The antibody titer (percentage inhibition [PI] titer) of the sow (gilt) after FMD vaccination was maintained at a level higher than 50 (P<0.05) for 5 months. A higher PI titer for the 1monthold growing pigs corresponded with greater inhibition of the PI titer of the vaccinated growing pigs (P<0.05). A negative correlation (P<0.05) between the PI titer of the 1monthold growing pigs and the PI titer of 3monthold growing pigs, 4monthold growing pigs after FMD vaccination at 2 months, 3months was identified, with a coefficient of determination (R 2 ) of Thus the PI titer of the growing pigs was inhibited to a greater degree when vaccination was performed at 2 months of age than at 3 months. However, many other factors likely influence growing pigs PI titer in addition to the PI titer of the sow and age at vaccination, given that the coefficient of determination was somewhat lower. FMD는동물에서전염성이강하고, 발생시발생축또는발생지역우제류가축의살처분, 백신접종, 교통통제, 국제교역상불이익발생등국가경제에미치는영향이매우커서세계동물보건기구 (OIE) 에서지정한 List A 질병이다 (Leforban, 1999; Park 등, 2008). 2 16 안길호ㆍ배진규ㆍ정광ㆍ왕영일ㆍ정준용ㆍ강순근ㆍ권혁무 FMD의원인바이러스인 FMDV는 7 종류의 serotype (O, A, C, SAT1, SAT2, SAT3, Asia1) 이각각의 serotype에면역학적으로영향을주지않고 (Callis 등, 1968), serotype 내에서도완전한방어를하지못하는 subtype이있으며 (Paton 등, 2005), 바이러스자체의빈번한변이, 다양한항원성으로인한백신개발의어려움 (Sur 등, 2000) 등의이유로질병근절이용이하지않다. 일반적으로 FMD의확산방지를위하여살처분과 ringvaccination을병행실시하는데 (Pattnaik 등, 2012), Asia 1형을제외한다양한 serotype의 FMD가발병하고있는우간다 (Ayebazibwe 등, 2010) 는발생지역가축이동제한과소에게만 ringvaccination을실시하였지만 (Mwiine 등, 2010), 백신프로그램과효능에대한검사를실시하지않았고지속적인 FMD 발생이보고되고있다 (Muleme 등, 2012). 우리나라에서 FMD는 1934년북한지역에서발생한이후비발생을유지하다가 2000년 3월파주에서다시발생하였으며살처분과백신접종정책으로 (Sur 등, 2000), 2002년과 2010년 1월, 4 5월의발생에서는강력한살처분정책등으로청정화를신속히유지하였으나, 2010년말안동에서발생한 FMD의경우전국적인확산으로백신접종을실시하여 2011년 3 월초부터급격히발생이감소하였고, 이후정기적인백신접종을추진중에있다. 