However, there is no study on the antimelanogenic effect of taurine. In this study, we investigated the whitening effect of taurine in B16F10 mouse melanoma cells. Cell viability was measured by MTT assay. We examined melanin contents and tyrosinase activity according to time and concentration. Extracellular signal regulated kinase (ERK) is an important regulator of melanogenesis. It has been reported that activated ERK induced microphthalmia associated transcription factor (MITF) phosphorylation and its subsequent degradation and thus reduced melanin synthesis. In our B16F10 cell culture system, taurine led to decrease melanin contents by 21% at 48 hr. We then observed taurine effects on ERK- R MITF and tyrosinase by Western blot. ERK was activated at 18 hr and 24 hr, whereas MITF reduced. We could not observe any differences in the levels of tyrosinase. These results suggested that taurine inhibited melanogenesis by ERK signal pathway via MITF degradation. We expect that taurine has potential skin whitening agents in cosmetics. Keywords taurine, ERK, MITF, tyrosinase, melanogenesis 타우린은 semiessential 아미노심 : 이지만단백질로는합성되지 않는다. 포유동물조직에서타우린은백혈구에 50 mm 농도로가 장많고, 심장망막, 근골격에도함유되어있으며조직에서여 러빈움에관여하는중요한성분으로알려져있다. 산화적손 상에조직방어기능 ^* 이있는것으로연구되어져있을뿐만아니 라면역학적측면,^* 산화억제작용, 항염작용 "' 등생물학적작 용기능이다양하게연구되고있어, 최근에는건강식품에도많 이첨가하쉬응용되고있다. 특히고양이과에서는꼭필요한 아미노산으로서타우린이부족하게되면망막의손실로시력에 영향을미친다고보고되있다. 또한향장학에서는타우린이보습 에효과가있는것으로발표되어있으나, 미백이나노화방지에 초점을둔타우린에대한연구는아직미미하다. 자외선이나환경오염그밖의외부요인의자극에대해피부 세포는방어기전으로델라닌을생성한다. 델라닌이많이생성 본논문에관한문의는저자에게로 ( 전화 ) ( 팩스 ) ( ) akkim(o;sookrnyung.ac.kr 되면결과적으로피부암을유발할수있다. 이처럼멜라닌이피부암을유발할수있다는점과사람둘의미적욕구증가로인해향장업계에서는멜라닌의생성의억제에대한관심이점점높어 " 지고있다. 따라서인체에부작용이적으면서델라닌을억제하는기능을가진새로운물질의개발이요구되고있는실정이다. 멜라닌은산화적스트레스나 NO 등에의한자국에의해서도생성이되는데, 이미밝혀진인체의다른부위에서티우린의항산화나항염증효과는피부의델라닌과도관련이있으리라관단되어티우린의델라닌생성억제에대하여실험을하고자하였다. 최근 melanogenesis의 signal로써 ERK pathway의억제는 MITF와 tyrosinase 의활성을증가시켜결과적으로델라닌의 pigmentation을높여주며, 한편으로 ERK의활성은 MITF의 ubiquitination(mrrf- -Ser73) 으로 MITF가 degradation되어 melanin의형성억제률유도?!: 다는연구결과가보고되었다. 델라닌형성과정중 ERK pathway는 Fig. 1에나타내었다. 이에본연구에서는타우린의델라닌억제효과를알아보기위하여 B16F10에서농도와시간에따라멜라닌의양을측정하고, tyrosinase activityl- 확인하고자하였다. 또한 ERK pathway 350

