1 JMLS [Journal of Marine Life Science] December 2018; 3(2): [ 보문 ] 수은노출에대한기수산물벼룩 Diaphnosoma celebensis 의항산화반응 배철희 이영미 * 상명대학교자연과학대학생명과학과 Antioxidant Responses in Brackish Water Flea Diaphanosoma celebensis - Exposed to Mercury Chulhee Bae, Young-Mi Lee * Department of Life Science, College of Natural Sciences, Sangmyung University, Seoul 03016, Korea Corresponding Author Young-Mi Lee Department of Life Science, College of Natural Sciences, Sangmyung University, Seoul 03016, Korea ymlee70@smu.ac.kr Received : November 30, 2018 Revised : December 05, 2018 Accepted : December 10, 2018 수은은다양한산업활동에사용되어해양환경내에지속적으로유입되며먹이사슬을따라축적되며생물체내로유입시해양생물의성장, 발생, 생식, 대사등에악영향을미칠수있다. 본연구에서는기수산물벼룩 Diaphanosoma clelbensis 에대한수은의급성독성과산화적스트레스지표 ( 총 glutathione 함량, GST, GR, GPx 활성 ) 를이용한항산화반응을조사하였다. 24 시간수은을노출시킨결과생존율이농도의존적으로감소하는양상을보였으며, 24 시간 LC 50 값은 mg/l (95% C.I ~0.655 mg/l) 로나타났다. 수은 (0.08 과 0.4 mg/l) 을 24 시간노출시킨 D. celebensis 에서총 glutathione 함량은유의하게감소하는양상을보였으며, GST, GR, GPx 활성은유의하게증가하는양상을보였다. 이러한결과는수은노출에의해 D. celebensis 에서산화적스트레스가유도되었음을의미하며, 이들산화적스트레스지표의변화는세포내에서방어기전으로작용하였음을나타낸다. 본연구는 D. celebensis 에서수은독성의분자메커니즘을이해하는데도움이되며, 중금속오염된해양생태계모니터링과해양생물의건강성평가에서이들분자지표의활용가능성을제시한다. Mercury (Hg) poses a threat to marine ecosystem due to continuous inflow from various industries and bioaccumulation to higher trophic level via food web. Mercury can adversely affect growth, development, reproduction and metabolism to aquatic organisms. In the present study, acute toxicity and oxidative stress markers (total glutathione content, and activities of GST, GR and GPx) were investigated in brackish water flea Disphanosoma celebensis exposed to HgCl 2 for 24 h. As results, Hg showed negative effect in survival of D. celebensis. 24 h-lc 50 value was determined as mg/l (95% C.I ~0.655 mg/l). After exposure to Hg (0.08 and 0.4 mg/l) for 24 h, total glutathione content was significantly decreased, whereas GST, GPx and GR activities were enhanced. These findings indicate that Hg induced oxidative stress in D. celebensis, and oxidative stress markers may be involved in cellular defense against Hg - mediated toxicity. This study provides a better understanding