긴급백신의경우 50% protective dose의 6배용량 (6PD 50) 으로만들어지고예방을위하여사용하는정기백신은보통 3PD 50 로만들어지는데 (Forman과 Garland, 2002), FMD의병원성과변이성등을고려하여 (De Clercq 등, 2008; Pay와 Hingley, 1986) 우리나라에서는현재까지 6PD 50 의백신을사용하고있다. FMD 확산을방지하기위하여그동안연구개발된백신으로는일반적으로사용되는 inactivated vaccine, novel attenuated vaccine, live vector vaccine, subunit vaccine, nucleic acid vaccine, synthetic peptide vaccine 등이있으나 (Zhang 등, 2011; Laporte, 1969; Rowlands 등, 1971; Wong 등, 2000; Pfaff 등, 1982; Acharya 등, 1989), 약독형이다시강독형으로돌아가는회귀현상때문에 attenuated virus 제작은성공하지못하고있다 (Sáiz 등, 2002). 아시아의중국, 베트남, 대만등은백신을접종하고있음에도불구하고 FMD가발생하고있고 (Paul 등, 2003), 남아프리카에서는국가생태공원등지속적으로 FMD가발생하고있는지역의소를대상으로 FMD 백신접종을하고있다 (Brückner 등, 2004). 북미및유럽지역은대부분청정국가로백신접종을하지않고있고, 남미는소에대한백신접종을통해질병을통제하고있어 (Cox와 Barnett, 2009; Paul 등, 2003), 돼지를대상으로백신접종에따른항체형성및역가의변화추이등에관한연구가부족한실정이다. 따라서이번연구는번식돈과자돈에서 FMD 백신접종후월별항체검사를통하여항체변화추이를조사하고, 모돈의항체역가와자돈의항체형성및역가등을조사하여실제돼지농가에서일어나는항체유지수준과백신접종효과향상방안을마련하고자수행하였다. 재료및방법농가선정이번연구는 2012년 1월부터 6월까지진행되었다. 경기도포천지역돼지사육농가중일관사육을하고, 번식돈 ( 모돈, 후보돈 ) 개체관리를실시하며, 분만후자돈을다른모돈에입양보내지않아모체이행항체를확인할수있고, 자돈개체관리를통해모돈과자돈간의상관관계를확인할수있는 4농가를선정하였다. 후보돈이없는 1농가 (A농가) 를제외한 3농가에대하여 5 6 두씩총 16두의후보돈을선정하였고, 모돈은 Intervet사에서제조한 FMD 백신 (Decivac R, Germany) 접종후 3주이상경과된개체를농가당 4두씩총 20두를선정하였다 (Table 1). Enzymelinked immunosorbent assay (ELISA) 를이용하여 FMDV의비구조단백질 (nonstructural protein, NSP) 과 FMD 항체 (structural protein, SP) 검사를실시하여 NSP ELISA 검사결과모두음성이었고, SP ELISA 검사결과에서후보돈의경우 1농가 (D 농가 ) 에서만전두수음성이었고, 나머지농가 (B, C 농가 ) 는항체양성률이 50% 이상이었으며, 모돈의경우 1농가 (B농가) 는전두수양성이었고, 1농가 (D농가) 는 1두가음성이었으나도태되었고, 나머지 2농가 (A, C농가 ) 는음성개체가확인되었다. 음성모돈 2두를포함하여총 10두가분만한 80두의자돈에이각및이표를부착하여개체별로고유번호를부여하였고, 모돈은반드시자신이분만한자돈에게만수유하도록별도사육함으로써실험군을농가에서구분할수있도록하였다.

3 돼지에서구제역백신접종후항체가변화조사 17 시료채취번식돈 : 월 1회농가를방문하여개체확인후경정맥에서혈액 5 ml를채취하여혈청분리후비동화하지않고검사전까지 20 o C에냉동보관하였다가실험에사용하였고, 폐사ㆍ도태ㆍ임신의이유로검사를 1회이상시행하지않더라도다음번에검사가능한경우검사를시행하였다. 자돈 : 월 1회씩총 6회에걸쳐개체번호확인후채혈하였으며, 시료채취과정은번식돈과동일하였다 (Table 2). 