2 B16F10 Cell 에서 Taurine 의 델라닌 생성 억제 효과 351 (H-50): SC-25386, tyrosinase(h-109): sc-15341, TRPi(H-90): SC-25543, TRP2(H-150) : sc-25544, actin(i-19), antibody는 Santa Cruz CA, 2차 antibody는 Cell Signaling에서 구입하였다. 기기로는 ELISA reader (Bio-Tek instrument Inc), cytofluor 2350 plate reader(millipore, Bedford, MA, USA), CO 2 incubator(forma Science), table top centrifuge(hanil Science Industrial Co. Ltd), centrifuge(supra 21K, Hanil Science Industrial Co. Ltd), inverted microscope(01ympus CK2), UV/ visible spectrophotometerdjltrospec 2000, Pharmacia Biotech), semi dry transfer ceil(bio Rad)을 사용하였다. ERKs CREB. CRE 세포배양 Mouse melanoma cell로부터 유래된 B16F10 cell은 ATCC 세포주 은행으로부터 분양받았다. 10% heat-inactivated fetal MITF- Scr73 bovine serum, 항생제(10,000 units/m/ penicillin G sodium, 10,000 ig/m/ streptomycin sulfate), 1 mm sodium pyruvate# MITF, Proleasomal degradation TRP,.THFa T y ro sin a se 포힘하는 DMEM 배지률 배양액으로 허여 s r c, humidified 5% CO2 incubator에서 배앙하였다. 25 cm크tissue culture flask나 Melanin Pigmentation 75 cm크tissue culture flask에서 계대 배양하고 confluent되었을 Fig, 1 - ERK signal pathway regulating melanogenesis. B16F10 cell suspension울 1 x I0'\:ells/m/의 농도로 96-well plate의 well에 KXM 씩 가하늬 배앙기에서 24시간 동안 안정화 시킨 후, 티우린을 농도별로 처러등M 실험 방범 48시간 동안 배양하였다. 2.5 mg/m/ MTT[3-(4,5-dimethylthiazol-2-yl)2,5- diphenyltetrazolium bromide]용액을 well당 50 나씩 넣어 4시간 동안 배양기 시약 및 기기 DMEM(4500 mg/l D-glucose, L-glutamine, 25 mm HEPES, 에 방치한다. 이후 상층액을 제거하고 DMSO률 well당 100 씩 sodium bicarbonate), FBS(fetal vobine serum), taurine(acros 가하여 1분간 shaking하여 formazan을 완전히 용해시킨 루 A )는 웰진에서 구입하였다. ELISA plate reader룰 이용하여 540nm에서 흡광도를 측정하 MTT(3-(4,5-dimethylthiazol'2-yl)2,5-diphenyltetrazoliumbromide), antibiotics(10,000 units/m/ 10,000 jig/m/ streptomycin sulfate), penicillin G L-DOPA, 였다.^ sodium, mushroom Tyrosinase 효소의 활성 촉정 및 Melanin 정량 tyrosinase^ Sigma(Saint, Louis, Mo), tryphan b lu e ^ Gibco Tyrosinase activity 측정 - 0.5X 10^ cells/m/의 B16F10 cell BRL Life Technologies Inc., RIPA buffer는 Sigma(R0278), suspension-i 60 tt tissue culture dish에 가하여 배잉"기에서 24 protease inhibitor cocktail은 Roche(complete mini) 제품을 사* 시간 동안 배양하여 안정화시켰다. 그 후 티우린을 농도벌로 처 용하였다. 라히여 48시간 배양& 후 배지를 제거하고 PBS로 2번 세척하였다. Antibody는 phospho-specific ERK 1/2(Thr 202/Tyr 204, 1% (w/v) Triton X-100을 10 mm sodium phosphate buffer number 9101s), tatalophosphorylated and non-phosphorylated ph 6.8로 만들어 150 m/로 cell을 모은 다옴에 2000 rpm에서 5 ERK l/2(number 9102>은: Cell Signaling Technology/ MITF 분간 centrifuge해서 각 각의 상층액 40 p/에 기 질인 L-DOPA Vol. 51, No. 5, 2007

