The samples were daily collected from a site of the Kumho River during 70 days from October 13 to December 21 in Total 126 diatom taxa collected in the study were classified into 114 species and 12 varieties belonging to 26 genera. About 80% of the taxa collected from the artificial substrata during the whole studying period were observed until tenth day from the start time of the study. In the study the successional process of the diatom community was divided into the three stages of the early, the middle and the later by cluster analysis based on the similarities in species composition among the communities. Cocconeis placentula Ehrenb. and C. placentula var. lineata (Ehrenb.) V.H., horizontally positioned species, would be divided into the early successional species, and Diatoma vulgre Bory, vertically positioned species, was the later successional species. Cell densities and species composition of the diatoms on the artificial substrata were closely similar to them of natural substrata after 40th day from the start time of the study. Key Words: artificial substrata, composition, density, epilithic diatom, succession 서론하천은시 공간적차이에따라많은환경요인들이변화하는복잡한생태계이며, 하천의물리 화학적그리고생물학적특성들은하천유역의기후, 토지사용, 지형학의조건등에의해영향받는다 (Richards et al. 1996; Stevenson and Pan 1999). 하천의수질상태를추론하기위해서는가능한많은환경요인들이측정되어야하지만 (Barbour et al. 1995; Norris and Norris 1995), 많은환경요인들중에서도 Stevenson and Pan(1999) 은생육시작시점부터채집시기까지복잡하게변하는수환경에적응하며살고있는생물군을이용한수질상태의추론이수환경을가장완전하게평가할수있다고보고하였다. Cox(1991) 는지표가되는생물군의선택시다음과같은기준에적합하여야한다고보고하였다. 첫째, 각분류군은 *Corresponding author (jungho@daegu.ac.kr) 정확한생태적범위를가져야한다. 둘째, 이범위는한정되어있어야한다. 셋째, 그분류군들은일관성있고정확하게동정되어져야한다. 이러한기준으로볼때, 지표종으로부착규조는매우적합하다. 왜냐하면부착규조는가장풍부하게생육하는군집들중하나이며 (Round et al. 1990) 쉽게채집할수있다. 또한그들의생태범위에대해서는 Knopp (1955), Zelinka and Marvan(1961), Sladecek(1973) 등에의해많은연구들이보고되었으며, 분류학적연구도다른생물군에비해상대적으로매우풍부한연구축적이이루어져있어동정의객관도가높은것으로보고되고있다 (Round et al., 1990). 그리고이미 Patrick(1973), Lange-Bertalot (1979), Wetzel(1983), Stevenson and Lowe(1986), Round (1991), Asai and Watanabe(1995) 등의부착규조를이용한생물학적수질평가가널리활용되고있다. 국내에서도부착규조의군집구조에대한조사와이를통한수질평가의연구들이많이이루어졌다 (Chung et al. 1985; 정 1987; Choi and Chung 1990; 홍과정 1990; 이와정 1991; 정등 1992; 이와정 1993; 정등 1993; 이와김 1996;

