The alcohol contents of tomato wines prepared from semi-mature and full-mature tomato were about 11% whereas that of immature tomato was slightly lower (10.6%). When characteristics of tomato alcohol fermentation were examined on pre-treatment conditions, the wine from full mature-crushed whole tomato contained lower alcohol at 10.6% than those of full mature-crushed tomato removed stem and full mature-tomato juice removed stem (about 11%). For the effect of initial sugar, the alcohol content and sugar content of tomato wine increased in proportion to the concentration of initial sugar (8.8~14.4% and 5.5~9.2 o Brix). However, initial titratable acidity and ph did not affect the alcohol contents during fermentation, and suggesting needed to be supplementation for sensory improvement of tomato wine. When the quality characteristics of tomato wine fermented under these optimal conditions were investigated, alcohol content, sugar content, titratable acidity and ph were found to be 11.1%, 6.1 o Brix, 0.68% and 3.7, respectively. For alcohol components, the total amounts of methanol and fusel oil were 346 ppm and 313 ppm. In addition, maltose showed the highest level of mg% in the free sugar analysis of tomato wine whereas sucrose contained 18.7 mg%. Key words: tomato, alcohol fermentation, wine, yeast, quality 서론토마토는가지과 (Lycopersicon esculentum Mill) 에속하는일년생작물로써우리나라기후풍토에도적합하여전국적으로재배되고있다 (1). 국내토마토의생산면적은 2001년이후급격히증가하여 2007년에는 7,353 ha로전년보다약 11% 증가하였고, 생산량도 10.7% 증가한 479,851톤으로나타났다 (2). 또한, 대구광역시의토마토면적은 2004년 116 ha에서꾸준히증가하여 2007년 225 ha로나타났으며, 생산량도 2004년 8,143톤에서 2007년 20,059톤으로 2배이상증가하였다 (2). 토마토는미국타임지선정 몸에좋은 10가지식품 중하나로라이코펜 (lycopene) 이세포의산화를막아노화를억제하고, 심혈관질환과각종암의예방효과가있다고보고되었으며 (3), 비타민 A가딸기에비하여 8배이상함유되어있고, 비타민 C는채소작물가운데가장많은양이함유되어있다. 토마토의유기산함량은 citric acid가 0.5~ 21% 를차지하며, 유리아미노산이 0.07~0.09% 함유되어독특한맛을주고노화방지, 퇴행성질환억제, 인체면역기능강화, 루틴성분으로인해혈관을튼튼하게하고혈압을내리는역할도한다 (4). 또한, 니코틴의해독과함께숙취에도효과적인것으로알려져있으며 (5), 풍부한식이섬유는육류등의지질을많이섭취하게되는현대인의건강유지에도대단히중요한역할을한다 (6). 그러나토마토는주로완숙되기전인 breaker 단계에서수확하여유통되고있다. Pink 단계이후에수확하는것이미숙과에비해품질이우수하나유통중품질변화가급격히일어나는특성으로인해완숙과를유통할경우부패및연화로인해품질이나빠지는문제가발생된다 (7). 토마토에관한연구는저장수명연장에관하여 (5,7) 활발히진행중이며, 토마토및토마토분말의기능성조사 (8-10), 토마토분말을첨가한설기떡제조 (4), 토마토의품종별주스제조적성 (1) 에관한연구등이보고되고있다. 토마토는서양에서는저장성이높은주스, 페이스트, Corresponding author. yjjeong@kmu.ac.kr Phone: , Fax:

2 444 장세영 우승미 조용준 김옥미 김이호 정용진 퓨레, 케첩등의음료및조미료개발로그용도가다양하고소비량도많으나, 우리나라에서는주로생식용으로소비되고있으며케첩, 주스로는소량이용되고있어서소비촉진과고부가가치지역브랜드상품으로개발하기위한다양한가공품개발이요구된다 (1). 국내산과실을이용한알코올발효및주류제조에관한연구는활발히이루어지고있으나 (11-14) 토마토의알코올발효에관한연구는없는실정으로토마토를이용한독창성있는주류의개발이기대된다. 토마토는당함량이낮고 ph는높아서알코올발효과정에서전처리조건, 보당, ph 조절등과같은발효조건과숙성과정에서의성분변화등에관한다양한연구기반의조사가요구된다. 본연구에서는토마토의전처리, 숙성정도및알코올발효조건에따른이화학적특성을비교조사하여독창성있는토마토과실주개발에기초자료로활용하고자한다. 재료및방법재료및시약본실험에사용된토마토는당도 4.5 o Brix, ph 4.2, 적정산도 0.45% 였으며대구광역시달성군에서 2009년수확된것을구입하여 4 o C에서냉장보관하면서사용하였다. 과실주제조에사용된혼합산 (acid blend) 및효모 Lalvin EC-1118 은 ( 주 ) 와인킷코리아 (Winekit Korea Co., Ltd., Seoul, Korea) 에서구입하여사용하였다. Starter 및주모배양 Starter는 YPD 배지 (yeast extract 1%, peptone 2%, glucose 2%, ph 6.0) 에시판되는 Saccharomyces cerevisiae 효모 Lalvin EC-1118을 0.02%(w/v) 접종하여 30 o C, 24시간배양한후사용하였다. 주모배양은토마토마쇄액 200 g에 10 o Brix가되도록설탕으로보당하여 autoclave(121 o C, 15 분 ) 한후 starter를 5%(v/v) 접종하였으며, 항온배양기 (HB-103-2H, Hanbaek Scientific Co., Bucheon, Korea) 에서 30 o C, 24시간동안정치배양시켜사용하였다. 토마토의발효조건에따른알코올발효토마토숙기에따른알코올발효특성을조사하기위해미숙, 중간및완숙된각각의토마토마쇄액을사용하였으며, 전처리조건에따른토마토알코올발효특성조사에서는완숙토마토마쇄액 (A), 꼭지제거된완숙토마토마쇄액 (B) 및꼭지제거된완숙토마토착즙액 (C) 으로나누어전처리하였다. 각각전처리한토마토 1 kg에설탕으로 18, 20, 22, 24 및 26 o Brix로보당하여초기당도에따른알코올발효특성을조사하였고, 초기산도에따른영향은혼합산 (acid blend) 을이용하여 0.0, 0.5, 0.7 및 0.9% 로각각보산후 starter 5%(v/v) 를접종하여항온배양기에서 30 o C, 3일동안정치배양시켰다. 발효종료후부직포로 1차여과한다음 13,000 rpm으로 5분동안원심분리시킨상등액을분석시료 로사용하였다. 최적조건에서의알코올발효상기설정된조건을토대로하여꼭지제거된완숙토마토마쇄액 30 kg에설탕으로 20 o Brix가되도록보당하고혼합산을이용하여초기산도를 0.5% 로조정한후 starter를 5% (v/v) 접종하여항온배양기에서 30 o C, 7일동안정치배양시켰다. 발효종료후부직포로 1차여과한다음 5 o C 이하에서 1개월간 racking 시킨후분석시료로사용하였다. 알코올함량및당도발효액의알코올함량은시료 100 ml를취하여증류한다음주정계를이용하여측정하였으며 Gay Luccac Table을이용하여 15 o C로보정하였고 (15), 당도는 digital refractometer(pr-101, ATAGO Co., Tokyo, Japan) 를사용하여측정하였다. 적정산도및 ph 적정산도는일정량의시료를취하여 1% phenolphthalein 지시약을 2~3방울떨어뜨린다음 0.1 N NaOH로중화적정하였으며 lactic acid(%) 로환산하였고, ph는 ph meter(metrohm 691, Metrohm, Herisau, Switzerland) 로실온에서측정하였다. 색도색도는 UV-visible spectrophotometer(uv-1601, Shimadzu Co., Kyoto, Japan) 를이용하여명도 (L), 적색도 (a) 황색도 (b) 값을측정하여 Hunter s color value로나타내었으며, 이때대조구는증류수 (L=99.99, a=0.06, b=-0.10) 를사용하였다. 