자돈에 FMD 백신접종 A와 B농가에서태어난실험대상자돈은 2개월령에, C와 D농가에서태어난실험대상자돈은 3개월령에귀뒤쪽목근육에 2 ml씩 Intervet사 FMD 백신을접종하였다. NSP ELISA 번식돈및자돈의 FMD 감염경험여부를조사하기위하여모돈은자돈분만전 1회, 자돈은출하전 1회채혈하여양성개체존재여부를조사하였다. NSP 항체검사는 VDPro R FMD NSP antibody ELISA kit (Median diagnostics, Korea) 를이용하여제조회사에서공급하는실험방법에따라실시하였다. ELISA reader (Sunrise, TECAN, Switzerland) 를이용하여 450 nm의파장에서 optical density (OD) 값을측정하였다. ELISA 결과판정은제조사계산식 (OD test sample/od negative cotrol) 에따라 SN값을얻어 0.60 이하일경우양성, 0.60 초과일경우음성으로판정하였다. SP ELISA 매회 SP 항체검사를실시하여측정한개체별항체가 (percentage inhibition titer, PI 수치 ) 를분석하여, 모돈과자돈간의모체이행항체간섭효과발생정도, 항체가지속정도, 백신접종시기별항체가형성정도를조사하였다. 결과는 simple linear regression analysis를사용하여검증하였다. SP 항체검사는 PrioCHECK R FMDV Type O ELISA kit (Prionics Lelystad B.V., Netherlands) 를사용하여제조사에서공급하는실험방법에따라실시하였다. ELISA 결과판정은제조사계산식 {100 (corrected OD 450 test sample/corrected OD 450 max) 100} 에따라 PI 수치를얻어 50 이상일경우양성, 50 미만일경우음성으로판정하였다. Table 1. 4 18 안길호ㆍ배진규ㆍ정광ㆍ왕영일ㆍ정준용ㆍ강순근ㆍ권혁무 통계분석통계분석은 Microsoft office excel 2007의통계데이터분석프로그램을이용하였다. 모든통계적검정은유의수준 α=0.05에서검정하였으며, descriptive statistics, correlation analysis, simple linear regression analysis의통계분석방법을이용하였다. 결과 NSP ELISA 검사결과모돈 4농가 20두, 후보돈 3농가 16두는최초채혈한개체에대하여 NSP ELISA 검사를, 자돈 4농가 51 두는마지막채혈한개체에대하여 NSP ELISA 검사를한결과모든개체가항체음성으로판정되어실험대상돼지는 FMD 감염경력이없는것으로확인되었다. 번식돈 ( 모돈, 후보돈 ) 에 FMD 백신접종후 PI 수치변화모돈 4농가 20두와후보돈 3농가 16두에대하여월별 FMD SP ELISA 검사를진행하였으나, 임신등으로해당개체를검사하지못하였더라도, 다음달에시료채취가가능한경우검사를진행하였다. FMD 백신접종후 5개월까지 PI 수치변화는 Table 1과같으며, 모돈이양성수준 (PI 수치 50 이상 ) 을유지하는지 descriptive statistics로분석한결과 95% 신뢰구간에서 PI 수치가 으로관찰되어양성수준 (PI 수치 50 이상 ) 의항체가를유지하여 FMD 백신접종후최소 5개월간양성수준의항체가를유지하는것으로관찰되었다 (P<0.05). 별항체양성률은 1개월령에 28.4%, 2개월령에 4.5%, 3개월령에 0%, 4개월령에 31.3%, 5개월령에 26.2%, 6 개월령에 41.2% 로관찰되었다. 모돈 PI 수치에따른백신접종전돼지의모체이행항체수준검사대상모돈 10두의 PI 수치와검사대상모돈에서분만된자돈 80두의 1개월령 PI 수치를비교하여모체이행항체를분석한결과모돈의평균 PI 수치는 62.4였고, 1개월령돼지의평균 PI 수치는 33.5이었으며, correlation analysis 결과상관계수는 0.858로 Rea와 Parker(2005) 의해석에따르면매우강한양의상관관계를보여 (P<0.05), 모돈 PI 수치가높을수록백신접종전돼지의 PI 수치가높은것으로관찰되었다 (Fig. 1) 1개월령돼지 PI 수치에따른백신접종후 PI 수치변화 3 4개월령에폐사한개체를제외한 1개월령돼지 67두의 PI 수치와그개체들을백신접종한지 1개월경과한후 PI 수치에대한 correlation analysis 결과상관계수는 0.524로비교적강한음의상관관계를보여 (P<0.05), 1개월령돼지의 PI 수치가높을수록백신접종후 PI 수치는낮은것으로관찰되었으며 (Fig. 2), 1개월령돼지의평균 PI 수치는 33.8이었고, 백신접종 1개월이지난돼지의평균 PI 수치는 39.8로관찰되었다. 