2 296 Algae Vol. 21(3), 2006 이 1998; 이 2003). 그러나위의연구들은규조의군집구조변화를조사간격이큰월별또는계절적으로만조사하여부착규조의군집형성에대한생태학적특성을이해하기에는부족하며, 특히군집형성과정에대한이해는자연적부착기질이아닌인공기질의사용이적극적으로권장된다는관점에서기존의연구들은많은제약을가지고있다 (Butcher 1932; Henrici 1933; Gause 1936; Castenholz 1960; Spoon and Burbanck 1967; Cairns et al. 1969; Tuchman and Blinn 1979; Biggs 1988; Danilov and Ekelund 2001). 국내에서는조 (1994) 와정등 (1996) 에의한인공기질을이용한소수의부착조류의연구들이있을뿐, 실질적인부착규조의착생및천이과정에대한연구는매우빈약한상태이다. 인공기질은부착규조의생태연구에다양한유용성을제공해준다. 특히, 부착규조에대한물리적또는생물학적현상이나특성을연구자의의도에따라조절하기에매우용이하며, 자연상태에서파악하기어려운군집형성이나천이과정을용이하게관찰할수있는이점이있다 ( 김 1987). 그러므로본연구에서는인공기질이용하여부착규조군집형성의초기단계에대한특성을밝힘으로서, 부착규조의기본적생태에관한이해를넓히고, 규조를이용한생물학적수질판정법의합리적활용법수립에기여하고자하였다. 재료및방법본조사는금호강본류중경상북도경산시진량읍내리리에위치한지점을대상으로실시하였다. 본지점은낙동강과접하는금호강하류로부터약 54 km 상류에위치하고있으며, 본류수역중유량등의환경조건의변화가비교적적은지역이다 (Fig. 1). 본조사정점의수심약 30 cm에 5cm 간격으로인공기질 ( 직육면체콘크리트불럭 ; 19.5 mm 8.0 mm 5.5mm) 600개를 2002년 10월 12일설치하여 2002년 10월 13일부터 12월 21일까지 70일간매일한차례씩채집하였다. 수온과 ph, 전기전도도는현장수질측정기인 D-24 (HORIBA) 로측정하였으며, BOD는수질오염공정시험방법 ( 환경부 1996) 에준하여 Winkler법으로측정하였다. 유속은 CR-7 유속계 (YOKOGAWA) 를사용하여세번이상측정한후평균값으로계산하였다. 부착규조는인공기질인콘크리트불럭의수면과평행한상면부를솔로서긁어채집하였다. 비교분석을위한시료로서본조사정점에서가장보편적인상태를지니고있는것으로추정되는지름 cm 정도의수면하의자연석을선정하여가로 10 cm, 세로 10 cm의고무판을사용하여상표면의 100 cm 2 를솔로긁어채집하였다. 부착규조의현존량산정을위하여인공기질의상면부를채집한정량시료를잘혼 Fig. 1. A map showing the sampling station of the Kumho River. 합한후 1mL를취해Sedgwick-Rafter Chamber를사용하여 Schoen(1988) 의방법으로계수하였다. 종의동정을위하여채집된시료를 KMnO 4 법 (Hendey, 1974) 으로세정하고 Pleurax로봉입하여영구표본을제작하였다. 제작된슬라이드는 OLYMPUS BX-50 광학현미경으로 배에서관찰하였고, OLYMPUS PM-20으로사진촬영하여분석하였다. 또한, 규조군집의상대빈도를위한계수는임의로선정된현미경하의시야에서피각의수가 500 개이상이되도록하였다. 부착규조군집구조를분석하기위하여전조사기간의출현종의상대빈도를기본자료로하여 Shannon and Weaver (1963) 의종다양성지수, Simpson(1949) 의우점도지수그리고 Pielou(1966) 의균등지수를구하였다. 집단분석 (cluster analysis) 과주성분분석은 MVSP 3.1을사용하여구하였다. 결 과 환경요인수온은 C의범위를보였으며, 전기전도도는 213 µs cm µs cm 1 를기록하였다. 조사기간동안전기전도도의평균치는 254 µs cm 1 로조사되었다. ph는 의범위를보였으며, 전조사기간의평균치는 8.1이었다. BOD는 1.2 mgl mgl 1 의범위를나타내었으며, 조사기간의평균치는 2.6 mgl 1 였다. 하천의플랑크톤및부착생물의생육의중요교란요인인유속 (Stevenson 1983) 은 6.2 cm sec cm sec 1 의범위로전조사시기에 17.5 cm sec 1 의평균치를나타내었다 (Fig. 2). 종조성과출현종본연구에서는 Aulacoseira 속, Caloneis 속, Cymbella 속, Fragilaria 속, Hantzschia 속, Navicula 속의각각 1종의총 6

Daily variation of environment factors at the sampling station of Kumho River during the studying period. 종의미동정종을포함한 125 taxa의규조가출현하였으며, 이들은 Simonsen(1979) 의분류체계에따라 2목 3아목 10과 26속의 107종 12변종으로분류되었다. 중심목규조는총 12 taxa로써전체의 9.5% 였으며, 우상목규조는총 114 taxa로전체의 90.5% 였다. 조사시기동안부착규조의일조사평균출현종수는 47종이었으며, 조사시작 2일째인 2002년 10월 14일에 30종으로가장빈약한출현종수를보였으며, 조사시작 38일째인 2002년 11월 19일에 55종으로가장많았다 (Fig. 3). 전조사기간에동정된전 체출현종중조사시작후10일사이에약 80% 가착생되는것으로확인되었다. 출현종의분포양상전체천이의과정을종조성의유사성을기초로한집단분석에따라 3단계 ( 초기, 중기, 후기 ) 로나누었으며, 각단계에서의인공기질과자연석의부착규조의상대빈도를비교하여, 출현종의양상을초기선호종 (early successional species) 과후기선호종 (later successional species) 으로나눌

