Early symptoms on fruit showed small, circular, light brown spots that eventually destroyed the entire fruit. Small sporodochia appeared on the fruit surface. Fruit susceptibility to brown rot increases during the 1 to 2 weeks period prior to harvest. The conidia were one-celled, hyaline, lemon-shaped, μm, and borne in branched monilioid chains. Based on the morphological characteristics and phylogenetic analysis of internal transcribed spacer (ITS), the fungus was identified as Monilinia fructicola. A BLAST search revealed that sequences of the fungus shared 100% identity to those of M. fructicola. Pathogenicity of a representative isolate was proved by artificial inoculation, fulfilling Koch s postulates. To our knowledge, this is the first confirmed report on the occurrence of M. fructicola on apricot in Korea. Keywords: Apricot, Brown rot, Monilinia fructicola, Prunus armeniaca 살구나무 (Prunus armeniaca L., Rosaceae) 는장미과벚나무 속에속하고, 식용이나약용, 정원수로이용된다. 중국이원 산지로한국, 일본, 몽골등아시아와미국, 유럽등지에널리 분포한다. 살구나무의과실인살구는구형의핵과이며, 융 모가있고 6 월에황색으로익어수확하게된다 (Kim, 1975). 국내에서살구나무에발생하는주요진균병으로는 Podosphaera tridactyla (powdery mildew), Cladosporium carpophilum (scab), Blastospora smilacis (rust), Phomopsis vexans (brown rot), Phyllosticta circumscissa, Septoria cerasina (leaf spot), Monilinia fructicola (brown rot), Taphrina mume (leaf curl) 등이보 Research in Plant Disease pissn , eissn 고되어있다 (The Korean Society of Plant Pathology, 2009). 특히, Monilinia균은살구를비롯하여복숭아, 자두, 매실, 체리, 사과, 배등여러과수류에잿빛무늬병을일으키는병원균으로전세계적으로 M. fructicola (G. Winter) Honey, M. fructigena (Aderhold & Ruhland) Honey, M. laxa (Aderhold & Ruhland) Honey 등 3종이알려져있다. 특히, M. fructicola는미국, 호주, 뉴질랜드와일부유럽과아시아에, 그리고 M. fructigena와 M. laxa는유럽에분포하는것으로보고되었다 (Fulton과 Brown, 1997; Hu 등, 2011; Shim 등, 2007). 또한, M. fructicola와 M. laxa 는꽃에감염되어꽃시들음병 (blossom blight) 과건전한과실및상처가있는과실에 brown rot을일으키지만 M. fructigena 는상처받은과실에만병을일으키는것으로보고되었다 (Rungjindamai 등, 2014). 3 124 히, 개화기와등숙기의잦은강우는심각한피해를초래하므로적절한방제대책이없을경우 50% 75% 까지피해를주는것으로보고되었다 (Rural Development Administration, 2008b). 병원균의분리및병원성검정. 병원균의단포자를분리하기위하여발병포장에서이병과를채집하여실험실에서분생포자덩어리를떼어내어멸균수가들어있는 1.5 ml eppendorf 튜브에넣고볼텍스믹서로 30초동안흔들었다. 분생포자가들어있는멸균수를루프로찍어감자한천배지 (Difco, Sparks, MD, USA) 에치상하였다. 병원균을 3일간암배양하여단포자의끝부분을떼어감자한천배지에옮긴후 25 o C 항온기에서 5일간배양한뒤병원균의균학적특성을조사하였다. 병원균이감염된표본은고려대학교표본실 (Korea University Herbarium, KUS) 에보존하였다. KUS-F29117 로부터대표균주가분리되어농업유전자원정보센터 (Korean Agricultural Culture Collection, KACC) 에기탁하였으며 (accession No. KACC48000), 병원성검정및염기서열분석에사용하였다. 또한병원균의특성비교를위하여잿빛무늬병이감염된다른살구에서같은방법으로단포자를분리하여 JBARES37 균주를확보하였다. 병원성검정은 Alexander 품종에서실시하였다. 