Enteritidis) and S. Typhimurium are the major causative agents of food-borne illness worldwide. Traditional detection methods for Salmonella are generally time-consuming and not so highly sensitive. Currently, a rapid detection system using multiplex real-time polymerase chain reaction (PCR) has been used as a highly sensitive, specific, and rapid test for the presence of pathogenic bacteria. In this study, a multiplex real-time PCR was used to detect Salmonella spp., S. Enteritidis and S. Typhimurium. We selected multiplex real-time PCR target genes, which were the inva, proto6e and inner membrane protein. The specificity of the multiplex real-time PCR assay, using 57 bacteria including 51 Salmonella isolates and 6 non-salmonella, was 100 %, 100 % and 100 % for Salmonella spp., S. Enteritidis and S. Typhimurium, respectively. This assay indicates that the specificity of the multiplex real-time PCR makes them potentially valuable tools for detection of Salmonella spp., S. Enteritidis and S. Typhimurium. Key Words : Salmonella, food-borne illness, multiplex real-time PCR. 서론 1) Salmonella속균은그람음성의통성혐기성세포내기생세균으로서사람과동물을비롯하여자연계에널리분포하고있다. 이들균에감염되면설사, 쇠약, 발열및패혈증등의전신성증상을일으키며, 특이성이있는몇몇균종을제외한대부분의균속이인수공통점염병의원인세균으로알려져있다 1,2). Salmonella enterica는수십년동안사람에서식중독의주요병원체로인식되어왔으며, 주로동물유래균으로오염된음식물섭취를통하여감염된것으로알려져있다 3). 살모넬라감염증은사람에서가장흔한식품매개질병으로서미국에서발생하는식중독의약 30 % 를차지하며, 국내에서도식중독원인세균중 가장높은분포를나타내고있다 4). Salmonella spp. 중 Salmonella enterica serotype Enteritidis (S. Enteritidis) 는사람과동물에감염하여주로급성장염을일으키고, 오염된계란이나식품을통하여폭발적인식중독발생을일으키고있으며 5,6), S. Typhimurium은전세계에널리분포하고있는균종으로, 사람을비롯하여소, 말, 양, 개, 가금, 설치류및조류등다양한숙주에감염되어장염, 패혈증, 유산및폐렴등을일으키며, S. Enteritidis와 S. Typhimurium은사람에서식품을매개로한살모넬라감염증의가장중요한원인체로서공중보건학적으로매우중요시되고있다 7,8). S. Enteritidis와 S. 2 232 이우원 정경태 이강록 이기흔 야한다. 또한 lipopolysaccharide (LPS, O항원 ) 와 flagella antigen (H 항원 ) 을이용하여 2,500여혈청형구분을위한검사가이루어져야한다 9,10). 전통적인배양방법은많은시간과복잡한절차가요구된다 11). 혈청형을동정하기위해서는항원에대한인자혈청을이용한응집반응법이이용되고있고, 다른방법으로 whole cell, LPS 및 OMP 항원을이용한 ELISA 기법은유용하고시간을절약할수있으나다른균종과의교차반응으로인한위양성반응이흔하여특이성에서문제시되고있다 12). Salmonella spp. 