of molecular mode of action of Hg toxicity in this specie and potent of molecular markers for heavy metal monitoring in marine ecosystem. Keywords: Acute toxicity( 급성독성 ), Antioxidant enzyme( 항산화효소 ) Diaphanosoma celebensis, Mercury( 수은 ), Oxidative stress( 산화적스트레스 ) 서론 수은은자연계에존재하며환경중에서순환하는데, 화석연료 의연소과정중에방출되며그외페인트, 전자기기, 펄프산업, 의료등다양한산업분야에사용되면서환경내로지속적으로유입되고있다 (Boening, 2000). 수은은화학적형태에따라금속형태 (Hg), 무기수은 (HgCl 2), 유기수은 (CH 3HgCl) 로분류하며, 환경중으로배출되는수은의형태는대부분금속원소형태나무기수

2 15 December 2018; 3(2): 수은노출에대한기수산물벼룩 Diaphnosoma celebensis 의항산화반응 75 은형태이다 (Joe et al., 2018). 해양환경내로유입되는수은은주로인접한육상을통해유입되어퇴적물내에축적되어미생물에의해독성이강한메틸수은 (MeHg) 으로변형될수있으며 (Domagalski, 2001; Valko et al., 2007), 해양생물체내에축적되어생리적, 생화학적변이를유발하여성장, 생식, 대사등에영향을미칠수있다 (Coen et al., 2001; Kidd and Batchelar, 2012; Hudspith et al., 2017). 한반도전연안표층퇴적물에대한수은의분포농도는 0.21~2,020 μg/kg( 평균 μg /kg) 으로넓은범위를나타냈다 (Joe et al., 2018). 수은과같은비필수금속 (non-essential metal) 은 redox active metals( 예, 구리, 철등 ) 과같이직접적으로활성산소종 (reactive oxygen species, ROS) 를생성하기보다는세포내 -SH (thiol group) 을갖는 GSH나단백질 (e.g. 항산화효소 ) 을공격하여활성을떨어뜨려간접적으로산화적스트레스를야기한다 (Buchwalter, 2001; Rooney, 2007). 장기적인산화적스트레스는세포내 DNA, 단백질및지질의산화를유도하여결과적으로생물의노화, 질병및죽음을초래할수있다 (Lushchak, 2011). GSH는비효소성항산화물질로 -SH기를통해활성산소종이나친전자성대사산물제거에중요한역할을담당하기때문에세포내에서 GSH의양은항산화능력에중요한지표가된다. GSH는이러한물질과결합하며산화형 GSSG 형태로바뀌고다시 glutathione reductase (GR) 에의해환원형 GSH의형태로변환된다. GSH는또한친전자성물질의제거에관여하는 glutathione S- transferase (GST) 와과산화수소 (H 2O 2) 의제거에관여하는 glutathione peroxidase (GPx) 의기질로사용될수있어세포내활성산소종및친전자성물질이지속적으로유지되경우 GSH 양의감소를유발하여세포의항산화효율이감소될수있다. 따라서 GSH 및 GSH 관련효소의활성변화는다양한생물에서산화적스트레스에대한지표로사용되어왔다 (Valavanidis et al., 2006). 무척추생물은해양생태계내에서 1차소비자로에너지전달자로, 특히수은과같이생물농축되어먹이사슬상위포식자에게전달되는물질을생태계내에전파하는데주요역할을담당한다. 그럼에도불구하고수은에대한독성은주된인간의식량원인어류와대형갑각류에집중되어있고, 반면해양무척추생물에대해서는일부이매패류 (Chen et al., 2013) 와요각류 (Lee et al., 2017) 등에서만일부보고되어있다. 지각목 (Cladoceran) 은다양한물질에대한독성평가에주로사용되어왔다 (Freitas and Rocha, 2011). 특히지각목의대표종인담수산물벼룩 (Daphnia magna) 는다양한화학물질에대한민감성때문에생태독성평가의모델종으로사용되어왔다 (Poynton et al., 2007; Jemec et al., 2016). 그러나해양환경을평가할수있는생물종은상대적으로부족한실정이다. 현연구에서사용된기수산물벼룩 Diaphanosoma celebensis 는지각목에속하며태평양연안에열대지역에분포하고광염성을나타내어연안생태계모니터링을위한모델종으로적합하다 (Korovchinsky, 1989). 