알코올성분알코올성분분석은국세청주류분석규정 (15) 에따라 gas chromatography(hewlett packard-6980, Hewlett packard Co., Palo Alto, CA, USA) 로분석하였으며, 표준물질로무수알코올 (99.9%) 을사용하여각각의알코올성분을일정량 (0~200 ppm) 첨가하여작성한 standard curve를이용하여정량하였다. 이때사용한 column은 fused silica capillary column(30 m 0.25 mm) 이었으며, detector는 FID, injector temperature는 250 o C, detector temperature는 260 o C, carrier gas는 N 2(30 ml/min) 를사용하였다. 유리당유리당분석은알코올발효액을 Sep-pak C 18 cartridge에통과시킨다음 0.45 μm membrane filter로여과하여 High Performance Liquid Chromatography(HPLC, Waters 2487, Waters Co., Milford, MA, USA) 로분석하였다 (16). 분석 column은 carbohydrate analysis column( mm, Waters Co.), mobile phase는 75% acetonitrile(j.t.baker Co., Phillipsburg, NJ, USA) 을사용하였고 flow rate는 1.0 ml/min, injection volume은 20 μl로하여 RI detector

3 알코올발효조건에따른토마토과실주의품질특성 445 (M410 RI, Waters Co.) 로분석하였다. 결과및고찰토마토의숙성정도에따른영향미숙, 중간및완숙된각각의토마토마쇄액을이용하여토마토숙기에따른알코올발효특성을조사한결과 Fig. 1, 2 및 Table 1과같다. 알코올함량에서는중간및완숙과가약 11% 로비슷한함량을나타내었고미숙과는 10.6% 로조금낮았다. 적정산도에서는숙성이많이될수록낮은수치를나타내어관능적으로큰차이를보일것으로생각되며, ph 는적정산도와상반되는경향을보였다. 색도중 L 및 a값은모든시료에서흡광도가각각 96.5 및 -3.5 전후로비슷하게나타났다. b값은미숙과에서 11.9로가장낮게나타났고완 숙과에서 13.8로가장높았으며, 갈색도는 b값과유사한경향을보여완숙과로제조된토마토알코올발효액이미숙과로제조된토마토알코올발효액에비하여노란색계통의색소성분을더많이함유하고있는것으로나타났다 (no data). 토마토에는항암작용이뛰어난라이코펜이함유되어있으나산화되기쉽고물에불용성이며에탄올및메탄올에도거의녹지않는것으로알려져있다 (17). 따라서토마토알코올발효액의여과과정중붉은색색소침출은어려운것으로판단되었다. 토마토전처리조건에따른영향완숙토마토마쇄액 (A), 꼭지제거된완숙토마토마쇄액 (B) 및꼭지제거된완숙토마토착즙액 (C) 으로나누어전처리조건에따른토마토알코올발효특성을조사한결과 Fig. 3, 4 및 Table 2와같다. 알코올함량에서는 (A) 가 10.6% 로낮게나타났고, (B) 및 (C) 가약 11% 로나타났다. 적정산도및 ph에서는모든시료에서각각 0.45% 및 4.4 전후로비슷한수치를보여전처리방법의차이가알코올발효에는큰영향을미치지않는것으로나타났다. 색도의 b값에서는 (A), (B), (C) 순으로높게나타났다. 4 446 장세영 우승미 조용준 김옥미 김이호 정용진 Table 2. s of tomato wine by pre-treatment conditions Pre-treatment condition A 1) B C 97.0±0.0 2) 96.8±0.0 1) Refer to Fig. 3. 2) Values are mean±sd (n=3). -4.2± ± ± ±0.0 인해쓴맛및풋내가나타나관능적으로부정적인영향을미칠것으로생각되며, (B) 및 (C) 에서는알코올발효능은비슷하였으나착즙액을원료로사용할경우색소침출능이조금떨어지는것으로나타나본연구에서는토마토전처리조건으로꼭지제거된완숙토마토마쇄액 (B) 을선정하였다. 초기당도및초기산도에따른영향 초기당도에따른토마토알코올발효특성을조사한결과 Fig. 5, 6 및 Table 3과같다. 적정산도및 ph는각각 0.45~ 0.52% 및 4.2~4.4의범위로초기당도에따른차이가크지않은것으로나타났다. 알코올함량및당도는초기당도가높아질수록각각 8.8~14.4% 및 5.5~9.2 o Brix로높게나타났다. Kim 등 (18) 이포도의초기당도를 24 o Brix로조정하여 9일동안알코올발효를시킨결과 14.7% 의알코올함량을 Table 3. Effect of initial sugar concentrations on Hunter's color values of tomato wine Initial sugar concentration ( o Brix) ±0.1 1) 97.1± ±0.0 1) Values are mean±sd (n=3). 