A fission yeast, Schizosaccharomyces pombe, SPBC gene encoding the ortholog of THOC5, a component of THO/TREX complex, was identified and characterized. The S. pombe thoc5 (spthoc5) is not essential for both growth and mrna export, but deletion of the spthoc5 gene caused growth defect and slight accumulation of poly(a) RNA in the nucleus. And the functional spthoc5-gfp protein is localized mainly in the nucleus. Co-immunoprecipitation analysis showed that the Hpr1(THOC1) protein, an evolutionally well-conserved component of THO/TREX complex, interacted with spthoc5 as well as Tho2(THOC2), another subunit of THO complex. These results suggest that S. pombe Thoc5 as a component of THO/TREX complex is also involved in mrna export from the nucleus. Key words: S. pombe, mrna export, THOC5, THO/TREX complex 진핵생물의핵안에서일어나는전사 (transcription) 과정은 pre-mrna의가공, 성숙한 mrnp (messenger ribonucleoparticle) 의포장 (packaging) 및세포질로의방출등과긴밀하게연계되어조절된다. 이와같은유전자발현의초기단계들은전사과정동안 RNA 중합효소와의상호작용을통해 premrna로옮겨가는많은단백질또는단백질복합체들에의해이루어지며, 각단계들은서로조절된다 (Perales and Bentley, 2009; Stewart, 2010). 출아효모인 Saccharomyces cerevisiae 에서전사의신장과정에관여하는복합체로처음알려진 THO 는 RNA 중합효소 II의가장큰소단위인 Rbp1의인산화된 C- 말단영역 (CTD) 과직접결합함으로써전사가활성화된유전자부위에모여든다 (Chavez et al., 2000; Meinel et al., 2013). THO 복합체를불활성화시키면전사신장에문제가생기는데, G/C가많거나길고반복서열이있는유전자의전사가특히감 *For correspondence. jhoyoon@sungshin.ac.kr; Tel.: ; Fax: 소한다 (Li et al., 2005; Voynov et al., 2006). 또한 THO 돌연변이는유전체의불안정성을야기하여재조합빈도가증가하는표현형 (hyper-recombination) 을보인다 (Aguillera, 2002; Jimeno et al., 2002; Dominguez-Sanchez et al., 2011). 이것은 THO 복합체에이상이생기면전사된 RNA가주형 DNA와결합된 DNA:RNA 혼성체인 R-loop가축적되고, 이로인해재조합수선기작이활성화되기때문이다 (Huertas and Aguilera, 2003; Stirling et al., 2012). 한편, THO 복합체는 mrna 방출에관여하는 mrna-결합단백질들인 Sub2 (DEAD box RNA helicase), Yra1 (mrna 방출인자인 Mex67의 adaptor), 그리고 SR-유사단백질인 Gbp2, Hrb1 등과결합하여 TREX (transcription and export) 라불리는복합체를형성한다 (Strasser et al., 2002; Zenklusen et al., 2002; Hurt et al., 2004). TREX는전사과정동안 pre-mrna로이동하여결합하는데, 전사된 mrna가적절한 mrnp로포장되는데관여하는것으로여겨진다 (Abruzzi et al., 2004). mrna에결합한 TREX는필수적인 mrna 방출

2 436 Koh and Yoon 운반체 (export receptor) 인 Mex67를불러들임으로써성숙한 mrna의핵에서세포질로의방출에관여한다 (Strasser et al., 2002; Zenklusen et al., 2002). 이외에도, TREX는스플라이싱과전사신장에관여하는 Prp19 복합체와도결합하며 (Chanarat et al., 2011), Pcf11과상호작용을통해 mrna의 3 말단가공에도관여한다 (Roug le et al., 2008; Johnson et al., 2011). 이와같이 THO/TREX 복합체는전사신장을 mrna 성숙및방출과정과연계하고조직화함으로써, 세포질에서번역될수있는성숙한 mrnp 생성에매우중요한역할을담당하고있다 (Luna et al., 2012). 위와같은 THO 복합체의기능에대한연구는처음발견된출아효모인 S. cerevisiae 에서주로연구되었지만, THO는식물과인간을포함한고등생물에도진화적으로잘보존되어있다. 