를비롯한 S. Enteritidis와 S. Typhimurium의신속한진단을위해서최근에는 plasmid profile, polymerase chain reaction (PCR), southern blot hybridization 기법을기초로한분자유전학적분석및제한효소처리에의한 DNA의절단양상등을분석하여역학관계를규명하고있다 13~15). 특히중합효소연쇄반응법 (PCR) 을기반으로개발되어진진단기법들은검체내소수의균이존재하더라도진단이가능하고신속한결과를얻을수있어조기진단에널리이용되고있으며민감도와특이도가높아이용이확대되고있다. 더불어 PCR보다민감도와특이성이높은것으로알려진 real-time PCR이개발되어진단방법으로연구되고있는데, 일반 PCR과달리 real-time PCR은증폭결과를실시간으로그래프를통해확인할수있어더신속하고민감도가더높은진단법으로알려져있다 16~18). Single real-time PCR 기법은 S. Enteritidis와 S. Typhimurium을비롯한주요 Salmonella spp. 특이적으로검출하기위하여적용되었으나 16,19,20), multiplex real-time PCR 기법은이들병원균의검출을위해서적용된예는흔하지않다. 이에본연구에서는 Salmonella spp., S. Enteritidis 와 S. Typhimurium의동시검출을위한 multiplex real-time PCR 검사법개발을시도하였다. 2주 ), E 1 (S. Westhampton 1주 ), E 4 (S. Senftenberg 1주 ), L (S. Ruiru 2주 ) 등 51주를공시하였다. 기타대조균주로는본연구원에서보관중인 E. coli, Staphylococcus aureus 및 Listeria monocytogenes를대조군으로사용하였다. Multiplex real-time PCR 1. DNA 추출 DNA 추출을위해공시균주들을 tryptic soy broth (Merk, Germany) 에서 37, 18 시간 ~ 24시간배양후, 배양액 2 ml을 microcentrifuge tube에옮긴다음 8,000 rpm으로 2분간원심분리하여상층액을제거하였다. 상층액이제거된침전물에서 genomic DNA를추출하기위해 PowerPrep TM Bacterial DNA Extraction Kit (Kogenebiotech, Korea) 를이용하여 DNA를추출하였다. 2. Primer와 probe 본연구에사용된 primer와 probe의정보는표 1에서와같이 primer는 Bioneer(Korea) 에합성의뢰하여사용하였고, probe는 ThermoFisher(USA) 에주문하여사용하였다. Primer와 probe 디자인은각각 target에특이적인유전자를사용하여디자인하였다. Salmonella spp. 는병원성인자중 invasion에관련된 inva 유전자를 target으로하여 primer와 probe를디자인하였고 probe의형광물질은 VIC으로설계하였다. S. Enteritidis 는편모와관련된 prot6e 유전자를 target으로하여 probe 형광물질을 FAM으로설계하였고 S. Typhimurium 은항원에관련된 viab 유전자를 target으로하여 probe 의형광물질을 Cy5로설계함으로써 multiplex real-time PCR 반응에서형광신호간에교차가발생하지않도록하였다. 재료및방법공시균주공시균주는 2005년이 21) 가소와돼지에서분리한 Salmonella속균 34종의 serotype 457주중 S. Enteritidis 10주, S. Typhimurium 25주, Salmonella serogroup B (S. Agona, S. Derby 및 S. Schwarzengrund 각 2주 ), C 1 (S. Ardwick, S. mbandaka 및 S. Rissen 각 3. Multiplex real-time PCR 반응조건 Real-time PCR 반응액은총 20 μl 로반응을수행하였다. 각각의 primer와 probe의최종농도는 500 nm, 100 nm이되도록혼합하였다. 2X Real-time PCR master mix 10 μl, 각각의 primer 및 probe는각각 100 um, 20 um로희석한후 3개 set의 primer, probe 각각 0.1 μl 씩사용하였다. D.W 4.1 μl 를첨가하여총 volume 15 μl 로고정한후 10 ng/μl 로희석한 template를 5 μl 첨가하여총 50 ng의 DNA가 PCR 반응에사용되도록하였다.

