5.9 mm), rotifer만공급한실험구 (5.5 mm) 순으로차이를보였으며체폭은 rotifer만공급한실험구가다른실험구보다작게나타났다 (P<.5; Fig. 1A). 부화 12 일째넙치자어의건중량은 P. nn만공급한실험구가.17 mg으로다른실험구보다높게나타났다 (P<.5; Fig. 1A). Different letters indicte significnt difference (P<.5) Dry weight (mg) 3.2 넙치자어의 Artemi nuplius 공급단계 넙치의크기 ( 전장및체폭 ) 는실험종료일인부화 3 일째, P. nn 단독공급구 (C) 가 16.7 mm 로가장크게 나타났으나 P. nn와 Artemi를혼합한실험구 (CA) 와유의적인차이는보이지않았다 (P>.5; Fig. 2). 넙치의건중량은부화 3일째 P. nn 단독공급구와 CA 공급구가 6.6 mg으로가장높게나타났다 (P<.5; Fig. 2). 넙치의생존율은모든실험구가통계적인차이는보이지않았다. Different letters indicte significnt difference (P<.5). 3.3 실험에사용한초기생먹이와넙치의지방산조성본실험에서넙치자어의초기생먹이로공급한 rotifer, Artemi, P. nn의지방산조성에서 EPA 함량은 rotifer (8.3%) 와 Artemi (11.1%) 가 P. nn (nuplius: 3.4%; C4-dult: 3.2%) 보다유의적으로높은함량비를보였고 DHA 함량은 P. nn가 rotifer와 Artemi보다높게나타났으며특히, P. nn의 nuplius (21.4%) 보다성체의 DHA의함량비 (28.9%) 가더높은것으로나타났다 (P<.5). n-3 HUFA의함량은 P. nn 성체가 32.1% 로가장높았고 (P<.5) rotifer 와 Artemi 및 P. nn nuplius는차이를보이지않았다 (P>.5). DHA/EPA는 P. nn가 rotifer와 Artemi보다유의적으로높게나타났다 (P<.5; Tle 1). 부화 12일째각생먹이가공급된넙치자어의지방산조성에서 EPA와 n-3 684

4 HUFA의함량비율은차이를보이지않았으나 (P>.5), DHA의함량에있어 copepod만단독으로공급한실험구가 1.8% 로 rotifer 단독실험구의 8.2% 보다높은비율을보였다 (P<.5; Tle 2). 부화 3일째각생먹이가공급된넙치자어의지방산조성에서 EPA는 Artemi 단독실험구 (A) 와 CA5 실험구가모두 23.2% 로 CA 실험구와 P. nn 단독공급구보다높게나타났으며 (P<.5), P. nn 단독공급구 (C) 가 16.2% 로가장낮은함량비를보였다. DHA는 C 실험구가 12.4% 로가장높게나타났고다음으로 CA 실험구가 6.8% 의함량비를보였으며 A 실험구와 CA5 실험구는각각 1.3% 와 2.4% 로낮게나타났다 (P<.5). n-3 HUFA 도 C 실험구가 28.7% 로다른실험구보다높게나타났다 (P<.5; Tle 3). Tle 1. Rotifer nd Artemi were enriched with A1 Super Selco.Copepod nuplii nd dults were enriched with Tetrselmis suecic. 4. 고찰 본실험에서넙치자어의초기먹이로써 P. nn의공급은넙치의 rotifer 공급단계와 Artemi 공급단계모두에서 rotifer나 Artemi를공급했을때보다넙치의성장에있어더효과적인것으로나타났다. Ft snook (Centropomus prllelus) 자어에게 Acrti tons 등을공급, pink snpper (Pgrus urtus) 와 West Austrlin dhufish (Glucosom hericum) 에게 Gldioferens impripes를공급그리고청소고기 (Elctinus figro) 에게채집한요각류를공급한결과 rotifer 공급구보다성 685