자돈에 FMD 백신접종후 PI 수치변화 A와 B농가는 2개월령돼지에, C와 D농가는 3개월령돼지에 FMD 백신접종후 6개월령까지월 1회 PI 수치를조사한결과는 Table 2와같으며, 월별자돈이양성수준을유지하는지 descriptive statistics로분석한결과 95% 신뢰구간에서 PI 수치가양성수준을보이는월령은없었다 (P<0.05). 또한, PI 수치 50 이상을양성기준으로볼때, 월 Fig. 1. 5 돼지에서구제역백신접종후항체가변화조사 19 자돈에서 FMD 백신접종시기에따른백신접종후 PI 수치변화 2개월령돼지에 FMD 백신접종한 A와 B농가는최초 34두를대상으로, 3개월령돼지에 FMD 백신접종한 C와 D농가는최초 46두를대상으로검사를진행하였으나, 폐사등으로검사대상두수가감소하여연속적인 PI 수치변화를확인하기위하여 4개월령이전에폐사한개체는이번통계분석에서제외하였다. 2개월령돼지백신접종실험군에서는접종후월별평균 PI 수치가 3개월령에 24.3, 4개월령에 36.4, 5 개월령에 28.7, 6개월령에 26.6으로, 백신접종후 1개월이경과한 3개월령에서 PI 수치가감소하다가 4개월령에 PI 수치가상승한후 5개월령부터 PI 수치가감소하기시작하였다. 또한, 3개월령돼지백신접종실험군에서는접종 Fig. 2. The result of linear regression analysis between 1monthold growing pig s PI titer and PI titer of the first month after FMD vaccination. 후월별평균 PI 수치가 4개월령에 46.8, 5개월령에 40.5, 6개월령에 49.4로, 백신접종후 1개월이경과한 4개월령에서 PI 수치가크게상승하여 3개월령돼지에 FMD 백신을접종하는것이 2개월령에접종하는것보다 PI 수치가높은것으로관찰되었다 (Table 3). 고찰이번연구에서는경기도포천지역돼지 4농가에대하여 FMD 백신접종후 SP ELISA를이용하여월별 PI 수치를조사하여, 번식돈및자돈의검사시기별적정항체가유지여부, 모돈의 PI 수치에따른 1 개월령돼지의 PI 수치변화, 백신접종시기별 PI 수치의상관관계에대하여 descriptive statistics, correlation analysis, simple linear regression analysis를시행하였다. 번식돈은월 1회 PI 수치를조사한결과분만 3주전백신접종으로 5개월경과시까지양성수준의항체가 (PI 수치 50 이상 ) 를유지하였다 (P<0.05). 그러나, 자돈의경우백신접종후월별 PI 수치검사에서 4개월령에 43.5, 5개월령에 36.7, 6개월령에 40.0으로양성수준의항체가 (PI 수치 50 이상 ) 를유지하지못하였고 (P<0.05), 특히 3개월령돼지에서항체양성률이 0% 를나타내어 FMD에대한방어능력이가장취약한시기인것으로관찰되었다. Rodriguez와 Grubman(2009) 은 FMD 불활화항원백신을동물에접종할경우 1 2주내에질병에대한방어력을가지게되나장기간면역방어력은유지하지못하기때문에집단면역의적정수준을유지하기위해서는여러번백신접종을하여야한다고하였 Table 3. 6 20 안길호ㆍ배진규ㆍ정광ㆍ왕영일ㆍ정준용ㆍ강순근ㆍ권혁무 다. 또한, Doel(2003) 은축종과기대수명에따라 4 6개월후추가접종이요구된다고하였다. 