4 298 Algae Vol. 21(3), 2006 수있었다. 본연구에서 Cocconeis placentula Ehrenb. 와 C. placentula var. lineata (Ehrenb.) V.H. 가초기선호종으로나타났다 (Fig. 4). 인공기질의착생초기에 C. placentula는 30% 이상의높은상대빈도보이다가, 그이후로 3% 이하의상대빈도를보였으며, 이에반하여자연석에서상대빈도는조사시기동안 % 사이로일정하게나타났다. C. placentula var. lineata도인공기질의착생초기에 16% 이상의높은상대빈도를보였다가그이후로 2% 이하의상대빈도를나타내었다. 비교분석을위한자연석조사에서 C. placentula var. lineata 상대빈도는평균 1.2% 로조사기간동안일정한상대빈도를나타내었다. Diatoma vulgare Bory는대표적천이후기선호종으로나타났다. 조사기간동안 D. vulgare는인공기질의착생초기에 0.5% 이하의낮은상대빈도를보이다가착생후기에는 3-8% 의일정한상대빈도를나타내었다. 비교를위한자연석조사에서의 D. vulgare의상대빈도는평균 3.5% 로조사기간동안거의일정하게나타났다 (Fig. 5). Fig. 3. Daily variation of species number on artificial substrata at the sampling station of the Kumho River during the studying period. 현존량본조사에서현존량은조사시기 1일째인 2002년 10월 13 일에 1,930 cells cm 2 로최저치를나타냈으며, 조사시기 63 일째인 2002년 12월 14일에 766,784 cells cm 2 로최고치를나타내었다 (Fig. 6). 종조성의유사성을기초로한집단분석에따라천이과정을 3단계 ( 초기, 중기, 후기 ) 로나누었으며, 이에따른현존량 Fig. 4. Variation of Diatoma vrlgare at the sampling station of the Kumho River during the studying period (A: artificial substrata, B: natural substrata). 전조사기간동안의우점종의변화는 Nitzschia amphibia Cocconeis placentula Nitzschia amphibia Nitzschia fonticola 순으로나타났다. 본연구에서가장높은상대빈도를나타낸 4종을그순서대로나열하면 Nitzschia amphibia, Nitzschia fonticola, Fragilaria construens var. venter 그리고 Cocconeis placentula 등이며, 각시기마다이들이차지하는비율을 Fig. 7에도시하였다. 이들은전조사기간중에서조사시작 2일째인 2002년 10월 14일에 30.2% 로가장낮은빈도를보였고, 조사시작 21일째인 2002년 11월 2일에 68.0% 로가장높았다. 이는종다양성지수의변화양상과유사함을보였다. Daily variation of standing crops on artificial substrata at the sampling station of the Kumho River during the studying period. 의변화는초기에 1,930 cells cm 2 에서 129,941 cells cm 2 사이로, 중기에는 48,889 cells cm 2 에서 497,895 cells cm 2 사이로, 후기는현존량이최대 766,784 cells cm 2 까지로거의일정한값을나타내었다. 우점종및아우점종의천이부착규조의군집형성의초기단계의생태학적특성을이해하기위해우점종및아우점종의천이를파악하고자하였다. 전조사기간중에1회이상우점종으로조사된 taxa는 Nitzschia amphibia Grun.(56회 ) 와 Nitzschia fonticola Grun. (11회), Cocconeis placentula(3회 ) 등 3 taxa였다. 또한 Nitzschia paleacea Grun. 와 Melosira varians C. Agardh, Fragilaria construens var. venter (Ehrenb.) Grun., Navicula capitatoradiata H. Germ. 등 4종이아우점종을이루었다 (Table 1). 종다양성및우점도지수종다양성지수는조사시작 21일째인 2002년 11월 2일에최저치 0.938을나타내었고, 조사시작 59일째인 2002년 12월 10일에최고치인 1.355를나타내었다. 또한균등도지수는종다양성지수와같이조사시작 21일째인 2002년 11월 2일에 0.578로최저치를, 그리고조사시작 59일째인 2002년 12월 10일에 0.794로최고치를나타내었다. 이는균등도지수가다양성지수에서산출된지수이므로종다양성지수와유사한경향을보이는것이다. 우점도지수는종다양성지수와균등도지수와는정반대로조사시작 21일째인 2002년 11월 2일에 0.274로가장높게나타났으며, 조사시작 59일째인 2002년 12월 10일에 0.070으로가장낮게조사되었다 (Fig. 8). 군집구조종조성의유사성으로부착초기 1일에서 5일까지와그이후의 5일간격으로집단분석을한결과, 3개집단 ( 초기, 중기, 후기 ) 으로나눌수있었다 (Fig. 9). 즉부착초기에수평형규조종 (horizontally positioned species) 과수직형규조종