5 한국산학기술학회논문지제 19 권제 11 호, 218 장과생존이우수한결과를얻었다 [11-13]. 또한 West Austrlin 해마, Hippocmpus suelongus에게 Gldioferens impripes를공급, 흰동가리 Amphiprion clrkii에게 Tise spp. 를공급, 대구 Atlntic cod에게채집한요각류를공급한결과 rotifer와 Artemi를공급했을때보다성장과생존이더우수한결과를보고한바있다.[2, 14-15] 초기생먹이의가치에대한잠재성평가는그들의화학적조성, 특히지질과지방산함량과관련되어있다 [5]. 지방산중 DHA는정상적인신경발달과기능에중요하며특히자어의망막발달과시력에중요한역할을하는것으로알려져있다 [3]. Yellowtil flounder (Limnd ferrugine) 의경우, DHA가높은먹이를먹은자어는성장과생존율이높게나타난반면, DHA 함량이매우낮은먹이를먹은자어는성장과생존율이유의적으로낮게나타난다 [4]. 본실험에서도 P. nn는 DHA 비율이 2% 이상으로 rotifer나 Artemi에비해상당히높을뿐아니라건조중량당 DHA 함량도 nuplius와 C4-성체각각 3.5% 와 4.8% 로 n-3 HUFA의대부분을차지하였다. 또한 P. nn의 n-3 HUFA는 nuplius와 C4-dult 각각 4.% 와 5.4% 로해산어류자어의정상적인성장을위한 n-3 HUFA함량인 3-4% 를초과한것으로나타났다 (Fig. 3). 일반적으로해산어류자어는자연산어란의난황과 copepod의극성지질및대부분해산어류자어자연먹이의 DHA:EPA 비인약 2:1인먹이를요구한다 [1]. 본실험에서 DHA:EPA 비는 A1 Super Selco R 로영양강화된 rotifer의경우, McEvoy et l. (1998)[3] 의실험에서.5와비슷한 1.으로낮은수치를보였고, Artemi의경우도 McEvoy et l. (1998)[3] 의실험에서.7-.8, Pyne nd Rippingle (2)[2] 의실험에서.2와비슷하게.7로낮게나타났다. 그러나 P. nn의경우, nuplius와 C4-성체각각 6.7과 9.으로매우높게나타났다. 일반적으로 copepod는극성지질이풍부한반면 Artemi는중성지질이풍부하다. 또한극성지질은자어의기초적인소화경로에서다른지질의소화를촉진시킬뿐아니라자어가가장쉽게소화할수있다. 따라서 copepod 내의 DHA 같은필수지방산은영양강화된 Artemi nuplius의중성지질내지방산보다자어가더쉽게소화시킬수있다 [3, 16-19]. 따라서본실험에서부화 3일째넙치의어체에대한지방산조성에서 DHA 의함량비는 P. nn 를연속적으로공급한실험구가 % 로 Artemi 단독구및 CA5 실험구보다높은함량비를보였다 (P<.5). 다른판형물고기처럼, 넙치는변태하는동안자세가수직에서수평형태로돌아가며눈이이동하는등완전한형태적변태와동시에유영생활에서저서생활로바뀌기때문에서식처나먹이섭취조건이완전히바뀌게된다 [2]. 본실험에서넙치의 Artemi 단계실험에서공급한 P. nn C4-성체는완전부유성인 nuplius와는달리일부부착하는습성도있다. 이때문에넙치사육수조내의먹이밀도의분포는대부분부유성인 Artemi에비해수조바닥에도고루분포하였다. 따라서넙치의변태시기는유영과착저를번갈아하는시기이므로완전부유성인 Artemi를공급한실험구의넙치는 P. nn를공급한실험구보다먹이섭취기회가더적었을수있으며이점은 Artemi 공급구의성장저해요인이되었을가능성이있는것으로판단된다. 5. 결론 본실험을종합해보면, P. nn를초기생먹이로이용한넙치의사육은 rotifer나 Artemi보다성장과생존측면에서더효과적인것으로나타났으며 P. nn를초기생먹이로공급시 rotifer나 Artemi와 5% 만혼합하여공급하여도충분할것으로판단된다.