이번연구에서자돈에비해번식돈이양성수준의항체가를장기간유지한것은번식돈의경우생후 2개월령에 1 차, 3개월령에 2차 FMD 백신접종을하고, 분만 3주전추가접종을실시하는등최소 3회이상정기적인백신접종을실시하고있는데반해, 자돈의경우생후 2 3개월령에 1회접종으로도축출하시까지추가접종을하지않는것에기인한다고추정된다. 모돈 PI 수치와 1개월령돼지 PI 수치를조사한결과모돈의 FMD PI 수치와백신접종전자돈의 PI 수치는매우강한양의상관관계가관찰되었다. 또한, 자돈의 FMD 백신접종시기별 PI 수치를조사한결과 2개월령돼지백신접종군에서는접종당시 PI 수치 31.5와비교하여 4개월령 (PI 수치 36.4) 을제외하고오히려낮은 PI 수치를나타내어백신접종에따른면역상승효과를보인시기를관찰할수없었으나, 3개월령돼지백신접종군에서는접종당시 PI 수치 15.2와비교하여모든검사월령에서높은 PI 수치를나타내어백신접종에따른면역상승효과가관찰되었다. Kitching과 Salt(1995) 는초유를통해신생가축에게전달된 FMDV의항체가일시적인방어효과를보이기도하지만신생가축에백신을접종할경우모체이행항체로인하여면역력증대효과가저해된다고보고하였다. 이번연구에서모돈과 1개월령돼지의 PI 수치가강한상관관계를보인것은 FMD 항체가모돈을통해자돈에게이행되기때문으로판단되며, 3개월령돼지에백신접종한 C와 D농가에대해상관분석을한결과모돈과 1개월령돼지에서 PI 수치의상관계수는 (P<0.05), 모돈과 2개월령돼지에서 PI 수치의상관계수는 (P<0.05), 모돈과 3개월령돼지에서 PI 수치의상관계수는 (P>0.05) 으로나타나 2개월령돼지의 PI 수치까지모돈의 PI 수치와통계적으로유의한상관관계를가지는것으로관찰되었다 (P< 0.05). FMD 백신접종시기에따라접종이후 PI 수치를비교한결과 2개월령돼지보다 3개월령돼지에백신접종하는것이높은항체양성률및 PI 수치를유지하는것으로관찰되었다. 이는 2개월령돼지까지모체이행항체의영향이남아있어 2개월령돼지에접종시에는면역력증대효과가저해되기때문으로추정되나, 2 개월령돼지의백신접종군과 3개월령백신접종군의 1개월령돼지의모체이행항체에서 PI 수치가 50.6과 26.2로비교적큰격차를보이는바, 보다정확한평가를위해서는추가확대검사가필요할것으로판단된다. 이번연구에서는모돈자돈사이의항체역가상관관계에대한연구를시행하여모돈의 PI 수치가자돈의항체형성에영향을주는것으로확인하였다. 앞으로더욱효과적인백신접종프로그램을설정을위하여백신제품별, 질병감염, 증체율, 3가 (O+A +Asia1 serotype) 항체검사등다양한요인에대하여도조사가필요할것으로생각된다. 결론이번연구는 FMD 백신접종후항체변화추이를조사하고, 모돈의 PI수치와자돈 PI 수치의상관관계등을조사하기위하여수행되었다. 번식돈 ( 모돈, 후보돈 ) 은 FMD 백신접종후 5개월경과시까지항체가 (PI 수치 ) 가양호하게유지되었으며 (P<0.05), 자돈은백신접종월령및모돈 PI 수치에따라다양한결과가관찰되었다. 백신접종전자돈의모체이행행체수준은모돈의 PI 수치가높을수록 1개월령돼지의 PI 수치가높은것으로관찰되었다 (P<0.05). 백신접종시모돈 PI 수치는자돈 PI 수치형성에영향을주며, 백신접종이전인 1개월령자돈의항체수치가높을수록백신접종이후에항체가잘형성되지않는것으로추정되었다 (P<0.05). 모돈의 PI 수치와자돈의 PI 수치는 2 개월령돼지까지통계적으로유의한상관관계를나타내었으나 (P<0.05) 3개월령에서는유의한상관관계가없었다 (P>0.05). PI 수치는 3개월령돼지에백신접종하는것이 2개월령돼지에접종하는것보다높게형성되었다. 이는모돈의 PI 수치가 2개월령돼지까지의 PI 수치에영향을미치는간섭효과때문으로판단된다. 그러나, 이번연구에서는많은개체를검사하지못하였고, simple linear regression analysis에서결정계수가다소낮은점을고려할때백신접종시기, 모돈의 PI 수치이외의다른요인이항체가형성에영향을주는것으로추정되는바앞으로효과적인 FMD 예방을위해백신제품별, 질병감염, 증체율, FMD 3가 (O+A+Asia1 serotype) 항체검사등다양한요인에대한연구가추가로진행되어야할것으로판단된다.