13.2± ± ± ± ±0.0 나타내어본결과의 13.9% 보다는높게나타났다. 이러한결과는발효기간에의한차이로생각되며본연구에서도발효기간을연장하면초기당도가높았던시료순으로알코올함량이더증가할것으로예상된다. 한편, Choi 등 (19) 은복분자발효주를제조할때관능검사를통한알코올의적정량은 11 13% 라고보고된바있어, 토마토를이용한알코올발효는초기당도 20~22 o Brix가적정할것으로생각되며, 이후숙성 저장및관능적인특성을고려하여초기당도를조정하고자하였다. 초기산도에따른토마토알코올발효특성을조사한결과 Fig. 7, 8 및 Table 4와같다. 알코올함량은대조구에서 11.2% 로가장높았으며다른시료들은 10.5% 전후로비슷한 Fig. 5. Free sugar components of tomato wine fermented under the optimal condition Components (mg%) Free sugar Fructose Glucose Sucrose Maltose Tomato wine 함량을나타내었고당도는 6.2 o Brix 전후로나타나초기산도에따른큰차이를보이지않았다. 적정산도는초기산도가높을수록, ph는초기산도가낮을수록증가하는경향이었다. 색도의 b값및갈색도는초기산도가높을수록조금증가하였다. 와인의총산은맛을결정하는중요한요소이며총산함량이낮은와인은맛이밋밋하고보관도중에쉽게상할수있는반면, 총산함량이높은와인은거칠고신맛이강하다 (20). 따라서토마토를이용한알코올발효의경우초기산도가알코올발효과정에는큰영향을미치지않았지만관능적인특성을고려할때초기산도 0.5% 이상은신맛이강하게나타나향후숙성과정등을고려한조건의보완이요구되었다. 최적조건에서의알코올발효특성상기설정된조건을토대로알코올발효후 5 o C 이하에서 1개월간 racking 시킨후발효액의품질특성을조사하였다. Table 5와같이알코올함량은 11.1%, 당도는 6.1 o Brix, 적정산도는 0.68%, ph는 3.7로나타났다. Table 6에서보는바와같이주요알코올성분으로는 acetaldehyde, methanol, ethanol, n-propanol, isoamylalcohol 등 5종이분석되었다. Acetaldehyde는 127 ppm으로나타났으며, methanol은 346 ppm으로나타났다. 국내식품공전규격 (21) 에서과실주의 methanol 함량 1,000 ppm 이하에는적합한것으로나타났다. 또한, n-propanol과 isoamylalcohol 등 fusel oil류총량은 313 ppm으로나타났다. 효모의아미노산발효로생성되는 fusel oil은원료중의아미노산함량및효모의종류에따라그함량이달라지며미량존재할경우에는주류의맛과향을높이는역할을함으로써품질에중요한요인이된다 (22,23). Table 7에서보는바와같이주요유리당함량은 fructose, glucose, sucrose 및 maltose 4종이분석되었다. Maltose가 mg% 로가장높게나타났으며 glucose, fructose 및 sucrose가각각 142.8, 71.5 및 18.7 mg% 로나타나보당에사용된설탕이발효에대부분이용된것을알수있었다. 이상의결과토마토의알코올발효조건을설정할수있었으며, 이를바탕으로토마토과실주의발효조건을확립할수있을것으로기대된다. 요약본연구에서는토마토를이용한알코올발효조건을조사하였다. 그결과, 토마토숙기정도에따른알코올함량은중간및완숙구간이약 11% 로비슷한함량을나타내었고미숙구간은 10.6% 로조금낮았다. 토마토전처리조건에따른토마토알코올발효특성을조사한결과, 알코올함량은완숙토마토마쇄액 (A) 이 10.6% 로낮게나타났고, 꼭지제거된완숙토마토마쇄액 (B) 및꼭지제거된완숙토마토착즙액 (C) 이약 11% 로각각나타났다. 초기당도에따른영향에서알코올함량은초기당도가높아질수록각각 8.8~ 14.4% 로높게나타났다. 초기산도및 ph는발효기간중알코올함량에는큰차이가없었으나향후토마토과실주의관능적특성을고려하여보완이요구되었다. 상기설정된조건으로알코올발효시킨발효액의품질특성을조사한결과, 알코올함량은 11.1%, 당도는 6.1 o Brix, 적정산도는 0.68%, ph는 3.7로나타났다. 알코올성분은 methanol이 346 ppm, fusel oil류총량이 313 ppm으로나타났다. 유리당은 maltose가 mg% 로가장높게나타났으며 sucrose 가 18.7 mg% 로나타났다. 이상의결과토마토를이용한알코올발효조건을설정할수있었으며, 향후다양한용도로활용이기대되었다.