또한생활사 가짧고, 실험실내에서사육이용이하며, 단성생식으로유전적동일성이높아독성실험에적합한종이다 (Marcial and Hagiwara, 2007). 최근 D. celebensis 전사체분석결과담수산물벼룩 D. magna 와유전자정보의유사성이매우높은것으로나타나향후유전자수준에서의다양한연구에활용가능하다고판단된다 (Kim et al., 2018). 따라서본연구에서는기수산물벼룩 D. celebensis 에무기수은을처리하여급성독성을조사하였고, 총 glutathione 함량및 GSH 관련효소들 (GR, GST, GPx) 의활성을측정하였다. 본연구의목적은수은에대한산화적스트레스기전을분자적수준에서이해하고자하며, 수은오염에대한모니터링을위한분자마커로서이들유전자의활용성을평가하는것이다. 재료및방법 1. 배양 기수산물벼룩 D. celebensis 는한국해양과학기술원이균우박사에게분양받아상명대학교유전학실험실에서배양되고있는것을사용하였다. 배양조건은여과된 15 psu 인공해수에서, 25, 12시간 :12시간의광주기를유지하였다. 배양수 1 ml당 1개체가되도록하며, 배양먹이는 Chlorella vulgaris 를 cells/l 씩일주일에 3회제공하였다. 2. 수은노출실험모든시약은분자생물학등급을사용하였으며특별한언급이없는한 Sigma-Aldrich Co. (St. Louis, Mo, USA) 으로부터구입하였다. 수은의 stock solution (1 g/l) 은 HgCl 2 를증류수에녹여서제조하였다. 아치사농도를결정하기위하여 OECD 가이드라인 TG202 (OECD, 2004) 에따라일부수정하여급성독성시험을실시하였다. 각각 0, 0.25, 0.5, 0.75, 1 mg/l 농도의수은이포함된배지를넣은 6-well plate 에 D. celebensis 성체를농도당 5마리씩넣고 24시간노출시켰고, 모든실험은 4반복 ( 총 20개체 / 농도 ) 으로수행하였다. 이때 10초간움직이지않는개체를죽은것으로간주하였다. 대조군의사망률은 5% 를넘지않았다. 결과값으로부터 ToxRat TM Professional software 를이용하여 lethal concentration (LC) values 와 95% 신뢰구간을결정하였다. 항산화물질의분석을위하여상기결과를토대로수은의처리농도는 LC 10 과 1/5 LC 10 으로선정하였다. 3. 총 glutathione 함량분석총 gluathione 함량은 OxiSelect TM Total Glutathione (SSG/GSH) assay kit (Cell Biolabs, INC, USA) 를사용하여측정하였다. 3 76 배철희 이영미 bensis 성체 300마리에 HgCl 2 를 0.08과 0.4 mg/l 를 24시간처리한후모아차가운 5% metaphosphoric acid (MPA) 에넣어균질화한시료를원심분리하여상층액만모아분석에사용하였다. Glutathione reductase 와 NADPH를추가한후 VersaMax ELISA Microplate Reader (molecular Devices, USA) 로 405 nm에서흡광도를측정하였다. 총 glutathione 함량은 glutathione standard curve를통해계산하였고, 대조군에상대적인값으로표시하였다. 4. GSH-related antioxidant enzyme activity 분석 GR 활성은 GR activity assay kit (BioVision, USA), 그리고 GPx 활성은 GPx colorimetric activity assay kit (BioVision, USA) 를사용하여분석하였다. D. celebensis 성체 300마리에 Hg를 0.08과 0.4 mg/l 를 24시간처리한후모아 assay buffer 를넣고균질화균질화한시료를원심분리하여상층액만모아분석에사용하였다. GR 분석의경우상층액을 3% H 2O 2 와혼합한후 5분간 incubation 시켰다. 이후 catalase 를추가처리한후다시 5분간 incubation 시켜준비하였다. 5,5'-Dithiobis (2-nitrobenzoic acid) (DTNB) 를기질로사용하였다. GPx 분석을위해상층액을 NADPH, GR, GSH와혼합한후 15분간 incubation 시켜준비하였다. Cumene Hydroperoxide 용액을 GPx 반응을개시하기위해추가하였다. 