병원성접종원은 KACC48000 균주를감자한천배지에서 1주일간배양한후멸균수를넣고메스 (scalpel) 로포자를수거한포자현탁액을 conidia/ml로준비하였다. 병원성검정은건전한과실 5개위에포자현탁액을분무하였으며, 대조과는멸균수를분무하였다. 접종과와대조과를밀폐된플라스틱박스에넣고박스안쪽표면에멸균수를분무하여상대습도를 100% 유지하였으며, 25 o C± 1 o C의인큐베이터에보관하였다. 접종 3일후포장에서와같은전형적인잿빛곰팡이병증상이나타나기시작하였으며, 열매가한쪽에서부터갈변되기시작하여 5일째에는전체로확산되었고, 흰색의분생포자덩어리가형성되어병원균이재분리되었다. 따라서살구잿빛곰팡이병을일으키는병원균은병원성이있는것으로판단되며, 대조구에서는병징이나타나지않았다. 병원균의균학적특징및염기서열분석. 병원균의형태적특징을관찰하기위해 KACC48000 균주를감자한천배지에치상하여 25ºC에서 1주일동안배양하였다. 형태적특징은 differential interference contrast (DIC) 광학현미경으로 200 1,000배에서관찰하였다. 감자한천배지에서균사생육은직경이 mm/4일이었으며, 균총은흰색 회색으로중앙부분에무수히많은분생포자가형성되었다 (Fig. 1F). 현 미경으로관찰된분생포자 (n=30) 는분지된염주상으로사슬모양을나타냈으며 (Fig. 1D), 단생, 투명하고, 난형 레몬형으로크기는 μm였다 (Fig. 1E). 이상과같이살구잿빛무늬병을일으키는병원균의형태적특징과병원성검정을기초로 M. fructicola (G. Winter) Honey로동정하였다 (Pellegrino 등, 2009). 형태적특징을기초로한병원균의동정을뒷받침하고자염기서열분석을실시하였다. M. fructicola 병원균 (KACC48000) 을감자한천배지에서 1주일동안배양한균사를 ribosomal DNA (rdna) 의 internal transcribed spacer (ITS) 영역의염기서열에사용하였다. Genomic DNA는 DNeasy Plant Mini kit (QIAGEN, Valencia, Canada) 를이용하여분리하였고, ITS1/ITS4 (White 등, 1990) primer를이용하여 PCR로증폭하였다. 증폭시킨 PCR 산물은 1.5% agarose gel에서전기영동하여 band를확인한후, PCR purification kit (CoreOne TM ; CoreBio, Seoul, Korea) 를이용하여정제하였다. 염기서열은 autosequencer (ABI 3030) 로분석하였으며, 최종획득한 ITS의염기서열들은 GenBank database (National Center for Biotechnology Information [NCBI] US National Institute of Health Bethesda; 에서확인하였다. 계통수분석은 GenBank 데이터베이스에올라와있는 ITS의염기서열들을이용하였다. 계통수는 MEGA 6.0 프로그램을이용하여 neighbor-joining 방법으로작성하였으며, sequence distance는 Tajima-Nei parameter model로계산하였다 (Saitou와 Nei, 1987; Tamura 등, 2013). Botrytis cinerea (GenBank accession No. KM016553) 를계통수분석 outgroup 으로사용하였다. 살구잿빛무늬병을일으키는 KACC48000 균주의 ITS sequence 크기는 558 bp로 NCBI의 GenBank에등록 (accession No. KU513386) 하였다. NCBI에서 BLAST search한결과 M. fructicola로등록된 GenBank accession Nos. KC544809, JN176564, FJ515894, KM 등과 100% 일치하였다. 또한계통수작성결과 KC544809의염기서열은 M. fructicola와같은계통군에속함을확인할수있었다 (Fig. 2). 최근에 ITS 염기서열을이용한 Monilinia종간구분을위하여, Hu 등 (2011) 이중국에서복숭아잿빛곰팡이병을일으키는 M. fructicola, M. yunnanensis, M. fructigena, M. laxa를분리하여 ITS 유전분석을한결과 100% 종간구분이되었으며, 4종간에상당한유전적변이가있음을확인하였다. 그러나과거에는 M. laxa와 M. fructigena에는존재하지않고, M. fructicola의 small subunit (SSU) rdna에만위치한 418 bp group-i intron을분석하여 M. laxa와 M. fructigena로부터 M. fructicola를구분하였다 (Fulton

M. fructicola에의한잿빛무늬병은세계적으로핵과류에발생하는중요한병으로알려져있으며 (Fulton과 Brown, 1997), 미국캘리포니아지역에서는이균의살균제저항성까지보고하였다 (Ma 등, 2003). 그러나우리나라의경우발생생태및병징에관한기록만확인된다. 따라서살구에서잿빛무늬증상을일으키는병원균을분리하여균학적특성, 병원성검정, ITS rdna 염기서열비교분석등의결과를바탕으로 M. fructicola로동정되었으며, 국내에보고된자료를좀더보완하는자료로제출하고자한다. 으키는병원균의형태적특징, 병원성검정, rdna의 ITS 영역의염기서열분석을기초로 M. fructicola로동정하였으며, NCBI에서 BLAST search한결과 M. fructicola로등록된 GenBank accession Nos.
