Sunshik basically was made of the heated cereals. Amount of spore-forming Bacillus cereus was detected and it has been caused some problem of food safety. B. cereus was isolate from 57 out of 161 Sunshik samples resulting in the isolation rate of 35.4 %. Quantitative analysis of 57 samples showed that 21 samples were less than 100 CFU/g, 33 samples were between 100 and 1,000 CFU/g and distinctively even 3 (1.9 %) samples had over 1,000 CFU/g. Typical morphology of B. cereus isolated from Sunshik was observed on MYP agar and then further characteristics was identified by using VITEK 2 (Biomeriux, France). 53 strains out of 57 strains isolated from Sunshik (about 93.0 %) produced diarrheal enterotoxin in brain heart infusion broth which was detected by the Bacillus cereus enterotoxin reversed passive latex agglutination test kit (Oxoid England). The D-values of the B. cereus spores were 75 o C (37.1mim), 80 o C (22.5mim), 85 o C (4.9mim), and 90 o C (3.1mim) respectively. The Z-value was calculated 12.8 o C in Sunshik sample inoculated with B. cereus. Therefore, the management of B. cereus in Sunshik is required for the food-safety. Key words: Bacillus cereus, Enterotoxin, D-values, Z-values Bacillus cereus는그람양성균으로서포자를형성하며, 운동성을지닌호기성균이나혐기적상태에서도잘자란다 1). 이균은토양, 물, 공기등자연계에서흔히발견되는미생물로식품에혼입될가능성이높아과채소류, 과채류가공품, 곡류건조제품, 향신료, 식육및식육가공품, 유제품그리고즉석조리식품등에오염될확률이매우높은것으로보고되고있다 2). B. cereus는일정한생육조건이되면독소를생성하며, 생성된독소에의하여식중독이발생한다. B. cereus에의하여생성된독소는 diarrhoeal type과 emetic type의두가지가있다. Emetic type의독소는식품에서 B. cereus가영양세포상태로성장하거나발아하는동안에생산된독소로쌀, 파스타, 국수등을튀기거나조리한전분질식품에의해서발생한다 4). 특히 B. cereus는불안전한조리과정을거친식품을 48 o C이하로유지한경우빠른증식을하여식중독을다수일으키나, B. cereus에의한식중독증상은미약하고보통 24시간내에치료가가능하므로식중독발생건수에비해보고가많이되어지고있지않다 5). 식생활의간편화추세에의거식사대용으로각광을받는선식은채소, 견과류나해조류등의식품류를볶아서건조후에분쇄혼합하여포장한것으로, 포장후더이상가열처리과정없이섭취하는식품이다. B. cereus는 endospore를형성하여제조열처리과정에서완전히사멸되지않으며생존한포자는일정조건이되면다시증식되므로선식의경우그위해성이문제시되고있다 6). 포자는 5-6 log spores/g의범위에서일반적인감염수치이며 343

2 344 Yong-Sun Cho, Eun-Young Jung, Myung-Ki Lee, Cheul-Young Yang, and Dong-Bin Shin 과거부터 B. cereus의포자를비활성화시키기위한여러가지방법이연구되어오고있다 7). 포자는수분활성도 (a w ) 에의해서열에대한저항성이영향을받으며식품에포함되어있는용질에의해서 a w 를감소시키면특이한내열성효과를나타낸다고보고되어있다 8). 최근의연구결과에의하면생식및선식에서포자를형성한 B. cereus가상당수검출되어안전성에문제화되어식품공전에서는관련식품에서 B. cereus에대한정량규격이설정되었다. 일부 B. cereus의효율적인관리및저감화를위한연구가이루어졌으나아직이에대한연구가더필요한실정이라고사료된다 9). 따라서본연구에서는시중에서유통되고있는선식을수거하여제품중의 B. cereus를정량함으로서오염수준을파악하고자하였으며, 선식에서분리한균주의생화학적특성을규명하고이에따른독소형태를파악하고자하였다. 또한저감화를위한방안의일환으로선식의제조과정에서 B. cereus의열처리과정에대한사멸정도를측정하였다. 재료및방법 시험재료국내시중에서판매하고있는선식을 2006년 5월부터 12월까지 70점, 2007년 5월부터 12월까지 91점을구입하여시료로사용하였다. 시료는매장에서임의적으로곡식을선정하여직접갈아서제조한제품을선정하였다. B. cereus (ATCC 11778) 은한국미생물보존센터 (KCCM) 에서분양하여사용하였다. 시험방법 B. cereus 정량시험시료 25 g을취하여 225 ml의멸균인산완충용액에가하고균질화하였다. 