반응은 VersaMax ELISA Microplate Reader (molecular Devices, USA) 를사용하여 GST와 GPx 는 340 nm, GR은 405 nm에서각각흡광도를측정하였다. 5. 통계분석데이터는 3반복의평균 ± 표준편차 (S.D.) 로나타내었다. 상대적인활성의차이는 Turkey's test 후 one-way analysis of variance (oneway ANOVA) 으로비교하였다. 통계분석은 PASW statistics 18.0 (SPSS Inc., Chicago, IL, USA) 을사용하였고, 0.05보다낮은 p값을통계적으로유의한것으로간주하였다. 결과 1. 수은의급성독성 본연구에서는수은에 24시간노출된기수산물벼룩 D. celebensis 에서생존률이농도의존적으로감소하는것을확인하였다 (Fig. 1). 이를토대로 24 h-lc 50 값은 mg/l (95% C.I.: ~0.655 mg/l) 였고, 24 h-lc 10 값은 mg/l (95% C.I.: ~0.481 mg/l) 로계산되었다. 이를토대로항산화물질의함량및활성분석을위한수은의노출농도는 LC 10 값과 1/5 LC 10 값으로결정하였다. 2. 수은노출에의한항산화물질의함량및활성변화수은에 24시간노출한 D. celebensis 에서총 glutathione 의함량은대조군과비교하여실험군에서유의하게감소하는양상을보였다 (p < 0.05; Fig. 2A). GR 활성의경우 0.4 mg/l 수은처리그룹에서유의하게증가되었다 (p <0.05; Fig. 2B). GST 활성의경우 0.08 mg/l의수은을처리했을때약 2.5배가량유의하게증가하는양상을보였다 (p < 0.05; Fig. 2C). 한편 GPx 활성은 0.08 mg/l 에서약간증가하였으나통계적으로유의한결과를얻지못했다 (p > 0.05; Fig. 2D). 고찰수은은인간의활동을통해환경내로끊임없이유출되고있어그위험성으로인해국제협약을통해수은사용및제조에대한규제를강화하였지만환경내에서지속적으로검출되고있다 (Klaassen et al., 1986; NIER, 2016). 인간에대한수은독성의관심이높아수서생물의경우어류와갑각류등의영향연구에주로집중되었다. 그러나해양생태계에서먹이생물로중요한역할을담당하는동물플랑크톤에대한수은의독성작용메커니즘에대한연구는상대적으로부족한실정이다. 본연구에서무기수은을기수산물벼룩에 24시간노출시킨결과 LC 50 값은 mg/l로나타났다 (Fig. 1). 다양한수서무척추동물의수은에대한 LC 50 값을 Table 1에비교하였다. 대부분의보고된갑각류 (Crustacean) 의경우성체는 3,300~10,000 μg/l (48 h

4 15 December 2018; 3(2): 수은노출에대한기수산물벼룩 Diaphnosoma celebensis 의항산화반응 77 LC 50; Brown shrimp), 930~2,200 μg/l (24 h LC 50; Crab) 등으로다양한범위를보였다 (WHO, 1989). 또한 Whiteleg Shrimp (Litopenaeus vannamei) 의 96 h LC 50 값도 1.23 mg/l로 (Frias-Espericueta et al., 2001) 다소크기가큰종들에비해서는 D. celebensis 의수은에대한민감도가상대적으로더높았다. 해양요각류인 Calanoid copepod 에속하는 Calanus glacialis 와 C. finmarchicus 의수은에대한 48시간 LC 50 값은각각 73.7 μg/l와 72.4 μg/l로나타났다 (Øverjordet et al., 2014). Verslycke et al. (2003) 은해양에분포하는 mysid Neomysis integer와 Americamysis bahia에대한수은의 96시간 LC 50 값을각각 6.9 μg/l와 3.5 μg/l로보고하였다. 한편서로다른조건에서 12~55 μg/l 의 24시간반수치사농도를보인같은지각류 (Cladoceran) 에속하는담수종인 Daphnia magna 보다는민감도가낮았다 (Tsui and Wang, 2006). 또다른 Cladocerans 에속하는담수종 4종 (Ceriodaphnia dubia, Daphnia laevis, D. pulex, D. similis) 들의수은민감도를비교한 Sarma와 Nandini (2006) 에따르면, 수은에대한 24시간 LC 50 값이가장높은종은 D. laevis 로 neonate 에서약 20 μg/l, adult 에서약 40 μg/l를보였으며 4종에서모두발생단계에따른민감도의차이를보였다.