고등생물의 THO/TREX 복합체도출아효모와마찬가지로 mrna 생성과정과방출에관여한다 (Katahira et al., 2009; Guria et al., 2011). 하지만 THO/TREX 복합체가 mrna에결합하는방식에는차이가있다. 전사과정중에 mrna에결합하는출아효모와는다르게, 사람의 THO/TREX 복합체는 mrna 가공과정인짜집기 (splicing) 와 5 -모자형성 (capping) 과정을통해 mrna의 5 말단쪽에결합한다 (Masuda et al., 2005; Cheng et al., 2006). 또한구성인자에도차이점이존재하는데, 출아효모의 THO 복합체는 Tho2 (184 kda), Hpr1 (88 kda), Tex1 (47 kda), Mft1 (45 kda), Thp2 (33 kda) 등의 5개소단위로구성되어있는반면 (Chavez et al., 2000; Peña et al., 2012), 고등생물에는 THOC1, THOC2, THOC3, THOC5, THOC6, THOC7 등의총 6개의소단위로구성되어있다 (Rehwinkel et al., 2004; Masuda et al., 2005). 즉, Hpr1 ( 포유동물에서는 THOC1), Tho2 (THOC2) 와 Tex1 (THOC3) 만연구된대부분의진핵생물에잘보존되어있고, Mft1과 Thp2는출아효모에특이적이고 THOC5, THOC6, THOC7 등은고등생물에특이적으로존재한다. 분열효모 Schizosaccharomyces pombe의유전체 database인 Pombase ( 에서 THO 복합체의구성인자의이종상동체 (ortholog) 를암호화하는유전자를찾아보았다. S. pombe에는진화적으로잘보존된 Hpr1 (THOC 1), Tho2 (THOC2), Tex1 (THOC3) 유전자는존재하였고, 그외에 SPCC24B10.11c, SPBC 유전자가존재하였다. SPCC24B10.11c는출아효모의 Mft1보다는사람, 생쥐, 초파리의 THOC7과더유사하였다 (Koh and Yoon, 2014). 본연구에서는 THO 복합체의구성인자로예측되는 open reading frame (ORF) 인 SPBC577.04에대해알아보았다. 이 ORF는인트론이 1개있으며, 200개아미노산 (aa) 으로이루어진예상분자량 23.5 kda, 등전점이 ph 9.29인단 백질을암호화하고있다. 이단백질은포유동물의 THOC5 (683 aa) 보다작고전체적인유사도도매우낮지만, 짧은보존서열이존재한다. 그러므로 THOC5의분열효모이종상동체 (spthoc5로명명 ) 를암호화하는것으로예측되는 SPBC 유전자가생장및 mrna방출에관여하는지, 세포내어디에위치하는지, 그리고분열효모의다른 THO 구성요소들과상호결합여부를조사하였다. 본실험에서는이전에기술된분열효모의유전학적방법과배양방법을사용하였다 (Moreno et al., 1991; Alfa et al., 1993). 먼저 one-step gene disruption 방법을사용하여 spthoc5 결실돌연변이균주를제작하였다. 선별유전자로 ura4 를사용하여 spthoc5::ura4 DNA 절편을 double-joint PCR 방법으로제작한후, 반수체야생형균주인 AY217 (h - leu1-32 ura4-d18) 에형질전환하였다. 우라실이빠진 EMM 최소배지에서자라는형질전환체들을얻은후, 이중 spthoc5 결실돌연변이를선별하기위해형질전환체의 DNA를추출하여 PCR을수행하였다. 대조군으로사용한야생형인균주에서는 2.4 kb의 DNA가증폭되는것에비해 3.5 kb의 DNA가증폭되는결실돌연변이를선별하였으며, ura4 에존재하는프라이머와 spthoc5 유전자의 5 쪽또는 3 쪽프라이머를각각사용하여야생형에서는아무것도증폭되지않지만결실돌연변이에서는각각 0.8 kb와 0.9 kb DNA가증폭되는것도확인하였다 (Fig. 1A). 이렇게반수체 spthoc5 결실돌연변이를얻은것은 spthoc5 유전자가생장에필수적이지않다는것을의미한다. S. pombe THO 복합체의구성요소중진화적으로잘보존된 hpr1, tho2 유전자는생존에필수적이지만 (Lee and Yoon, 2012; Koh and Yoon, 2015), 출아효모보다고등생물과더유사한 spthoc7처럼 spthoc5는필수적이지않았다 (Koh and Yoon, 2014). 이러한결과는분열효모에서 THO 복합체의모든구성요소들이하나의복합체로항상동일한기능에관여하지않거나, 구성요소각각의고유기능이존재함을암시한다. 한편출아효모 S. cerevisiae의 THO 복합체의구성요소들은모두최적생장온도에서는생장에아무런영향을미치지않지만, 높은온도에서는온도민감성 (temperature-sensitivity) 을보인다. 그러므로다양한온도에서 spthoc5 결실돌연변이의생장을 spot assay로확인하였다. 대조군인야생형 (WT) 에비해 spthoc5 결실돌연변이는측정한모든온도 (22, 28, 34 C) 에서느린생장속도를보였다 (Fig. 1B). 이러한결과는 S. cerevisiae의 THO복합체와는다르게분열효모의 spthoc5 는생장에필수적이지는않지만, 모든온도에서정상적인생장에필요하다는것을의미한다. 다음으로 spthoc5 단백질이 mrna의핵에서세포질로의방 미생물학회지제 51 권제 4 호