3 Real-time PCR 을이용한살모넬라검출에대한연구 233 Real-time thermal cycler는 7500 Fast Real-time PCR system (Applied Biosystems, USA) 을사용하였다. 반응조건은 50 에서 2분, 95 에서 10분 predenaturation 시킨후, 95 에서 15초 denaturation, 60 에서 60 초 annealing&extension 주기를 40회반복하였다. 증폭결과는 7500 software (Applied Biosystems, USA) 프로그램을이용하여확인하였다 22). 결과및고찰 Multiplex real-time PCR 특이도공시균주로부터 DNA를추출한다음 primer와 probe 를이용하여 multiplex real-time PCR의특이도를확인한결과표 2에서와같이 Salmonella spp., S. Enteritidis와 S. Typhimurium에특이적인 primer와 probe는모두해당균에만반응하였고, 다른세균에서는 Table 1. 4 234 이우원 정경태 이강록 이기흔 교차반응이관찰되지않았다. 이는 Hadjinicolaou 등 23) 이환경재료와임상시료에서 S. Typhimurium과 S. Enteritidis 분리를위한 real-time PCR 기법을이용한결과및 Pochop 등 24) 이우유와식육에서, Lee 등 25) 이식육에서 Salmonella spp., S. Typhimurium과 S. Enteritidis 분리를위하여 real-time PCR 기법을이용하여연구보고한결과와유사하였다. 민감도민감도확인을위해표 1에서제시한 target gene을플라즈미드형태로클로닝하여각각의민감도를확인하였다. 플라즈미드의최고농도는 10 5 copies로제작하고순차적으로 10배씩희석하여 1 copy 까지희석하여민감도를확인하였다. 그림 1 ~ 그림 3까지검출민감도의정확 성확인위해 3회반복하여민감도를확인하였다. 세종류모두 10 1 까지검출되는것을확인하였다.( 그림 1 ~ 그림 3) 이는 Fey 등 26) 이 S. Typhimurium의검출을위한 real-time PCR의검출한계는 (0.54 ± 0.09) log 10 CFU/mL, Seo 등 19) 은 S. Enteritidis의검출을위한 real-time PCR의검출한계는 (0.65 ± 0.07) log 10 CFU/mL 이라고보고한성적보다는민감도가낮았다. 또한 Salmonella spp., S. Enteritidis 와 S. Typhimurium 의동시검출을위한 multiplex real-time PCR 결과에서도민감도의감소없이각각의 real-time PCR 결과와동등한결과를확인하였다.( 그림 4) 표 3은각종류별로농도에따른 C T 값 (cycle threshold) 의변화를표기한내용이다. 세종류의시스템모두검출한계는 10 copies 까지검 Fig. 1. Real-time PCR 을이용한살모넬라검출에대한연구 235 출한계를확인하였다. 본연구에서도입한 real-time PCR 기법은일반 PCR 과비교하였을때결과를확인하기위하여 agarose gel에 loading하고 PCR product를염색하는과정을생략함으로써검출시간을단축할수있었다.

5 Real-time PCR 을이용한살모넬라검출에대한연구 235 출한계를확인하였다. 본연구에서도입한 real-time PCR 기법은일반 PCR 과비교하였을때결과를확인하기위하여 agarose gel에 loading하고 PCR product를염색하는과정을생략함으로써검출시간을단축할수있었다. 뿐만아니라 probe라는형광표지 DNA 절편을이용함으로서각각의 target에대한민감도와특이성을향상시킬수있었다. 이와같은검사법의응용으로 Salmonella spp., S. 6 236 이우원 정경태 이강록 이기흔 결론사람의주요식중독균으로알려진 S. Enteritidis와 S. Typhimurium을비롯한 Salmonella spp. 의 multiplex real-time PCR 검사법을실시한결과다음과같은결론을얻었다. 1. Multiplex real-time PCR의특이도를확인한결과 Salmonella spp., S. Enteritidis와 S. Typhimurium 에특이적인 primer와 probe는해당균에만반응하였고, 다른세균에서는교차반응이관찰되지않았고, 각각의 target에대한검출민감도를확인한결과 10 1 까지검출되는것을확인하였다. 2. 본연구에서싱글플렉스와멀티플렉스반응에서민감도의변화없이동등한결과를나타내었다. 이와같은검사법의응용으로 Salmonella spp., S. Enteritidis 와 S. Typhimurium의구별이가능함을확인하여검사의편리성을확대하였다. 참고문헌 1.