Redox inactive metal 인수은은세포내에서 sulfhydryl 기 (-SH) 를갖는 GSH 및항산화효소와결합하여이들의방어기작을방해하는것으로알려져있다 (Buchwalter, 2001). 본연구결과에서는 24시간아치사농도의수은에노출된 D. celebensis 에서총 glutathione 함량은감소한반면, GR와 GST 활성은증가하는양상을보였다 (Fig. 2). 그러나 glutathione 함량과 GSH-related antioxidant enzymes 의활성은항상일관된양상을보이지는않는것같다. 예를들어, 수은에노출 된 golden grey mullet (Liza aurata) 에서는총 glutathione 함량과 GPx, GR, 그리고 GST의활성이모두유의하게감소했다 (Mieiro et al., 2010). 저자들은이에대해수은이항산화시스템을저해함으로써 L. aurata 의뇌에서나타나는신경독성을유발했을것으로추측하였다. 특히본연구에서총 glutathione 함량의감소는수은의타겟이 -SH기를갖는분자라는점과일치하는결과라고판단되며, 추가적인요인으로 glutathione 을기질로사용하는 GST 활성의증가가 glutathione 함량의고갈에기여했을것이라생각된다. 또한 glutathione 의감소는사용된환원형 glutathione 을회복하기위해 GR 활성을증가시켰을것으로보인다. 반면, 수은을 96시간노출시킨 crab Carcinus maenas (Elumalai et al., 2007) 에서 GST 활성이유의하게증가하였고, tropical freshwater fish matrinxã 에서는 GSH 함량이증가하였다 (Monteiro et al., 2010). 한편, 48시간노출시킨 D. magna 에서도총 GSH 함량과 GST 활성이모두유의하게증가하는양상을보였다 (Kim et al., 2017). 특히이들은 48시간수은처리농도 (0.08과 2 μg/l) 에서모두활성산소수준이유의하게감소하는양상을보고하였다. 이

6 15 December 2018; 3(2): 수은노출에대한기수산물벼룩 Diaphnosoma celebensis 의항산화반응 79 러한결과와관련하여저자들은세포내 GSH:GSSG 비율의균형을유지하기위해 GSH가증가한것으로보이며 (Monteiro et al., 2010), GSH 함량과 GST 활성의증가는 ROS 수준을감소시키는데관여했을것이라제시하였다 (Kim et al., 2017). 상기결과들을토대로수은에대한 GSH와 GSH 관련항산화효소의관계는수은에대한종의민감도와실험조건의차이등에따라서로다른결과를나타낼수있는것으로판단된다. 본연구에서는기수산물벼룩 D. celebensis 에수은을노출시킨결과생존율에영향을미쳤으며이를토대로수은노출에따른 24시간반수치사농도를결정하였다. 아치사농도의수은처리후총 glutathione 함량은감소하는경향을보였고, GST와 GR의활성은증가되는양상을보였다. 결과에서산화적스트레스지표의변화는수은노출에의해 D. celebensis 에서산화적스트레스가유발되었음을의미하며이들항산화물질이세포내에서방어기전으로작용하였음을나타낸다. 또한이러한분자지표는수은과같은중금속에오염된해양생태계의생물건강성을평가하는데활용될수있을것으로생각된다. 참고문헌 Boening DW Ecological effects, transport, and fate of mercury: a general review.