(vertically positioned species) 이혼재되어나타난 1일에서 5 일까지의집단과 Nitzschia amphibia와 Cocconeis placentula, Nitzschia fonticola가우점종으로출현하는 10일째에서 20일째까지의집단, Nitzschia amphibia와 Nitzschia fonticola가우점종으로출현하는 35일째에서 70일째까지등 3개집단으로구분되는결과를나타냈다. 초기단계의군집유사성은 30-60% 로군집들간의유사성이낮게나타났으며, 중기와후기단계 70 로진행될수록군집유사성이최고 85% 로높게나타났다. 또한, 주성분분석 (PCA) 을통한결과에서도시간이경과함에따라군집간의구조가유사해짐을알수있었다. Fig. 10 의주성분분석의결과에서나타난것처럼 Axis 1을기준으로시간의경과에따라초기단계와중기단계, 후기단계로구분할수있었으며, 초기단계의시기들은넓게산재되어있는반면, 후기로갈수록시기들이조밀하게표시되어시간의경과에따라군집들간의종조성및군집구조가유사해짐을나타내고있다. 고찰현존량은조사시작 1일째인 2002년 10월 13일에 1,930 cells cm 2 로최저치를나타냈으며, 조사시작 63일째인 2002 년 12월 14일에 766,784 cells cm 2 로최고치를나타내었다. Stevenson(1983) 은물속의부유성플랑크톤이기질에유입된군집의근원이라고보고하였으며, 유입의범위를 30-1,500 cells cm 2 d 1 로나타내었다. 그러나본연구에서는조사 1일째에 1,930 cells cm 2 d 1 로 Stevenson(1983) 이보고한유입범위를초과하였는데, 이는유속의평균치가 17.5 cm sec 1 로낮게나타났기때문에착생초기에다양하고많은종들이유입되어졌다고사료되어진다. 착생초기에인공기질에상대적으로높은상대빈도를나타낸종은수평형규조종 (horizontally positioned species) 인 Achnanthes convergens, Cocconeis placentula, C. placentula var. lineata, C. placentula var. euglypta와수직형규조종 (vertically positioned species) 인 Nitzschia palea, N. amphibia, N. paleacea, Navicula viridula var. rostellata로나타

Horne and Goldman(1994) 은호수와하천의새로운기질에대한조류의천이과정에서착생초기에는수평형규조종의부착규조로시작된다고보고하였으나, 본연구에서는수평형규조종과수직형규조종이착생초기에기질에혼재하여나타났는데, 이는비교적느린유속으로인하여착생초기의기질에수평형규조종과수직형규조종이혼재하여 나타난현상으로추측된다 (Stevenson 1983; McIntire 1966; Horner and Welch 1981). 본연구에서는부착규조의천이과정을집단분석에준하여초기와중기, 후기로나누었는데, 천이초기선호종은 Cocconeis placentula와 C. placentula var. lineata가, 후기선호종은 Diatoma vulgare였다. 이는초기유속등의환경조건에서기질에부착하기유리한부착형태인수평형규조종이초기선호종으로, 천이과정에있어서는빛의경쟁에유리한부착형태인수직형규조종이후기선호종으로나타났다고판단되어진다. 그리고 Achnanthes convergens와 Cocconeis placentula var. euglypta, Nitzschia palea, N. amphibia, N. fonticola, N. paleacea, Navicula viridula var. rostellata, Synedra ulna 등은인공기질과자연석에서유사한변화양상을보였다. 종조성의유사성을기초로집단분석의결과, 크게 3개의집단 ( 초기, 중기, 후기 ) 으로나눌수있었다. 즉부착초기에수평형부착규조와수직형부착규조가혼재되어나타난 1일째에서 5일째까지의집단과 Nitzschia amphibia와 Cocconeis placentula가우점종으로출현하는 10일째에서 25일째까지의집단, N. amphibia와 N. fonticola가우점종으로출현하는 30일째에서 70일째까지등 3개집단으로구분되는결과를나타냈다. 그리고초기단계의군집들간의유사성은 30-60% 로낮았으며, 중기와후기단계의군집들간의유사성은 60% 에서최고 85% 로높게나타났다. 이는군집구조가시간의경과에따라변화하며, 후기로갈수록군집구조가더욱유사해짐을나타내는것으로이해된다. 주성분분석결과에서도 Axis 1을기준으로시간경과에따라초기단계와중기, 후기단계로구분할수있었으며, 초기단계의시기들은넓게산재되어있는반면, 후기로갈수록시기들이조밀하게표시되어시간의경과에따라군집들간의종조성및군집구조가유사해짐을알수있었다.