균질화한시험액을 MYP agar (Merck, Germany) 5매에 200 µl씩도말한뒤 30 o C에서 24시간배양한후혼탁한환을가지며 lecithinase를분해한분홍색집락을선별계수하였다. 계수한평판에서 5개이상의전형적인집락을선별하여 Nutrient agar (Merck, Germany) 에접종하고 30 o C에서 24시간배양한후 VITEK2 (Biomerieux, France) 를이용하여동정하였다. 동정이확인된균수에희석배수를곱하여최종집락수를산정하였다 10). (Oxoid England) 를사용하여각각감작 latex와대조 latex 를첨가하여 10분간교반한후, 20 o C에서 18~20시간방치후 latex의침강상을육안으로관찰하여결과를판정하였다 11). Starch 가수분해시험 B. cereus의전분가수분해를알아보기위하여 Nutrient agar (Merck, Germany) 에가용성전분을 0.2 %(w/v) 첨가하고이에분리동정한균주를도말하여 30 o C에서 48시간배양하였다. Lugol's liquor를이용하여자란집락주위에가하여 clear zone이있는균주를전분가수분해양성으로판정하였다 4). 혈액배지에서 Hemolysis 시험 0.85 % sodium chloride와 5 %(v/v) defibrinated나 oxalated blood가함유된 Nutrient agar (Merck, Germany) 에분리균주를도말하여 30 o C에서 48시간배양후 β-hemolysis를확인하였다 12). B. cereus의 tryptic soy broth에서의사멸률시험 Tryptic soy broth (Merck, Germany) 에 B. cereus ATCC 균주를 30 o C에서 1주일배양한후 log 포자가되도록하였다. 만들어진포자는 tryptic soy broth (Merck, Germany) 에접종하여 75 o C, 80 o C, 85 o C, 90 o C 의각온도에서시간대별로 water bath (Vison, Korea) 에서열처리한후에바로냉각하였다. 냉각후포자시험액을일정농도로희석후 Plate count agar (Merck, Germany) 에 3 평판씩분주하여혼합희석평판배양법을이용하였다. 시험평판은 30 o C에서 24시간배양한후에집락수를산정하였다. 산정한집락수로부터각온도별 D-value 및 Z- value를산출하였다. 선식에서의 B. cereus 사멸률시험 선식을이용한사멸률시험은정성시험을통해 B. cereus 가검출되지않은선식을 UV하에서 20분간방치한후이에일정량의포자를접종하여충분히균질화한다음초기접종균수를확인하고 tryptic soy broth 방법과동일한온도와시간조건에따라저감화시험을하였다. 선식에포함되어있는다른균주를고려해서 MYP (Merck, Germany) 평판을이용하여집락수를산정하였다. Enterotoxin 확인시험분리동정한균주를 Brain heart infusion broth (Merck, Germany) 에접종하여 35 o C에서 6-18시간배양하였다. 배양액을 3,000 rpm에서 20분간원심분리하여상등액을시험용액으로사용하였다. 시험용액은 Bacillus cereus enterotoxin reversed passive latex agglutination(bcet-rpla)kit 결과및고찰 B. cereus 정량시험시중유통중인선식의 B. cereus의오염정도를파악하기위해 161점에대해정량시험을한결과 Table 1과같다. Table 1에서보는바와같이 161점중 57점 (35.4 %) 에

Characterization of B. cereus strains isolated from Sunshik VITEK BCET-RPLA Starch hydrolysis hemolysis No. of samples B.crerus/B.thuringiensis B.crerus/B.thuringiensis B.crerus/B.thuringiensis B.crerus/B.thuringiensis B.crerus/B.thuringiensis B.crerus/B.thuringiensis 서 B. cereus가검출되었다. 검출된시료 57점중에 100 CFU/g 미만은 21점이고 100-1,000 CFU/g 미만은 33 점, 1,000 CFU/g 이상은 3점으로검출되었다. 이는박등 13) 이보고한생식의 CFU/g 정도의오염도와는유사한수준으로확인되었으나조등 14) 이보고한생식의양성률 43 % 보다는낮은수치를나타냈다. 이는선식의경우제조과정상가열처리과정을거치므로생식에비해낮은양성수치를나타나는것으로사료된다. 그러나본연구에서정량한시료중 3점에서 1,000 CFU/g 이상이검출되어 B. cereus가가열처리에서도사멸하지않음을알수있었다. 또한가열처리후건조를통해서낮은수분활성도를가지는선식의특성에도불구하고 B. cereus가사멸하지않는것은포자를형성하고있다가발아한것으로사료된다. B. cereus 분리균주의특성선식에서분리한 B. cereus의생화학적특성을통해서독소형태를알아보고자하였다. Table 2와같이 BCET- RPLA를이용하여 enterotoxin을시험한결과총 57점중 42점에서양성 (90.3 %) 으로대부분의분리균주는 enterotoxin을생성함을알수있었다. 이는 Sarrias 등 11) 이 spanish 쌀인곡류에대해서실험한결과와유사하였다. Starch hydrolysis양성은전체중 86.0 % 이며 hemolysis는양성은 93.0 % 였다. Emetic toxin을가진 B. cereus로예상되는균주의생화학적특성은 enterotoxin 음성이며 starch hydrolysis 와 hemolysis 음성인균주로 15-17) 총 57점중 3점 (5.3 %) 이검출되었다. 