AllInOneCycler PCR System Interchangeable thermal block system: AllInOneCycler TM 는재질에따라일반 thermal block 과특수합금 thermal block 을선택적으로사용할수있으며, 샘플에따라 96-well, 384- well, slide block 을선택할수있습니다. 총 6 가지 block 에서실험목적에맞게 block 을교체하여사용할수있으며, 신규개발 block 을계속추가할예정입니다. A reflection light (Unpolarized) A reflection light (Polarlized) Description Exicycler TM 96 (Ver.4) 는 96 well 동시검출방식의 Real-Time qpcr 장비입니다. 바이오니아의특허인 LT (Light Tunnel) 기술과편광을이용한형광검출기술로광학적민감도와정확도를높이고 96 well 간의편차를최소화하여 reference dye 에의한 normalization 이없이각 well 당 5 채널의형광값을정밀하게분석할수있습니다. 또한최적의 PCR annealing 온도를한번에찾을수있는 thermal gradient block 기능으로실험조건설정시시간과소모품비용을최소화할수있습니다. 분석소프트웨어는 Absolute Quantification, Relative Quantification, SNP genotyping, Existence/Nonexistence 모드를제공하여간편하게 viral load analysis, 유전자발현분석, SNP genotyping 등다양한분야의실험분석이가능합니다. Features and Benefits 민감하고정확한실험을위한최고의광학계편광을이용한형광검출기술로반사광에의한노이즈 * 를최소화하여실험민감도와정확도가향상되었습니다 (Figure 1). * 반사광에의한노이즈 : qpcr에이용되는형광물질은여기광 (Excitation energy) 을받아여기상태가되었다가기저상태로떨어지면서형광을발하게됩니다 (emission). 일반적인 qpcr 장비에서는형광물질 (fluorescence dye, reporter dye 등 ) 을여기시키기위해시료로조사된여기광의일부가 plate 및기구내부표면에서반사되면서시료에서발생된형광과함께형광검출장치로입사됩니다. 일반적으로형광필터 (band pass filter) 로반사광을제거하게되지만, 여기광의광량이형광량에비해아주크기때문에, 이에비례하여반사광의광량또한상당히커서형광필터로완벽하게제거하지못하여시료의형광검출시제거되지못한반사광이노이즈를발생시켜민감도와정확도를떨어뜨리는요인으로작용합니다. Reference dye 사용이불필요하여 5-multiplex PCR 가능 Well 간광학편차를기구적으로해결하는바이오니아의 LT 기술로일반적으로편차보정을위해사용되는 reference dye 가필요하지않으므로 5 채널의형광을모두시료에활용할수있어 real 5 multiplex PCR 이가능합니다. Exicycler 96 (Ver.4) Real-Time Quantitative Thermal Block 정확한온도제어와 Ct 값오차최소화로높은실험재현성구현 정확한온도제어를통해전체 well에대한온도정확성과균일성을향상시켰습니다. 온도편차가 ±0.3 이내로가장자리와중앙의온도차이가매우작습니다 (Figure 3). 또한빠르고정확한분석을위해모든 well의형광 data를 2D CCD sensor를통해동시에측정하여 96 well plate의 well간의측정시간차이에따른편차 (Time-lag) 를없앴습니다. 광학계에 LT 특허기술을적용하여모든 well에조사되는빛의양이균일하기때문에, 어떤 well에서 PCR 하여도 Ct 값편차가 0.3 cycle 이내로유지됩니다 (Figure 4).