The DNA was coincidentally stained with 4,6-diamidino-2phenylindole (DAPI) and shown in the right panels. spthoc5 단백질의 세포 내 위치를 알아보기 위하여 GFP 유 r 전자를 spthoc5의 ORF의 3 말단에 붙인 spthoc5-gfp::kan DNA 절편을 제작한 후 야생형 반수체 균주인 AY217에 형질 전환하여 spthoc5 유전자 위치에 삽입하였다. 이렇게 제작한 균주는 생장과 poly(a) RNA 분포가 야생형 균주와 거의 차이 가 없기 때문에 spthoc5-gfp 융합단백질이 정상적인 기능을 하는 것으로 여겨진다(자료 미제시). 이 균주에서 spthoc5-gfp 의 세포 내 위치를 형광현미경으로 관찰한 결과, 세포질에서 도 약간 존재하였지만 대부분 핵 안에서 관찰되었다(Fig. 2). 이러한 결과는 핵 안에서 역할을 하는 THO/TREX 복합체를 고려하면 spthoc5가 THO/TREX 복합체의 구성요소라는 가 정과 잘 부합한다. 출아효모의 THOC5의 상동체가 핵 안에 주로 존재하고 정 상적인 생장과 mrna 방출에 관여하므로, 실제로 다른 THO/TREX 구성요소와 상호결합 하는지를 알아보기 위해 출에 관여하는지 알아보고자 fluorescence in situ hybridization TAP (Tandem Affinity Purification) 방법을 사용하였다(Puig (FISH) 방법을 사용하였다(Yoon et al., 2000). Fig. 1C에서 보 et al., 2001). 이를 위해 주요 THO/TREX 구성요소인 sphpr1 듯이 세포 내의 poly(a) RNA의 분포는 야생형 균주에서는 정 ORF의 3 말단에 TAP tag을 붙인 sphpr1-tap::kan DNA 절편 상적으로 세포 전체에 거의 균일하게 분포하였다. 반면, 을 제작한 후, sphpr1 유전자 위치에 삽입하여 sphpr1-tap 균 r spthoc5 결실돌연변이 균주에서는 핵 안에 poly(a) RNA가 주를 제작하였다(Gould et al., 2004). TAP tag에는 Protein A 약간 더 축적되는 것이 관찰되었다. 이러한 결과는 분열효모의 와 CBP (Calmodulin binding peptide) tag이 연속적으로 존재 THOC5 상동체도 정상적인 bulk mrna의 방출에 필요하다 한다(Rigaut et al., 1999). 한편 spthoc5를 포함하여 THO/ 는 것을 의미한다. 이렇게 spthoc5 유전자가 결실되면 생장과 TREX 구성요소로 알려진 sptho2와 spthoc7 단백질의 N 말 mrna 방출에 결함을 보이므로, 반대로 과발현(over-expression) 단에는 HA tag을 붙이기 위해 pslf273 벡터에 각 유전자의 되는 경우에도 표현형에 영향을 미치는지를 알아보았다. 강 ORF를 클로닝하였다(Forsburg and Sherman, 1997). 제작된 력한 야생형 nmt1 프로모터를 이용하여 spthoc5 단백질이 정 균주와 벡터들은 DNA 염기서열 분석을 통해 이상이 없음을 Korean Journal of Microbiology, Vol. 51, No. 4

Input whole cell extracts (WCE), supernatants (Sup), and eluents from IgG bead (IgG Elute) are shown as indicated. 확인하였다. 이렇게확인된벡터들을 sphpr1-tap 균주그리고대조군으로사용될 sphpr1-tap 단백질이발현되지않은균주 (Tap) 에각각형질전환하였다. 형질전환체로부터얻은세포추출물을사용하여 sphpr1-tap 단백질을 IgG-Sepharose beads로포획하였다. 포획된 sphpr1-tap 단백질과결합한 HA-단백질을 Western blotting으로분석함으로써, 각각의단백질들이서로물리적으로상호작용을하는지를알아보았다. 대조군에는 sphpr1-tap 단백질이발현되지않으므로, HAtagged 단백질은정상적으로발현되었지만어느단백질도 IgG-Sepharose beads로포획되지않았다 (Fig. 3, lanes 4 6). 반면, sphpr1-tap 단백질이 IgG beads에포획된경우, HAspThoc7는같이포획되지는않았지만 HA-spTho2와 HAspThoc5는같이포획되었다 (Fig. 3, lanes 1 3). 특히 HA-spThoc5 단백질이 HA-spTho2보다더많이포획되었다. 이결과들은분열효모의 spthoc5 단백질은 sphpr1 단백질과강하게결합한다는것을의미한다. 앞선모든결과들을종합하면 spthoc5 는분열효모에서도 THO/TREX 복합체의구성요소로서생장과 mrna 방출에관여한다는것을보여준다. 적요 THO/TREX 복합체는전사신장, mrna 가공및방출, 그리고유전체안정성에중요한역할을담당한다. 분열효모 Schizosaccharomyces pombe에서 THO/TREX 복합체의한구성요소인 THOC5의이종상동체를암호화하고있는 SPBC 유전자를찾아, 그것의기능을분석하였다.