3 정효숙 송경희 김인근 200 ^ 가태 37X 에서 1 시간배양하였다. 여기에서생성된 dopa chrome의양을 475nm에서흡광도를측정하였다.^가 Melanin 정량 - 원심분리한 pellet을이용하여농도별로 1N NaOH 100 i/ 에증류수 200 m/ 를가해서 6(fC에서 1시간배양한다옴완전히녹인후 405 nm에서멜라닌의양을측정하였다 Western blol으로단백질발현측정 - B16F10 cell suspension 을 tissue culture dish 에각 well당 0.5 x 10 cells/m/ cells 로가한후 24시간동안배양하여 cell을안정화시켰다. 배지률제거한후 40mM 타우린을처리하식 18 시간, 24시간, 48시간벌로배양한후또다시배지를제거하고 PBS로 2번세척해주었다. lysis buffer(ripa buffer 10 m/ 에 complete mini 1 tab률가함 ) 100 nz 로용해해서 4 C 14,000 rpm에서 15 분간원심분리하였다. 원심분리하여얻은상층액은 Bradford assay로정량하여단백질 (20 ng) 을 7.5~15% 의 SDS-PAGE상에서전기영동하여분리하였다. 분리된단백질은 semi dry transfer cell 기기 (Bio Rad) 를이용하여 nitrocellulose membrane 에옮겨, 실온에서 2시간동안 blocking buffer(5% skim milk in TBST) 에서 incubation 시켰다. 10 분간격으로 TBST 로 3회 washing 하고 1 차항체를 1:500으로희석하여 over night한다옴, 다시 10 분간격으로 TBST로 3회 washing 하고 2차항체룰 1:1000으로희석하여실온에서 2시간동안 incubation 시켰다. 3회 washing 하고 vorsadoc으로농도률측정하였다 Taurine(mM) Fig. 2 - Cell viability of B16F10 cells after treatment with taurine. Values are means ±SD and were obtained from three different experiments. *p<0.05, **p<0.001: Significantly different from untreated control cells. 세포생존율 B16F10 cell에서티우린의농도별세포생존율을알아보기위해 MTT assay를실행하였다. 배양기간은 2일로고정하였으며, 타우린의농도는 20~60mM 범위에서실험을시행하였다. 40 mm 농도까지는세포생존율에크게영향을미처지못하였으나타우린의농도가중가함쳐 1 따라세포생존율이농도의존적으로감소하여 taurine 의농 :E 가 60(mM) 에서는세포생존율이 66.3% 로나타났다. 이에타 f 린의농도가 40 mm일때최고능도로하여실험을시행하였다 (Fig. 2). Melanin 정량 B16F10 에서타우린과 kojic acid를농도별로처러하식 48시간 투멜라닌의양을즉정하였다. 멜라닌의앙은 control 대비타우린 20 미보에서는 85%, 40mM 에서는 79% 로측정되어각각 15%, 21% 의델라닌의억제효과를나타내었다. 대조군인 kojic acid의결과는 10 _im 에서는영향이없었고, 100 nm은 91.8% 의멜라닌이측정되어약 10% 의억제율을나타내었다 (Fig. 3). Tyrosinase activity Tyrosinase는델라닌형성의 cascade 반응에관여히는스?. 서전사인자인 MITF 와도밀접한관계가있다. 따라서타우린이델라닌형성의과정중어느단계에서작용하여억제효과를나타내는지규명하기위해서 tyrosinase 의활성을확인하여보 J. Pharm. Soc. Korea

B16F10 Cell 에서 Taurine 의 델타닌 생성 억제 효파 353 120 린(40mM)을 시간벌로 처러한 후 Western blot으로 단백질의 발 100 현 양상을 측정하였다. 그 결과, 18시간과 24시간에 ERK가 activation 되어 ERK-P 80 가 증가 되었으며, 이에 따른 MITF의 발현은 감소 하였다.

Equal protein loadings were confirmed by reaction using phosphorylationindependent ERK antibodies and beta-actin antibody. 구가 선행되어져야 하며, 델라닌 억제 기견에 대해서도 추가로 깊이 있는 실험이 요구된다. 결 론 본 연구에서는 머백 효과를 알아보기 위한 일환으로 침습성이 강하고 전이가 빠른 악성 흑색종 세포에서 티우린의 델라닌 생 성 억제 효파와 그 분자적 기전을 알아보고자 하였다. B16F10 았다. B16F10 cell에서 타우린을 능도별로 처러하여 48시간 후 murine melanoma cell에서 농도와 시간에 따라 델라닌의 양을 측정 하였으며, 대조군으로는 kojic add률 사용하였고, 그 결과 측정하고, tyrosinase 활성을 확인하였으며, ERK-R MITF, 는 control 대비 백분율로 나타내었다. 티우린은 tyrosinase에 오 tyrosinase의 gene 발현을 관찰하여 비교하였다. 그 결과 다옴파 히려 약간의 활성이 있는 것으로 나타났으나 티우린의 델라닌 생 같은 결론을 얻을 수 있었다. 성 억제에는 크게 영향을 머치지 못하는 것으로 고>려 된다. 티우린 40mM 농도로 처리한 세포에서 델라닌 억제 효과가 있었고, 델라닌 형성 과정 중 ERK pathway와 관련이 있으며, 단백질 발현에 미처는 영향 tyrosinase 활성을 약간 상승 시키는 것으로 보아 타우린의 효과 Taurine이 델라닌 형성과정의 signal 인 ERK-R ERK, MITE 는 MITF degradation 이나 MITF gene 발현 억제에 의한 것으 tarosinase 에 어떠한 영향을 나타내는지 알아보기 위하셔 타우 Vol. 51, No. 5, 2007 로 추정된다.