이 3점의시료의정량결과가두점의시료는 80 CFU/g이검출되었고다른한점의시료는 160 CFU/g 이검출되어 Infective dose가 cells/g에 emetic toxin 에의한식중독이발생하므로상대적으로발병률은낮은것으로보였다. 하지만제조과정에서 48 o C이하로온도가 Table 3. Mean D-values (min) of B. cereus ATCC strains suspended in tryptic soy broth and heated at different temperatures Temperature ( o C) D-value (min) correlation coefficient 떨어지면포자가발아해서 B. cereus 가잘자라게되므로이에대한관리가요구된다 5). 온도에따른 B. cereus의 D-value B. cereus에대한저감화방안을모색하고자온도에따른 90 % 사멸률인 D-value를시험하였다. Table 3과같이 tryptic soy broth에포자를접종하여 D-value를산출한결과 75 o C에서는 40.3분, 80 o C에서는 8.8분, 85 o C에서는 3.3 분, 90 o C에서는 1.1분으로나타났다. Sarrias 11) 는 Spanish 쌀에서분리한 B. cereus에대해서 90 o C에서 D-value를산출한결과 4.6분에서 14.0분의범위라고보고하였다. 본연구결과와의차이는균주의특성에기인한것으로사료된다. 가열처리하는과정중에 B. cereus는 90 o C에서 10 분이상가열해도완전하게사멸을하지않았는데이는선식의제조공정과정중에고온으로단시간재료를가열하는과정에서는 B. cereus가완전히사멸하지않았기때문인것으로사료된다. 특히 Kim 7) 등에의하면고추장에서분리한균주의경우는 85 o C에서 D-value가 45.05분이라고보고하였는데이는고추장에들어있는소금때문에열저항성이상승한것이라고하였다. Margarita 8) 의연구결과에의하면식품중의소금은수분활성도를떨어뜨리고수분활성도가낮아지면열에대한저항성이높아진다고하였다. 따라서수분활성도가낮고용질을첨가하

4 346 Yong-Sun Cho, Eun-Young Jung, Myung-Ki Lee, Cheul-Young Yang, and Dong-Bin Shin 는선식에서의 B. cereus가오염되었을때열에대한저항성을비교해보기위해선식시료에일정량의포자를접종한후온도에따른 D-value를측정한결과는 Table 4와같다. Table 4에서보는바와같이 75, 80, 85 및 90 o C 에서의 D-value는각각 37.1분, 22.5분, 4.9분및 3.1분이였다. 75 o C와 80 o C의경우는 tryptic soy broth와선식에서현격한차이를보였는데이는수분활성도가낮은선식으로인해서 D-value가커진것으로사료되며, 이로인해서포자가발아하는온도와비슷해져서높은 D-value가측정되어진것으로예측되어진다. Mean D-values (min) of B. cereus ATCC strains spiked in Sunshik and heated at different temperatures Temperature ( o C) D-value (min) correlation coefficient 온도에따른 B. cereus의 Z-values 결과온도별 B. cereus의 D-value를이용하여 Z-value를구한결과 Fig 1과같이 tryptic soy broth에서 9.8 o C이며선식에접종한경우는 Fig 2와같이 12.8 o C로나타났다. Z- value 온도는 Sarrias 11) 등이쌀에서분리한 B. cereus의 8종에대해 Z-value를산출한결과 7.42 o C에서 8.20 o C로보고하였으며, Johnson 등 18) 은 6.8에서 13.9 o C로보고한바있다. 본연구결과와이들의연구결과를보면 Z-value는균의상태에따라서차이가있는것으로보인다. 그러나선식에직접균주를접종하고얻은수치가아니므로직접적인비교는어려운것으로사료된다. 이러한결과를보았을때이미선식원재료에서오염된 B. cereus는가열처리로서저감화하기어려운것으로보인다. 따라서선식을제조할때원재료에서 B. cereus에대한오염을제거시키는방안을모색하여야할것으로사료된다. Least-squares regression analysis of the thermal death time curve obtained for B. cereus ATCC spores suspended in tryptic soy broth. 요약 최근식사대용으로각광을받는선식은포장후더이상가열처리과정없이섭취하는식품으로포자를형성하여열처리과정에서완전히사멸되지않으며생존하는 B. cereus에의한위해성이문제시되고있다. 따라서시중에서유통되고있는선식에대한 B. cereus정량을통해오염수준을파악한결과선식 161점중 57점 (35.4 %) 에서 B. cereus가검출되었고검출된시료 57점중에 100 CFU/g 미만은 21점, CFU/g 미만은 33점, 1,000 CFU/g 이상은 3점이검출되었다. 선식에서분리한 B. cereus의생화학적특성을통해독소형태를알아본결과는 enterotoxin 양성이 93 % 이며 emetic toxin 의심균주는 5.3 % 였다. 이러한 B. cereus의저감화를위한방안의일환으로선식의제조과정에서 B. cereus의열처리과정에대한사멸정도를측정하였다측정한결과 tryptic soy broth에포자를접종한결과는 75, 80, 85, 90에서 40.3분, 8.8분, 3.3분, 1.1 분이였다. 용질에따라수분활성도가낮아지면열에대한저항성이높아지므로선식에시료에대한 D-value를시 Fig. 2.
